Autobiographical memory (AM) represents the ability to remember personal experiences. There are s... more Autobiographical memory (AM) represents the ability to remember personal experiences. There are several laboratory or neuropsychological tasks to assess different aspects of memory function. However, there has been little research on self-reported AM ability. The Survey of Autobiographical Memory (SAM) is a self-report questionnaire, developed to assess individual differences in AM. Evidence for the factor structure and network properties of the SAM is still needed. Here, using an integral approach, we validated the SAM using a Multiple Correspondence Analysis (Study 1) as in the original study, a Confirmatory Factor Analysis (Study 2), and a Network Analysis (Study 3) using two large independent samples (total n = 2896). We first replicated the original findings, then confirmed the existence of 4 factors and found that the Episodic and Semantic categories were the most central ones. Finally, we found between "groups" differences for Gender and Anxiety. Overall, three different methods revealed a robust profile of the SAM.

It represents a unifier concept within the memory field, as it is the driving force of memory acquisition and updating. Prediction error induces updating of consolidated memories in strength or content by memory reconsolidation. This process has two different neurobiological phases, which involves the destabilization (labilization) of a consolidated memory followed by its restabilization. The aim of this work is to emphasize the functional role of PE on the neurobiology of learning and memory, integrating and discussing different research areas: behavioral, neurobiological, computational and clinical psychiatry.

Especially in agonistic encounters, remembering an opponent and the previous fight could allow avoiding new conflicts. Considering this, we hypothesized that this type of social interaction forms a long-term recognition memory lasting several days. It has been shown that a second encounter 24 hours later between the same pair of zebrafish males is resolved with lower levels of aggression. Here, we evaluated if this behavioral change could last for longer intervals and a putative mechanism associated with memory storage: the recruitment of NMDA receptors. We found that if a pair of zebrafish males fight and 48 or 72 hours later fight again, they resolved the second encounter with lower levels of aggression. We present a study of the CNS of Neohelice granulata performed at both the neuronal cluster and t... more We present a study of the CNS of Neohelice granulata performed at both the neuronal cluster and the neuropil level, describing eyestalk ganglia, the thoracic ganglion, and the central brain, with special focus on the latter. This characterization is based on the functional study of the crab´s nervous system during two main phases: visual information processing, and storage of memory. The nervous system delineation uses classical histological techniques as well as immunohistochemistry to characterize cellular clusters and neuropils. This is the first general description of N. granulata nervous system as a whole, at neuropil and cellular level. A Prediction-Error (PE) is a mismatch between expected and current events. It was proposed as the process by which memories are acquired. But, our memories like ourselves are subject to change. Thus, an acquired memory can become active and update its content or strength by a labilization-reconsolidation process. Within the reconsolidation framework, PE drives the updating of consolidated memories. Moreover, memory features, such as strength and age, are crucial boundary conditions that limit the initiation of the reconsolidation process. In order to disentangle these boundary conditions, we review the role of surprise, classical models of conditioning, and their neural correlates. Several forms of PE were found to be capable of inducing memory labilization-reconsolidation. Consolidation theory assumes that memories are labile during a limited time window after acquisit... more Consolidation theory assumes that memories are labile during a limited time window after acquisition, but as time passes, memories become stable and resistant to amnesic agents. However, the vision of immutable memories after consolidation has been challenged. Thus, after the presentation of a reminder, the reactivated old memories become labile and again susceptible to amnesic treatments. This process implies a re-stabilization phase, usually referred to as reconsolidation. Gamma-aminobutyric acid (GABA) is the major inhibitory neurotransmitter both in the Central nervous system (CNS) and in the periphery. A considerable amount of evidence has arisen from different studies regarding the role of the GABA(A) receptor in diverse behavioral paradigms and tasks. Here, we investigate the role of the GABAergic system on both memory consolidation and reconsolidation phases by using the memory paradigm of the crab Chasmagnathus. In order to achieve such a goal, we design pharmacological-behavioral experiments, which include the administration of classic agonist (muscimol) and antagonist (bicuculline) of the mammals GABA(A) receptors. The current results show that the systemic administration of muscimol impairs the consolidation and reconsolidation processes. In contrast, the administration of bicuculline improves the consolidation and reconsolidation processes. Furthermore, the co-administration of both drugs blocks the agonist amnesic effect on the consolidation phase. The ubiquity of the neurotransmitter and its receptors in the animal taxa allows us to use the classic agonist-and-antagonist administration procedure in this invertebrate. Thus, all the results reported in this paper can be judged as a result of the modulation exerted by the functional state of the GABAergic system in the CNS. To conclude, the results obtained in this report with an invertebrate model represent additional evidences supporting the view that some molecular mechanisms subserving different memory phases could be the basic tools employed by phylogenetically disparate animals.

Robust retention is shown at 24 h after 15 trials of shadow presentation or at 120 h after 30 trials, but no significant retention is disclosed at 24 h after 5 trials or at 72 h after 15. A cocktail of the cAMP membrane permeable analog 8-(4-chlorophenylthio)-cAMP (CPT-cAMP), plus the phosphodiesterase inhibitor isobutyl methylxanthine (IBMX), was given by systemic administration. Pretraining injection of the cocktail (25 or 50/~M, 15 min before a 5-trial session) failed to affect short-term habituation, but induced significant retention when tested at 24 h. This facilitatory effect was not shown when a lower dose (5 pM) was used. A post-training injection of 25 pM, immediately after a 5-or 15-trial session, induced retention when tested at 24 or 72 h, respectively. Thus, the administration of CPT-cAMP + IBMX during acquisition of a habituated response or immediately after, improves long-term habituation, a result supporting the view that an increase in the cAMP level is one of the steps in long-term memory consolidation.

Deregulation of consolidation and reprocessing of mnemonic traces that encode fearful experiences might result in fear-related psychopathologies. Here, we assessed how pre-established memories change with experience, particularly the labilization/reconsolidation of memory, using the whole-brain analysis technique of positron emission tomography in male mice. We found differences in glucose consumption in the lateral neocortex, hippocampus and amygdala in mice that underwent labilization/reconsolidation processes compared to animals that did not reactivate a fear memory. We used chemogenetics to obtain insight into the role of cortical areas in these phases of memory and found that the lateral neocortex is necessary for fear memory reconsolidation. Re-exposure to specific learned cues during sleep or awake states, namely targeted memory reactivation, has been shown to improve memory retention at long delays. Manipulation of memory reactivation could have potential clinical value in populations with memory deficits or cognitive decline. However, no previous study investigated a target memory reactivation approach on those populations. Once consolidated they can enter a new labile state after the presentation of a reminder of the original memory, followed by a period of re-stabilization (reconsolidation). During these periods of lability the memory traces can be modified. Currently, some studies show a rapid stabilization after 30 min, while others show that stabilization occurs after longer periods (e.g. > 6 h). Here we investigate the effect of an interference treatment on declarative memory consolidation, comparing distinct time intervals after acquisition. On day 1, participants learned a list of non- syllable pairs (List 1). 5 min, 30 min, 3 h or 8 h later, they received an interference list (List 2) that acted as an amnesic agent. However, the dynamics of the word memory formation is still unclear. Here, we addressed the temporal profile of two key cognitive markers of memory consolidation in the domain of word learning: i) the susceptibility of recently learned novel words to memory interference; ii) their lexical integration using a semantic judgment task while recording the ERPs responses. Young adults acquired a set of novel picture-label-meaning associations. In a first experiment, we performed a temporal gradient of retroactive interference (5 min, 30 min, 4 h and 24 h) and evaluated the memory retention 48 h after learning. However, this approach is focused on implicit responses evaluated in a single day. Here, we evaluated negative-valence, positive-valence and cognitive-systems in order to evaluate the extent to which threat conditioning models anxiety disorders. Subjects underwent threat conditioning and five-minutes (Short-term evaluation) or 48 hs (Long-term evaluation) later, both groups performed several tasks targeting cognitive-systems and valenced-systems. In the short-term evaluation, successful conditioning maintained state-anxiety and increased the aversiveness representation of the CS+ and the valuation for negative events. Reaction-times for the CS+ were faster, reflecting an attentional bias toward threat. In the long-term evaluation, participants represented the CS+ as more aversive and generalized to all stimuli. During this process of labilization/restabilization memory traces can be either impaired, strengthened or updated in content. Here, we study if a consolidated memory can be updated by linking one original cue to two different outcomes and whether this process was modulated by the GABAergic system. To aim that, we designed two experiments carried out in three consecutive days. All participants learned a list of non-sense syllable pairs on day 1. On day 2 the new information was introduced after the reminder or no-reminder presentation. Participants were tested on day 3 for the updated or original list (Exp. 1). The most common behavioral tool used to reveal this process is interference produced by new learning shortly after memory reactivation. Memory interference is defined as a decrease in memory retrieval, the effect is generated when new information impairs an acquired memory. In general, the target memory and the interference task used are the same. Here we investigated how different memory systems and/or their valence could produce memory reconsolidation interference. Memory storage is based on alterations in the strength of synaptic connections between neurons. The glutamatergic synapses undergo various forms of N-methyl-D aspartate receptor (NMDAR)-dependent changes in strength, a process that affects the abundance of other receptors at the synapse and underlies some forms of learning and memory. Here we propose a direct regulation of the NMDAR. Changes in NMDAR's functionality might be induced by the modification of the subunit's expression or cellular trafficking. This trafficking does not only include NMDAR's movement between synaptic and extra-synaptic localizations but also the cycling between intracellular compartments and the plasma membrane, a process called surface expression. Consolidated memories return to a labile state after the presentation of cues (reminders) associa... more Consolidated memories return to a labile state after the presentation of cues (reminders) associated with acquisition, followed by a period of stabilization (reconsolidation). However not all cues are equally effective in initiating the process, unpredictable cues triggered it, predictable cues do not. We hypothesize that the different effects observed by the different reminder types on memory labilization-reconsolidation depend on a differential neural involvement during reminder presentation. To test it, we developed a declarative task and compared the efficacy of three reminder types in triggering the process in humans (Experiment 1). Finally, we compared the brain activation patterns between the different conditions using functional magnetic resonance imaging (fMRI) (Experiment 2). We confirmed that the unpredictable reminder is the most effective in initiating the labilization-reconsolidation process. Furthermore, only under this condition there was differential left hippocampal activation during its presentation. We suggest that the left hippocampus is detecting the incongruence between actual and past events and allows the memory to be updated.

A Prediction-Error (PE) is a mismatch between expected and current events. It was proposed as the process by which memories are acquired. But, our memories like ourselves are subject to change. Thus, an acquired memory can become active and update its content or strength by a labilization-reconsolidation process. Within the reconsolidation framework, PE drives the updating of consolidated memories. Moreover, memory features, such as strength and age, are crucial boundary conditions that limit the initiation of the reconsolidation process. In order to disentangle these boundary conditions, we review the role of surprise, classical models of conditioning, and their neural correlates. Several forms of PE were found to be capable of inducing memory labilization-reconsolidation. Consolidation theory assumes that memories are labile during a limited time window after acquisit... more Consolidation theory assumes that memories are labile during a limited time window after acquisition, but as time passes, memories become stable and resistant to amnesic agents. However, the vision of immutable memories after consolidation has been challenged. Thus, after the presentation of a reminder, the reactivated old memories become labile and again susceptible to amnesic treatments. This process implies a re-stabilization phase, usually referred to as reconsolidation. Gamma-aminobutyric acid (GABA) is the major inhibitory neurotransmitter both in the Central nervous system (CNS) and in the periphery. A considerable amount of evidence has arisen from different studies regarding the role of the GABA(A) receptor in diverse behavioral paradigms and tasks. Here, we investigate the role of the GABAergic system on both memory consolidation and reconsolidation phases by using the memory paradigm of the crab Chasmagnathus. In order to achieve such a goal, we design pharmacological-behavioral experiments, which include the administration of classic agonist (muscimol) and antagonist (bicuculline) of the mammals GABA(A) receptors. The current results show that the systemic administration of muscimol impairs the consolidation and reconsolidation processes. In contrast, the administration of bicuculline improves the consolidation and reconsolidation processes. Furthermore, the co-administration of both drugs blocks the agonist amnesic effect on the consolidation phase. The ubiquity of the neurotransmitter and its receptors in the animal taxa allows us to use the classic agonist-and-antagonist administration procedure in this invertebrate. Thus, all the results reported in this paper can be judged as a result of the modulation exerted by the functional state of the GABAergic system in the CNS. To conclude, the results obtained in this report with an invertebrate model represent additional evidences supporting the view that some molecular mechanisms subserving different memory phases could be the basic tools employed by phylogenetically disparate animals.

Robust retention is shown at 24 h after 15 trials of shadow presentation or at 120 h after 30 trials, but no significant retention is disclosed at 24 h after 5 trials or at 72 h after 15. A cocktail of the cAMP membrane permeable analog 8-(4-chlorophenylthio)-cAMP (CPT-cAMP), plus the phosphodiesterase inhibitor isobutyl methylxanthine (IBMX), was given by systemic administration. Pretraining injection of the cocktail (25 or 50/~M, 15 min before a 5-trial session) failed to affect short-term habituation, but induced significant retention when tested at 24 h. This facilitatory effect was not shown when a lower dose (5 pM) was used. A post-training injection of 25 pM, immediately after a 5-or 15-trial session, induced retention when tested at 24 or 72 h, respectively. Thus, the administration of CPT-cAMP + IBMX during acquisition of a habituated response or immediately after, improves long-term habituation, a result supporting the view that an increase in the cAMP level is one of the steps in long-term memory consolidation.

Deregulation of consolidation and reprocessing of mnemonic traces that encode fearful experiences might result in fear-related psychopathologies. Here, we assessed how pre-established memories change with experience, particularly the labilization/reconsolidation of memory, using the whole-brain analysis technique of positron emission tomography in male mice. We found differences in glucose consumption in the lateral neocortex, hippocampus and amygdala in mice that underwent labilization/reconsolidation processes compared to animals that did not reactivate a fear memory. We used chemogenetics to obtain insight into the role of cortical areas in these phases of memory and found that the lateral neocortex is necessary for fear memory reconsolidation. Here, we addressed the temporal profile of two key cognitive markers of memory consolidation in the domain of word learning: i) the susceptibility of recently learned novel words to memory interference; ii) their lexical integration using a semantic judgment task while recording the ERPs responses. Young adults acquired a set of novel picture-label-meaning associations. However, this approach is focused on implicit responses evaluated in a single day. Here, we evaluated negative-valence, positive-valence and cognitive-systems in order to evaluate the extent to which threat conditioning models anxiety disorders. Subjects underwent threat conditioning and five-minutes (Short-term evaluation) or 48 hs (Long-term evaluation) later, both groups performed several tasks targeting cognitive-systems and valenced-systems. In the short-term evaluation, successful conditioning maintained state-anxiety and increased the aversiveness representation of the CS+ and the valuation for negative events. Reaction-times for the CS+ were faster, reflecting an attentional bias toward threat. In the long-term evaluation, participants represented the CS+ as more aversive and generalized to all stimuli. Consolidated memories return to a labile state after the presentation of cues (reminders) associa... more Consolidated memories return to a labile state after the presentation of cues (reminders) associated with acquisition, followed by a period of stabilization (reconsolidation). However not all cues are equally effective in initiating the process, unpredictable cues triggered it, predictable cues do not. We hypothesize that the different effects observed by the different reminder types on memory labilization-reconsolidation depend on a differential neural involvement during reminder presentation. To test it, we developed a declarative task and compared the efficacy of three reminder types in triggering the process in humans (Experiment 1). Finally, we compared the brain activation patterns between the different conditions using functional magnetic resonance imaging (fMRI) (Experiment 2). We confirmed that the unpredictable reminder is the most effective in initiating the labilization-reconsolidation process. Furthermore, only under this condition there was differential left hippocampal activation during its presentation. We suggest that the left hippocampus is detecting the incongruence between actual and past events and allows the memory to be updated.
