Surgery is the mainstay for treatment, which is accompanied by rehabilitative and psychological support. Despite the advances in surgical techniques, the recurrence of TMJA post-surgery has been reported as a common complication. Therefore, it becomes essential to investigate and understand the histo-morpho-pathological processes governing these ankylotic changes. Given the lack of such studies in the literature, herein we present a case of a girl child who underwent primary surgery at the age of six years, followed by a second surgery at the age of twelve years. Ankylotic tissue samples collected during both surgeries were studied using various immunohistochemical markers for tissue remodeling, inflammation, antimicrobial activity, and transcriptional regulation. Cleft lip and palate are the most common congenital malformations which require early primary cle... more Cleft lip and palate are the most common congenital malformations which require early primary cleft surgery in all infants. The surgical treatment of cleft lip and palate deformities results in the formation of scar tissue. However, the scarred tissue has sparsely been immunohistochemically evaluated to date. Herein, we report the differences in the cellular expression of various proteins in the scar and nearby healthy tissue of a 35-year-old patient who underwent cleft correction surgery during infancy. The scar tissue showed basal cell proliferation, prominent cysts, and fibrotic connective tissue. An increased expression of both interleukin 1a (IL-1a) and IL-10 in the scar tissue was noted, although the balance favored an anti-inflammatory environment. No differences in the expression of matrix metallopeptidase-2 (MMP-2) and TIMP-2 were found whilst an increased expression of PAX-9 and MSX-1 was observed in the scar tissue, which colocalized with RYK expression. Temporal studies like the present one can aid in advancing our understanding of the longitudinal processes governing wound healing morpho-pathogenesis in cleft-affected patients.

Previous research has been primarily focused on the genetical aspects of its complex and multifactorial etiology. In the present study, we investigated the role of cytokines as mediators of epithelial–mesenchymal crosstalk and local site inflammation in cleft affected infants. Lip material was obtained from 12 children aged before primary dentition who suffered from orofacial clefting. The quantification of 12 cytokines (Interleukin-2,4,5,6,10,12,13,17A, Tumor Necrosis Factor-α, Interferon-γ, Transforming Growth Factor beta-1 and Granulocyte-Colony Stimulating Factor) was done using ELISA. Nonparametric Spearman Rho was used to ascertain the correlation between the expression levels of different cytokines. Though phonetic trauma/abuse is thought to be the main cause of the development of vocal nodules, the exact etiopathogenesis remains unknown. Hence, we compared the immunohistochemical markers for proliferation (Ki-67), apoptosis (TUNEL), growth (EGFR), ischemia (VEGF), inflammation (IL-1α and 10), and immunoreactive innervation (PGP 9.5), in vocal nodule tissue samples obtained from 10 females (17–56 years) and vocal cord tissue from seven controls. A statistically significant increase in Ki-67, TUNEL, EGFR, VEGF and IL-1α expression was noted (p < 0.05) between nodule tissue and control tissue in both epithelial and subepithelial layers. The human keratinocytes obtained from psoriasis lesions are a very rich source to various antimicrobial peptides. Both Th 17 and Th 1 pathways play a role in the pathogenesis of psoriasis. In the present work we have studied the prevalence of fungal flora in the liver of 18 dark brown ... more In the present work we have studied the prevalence of fungal flora in the liver of 18 dark brown minks. We isolated 12 genus of moulds. Members of the genus Candida, Aspergillus and Penicillium were the most frequently isolated strains. When mink are infected with fungi, the cell recruitment is impaired at the site of the infection, the tissue reaction is highly disorganized in target organs and the infection s evolution is more severe. We studied HGF expression by immunohistochemistry. Our results demonstrated that the distribution of hyphae were between hepatocytes in all areas of liver acinus, but fungal blastospores mainly in interlobular bile ducts. Allocation of HGF in hepatocytes was observed uniform or non uniform, frequently around bile-ducts and portal tracts. In conclusion, detection of HGF simultaneously to inflammation liver might be connected to the tissue ischemia and damage realized by the same inflammatory cells.

The aim of our work was a histomorphometrical and immunohistochemical research on bone morphology and factors influencing it in healthy dairy cows fed with self-produced grain and with rapeseed cakes. The bone of self-produced grain-fed cows demonstrated statistically significant difference in the number of osteocytes from the bone of rapeseed cakes-fed cows. The rapeseed cakes-fed cows didn't show any bone cell positive for BMP2/4, while FGFR1 increased significantly in their supportive tissues. The number of bFGF- and apoptosis-containing structures varied in cows of both groups. Biopsy samples from cows uterus were taken in winter 2004/2005 on the research and training farm “Vecauce”. Histological investigations were performed at the Institute of Anatomy and Anthropology of Riga Stradins University. Nine cows were biopsied twice – in the first and fifth week after parturition. The aim of the work was to investigate vascular endothelial growth factor (VEGF), nerve growth factor receptors p75 (NGFR p75), protein gene product 9.5 (PGP 9.5), tumor necrosis factor α (TNF–α), and apoptosis in the cow endometrium in post partum period. The effectiveness of PRF was demonstrated in various good quality randomized control studies, but mechanisms of action are still unclear. The aim of our study is to analyse the histological effects of PRF on the domestic porcine dorsal root ganglion (DRG), and evaluate the expression of biomarkers in gangliocytes. 3 domestic porcines were investigated. Under general anaesthesia and X-ray control, DRG PRF was performed. Four lumbar DRGs (L1, L2, L3, L4) were randomly treated. The opposite side DRGs was used as control. One month after the procedure the animal was euthanized. The lumbar region of the spine was placed in 10% formaldehyde for a month. After this fixation DRG samples were prepared for slide analysis. Cleft lip and palate (CLP) is a common congenital anomaly with a complex etiology which has not b... more Cleft lip and palate (CLP) is a common congenital anomaly with a complex etiology which has not been elucidated yet. This study investigated whether expression of osteopontin (OPN), osteoprotegerin (OPG), and osteocalcin (OC), which are essential for the normal craniofacial bone remodelling, is not regulated in children with CLP. Alveolar bone tissue samples were obtained from patients with complete bilateral (CB) CLP (n = 14) during corrective plastic surgery and unaffected control subjects (n = 9). OPN, OPG, and OC expression was assessed by immunohistochemistry, and data were analyzed with the Mann-Whitney test. OPN expression was observed only sporadically in the alveolar bone of 3 patients, in contrast to the control group (z = − 2.962; P < 0.003). The number of OPG-positive bone cells varied from occasional to moderate, in contrast to the control group (z = − 2.247; P = 0.025). OC-positive osteocytes were present in moderate to numerous numbers in both patients and controls, with no significant difference between them (z = − 1.356; P < 0.175). The prominent expression of OC characteristic for CBCLP affected hard tissue indicates a high potential of bone mineralization. Few OPG-positive osteocytes in the bone tissue implicate the disregulation of osteoclast differentiation, maturation, and activity, but few OPN-containing cells may prove the common disregulation of bone remodelling during cleft morphopathogenesis.

Alveolar bone tissue samples were obtained from patients with complete bilateral (CB) CLP (n = 14) during corrective plastic surgery and unaffected control subjects (n = 9). OPN, OPG, and OC expression was assessed by immunohistochemistry, and data were analyzed with the Mann-Whitney test. OPN expression was observed only sporadically in the alveolar bone of 3 patients, in contrast to the control group (z = − 2.962; P < 0.003). The number of OPG-positive bone cells varied from occasional to moderate, in contrast to the control group (z = − 2.247; P = 0.025). OC-positive osteocytes were present in moderate to numerous numbers in both patients and controls, with no significant difference between them (z = − 1.356; P < 0.175). The prominent expression of OC characteristic for CBCLP affected hard tissue indicates a high potential of bone mineralization. Few OPG-positive osteocytes in the bone tissue implicate the disregulation of osteoclast differentiation, maturation, and activity, but few OPN-containing cells may prove the common disregulation of bone remodelling during cleft morphopathogenesis.

Biopsy samples from cows uterus were taken in winter 2004/2005 on the research and training farm "Vecauce".
