For the characterization of posttranslational modifications of the sarcoplasmic/endoplasmic retic... more For the characterization of posttranslational modifications of the sarcoplasmic/endoplasmic reticulum Ca-ATPase (SERCA), we developed a two-dimensional separation protocol based on reversed-phase HPLC followed by SDS-PAGE and LC-MS/MS analysis of in-gel tryptic digests. Representative experiments are shown for the rabbit fast-twitch skeletal muscle isoform SERCA1. Matrixassisted laser desorption-ionization and electrospray ionization-mass spectrometry analyses of SERCA1 tryptic digests revealed ca. 66% coverage of the protein sequence. This approach was used for the detection and quantitation of nitrotyrosine formation after exposure of SERCA1 to peroxynitrite in vitro. At molar ratios of nitrotyrosine to protein of 0.23 we confirmed by LC-MS/MS the nitration of predominantly Tyr 122 in the SERCA1 sequence. Ó

Several nuclear receptor (NR) superfamily members are known to be the molecular target of either ... more Several nuclear receptor (NR) superfamily members are known to be the molecular target of either the SUMO- or ubiquitin-signaling pathways. However, little is currently known regarding how these two post-translational modifications interact to control NR biology. We show that the SUMO and ubiquitin circuitry coordinately modifies pregnane x receptor (PXR, NR1I2) to play a key role in regulating PXR protein stability, transactivation capacity, and transcriptional repression. The SUMOylation and ubiquitylation of PXR is increased in a ligand- and tumor necrosis factor-alpha (TNFα)-dependent manner in hepatocytes. The SUMO-E3 ligase enzymes PIAS1 and PIASy drive high levels of PXR SUMOylation. Expression of PIAS1 selectively increases SUMO(3)ylation, as well as PXR-mediated induction of CYP3A and the xenobiotic response. Previous studies of chemoreceptive behavior in vipers suggest that snakes focus on the scent of e... more Previous studies of chemoreceptive behavior in vipers suggest that snakes focus on the scent of envenomated tissue to track their prey following envenomation. Other studies have indicated a correlation between qualitative differences in venom biochemistry and geographic variation in diet. The North American copperhead (Agkistrodon contortrix) varies geographically in diet and venom biochemistry; snakes were collected from three populations (Kansas, Texas, and Louisiana) that are known to have different prey preferences. Behavioral experiments were conducted to assess whether copperheads preferred envenomated prey more than nonenvenomated prey, as do other species of vipers studied thus far. Additional experiments tested the ability of copperheads to distinguish between envenomated prey from different geographic populations, and between geographic populations of copperheads and two other species of viper. Potential roles of host cell components in phage assembly haven't been well understood. It was previously reported that two unidentified proteins were present in bacteriophage Sf6 virion (Casjens et al, 2004, J.Mol.Biol. 339, 379-394, Fig. 2A). Using tandem mass spectrometry, we have identified the two proteins as outer membrane proteins (OMPs) OmpA and OmpC from its host Shigella flexneri. The transmission electron cryo-microscopy structure of Sf6 shows significant density at specific sites at the phage capsid inner surface. This density fit well with the characteristic beta-barrel domains of OMPs, thus may be due to the two host proteins. Locations of this density suggest a role in Sf6 morphogenesis reminiscent of phage-encoded cementing proteins. These data indicate a new, OMP-related phage:host linkage, adding to previous knowledge that some lambdoid bacteriophage genomes contain OmpC-like genes that express phage-encoded porins in the lysogenic state.

Direct infusion electrospray ionization triple quadrupole precursor scanning for three oxidized f... more Direct infusion electrospray ionization triple quadrupole precursor scanning for three oxidized fatty acyl anions revealed 86 mass spectral peaks representing polar membrane lipids in extracts from Arabidopsis (Arabidopsis thaliana) infected with Pseudomonas syringae pv tomato DC3000 expressing AvrRpt2 (PstAvr). Quadrupole time-of-flight and Fourier transform ion cyclotron resonance mass spectrometry provided evidence for the presence of membrane lipids containing one or more oxidized acyl chains. The membrane lipids included molecular species of phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol, digalactosyldiacylglycerol, monogalactosyldiacylglycerol, and acylated monogalactosyldiacylglycerol. The oxidized chains were identified at the level of chemical formula and included C 18 H 27 O 3 (abbreviated 18:4-O, to indicate four double bond equivalents and one oxygen beyond the carbonyl group), C 18 H 29 O 3 (18:3-O), C 18 H 31 O 3 (18:2-O), C 18 H 29 O 4 (18:3-2O), C 18 H 31 O 4 (18:2-2O), and C 16 H 23 O 3 (16:4-O)

Brain aging is associated with a progressive decline in cognitive function though the molecular m... more Brain aging is associated with a progressive decline in cognitive function though the molecular mechanisms remain unknown. Functional changes in brain neurons could be due to age-related alterations in levels of specific proteins critical for information processing. Specialized membrane microdomains known as 'lipid rafts' contain protein complexes involved in many signal transduction processes. This study was undertaken to determine if two-dimensional fluorescence difference gel electrophoresis (2D DIGE) analysis of proteins in synaptic membrane lipid rafts revealed agedependent alterations in levels of raft proteins. Five pairs of young and aged rat synaptic membrane rafts were subjected to DIGE separation, followed by image analysis and identification of significantly altered proteins. Of 1046 matched spots on DIGE gels, 94 showed statistically significant differences in levels between old and young rafts, and 87 of these were decreased in aged rafts. The 41 most significantly altered (p < 0.03) proteins included several synaptic proteins involved in energy metabolism, redox homeostasis, and cytoskeletal structure. This may indicate a disruption in bioenergetic balance and redox homeostasis in synaptic rafts with brain aging. Differential levels of representative identified proteins were confirmed by immunoblot analysis. Our findings provide novel pathways in investigations of mechanisms that may contribute to altered neuronal function in aging brain.
