This study described a case of renal cell carcinoma in an 8-year-old mare, which presented with an enlargement in the left flank, decreased appetite and straining to defecate. Clinically, a tumor in the left retroperitoneal space was detected by rectal examination and ultrasonography. The animal died after one month of clinical evolution. At necropsy, the left kidney was completely effaced by a 50cm in diameter, irregular, round, lobulated cystic mass with necro-hemorrhagic areas. Within the peritoneal cavity, there was severe hemoperitoneum and numerous sessile and pedunculated masses ranging from 2.0 to 15.0cm diameter that were attached to the visceral and parietal peritoneum. Histologically, the primary neoplasia and its metastasis were composed by a proliferation of epithelial cells, which were arranged in a tubulopapillary pattern. Diagnosis of renal cell carcinoma with peritoneal carcinomatosis and hemoperitoneum was based on macroscopic aspects, and mainly on the histological features.

The objective of this experiment was to macroscopically and microscopically investigate the process of healing by second intention in skin wounds of eight healthy gelding crossbred horses treated (T) or untreated (UT) with a single dose of leukocyte-poor PRP (LP-PRP). Three squareshaped wounds were induced in both gluteal regions. After 12 h of wound induction, 0.5 mL LP-PRP was administered in each border of the lesion in one of gluteal region. Contralateral wounds were UT. Six skin biopsies were obtained with a 6-mm Punch. Macroscopic variables of one of the non-biopsied wounds were evaluated. Samples were processed for histomorphometric evaluation. No difference was observed between the time required for wound closure in the two groups. Histomorphometric analysis performed 14 days after wound induction revealed higher (p = 0.034) angiogenesis and number (p = 0.0179) of total leukocytes in T wounds. Fibrocytes numbers increased significantly (p = 0.023) on day 7 after injury in the UT group. General microscopic evaluation performed independently of scores and morphometric analysis revealed that the majority of the T wounds showed better healing variables in the sections analyzed after complete macroscopic closure of the wound. A single injected dose of LP-PRP 12 h after wound induction in horses does not interfere with the wound healing process but reveals that majority of the T wounds exhibit better healing histomorphometric characteristics.

Research paper thumbnail of The pathogenicity of two Neospora caninum goat strains in a BALB/c mouse model

Experimental Parasitology, Oct 1, 2019

Goats are frequently described as an intermediate host for the protozoan Neospora caninum, manife... more Goats are frequently described as an intermediate host for the protozoan Neospora caninum, manifesting the disease mainly by recurrent abortions with placentitis and encephalitis in fetuses. Several reports of natural and experimental infections in cattle and mice show differences in the immune response, and the outcome of the infection can be variable depending on the species affected and by the behavior of the infective strain. This study describes for the first time two Neospora caninum strains isolated from naturally infected goats from the state of Minas Gerais, Brazil. One placenta and one brain from different goats were processed for a first bioassay in gerbils (Meriones unguiculatus). Subsequently, a second bioassay was performed by inoculating the processed brain samples from gerbils into Interferon gamma (IFN-γ) knockout mice (KO mice). Tachyzoites collected from the peritoneal fluid of the KO mice were inoculated into VERO cell monolayers, where they presented a very slow growth rate. The tachyzoites were also inoculated into BALB/c mice with a dose of 10 6 tachyzoites per animal. After a 5-week follow up, the animals infected with both of the strains developed a strong polarized Th1 response with increased serum and spleen gene expression levels of pro-inflammatory cytokines (mainly IFN-γ and TNF-α) in the first week. Tissue lesions were mild in the animals infected with both strains. Despite the strong immune response preventing an infection in the visceral organs, the parasite was able to reach the brain, causing progressive brain lesions from the second to fifth week post infection. The NC-goat1-infected mice presented with severe meningoencephalitis, but the NC-goat2-infected animals had considerable histological brain lesions only at week 5. Immunohistochemical analysis of the mouse brains revealed a different pattern of inflammatory cells compared to the naturally infected goats. A severe inflammatory infiltrate of CD3 + T lymphocytes was found in the NC-goat1-infected mice. A more discrete infiltrate of CD3 + T cells was found in the NC-goat2-infected animals. Additionally, IBA1 IHC revealed an intense microglial reaction and monocyte perivascular cuffs in the NC-goat1-infected animals and lower microglia/monocyte infiltrates in the NC-goat2infected mice. This work contributes knowledge on the pathogenicity of new Neospora caninum strains in mice, comparable with other well-established mouse models of the disease, and demonstrates the importance of studying goats as an intermediate host of this parasite.

Immunohistochemistry has been used for rabies diagnosis in formalin-fixed tissue with satisfactory results, although the time to diagnosis is considerably longer than that with direct immunofluorescence. The protocol provided a provisory histopathological rabies diagnosis in approximately three and half hours and the immunohistochemical diagnosis was available after six hours. The protocol achieved 100% correlation with direct immunofluorescence and is a promising method, particularly in situations in which only material in formalin is available for diagnosis or when the refrigeration or transportation of biological material is difficult.

The goats transmitted the parasite to their offspring in previous pregnancies. Histopathological examination showed a mild lymphoplasmacytic inflammatory infiltrate in skeletal muscles (10/168 muscles; 1.68%) and myocardium in two goats. Moderate to severe lymphoplasmacytic perivascular cuffs were the most significant lesion in the nervous system and were observed in 62.5% of the goats. Mild to moderate gliosis also affected 37.5% of them. Parasitic structures were not found in the tissues by histopathology and IHC labeling for N. caninum Polymerase chain reaction (PCR) analysis of 359 tissue samples showed 133 (37%) positive samples. The highest percentage of positive results was found in the nervous system (28/63; 44.4%) and skeletal muscle (72/168; 42.8%). Statistical analysis of the PCR results did not show parasitic preference for any particular muscle group (head, hindlimbs, neck, thoracoabdominal or forelimb muscles) or region of the nervous system. Yet, the highest percentage of PCR-positive samples was from the cleido-occipital, brachial triceps and semimembranosus muscles and the cervical segment of the spinal cord (62.5% each). These data suggest that PCR diagnosis of N. caninum infection should include analysis of these tissues. This is the first report of N. caninum distribution in adult female goat tissues.

It used 100 Nelore cattle, being divided into two groups of 50 animals, which received a cottonseed (CS) in grain form by the diet in the proportion of 15% and the other group was provided with control diet without CS in the feedlot for 88 days. The diets were isoprotein and isoenergetic and concentrate:roughage ratio of 60:40. The animals were slaughter in the end of experimental time and the collection of liver samples for histological analysis. After cooling 24 hours at ± 1°C were conducted evaluation of carcass and major cuts, final pH and collected muscle samples longissimus to carry out the physicochemical analyzes (Color, Cooking Loss, Pigments and Shear Force), chemical composition, fatty acid profile and oxidation. The inclusion of CS in the diet provided animals with higher slaughter weight, daily weight gain, Hindquarter, however, the animals showed reduced carcass yield, forequarter weight and Thin flank, without changing the parameters as rib-eye area and backfat thickness. There was an increase of the yellow values (b*) and moisture with the final pH reduction. There was no influence of the use of cottonseed in relation to histological parameters evaluated in the liver. The cottonseed in the feeding feedlot cattle promoted improvement in animal performance, despite the reduction of the forequarter weight, with appropriate aspects of meat quality, fatty acid profile and conservation.

In the cat echocardiography examination was observed cardiomegaly, decreased systolic function and thinning of the left ventricle wall. The electrocardiogram showed the occurrence of supraventricular tachycardia. The treatment was initiated with digoxin, furosemide and taurine supplementation, with an improvement of the clinical signs, however the animal died in a fall. The diagnosis of cardiomyopathy associated with excessive moderator band, was based on the multiple cardiac bunds which connect the septum to the free wall of the left ventricle observed in the cat necropsy.

For this purpose, blood samples from 703 dogs were collected and questionnaires addressing epidemiological aspects were completed by dog-owners. The sera were analysed for anti-N. caninum antibodies by indirect fluorescent antibody tests (IFAT 5 1:50). Association between epidemiological aspects and seropositivity in dogs was evaluated with multivariate logistic regression models. A total of 80 (11•4%) dogs tested positive for N. caninum. In the multivariate logistic regression models, dogs aged >4 years, dogs used as guard dogs, dogs that spontaneously hunt, and history of bovine abortion were found to be greater risk factors for canine N. caninum infection. When we considered only dogs from rural areas, an association with seroprevalence was seen for milk farms, dogs not fed with commercial food, dogs that hunt, and dogs used as guard dogs.

Um dos componentes principais da região medular são as células cromafins, as quais são neurônios simpáticos modificados que sintetizam epinefrina e norepinefrina distintamente (Hullinger e Andrisani, 2006).

Research paper thumbnail of Risk factors associated with seroprevalence of Neospora caninum in dogs from urban and rural areas of milk and coffee production in Minas Gerais state, Brazil
