This article presents clinical recommendations for using ziprasidone based on information from a ... more This article presents clinical recommendations for using ziprasidone based on information from a year of post-marketing experience. The recommendations are based on the clinical literature, the package insert, presentations at recent meetings, data on file with the manufacturer, and the consensus of a panel of expert psychiatrists. The article provides updates on efficacy and safety data and gives recommendations for dosing and switching strategies. With regard to the QTc issue, there has not been any case of torsades de pointes reported in the more than 150,000 patients who have received ziprasidone since its approval. Ziprasidone is weight neutral and does not appear to cause increases in glucose or lipid levels or in insulin resistance. The panel generally recommends beginning with an initial dose of 80 mg/day (40 mg b.i.d.) rather than the 40 mg/day dose recommended in the package insert. The ability to begin with a therapeutic dose and to titrate up rapidly is an advantage of z...

- The effects of antipsychotic agents in blocking dopamine (DA) receptors and the similarity bet... more 1) The effects of antipsychotic agents in blocking dopamine (DA) receptors and the similarity between paranoid schizophrenia and amphetamine-induced psychosis have opened new research areas in schizophrenia. (2) This paper reviews studies on the hypothesized role for DA in schizophrenia, specifically defects in the DA system including receptor supersensitivity and other dysfunctions. While clinical studies seem to support the DA hypothesis, evidence from the evaluation of spinal fluid homovanillic acid (HVA), dopamine~13-hydroxylasc (DBH) and plasma prolactin has been inconclusive. (4) Schizophrenic symptoms fluctuate with central DA activity and some positive brain autopsies appear to support the notion of dysfunctional DA systems. (5) Increasing evidence suggests that other neurotransmitters or modulators of DA activity, such as amino acids (e.g. GABA), amines (norepinephrine) or peptides (endorphins), may be involved in the regulation of psychosis. The author suggests that more specific presynaptic DA agonists and agents that specifically affect mesolimbic DA systems should be developed to test the DA hypothesis further. The mechanism of hgw DA activity affects schizophrenia has not yet been elucidated.

It is widely known that hepatitis and its complications such as cirrhosis or hepatocellular carci... more It is widely known that hepatitis and its complications such as cirrhosis or hepatocellular carcinoma are one of the major health problems of the world especially since no specific treatment is available. In the present study we investigated the hepatoprotective potential of the methanolic extract of the whole plant of Dodonaea viscosa and its ethyl acetate, aqueous, butanol and n-hexane fractions against carbon tetrachloride (CCl 4 ) induced hepatoxicity in rats. Hepatoprotection was assessed in terms of reduction in serum enzymes (ALT, AST, and ALP) that occur after CCl 4 injury, and by histopathology and immunohistochemistry. The methanolic extract reduced the serum enzyme level (ALT, AST, and ALP) down to control levels despite CCl 4 treatment. It also reduced the CCl 4 -induced damaged area to 0% as assessed by histopathology. The CD68+ macrophages were also reduced in number around the central vein area by the methanolic extract. These hepatoprotective effects were better than the positive control silymarin. Similar hepatoprotective activities were found with the ethyl acetate, and aqueous fractions of the methanolic extract. The butanol and n-hexane fractions showed elevated levels of ALT, AST and ALP as compared to the positive control silymarin. Histopathology showed ∼30% damage to the liver cells with the butanol and n-hexane fractions which still showed some protective activity compared to the CCl 4 treated control. HPLC fingerprinting suggested that hautriwaic acid present in the methanolic extract and its ethyl acetate, and aqueous fractions may be responsible for this hepatoprotective activity of Dodonaea viscosa which was confirmed by in vivo experiments.

Two salinity levels of 0 and 10 dS m−1 were created with sodium chloride (NaCl) and foliar spray of Zn (0.05%; Chelated-Zn) was applied. A decrease in growth and yield related parameters were observed under salt stress, which was ameliorated in plants that received Zn foliar spray. Similar effects of salinity and Zn foliar spray were noted on photosynthetic rate, transpiration rate, stomatal conductance, water use efficiency, and water relations of plants. Salt induced increase in sodium (Na) content and decrease in other macro- and micronutrients contents were also reversed by Zn. Other salt tolerance indicating parameters likSe total free amino acids and total soluble sugars increased under Zn spray, clearing its role in improving salt tolerance.

A fast, simple, and a reliable high-performance liquid chromatography linked with electrochemical... more A fast, simple, and a reliable high-performance liquid chromatography linked with electrochemical detector (HPLC-ECD) method for the assessment of lipoic acid (LA) and dihydrolipoic acid (DHLA) in plasma was developed using naproxen sodium as an internal standard (IS) and validated according to standard guidelines. Extraction of both analytes and IS from plasma (250 μl) was carried out with a single step liquid-liquid extraction applying dichloromethane. The separated organic layer was dried under stream of nitrogen at 40°C and the residue was reconstituted with the mobile phase. Complete separation of both compounds and IS at 30°C on Discovery HS C18 RP column (250 mm × 4.6 mm, 5 μm) was achieved in 9 min using acetonitrile: 0.05 M phosphate buffer (pH 2.4 adjusted with phosphoric acid) (52:48, v/v) as a mobile phase pumped at flow rate of 1.5 ml min(-1) using electrochemical detector in DC mode at the detector potential of 1.0 V. The limit of detection and limit of quantification for lipoic acid were 500 pg/ml and 3 ng/ml, and for dihydrolipoic acid were 3 ng/ml and 10 ng/ml, respectively. The absolute recoveries of lipoic acid and dihydrolipoic acid determined on three nominal concentrations were in the range of 93.40-97.06, and 93.00-97.10, respectively. Similarly coefficient of variations (% CV) for both intra-day and inter-day were between 0.829 and 3.097% for lipoic acid and between 1.620 and 5.681% for dihydrolipoic acid, respectively. This validated method was applied for the analysis of lipoic acid/dihydrolipoic acid in the plasma of human volunteers and will be used for the quantification of these compounds in patients with oxidative stress induced pathologies.
