An early bottleneck in the rapid isolation of new antibody fragment binders using in vitro librar... more An early bottleneck in the rapid isolation of new antibody fragment binders using in vitro library approaches is the inertia encountered in acquiring and preparing soluble antigen fragments. in this report, we describe a simple, yet powerful strategy that exploits the properties of the Spycatcher/ SpyTag (SpyC/SpyT) covalent interaction to improve substantially the speed and efficiency in obtaining functional antibody clones of interest. We demonstrate that Spyc has broad utility as a protein-fusion tag partner in a eukaryotic expression/secretion context, retaining its functionality and permitting the direct, selective capture and immobilization of soluble antigen fusions using solid phase media coated with a synthetic modified SpyT peptide reagent. In addition, we show that the expressed SpyCantigen format is highly compatible with downstream antibody phage display selection and screening procedures, requiring minimal post-expression handling with no sample modifications. To illustrate the potential of the approach, we have isolated several fully human germline scfvs that selectively recognize therapeutically relevant native cell surface tumor antigens in various in vitro cell-based assay contexts.

Derivation of tissue-engineered valve replacements is a strategy to overcome the limitations of t... more Derivation of tissue-engineered valve replacements is a strategy to overcome the limitations of the current valve prostheses, mechanical, or biological. In an effort to set living pericardial material for aortic valve reconstruction, we have previously assessed the efficiency of a recellularization strategy based on a perfusion system enabling mass transport and homogenous distribution of aortic valve-derived "interstitial" cells inside decellularized pericardial material. In the present report, we show that alternate perfusion promoted a rapid growth of valve cells inside the pericardial material and the activity of a proliferation-supporting pathway, likely controlled by the YAP transcription factor, a crucial component of the Hippo-dependent signaling cascade, especially between 3 and 14 days of culture. Quantitative mass spectrometry analysis of protein content in the tissue constructs showed deposition of valve proteins in the decellularized pericardium with a high variability at day 14 and a reproducible tissue maturation at 21 days. These results represent a step forward in the definition of strategies to produce a fully engineered tissue for replacing the calcified leaflets of failing aortic valves.

This exploratory retrospective study aims to investigate the thermal changes in the thyroid gland... more This exploratory retrospective study aims to investigate the thermal changes in the thyroid gland region of patients with hypothyroidism and fibromyalgia by analyzing the temperature of the brown adipose tissue (BAT). A total of 166 individuals from 1000 thermographic electronic medical records were classified into four groups: Group HP + FM-50 individuals with hypothyroidism and fibromyalgia; Group FM-56 individuals with fibromyalgia only; Group HP-30 individuals with hypothyroidism only, and Group Control-30 healthy individuals. The thermal images from the electronic medical records were acquired by a FLIR T650SC infrared camera (used for thermometry) and the temperature data for each group were statistically analyzed. Group HP + FM showed r = 0, meaning that the average temperatures of the thyroid and BAT are independent of each other. Groups FM, HP and Control showed r = 1, meaning that the average temperatures of the thyroid and BAT were directly related. Our findings showed that the average temperatures of the thyroid and BAT regions are similar. Also, there was no correlation between thyroid gland temperature and the presence of hypothyroidism or fibromyalgia using thermometry. Medical infrared thermography (MIT) is a noninvasive and nonradioactive analysis method capable of analyzing physiological functions related to the control of skin temperature, an important organ for controlling body temperature 1,2. This thermography´s technique allows the evaluation of physiological changes 2,3 , with applications in the field of medicine to identify neurological, rheumatological and dermatological disorders, vascular diseases, urologic, and gynecological and orthopedic pathologies 3-7 , and this method can provide support for sports medicine 3. Fibromyalgia (FM) is a rheumatologic disorder, of undefined cause, but its development is associated with the central nervous system's regulation of pain 8,9 , neurosensory, neuroendocrine and neurotransmitter-related disorders, as well as a genetic predisposition 10,11. FM is characterized by musculoskeletal pain in diffuse to chronic intensity and is associated with symptoms such as fatigue, sleep disturbances, palpebral venous congestion, morning stiffness, diffuse paresthesia, subjective sensation of edema, cognitive disorders, depression and anxiety 10,12,13. Infra-Vermelha da Difusão do Óxido Nitroso Via Inalatória para a Cavidade Abdominal, em Ratos Sub... more Infra-Vermelha da Difusão do Óxido Nitroso Via Inalatória para a Cavidade Abdominal, em Ratos Submetidos a Pneumoperitônio JUSTIFICATIVA E OBJETIVOS: O óxido nitroso (N 2 O), por ser uma estrutura tri-atômica assimétrica, assume características de alta emissão e absorção de energia no espectro infra-vermelho, com um pico característico de absorção em 4,5 µm, o que o torna visível ao infra-vermelho curto, quando contrastado com uma fonte emissora de calor (anteparo quente). Diversos autores têm descrito a difusão do N 2 O para cavidades fechadas por métodos como cromatografia gasosa e analisador de gases, que não permitem um estudo macroscópico detalhado do gás. O presente estudo teve como objetivo a filmagem macroscópica no espectro infra-vermelho da difusão de N 2 O, utilizado em anestesia inalatória, para a cavidade peritoneal de ratos submetidos a pneumoperitônio de 20 mmHg com ar ambiente. MÉTODO: Dividiu-se os animais em três grupos, de acordo com o anestésico utilizado: I-controle venoso: tiopental intra-peritoneal; II-controle inalatório: isoflurano a 1,2% em O 2 100%; III-óxido nitroso: N 2 O 66% em oxigênio e isoflurano a 0,6%. Os termogramas provenientes da descompressão abdominal foram obtidos, por meio de um radiômetro AGEMA 550 filmados a 7 quadros por segundo. RESULTADOS: O N 2 O demonstrou-se visível ao infra-vermelho. No momento da descompressão abdominal, não houve nos grupos I e II termogramas com rastros de gases visíveis ao infra-vermelho. Houve, porém, rastros de gases visíveis ao infra-vermelho no grupo III. CONCLUSÕES: Conclui-se que o óxido nitroso inalatório a 66% difundiu-se para a cavidade peritoneal de ratos submetidos a pneumoperitônio de 20 mmHg com ar ambiente, sem aumento de pressão intra-abdominal.

Infrared thermal imaging has been used to assess possible complications, such as necrosis and changes in micro- and macro-circulation, based on perforator mapping techniques, among others. The objective of this study was to present two clinical cases involving thermal imaging monitoring of the healing process of lipoabdominoplasty in the immediate postoperative evaluation and its preliminary results. Infrared thermography was performed 24 hours after the operation and on postoperative days 5, 25, and 27. In clinical case 1, it was found that the delta-R (∆TR)–defined as the difference in minimum temperature between the highest and lowest points in the SA3 region (caution suction area) following the classification established by Matarasso–was 0.4°C at 24 hours after surgery and decreased to 0.1°C on a postoperative day 5. BACKGROUND Low back pain during pregnancy is very common and thermography seems to be a promising... more BACKGROUND Low back pain during pregnancy is very common and thermography seems to be a promising method of evaluation for pregnant women, because it is painless and safe. The aim of the present study was to evaluate low back pain, during pregnancy, using thermography together with artificial intelligence. METHODS A cross-sectional study was carried out with pregnant women recruited from a university hospital. The following data were collected: (a) clinical data; (b) physical assessment with mobility and low back pain provocation tests; and (c) thermograms acquisitions, in a controlled environment. Artificial intelligence and the statistical tests were used to compare the groups' mean: with low back pain (LBP) and without low back pain (WLBP). RESULTS Thirty pregnant women took part, with fifteen in each group. The mean ± Standard Deviation temperature of the lumbar region in both groups were 32.7 ± 1.05 °C and 32.6 ± 1.01 °C for LBP and WLBP, respectively. There was not any difference in temperature between the groups; however, the artificial intelligence software found thermogram differences between groups; furthermore, the correlation between pain intensity and functionality was found. CONCLUSION Thermography associated with artificial intelligence analyses demonstrated to be a promising method as an adjunct to clinical evaluation.

Summary Tumor endothelial marker 1 (TEM1) is an emerging cancer target with a unique dual express... more Summary Tumor endothelial marker 1 (TEM1) is an emerging cancer target with a unique dual expression profile. First, TEM1 is expressed in the stroma and neo-vasculature of many human carcinomas but is largely absent from healthy adult tissues. Second, TEM1 is expressed by tumor cells of mesenchymal origin, notably sarcoma. Here, we present two fully human anti-TEM1 single-chain variable fragment (scFv) reagents, namely, 1C1m and 7G22, that recognize distinct regions of the extracellular domain and possess substantially different affinities. In contrast to other, well-described anti-TEM1 binders, these fragments confer cytolytic activity when expressed as 2nd generation chimeric antigen receptors (CARs). Moreover, both molecules selectively redirect human T cell effector functions toward TEM1+ tumor cells when incorporated into experimental soluble bispecific trivalent engagers that we term TriloBiTEs (tBs). Furthermore, systemic delivery of 1C1m-tB prevents the establishment of Ewing sarcoma tumors in a xenograft model. Our observations confirm TEM1 as a promising target for cancer immunotherapy and illustrate the prospective translational potential of certain scFv-based reagents.

Human serum albumin (HSA) is associated with several physiological functions, such as maintaining... more Human serum albumin (HSA) is associated with several physiological functions, such as maintaining oncotic pressure and microvascular integrity, among others. It also represents the major and predominant antioxidant in plasma due to the presence of the Cys34 sulfhydryl group. In this study, we assessed qualitative and quantitative changes in HSA in patients with heart failure (HF) and their relationship with the severity of the disease. We detected by means of mass spectrometry a global decrease of the HSA content in the plasma of HF patients in respect to control subjects, a significant increase of thio-HSA with a concomitant decrease in the reduced form of albumin. Cysteine and, at a lesser extent, homocysteine represent the most abundant thiol bound to HSA. A strong inverse correlation was also observed between cysteine-HSA and peak VO2/kg, an index of oxygen consumption associated with HF severity. Brown adipose tissue (BAT) is an endocrine adipose tissue with attributes to dissipate energy as ... more Brown adipose tissue (BAT) is an endocrine adipose tissue with attributes to dissipate energy as heat in response to changes in temperature and diet. Infrared thermography (IRT) has been studied in recent years in the assessment of BAT thermogenesis, as an option to positron emission tomography-computed tomography (PET-CT), because of several advantages. We performed a systematic review on the use of IRT in BAT assessment. Comprehensive online search was performed in different databases. The QUADAS 2 tool was used to evaluate studies' quality. 12 studies fit the inclusion criteria, whereas only one of these was considered of low risk of bias. 10 studies were favorable to IRT appliance in BAT evaluation, observing elevation of supraclavicular skin temperature correlated with BAT activity. Studies were heterogeneous in design, and a meta-analysis was precluded. Further studies with similar methodologies are needed. Conclusion: Despite the large number of published methodologies, IRT is a promising method for detecting BAT activation. Current knowledge already allows a better understanding of thermography to improve and standardize the technique.

Introduction Hip magnetic resonance arthrography (MRA) is the pre-operative imaging modality of c... more Introduction Hip magnetic resonance arthrography (MRA) is the pre-operative imaging modality of choice in patients with labral damage, with several classifications of labral tears having been reported based on MRA findings. Nevertheless, none of the available classification systems allows the surgeon to predict before surgery how a labral tear could be treated. Our purpose was to develop a new MRA-based scoring system of labral tears to predict before surgery the treatment option more suitable for labral tears. Materials and methods Forty-seven patients (29 males and 18 females; mean age: 35.9 ± 12.4) performed hip MRA for suspicious of femoroacetabular impingement and were afterwards subjected to arthroscopic treatment. Two musculoskeletal radiologists reviewed all pre-operative examinations and provided the Extension-Thickness-Damage score for each patient, based on Extension of tear, Thickness of labrum, and type of Damage. Chondral lesions grading was based on the arthroscopic findings according to Konan classification. For statistical purposes, patients were divided into two groups, depending on the type of treatment: labral repair or debridement. Mann-Whitney U, Chi-square, receiver operator curves, and Cohen kappa statistics were used. Results 35/47 underwent repair, while 12/47 were debrided. In both groups, the median chondral damage was grade III, with no significant differences (p = 0.439). The median Extension-Thickness-Damage score in the repair group (6) was significantly lower (p < 0.001) than that in the debridement group (8). The highest diagnostic performance (area under the curve) of Extension-Thickness-Damage was 0.819. The inter-observer agreement was substantial in the evaluation of Extension (k = 0.626) and Thickness (k = 0.771), and almost perfect for Damage (k = 0.827). Higher scores of Extension and Thickness were more frequently associated with debridement (p < 0.001; p = 0.0016, respectively), with no significant differences on the basis of Damage parameter (p = 0.284). Conclusions The MRA-based Extension-Thickness-Damage score could represent a helpful pre-operative tool, expressing the extent of the damage and its reparability before arthroscopy.

We previously observed that AML blast cells, cultured in vitro from a KIT D816Y patient, give rise to adherent cells with mast-cell like phenotype and tryptase was released in the serum-free medium. To correlate total serum tryptase (ts-try) levels with cytogenetic features and KIT mutational status, we analyzed serum samples from AML patients at diagnosis. In an effort to set living pericardial material for aortic valve reconstruction, we have previously assessed the efficiency of a recellularization strategy based on a perfusion system enabling mass transport and homogenous distribution of aortic valve-derived "interstitial" cells inside decellularized pericardial material. In the present report, we show that alternate perfusion promoted a rapid growth of valve cells inside the pericardial material and the activity of a proliferation-supporting pathway, likely controlled by the YAP transcription factor, a crucial component of the Hippo-dependent signaling cascade, especially between 3 and 14 days of culture. Quantitative mass spectrometry analysis of protein content in the tissue constructs showed deposition of valve proteins in the decellularized pericardium with a high variability at day 14 and a reproducible tissue maturation at 21 days. These results represent a step forward in the definition of strategies to produce a fully engineered tissue for replacing the calcified leaflets of failing aortic valves.

This exploratory retrospective study aims to investigate the thermal changes in the thyroid gland... more This exploratory retrospective study aims to investigate the thermal changes in the thyroid gland region of patients with hypothyroidism and fibromyalgia by analyzing the temperature of the brown adipose tissue (BAT). A total of 166 individuals from 1000 thermographic electronic medical records were classified into four groups: Group HP + FM-50 individuals with hypothyroidism and fibromyalgia; Group FM-56 individuals with fibromyalgia only; Group HP-30 individuals with hypothyroidism only, and Group Control-30 healthy individuals. The thermal images from the electronic medical records were acquired by a FLIR T650SC infrared camera (used for thermometry) and the temperature data for each group were statistically analyzed. Group HP + FM showed r = 0, meaning that the average temperatures of the thyroid and BAT are independent of each other. Groups FM, HP and Control showed r = 1, meaning that the average temperatures of the thyroid and BAT were directly related. Our findings showed that the average temperatures of the thyroid and BAT regions are similar. Also, there was no correlation between thyroid gland temperature and the presence of hypothyroidism or fibromyalgia using thermometry. Medical infrared thermography (MIT) is a noninvasive and nonradioactive analysis method capable of analyzing physiological functions related to the control of skin temperature, an important organ for controlling body temperature 1,2. This thermography´s technique allows the evaluation of physiological changes 2,3 , with applications in the field of medicine to identify neurological, rheumatological and dermatological disorders, vascular diseases, urologic, and gynecological and orthopedic pathologies 3-7 , and this method can provide support for sports medicine 3. Fibromyalgia (FM) is a rheumatologic disorder, of undefined cause, but its development is associated with the central nervous system's regulation of pain 8,9 , neurosensory, neuroendocrine and neurotransmitter-related disorders, as well as a genetic predisposition 10,11. Infra-Vermelha da Difusão do Óxido Nitroso Via Inalatória para a Cavidade Abdominal, em Ratos Sub... more Infra-Vermelha da Difusão do Óxido Nitroso Via Inalatória para a Cavidade Abdominal, em Ratos Submetidos a Pneumoperitônio JUSTIFICATIVA E OBJETIVOS: O óxido nitroso (N 2 O), por ser uma estrutura tri-atômica assimétrica, assume características de alta emissão e absorção de energia no espectro infra-vermelho, com um pico característico de absorção em 4,5 µm, o que o torna visível ao infra-vermelho curto, quando contrastado com uma fonte emissora de calor (anteparo quente). Diversos autores têm descrito a difusão do N 2 O para cavidades fechadas por métodos como cromatografia gasosa e analisador de gases, que não permitem um estudo macroscópico detalhado do gás. O presente estudo teve como objetivo a filmagem macroscópica no espectro infra-vermelho da difusão de N 2 O, utilizado em anestesia inalatória, para a cavidade peritoneal de ratos submetidos a pneumoperitônio de 20 mmHg com ar ambiente. MÉTODO: Dividiu-se os animais em três grupos, de acordo com o anestésico utilizado: I-controle venoso: tiopental intra-peritoneal; II-controle inalatório: isoflurano a 1,2% em O 2 100%; III-óxido nitroso: N 2 O 66% em oxigênio e isoflurano a 0,6%. Os termogramas provenientes da descompressão abdominal foram obtidos, por meio de um radiômetro AGEMA 550 filmados a 7 quadros por segundo. RESULTADOS: O N 2 O demonstrou-se visível ao infra-vermelho. No momento da descompressão abdominal, não houve nos grupos I e II termogramas com rastros de gases visíveis ao infra-vermelho. Houve, porém, rastros de gases visíveis ao infra-vermelho no grupo III. CONCLUSÕES: Conclui-se que o óxido nitroso inalatório a 66% difundiu-se para a cavidade peritoneal de ratos submetidos a pneumoperitônio de 20 mmHg com ar ambiente, sem aumento de pressão intra-abdominal.

Infrared thermal imaging has been used to assess possible complications, such as necrosis and changes in micro- and macro-circulation, based on perforator mapping techniques, among others. The objective of this study was to present two clinical cases involving thermal imaging monitoring of the healing process of lipoabdominoplasty in the immediate postoperative evaluation and its preliminary results. Infrared thermography was performed 24 hours after the operation and on postoperative days 5, 25, and 27. The aim of the present study was to evaluate low back pain, during pregnancy, using thermography together with artificial intelligence. METHODS A cross-sectional study was carried out with pregnant women recruited from a university hospital. The following data were collected: (a) clinical data; (b) physical assessment with mobility and low back pain provocation tests; and (c) thermograms acquisitions, in a controlled environment. Artificial intelligence and the statistical tests were used to compare the groups' mean: with low back pain (LBP) and without low back pain (WLBP). RESULTS Thirty pregnant women took part, with fifteen in each group. The mean ± Standard Deviation temperature of the lumbar region in both groups were 32.7 ± 1.05 °C and 32.6 ± 1.01 °C for LBP and WLBP, respectively. There was not any difference in temperature between the groups; however, the artificial intelligence software found thermogram differences between groups; furthermore, the correlation between pain intensity and functionality was found. CONCLUSION Thermography associated with artificial intelligence analyses demonstrated to be a promising method as an adjunct to clinical evaluation.

First, TEM1 is expressed in the stroma and neo-vasculature of many human carcinomas but is largely absent from healthy adult tissues. Second, TEM1 is expressed by tumor cells of mesenchymal origin, notably sarcoma. Here, we present two fully human anti-TEM1 single-chain variable fragment (scFv) reagents, namely, 1C1m and 7G22, that recognize distinct regions of the extracellular domain and possess substantially different affinities. In contrast to other, well-described anti-TEM1 binders, these fragments confer cytolytic activity when expressed as 2nd generation chimeric antigen receptors (CARs). Moreover, both molecules selectively redirect human T cell effector functions toward TEM1+ tumor cells when incorporated into experimental soluble bispecific trivalent engagers that we term TriloBiTEs (tBs). Furthermore, systemic delivery of 1C1m-tB prevents the establishment of Ewing sarcoma tumors in a xenograft model. Our observations confirm TEM1 as a promising target for cancer immunotherapy and illustrate the prospective translational potential of certain scFv-based reagents.

It also represents the major and predominant antioxidant in plasma due to the presence of the Cys34 sulfhydryl group. In this study, we assessed qualitative and quantitative changes in HSA in patients with heart failure (HF) and their relationship with the severity of the disease. We detected by means of mass spectrometry a global decrease of the HSA content in the plasma of HF patients in respect to control subjects, a significant increase of thio-HSA with a concomitant decrease in the reduced form of albumin. Cysteine and, at a lesser extent, homocysteine represent the most abundant thiol bound to HSA. Infrared thermography (IRT) has been studied in recent years in the assessment of BAT thermogenesis, as an option to positron emission tomography-computed tomography (PET-CT), because of several advantages. We performed a systematic review on the use of IRT in BAT assessment. Comprehensive online search was performed in different databases. The QUADAS 2 tool was used to evaluate studies' quality. 12 studies fit the inclusion criteria, whereas only one of these was considered of low risk of bias. 10 studies were favorable to IRT appliance in BAT evaluation, observing elevation of supraclavicular skin temperature correlated with BAT activity. Studies were heterogeneous in design, and a meta-analysis was precluded. Further studies with similar methodologies are needed. Conclusion: Despite the large number of published methodologies, IRT is a promising method for detecting BAT activation. Current knowledge already allows a better understanding of thermography to improve and standardize the technique.

Nevertheless, none of the available classification systems allows the surgeon to predict before surgery how a labral tear could be treated. Our purpose was to develop a new MRA-based scoring system of labral tears to predict before surgery the treatment option more suitable for labral tears. Materials and methods Forty-seven patients (29 males and 18 females; mean age: 35.9 ± 12.4) performed hip MRA for suspicious of femoroacetabular impingement and were afterwards subjected to arthroscopic treatment. Two musculoskeletal radiologists reviewed all pre-operative examinations and provided the Extension-Thickness-Damage score for each patient, based on Extension of tear, Thickness of labrum, and type of Damage. Chondral lesions grading was based on the arthroscopic findings according to Konan classification. For statistical purposes, patients were divided into two groups, depending on the type of treatment: labral repair or debridement. Mann-Whitney U, Chi-square, receiver operator curves, and Cohen kappa statistics were used. Results 35/47 underwent repair, while 12/47 were debrided. In both groups, the median chondral damage was grade III, with no significant differences (p = 0.439). The median Extension-Thickness-Damage score in the repair group (6) was significantly lower (p < 0.001) than that in the debridement group (8). The highest diagnostic performance (area under the curve) of Extension-Thickness-Damage was 0.819. The inter-observer agreement was substantial in the evaluation of Extension (k = 0.626) and Thickness (k = 0.771), and almost perfect for Damage (k = 0.827). Higher scores of Extension and Thickness were more frequently associated with debridement (p < 0.001; p = 0.0016, respectively), with no significant differences on the basis of Damage parameter (p = 0.284). Conclusions The MRA-based Extension-Thickness-Damage score could represent a helpful pre-operative tool, expressing the extent of the damage and its reparability before arthroscopy.

We previously observed that AML blast cells, cultured in vitro from a KIT D816Y patient, give rise to adherent cells with mast-cell like phenotype and tryptase was released in the serum-free medium. To correlate total serum tryptase (ts-try) levels with cytogenetic features and KIT mutational status, we analyzed serum samples from AML patients at diagnosis.
