Coleus forskohlli Briq., an endangered medicinal plant, has been successfully micropropagated und... more Coleus forskohlli Briq., an endangered medicinal plant, has been successfully micropropagated under in vitro conditions. Best adventitious shoots were developed in solid Murashige and Skoog's medium supplemented with 1.5 mg L -l 6-benzylaminopurine, while best rhizogenesis was achieved without growth regulator. The second event of success was in vitro conservation of the species by slow growth technique employing osmotic regulators (sorbitol and mannitol). 3 M mannitol showed best performance for reducing growth. This study elucidated an efficient method of ex situ conservation of germplasm and sustainable management of this medicinally important plant. Introduction Coleus forskohlii Briq. The present study describes the role of different exogenous phytohormones, polyamines and sucrose... more The present study describes the role of different exogenous phytohormones, polyamines and sucrose on growth and rosmarinic acid (RA) production in whole plant culture of Solenostemon scutellarioides. It was further aimed to conserve elite clones via synthetic seed technology. S. scutellarioides was treated either singly or in combination with different phytohormones. Cultures incubated with NAA (0.5 mg L(-1)) yielded the highest RA accumulation (g(-1FW)), but negatively affected the growth. So, overall RA content was insignificant. Cultures incubated with IBA, BAP and GA3 at low concentration significantly improved growth and RA bioaccumulation. In the combinatorial study, IBA+BAP+GA3 (0.5 mg L(-1) each) was found optimum for plant biomass and RA production (65.2% improvement of total RA). An attempt was made to develop a protocol for the preparation of synthetic seeds using in vitro r... more An attempt was made to develop a protocol for the preparation of synthetic seeds using in vitro regenerated, genetically identical bulbs of Zephyranthes grandiflora. Encapsulation was standardized with 4% sodium alginate and 1% sucrose for uniform bulb size with high conversion potential. An optimum storage temperature was found to be 4 degrees C. Synthetic seeds were germinated in MS medium supplemented with benzyladenine (2 mg dm(-3)) and rooted in the presence of indole-3-butyric acid (1 mg dm(-3)) in MS medium. Well-rooted plants were transferred to the experimental field with 80% survival after hardening. This study elucidated an efficient technique for exchange of germplasm and ex situ conservation method.

The aim of the present study was to determine the effect of three strains of Agrobacterium rhizog... more The aim of the present study was to determine the effect of three strains of Agrobacterium rhizogenes (ATCC 15834, A4 and LBA 9402) and the nature of explants (leaf and stem) on hairy root induction, growth and plumbagin production in Plumbago indica. The first appearance of hairy roots, the transformation frequency, dry root biomass and plumbagin accumulation were found to be maximum in hairy roots induced in leaf explants infected with A. rhizogenes ATCC 15834 as compared with the other two bacterial strains. The hairy roots generated from stem explants infected with all three strains were not found to be productive in terms of the selected parameters. Finally, the insertion of the rolB gene of A. rhizogenes ATCC 15834 in hairy roots of P. indica derived from leaf explants was confirmed by PCR analysis.

The present study was designed to explore the therapeutic role of aqueous extract of I. aquatica (AEIA) against As-intoxication. Methods: AEIA was chemically standardized by spectroscopic and chromatographic analysis. The cytoprotective role of AEIA was measured on isolated murine hepatocytes. The effect on redox status were measured after incubating the hepatocytes with NaAsO 2 (10 μM) + AEIA (400 μg/ml). The protective effect of AEIA (400 μg/ml) in expressions of apoptotic proteins were estimated in vitro. The protective role of AEIA was measured by in vivo assay in mice. Haematological, biochemical, As bioaccumulation and histological parameters were evaluated to ensure the protective role of AEIA (100 mg/kg) against NaAsO 2 (10 mg/kg) intoxication.

Bioautography is a mean of target-directed isolation of active molecules on chromatogram. Organic solvents employed in chromatographic separation process can be completely removed before biological detection because these solvents cause inactivation of enzymes and/or death of living organisms. It offers a rapid and easy identification of bioactive lead/scaffolds in complex matrices of plant extracts. Bioautography is a technique to isolate hit(s)/lead(s) employing a suitable chromatographic process followed by a biological detection system. This review critically describes the methodologies to identify antimicrobial, antioxidant, enzyme inhibitor lead/scaffolds employing bioautography. Significant number of examples has been incorporated to authenticate the methodologies.

(Malvaceae) leaf is traditionally used to treat diabetes in India and Southern Asia. Therefore, current study was performed to evaluate the protective effect of defatted methanol extract of A. augusta leaves (AA) against type 2 diabetes mellitus (T2DM) and its associated nephropathy and cardiomyopathy in experimental rats. Methods: Antidiabetic activity of AA extracts (100 and 200 mg/kg, p.o.) was measured in streptozotocin-nicotinamide induced type 2 diabetic (T2D) rat. Fasting blood glucose level (at specific interval) and serum biochemical markers (after sacrifice) were measured. Redox status, transcription levels of signal proteins (NF-κB and PKCs), mitochondria dependent apoptotic pathway (Bad, Bcl-2, caspase cascade) and histological studies were performed in kidneys and hearts of controls and AA treated diabetic rats.

The aim of this study was to employ precursor-feeding strategy for the improved production of ros... more The aim of this study was to employ precursor-feeding strategy for the improved production of rosmarinic acid (RA) in Solenostemon scutellarioides in vitro. The cultures were fed with precursors, namely L-phenylalanine (Phe), L-tyrosine (Tyr) and cucumber juice (CJ), at different concentrations. Phe (100 mg L 21 ) and Tyr (400 mg L 21 ) caused ,1.5and 2.1-fold increase in RA accumulation within 48 h. CJ (50 mg L 21 ) feeding displayed highest RA content (, 1.6-fold) in 72 h. In this study, we focused on the function of individual precursor on key enzymes involved in RA biosynthesis. The phenylalanine ammonia lyase activity was significantly upregulated after Phe (100 mg L 21 ) feeding, while tyrosine aminotransferase and hydroxyphenylpyruvate reductase activities were improved with Tyr (400 mg L 21 ) treatment. However, rosmarinic acid synthase activity was significantly enhanced by all three precursors. In synergy study, Phe (100 mg L 21 ) þ Tyr (400 mg L 21 ) could enhance (, 3.1-fold) RA biosynthesis within 48 h.

The matured fruits of Diospyros peregrina possess significant antidiabetic activity. The present ... more The matured fruits of Diospyros peregrina possess significant antidiabetic activity. The present investigation was undertaken to evaluate the target of action of the hydroalcoholic extract of D. peregrina fruit in diabetic pathophysiology. The extract was tested in vitro for inhibition of glucose absorption by -amylase inhibition assay, glucose uptake study by isolated rat hemidiaphragm method and free radical scavenging activity by DPPH method. The extract exhibited significant -amylase inhibition (IC 50 136.5 mg mL À1 ), peripheral glucose utilisation and radical scavenging (IC 50 167.5 mg mL À1 ) activity. From the results, it may be concluded that the hydroalcoholic extract of D. peregrina fruit can counteract diabetes by multiple mechanisms, namely inhibition of carbohydrate absorption, improving peripheral glucose utilisation and antioxidant defence.

Elicitation of Plumbago indica hairy roots with yeast carbohydrate fraction, chitosan, manganese ... more Elicitation of Plumbago indica hairy roots with yeast carbohydrate fraction, chitosan, manganese chloride, copper chloride and methyl jasmonate exhibited significant elevation (~1.2 to 2 fold) of plumbagin production in shake flask culture as compared with control. Chitosan and methyl jasmonate elicitation also caused simultaneous plumbagin leaching into culture media. Three days' exposure of chitosan (200 mg l −1 ) and methyl jasmonate (80 μM) together synergized total plumbagin yield to its maximum 11.96 ± 0.76 mg g −l DW in shake flask culture. In bioreactor cultivation, a significant raise in fresh root biomass was recorded on day 20 as compared with control shake flask culture. Three days' exposure of chitosan (200 mg l −1 ) and methyl jasmonate (80 μM) with 20 days old bioreactor-culture significantly improved total plumbagin production to 13.16 ± 1.72 mg g −l DW with simultaneous plumbagin leaching into bioreactor media.

The present study was undertaken to evaluate the protective effect of aqueous extract of Corchorus olitorius leaves against Pb-acetate induced toxic manifestation in blood, liver, kidney, brain and heart of Wistar rats. The Pb-acetate (5 mg/kg body weight) treated rats exhibited a significant inhibition of co-enzymes Q, antioxidant enzymes and reduced glutathione levels in the tissues. In addition, the extent of lipid peroxidation, DNA fragmentation and haematological parameters were significantly altered in the Pb-acetate treated rats as compared to control. Simultaneous administration of test extract (25, 50 and 100 mg/kg body weight), could significantly restore the biochemical and haematological parameters near to the normal status through antioxidant activity and/or by preventing bioaccumulation of Pb within the tissues of experimental rats. In vitro bioassay on isolated mice hepatocytes confirmed dose dependent cytoprotective effect of AECO. The CdCl 2 (30 lM) exhibited a significantly increased levels of lipid peroxidation, protein carbonylation along with the reduction of antioxidant enzymes and reduced glutathione levels in hepatocytes. AECO (200 and 400 lg/ml) + CdCl 2 (30 lM) could significantly restore the aforementioned oxidation parameters in hepatocytes. Beside this, AECO could significantly reduce Cd-induced increase in Bad/Bcl-2 ratio and the over-expression of NF-jB, caspase 3 and caspase 9. In in vivo assay, CdCl 2 (4 mg/kg body weight, for 6 days) treated rats exhibited a significantly increased intracellular Cd accumulation, oxidative stress and DNA fragmentation in the organs. In addition, the haematological parameters were significantly altered in the CdCl 2 treated rats. Simultaneous administration of AECO (50 and 100 mg/kg body weight), could significantly restore the biochemical, antioxidant and haematological parameters near to the normal status. Histological studies of the organs supported the protective role of jute leaves. Presence of substantial quantity of phenolic compounds and flavonoids in extract may be responsible for overall protective effect.

Zephyr lily (Zephyranthes grandiflora), an important ornamental plant has been micropropagated in... more Zephyr lily (Zephyranthes grandiflora), an important ornamental plant has been micropropagated in vitro after controlling microbial contamination by a pretreatment with 0.2 % Bavistin and 0.1 % Pantomycin for 4 h before final sterilization with 0.1 % mercuric chloride. In 67 % of the sterile cultures, 11 shoots on average were regenerated directly from basal half of bulb scales in Murashige and Skoog (MS) medium containing 3 % sucrose and 2 mg dm -3 benzylaminopurine (BAP). Shoots emerged in bunches on a basal achlorophyllous bulbous part. Combination of 2 mg dm -3 BAP with 1 mg dm -3 gibberellic acid (GA 3 ) enhanced shoot growth. Stout roots (maximum of 5 -6 per shoot) were developed in presence of 1 mg dm -3 indole-3-butyric acid (IBA). Micro-bulbs showed potential of regeneration and could be used as secondary explants. The morphologically identical plants derived by in vitro propagation were genetically identical as shown by PCR based ISSR marker analysis of genomic DNA.

Beside this, AECO could significantly reduce Cd-induced increase in Bad/Bcl-2 ratio and the over-expression of NF-jB, caspase 3 and caspase 9. In in vivo assay, CdCl 2 (4 mg/kg body weight, for 6 days) treated rats exhibited a significantly increased intracellular Cd accumulation, oxidative stress and DNA fragmentation in the organs. In addition, the haematological parameters were significantly altered in the CdCl 2 treated rats. Simultaneous administration of AECO (50 and 100 mg/kg body weight), could significantly restore the biochemical, antioxidant and haematological parameters near to the normal status. Histological studies of the organs supported the protective role of jute leaves. Presence of substantial quantity of phenolic compounds and flavonoids in extract may be responsible for overall protective effect.

Zephyr lily (Zephyranthes grandiflora), an important ornamental plant has been micropropagated in... more Zephyr lily (Zephyranthes grandiflora), an important ornamental plant has been micropropagated in vitro after controlling microbial contamination by a pretreatment with 0.2 % Bavistin and 0.1 % Pantomycin for 4 h before final sterilization with 0.1 % mercuric chloride. In 67 % of the sterile cultures, 11 shoots on average were regenerated directly from basal half of bulb scales in Murashige and Skoog (MS) medium containing 3 % sucrose and 2 mg dm -3 benzylaminopurine (BAP). Shoots emerged in bunches on a basal achlorophyllous bulbous part. Combination of 2 mg dm -3 BAP with 1 mg dm -3 gibberellic acid (GA 3 ) enhanced shoot growth. Stout roots (maximum of 5 -6 per shoot) were developed in presence of 1 mg dm -3 indole-3-butyric acid (IBA). Micro-bulbs showed potential of regeneration and could be used as secondary explants. The morphologically identical plants derived by in vitro propagation were genetically identical as shown by PCR based ISSR marker analysis of genomic DNA.
