Results from verification of the conversion of BactoScan FC counts (dimension IBC/μl) onto the sc... more Results from verification of the conversion of BactoScan FC counts (dimension IBC/μl) onto the scale of the anchor method (colony count reference method, dimension: cfu/ml) for the purpose of raw cow milk testing are described. The study was performed in Slovakia in two phases (preliminary set-up and verification) during 2002/2003 using 1281 pairs of measurements. The relationship between BactoScan FC results and anchor methods was log10 cfu/ml = 0.935 log 1 0 IBC/μl + 2.812, with an accuracy of the estimate of s y x = 0.25 log 1 0 cfu/ml.

Žinčica is a popular Slovak dairy product made from ewes’ milk. It is a by-product resulting duri... more Žinčica is a popular Slovak dairy product made from ewes’ milk. It is a by-product resulting during ewes’ lump cheese processing. Microbiota in Žinčica have rarely been studied, especially enterococci; however, they can produce beneficial substances – bacteriocins. In this study, the presence of Enterocins (Ents) genes were analysed in enterococci from Žinčica and partially also the inhibition activity. Samples of Žinčica were collected from different agrofarms producing ewes’ lump cheese (34) in Central Slovakia. In the enterococci tested, Ent P gene was the most frequently detected (in 6 out of 7 enterococci), followed by Ent A and Ent L50B genes. Ent B gene was detected only in E. faecium 30E1. On the other hand, E. faecalis 31E2 did not contain Ent genes, although it showed inhibition activity against the indicator strains Enterococcus avium EA5, Staphylococcus aureus SA5, Listeria monocytogenes CCM4699 (inhibition zone sizing up to 20 mm). In contrast, the temperature optimum (50-52°C) was not affected significantly by the intracellular localization. Abstract The influence of several parameters, such as temperature, pH, and concentration of buffe... more Abstract The influence of several parameters, such as temperature, pH, and concentration of buffer and solvent, on the release of β-galactosidase from Kluyveromyces marxianus cells was studied. In optimal conditions (37°C, pH 9.5–10.5) greater than 90% of the intracellular β-galactosidase activity was released into 0.1-0.5 m phosphate buffer after 1.5-2.0 h treatment with 1% chloroform. The described method is simple, effective, relatively fast, and selective.

Experiences with the introduction of the BactoScan FC method (BSc) in Slovakia are described. The... more Experiences with the introduction of the BactoScan FC method (BSc) in Slovakia are described. The emphasis is put on the conversion establishment. The ex-farm cow-milk samples (n=910) were collected from August to December 2002 and they were analyzed by BSc and by an anchor method - a standard plate-count method with colony-forming units (CFU)/ml as measuring units. Data from 827 samples were used for the calculation of the relationship between the results of both methods. Setting log 10 Individual Bacterial Cells (IBC) as the independent variable x, the linear regression log 10 CFU/ml = 0.936 log 10 IBC + 2.818 with an accuracy of the estimate of s y,x = 0.263 log 10 CFU/ml was computed. However, the reliability of this conversion should be verified permanently.

Rapid lactose hydrolysis by small amounts of permeabilized cells following the fermentation of released glucose and galactose by S. cerevisiae resulted in a twofold enhancement of the overall volumetric productivity (1.03 g/l × h), compared to the fermentation in which the lactose was directly fermented by K. marxianus.

The aim of this study was the analysis of measurement of abilities and properties of RT (Real Time) system (AfiLab = AfiMilk (NIR measurement unit (near infrared spectroscopy) and electrical conductivity (C) of milk by conductometry) + AfiFarm (calibration and interpretation software)) for the analysis of individual milk samples (IMSs). There were 2 × 30 IMSs in the experiment. Results are important for inheritance check. The occurrence of errors in the data may compromise the efficiency of breeding of dairy cows. The aim was possibility to reduce the incidence of MR database errors. Analyses of frequency distribution of MR data deviations from different sources and estimations of limits of difference acceptability in milk recording were performed. The results of MR control days of flowmeter in parlor (DMY) were paired to the AVG7 results (average for 7 days) from the same flowmeter (n = 16,247, original recordings of complete lactations). The individual differences in milk yield indicators were calculated between successive MR control days (DMY – R, monthly interval, the reference value (R) = previous DMY) for MR data file. Strains potential such as bacteriocin activity, biofilm formation ability, growth in skim milk, s... more Strains potential such as bacteriocin activity, biofilm formation ability, growth in skim milk, susceptibility to antibiotics, tolerance to bile and low pH as well as enzyme production was tested in the species Lacticaseibacillus paracasei and Lactococcus lactis detected in Slovak raw goat milk. The strains showed mostly low-grade biofilm formation ability, susceptibility to antibiotics and sufficient tolerance to oxgall/bile. Lacticaseibacillus (Lcb.) paracasei ZM-1, ZM-2 and Lactococcus (Lc.) lactis PD MO 1/8 showed high tolerance to pH 3 (67 %, 83 % and 63 %, respectively). The strains showed bacteriocin activity against the principal indicator Enterococcus avium EA5 (inhibition zone ranging 5–24 mm). A concentrated substance of Lcb. paracasei LPa ML 12/1 (pH 6.3) inhibited EA5 strain (inhibition activity 100 AU ml–1). Seventeen staphylococci isolated from 54 Slovak local lump cheeses made from ewes’ milk were taxo... more Seventeen staphylococci isolated from 54 Slovak local lump cheeses made from ewes’ milk were taxonomically allotted to five species and three clusters/groups involving the following species: Staphylococcus aureus (5 strains), Staphylococcus xylosus (3 strains), Staphylococcus equorum (one strain) Staphylococcus succinus (5 strains) and Staphylococcus simulans (3 strains). Five different species were determined. The aim of the study follows two lines: basic research in connection with staphylococci, and further possible application of the bacteriocins. Identified staphylococci were mostly susceptible to antibiotics (10 out of 14 antibiotics). Strains showed γ-hemolysis (meaning they did not form hemolysis) except for S. aureus SAOS1/1 strain, which formed β-hemolysis. S. aureus SAOS1/1 strain was also DNase positive as did S. aureus SAOS5/2 and SAOS51/3. The other staphylococci were DNase negative. The work is focused on the evaluation of seasonal dynamics of the total count of microorganisms i... more The work is focused on the evaluation of seasonal dynamics of the total count of microorganisms in sheep’s milk and on the proposal of developing cut-off values for standard limitation for the next fifteen years for hygienic safety support. The total count of microorganisms was measured between years 2012–2014 (n = 4,746). The results were statistically evaluated using medians, geometric means, arithmetic means, means of log values (log10) and standard deviations for the cut-off limit determination. This model was patterned on the maximum of total count of microorganisms in the tested percentiles 95 (statistical conventional interval), 91, 90, 80 and 70% and on the result of medians (from 109 to 148 × 103 cfu/ml). Stored ewe’s milk lump cheese is a local product that can be a source of autochthonous beneficial... more Stored ewe’s milk lump cheese is a local product that can be a source of autochthonous beneficial microbiota, especially lactic acid bacteria. The aim of this study was to show the antimicrobial potential of Lactiplantibacillus plantarum LP17L/1 isolated from stored ewe’s milk lump cheese. Lpb. plantarum LP17L/1 is a non-hemolytic, non-biofilm-forming strain, susceptible to antibiotics. It contains genes for 10 bacteriocins—plantaricins and exerted active bacteriocin with in vitro anti-staphylococcal and anti-listerial effect. It does not produce damaging enzymes, but it produces β-galactosidase. It also sufficiently survives in Balb/c mice without side effects which indicate its safety. Moreover, a reduction in coliforms in mice jejunum was noted. LP17L/1 is supposed to be a promising additive for Slovak local dairy products.

The total count of microorganisms was measured between years 2012–2014 (n = 4,746). The results were statistically evaluated using medians, geometric means, arithmetic means, means of log values (log10) and standard deviations for the cut-off limit determination. This model was patterned on the maximum of total count of microorganisms in the tested percentiles 95 (statistical conventional interval), 91, 90, 80 and 70% and on the result of medians (from 109 to 148 × 103 cfu/ml). Stored ewe’s milk lump cheese is a local product that can be a source of autochthonous beneficial... more Stored ewe’s milk lump cheese is a local product that can be a source of autochthonous beneficial microbiota, especially lactic acid bacteria. The aim of this study was to show the antimicrobial potential of Lactiplantibacillus plantarum LP17L/1 isolated from stored ewe’s milk lump cheese. Lpb. plantarum LP17L/1 is a non-hemolytic, non-biofilm-forming strain, susceptible to antibiotics. It contains genes for 10 bacteriocins—plantaricins and exerted active bacteriocin with in vitro anti-staphylococcal and anti-listerial effect. It does not produce damaging enzymes, but it produces β-galactosidase. It also sufficiently survives in Balb/c mice without side effects which indicate its safety. Moreover, a reduction in coliforms in mice jejunum was noted. LP17L/1 is supposed to be a promising additive for Slovak local dairy products.
