Une première partie revient sur la démarche suivie et pointe l’enjeu de l’évaluation de la qualité de données hétérogènes issues d’une méthode mixte et longitudinale de collecte (suivis GPS, enquêtes, observations). Une deuxième partie établit un diagnostic qualité selon l’origine (GPS, algorithme, enquête) et la nature des erreurs (complétude, précision, cohérence). Ces typologies permettent, dans une troisième partie, de définir d’une chaîne de traitements reproductible visant à améliorer la qualité interne et externe des données.

Through collaboration with Inuit hunters, we examined the stomach contents of 142 seals (ringed s... more Through collaboration with Inuit hunters, we examined the stomach contents of 142 seals (ringed seals [Phoca hispida; n = 135], bearded seals [Erignathus barbatus; n = 6], and one harbour seal [Phoca vitualina; n = 1]) hunted between 2007 and 2019 from communities around Nunavut to assess whether seals in the eastern Canadian Arctic ingest and retain plastics in their stomachs. The seals in this study ranged from juveniles to adults of up to 30 years of age, and 55% of the seals were males. We found no evidence of plastic ingestion in any of the seals suggesting that seals in Nunavut are not accumulating plastics (> 425 μm) in their stomachs. These data provide important baseline information for future plastic pollution monitoring programs in the Arctic.

Thoroughbred horses (TB) have no ventilatory response to added CO2 during near-maximal exercise. ... more Thoroughbred horses (TB) have no ventilatory response to added CO2 during near-maximal exercise. To see whether that reflects mechanical limits to ventilation or the control of breathing, we examined the effects of varying inspired O2 fraction (0.16, 0.21, or 0.30) in five normal TB standing quietly and galloping at 10 and 14 m/s on a level treadmill. We measured gas exchange (O2 consumption and CO2 production) and ventilation with a flow-through mask system. We also measured PO2, PCO2, and O2 contents in arterial and mixed venous blood and calculated cardiac output by using the Fick equation. Low inspired O2 fraction (0.16 vs. 0.21) had significant effects in TB galloping at 14 m/s. Arterial PO2 then was 38 Torr compared with 56 Torr for horses on air. Permafrost in peatlands strongly influences ecosystem characteristics, including vegetation compo... more Permafrost in peatlands strongly influences ecosystem characteristics, including vegetation composition, hydrological functions, and carbon cycling. Large amounts of organic carbon are stored in permafrost peatlands in northwestern Canada. Their possible degradation into permafrost-free wetlands including thermokarst bogs may affect carbon (C) stocks, but the direction and magnitude of change are uncertain. Using peat core reconstructions, we characterized the temporal and spatial variability in vegetation macrofossil, testate amoebae, C content, and peat decomposition along a permafrost thaw chronosequence in the southern portion of the Scotty Creek watershed near Fort Simpson, Northwest Territories. The accumulation of limnic and minerotrophic peat prevailed at the site until permafrost formed around 5000 cal. yr BP. La proteomique ouvre de nouvelles perspectives dans la caracterisation des facteurs de virulence ... more La proteomique ouvre de nouvelles perspectives dans la caracterisation des facteurs de virulence des bacteries associees au bio-terrorisme. L’electrophorese en 2 dimensions (2-DE) est une des techniques les plus couramment utilisee permettant une separation tres resolutive des proteines. Cependant au cours de mes travaux de these nous avons developpe au laboratoire une nouvelle technique plus performante, la 2D-DIGE (Two-Dimensional Differential Gel Electrophoresis) afin d’etudier plus precisement les proteines synthetisees par ces agents du bio-terrorisme. En effet, cette technique permet de comparer plusieurs conditions dans un meme gel avec une meilleure sensibilite, reproductibilite et efficacite que la 2-DE. L’utilisation de cette technique pour la comparaison de souches virulentes et avirulentes chez Francisella tularensis et Rickettsia prowazekii est une premiere, et a permis d’identifier de nouvelles proteines presentant des niveaux d’expressions differents ainsi que confirmer des resultats obtenus avec d’autres approches (genomique, transcriptomique). D’autre part l’etude de l’immunogenicite de F. Tularensis, avec l’utilisation de la 2-DE associee aux Western blots, a permis d’identifier de nouvelles proteines immunoreactives et de comprendre l’origine de la reaction croisee avec Mimivirus. De plus, l’etude proteique presente un avantage considerable par rapport aux approches geniques ou transcriptionnelles, qui est de pouvoir apprecier les modifications post-traductionnelles. Nous avons donc decide d’etudier les methylations lors de l’analyse du proteome de R. Prowazekii. L’approche proteomique de ces bacteries potentielles du bio-terrorisme est donc essentielle pour une meilleure comprehension des mecanismes responsables de la virulence et de la maladie. Ces nouvelles pistes ouvrent des perspectives interessantes dans le developpement de nouveaux outils diagnostics et therapeutiques. Des etudes plus precises sur les modifications posttraductionnelles mais aussi sur des fractions enrichies en proteines membranaires pourraient permettre de mieux comprendre ces bacteries et leurs interactions avec l’hote

Horses have high mean pulmonary artery pressure (Ppa) both at rest and during exercise (~ 30 and ... more Horses have high mean pulmonary artery pressure (Ppa) both at rest and during exercise (~ 30 and > 80 mmHg, respectively). The mechanisms are unknown. To see if hypoxic pulmonary vasoconstriction (HPV) plays a role, we compared pulmonary artery pressure-flow (Ppa-Q) curves when inspired 02 fraction (FIo2) was 0.16, 0.21, and 0.30, in 5 normal Thoroughbred horses standing quietly and while galloping at 10 and 14 m/sec on a level treadmill. We calculated 02 consumption (Xto2) from measurements of respired gas composition and flow, and calculated 0 from ~Zo: and measurements of oxygen content in arterial and mixed venous blood (Cao2 and Crlo2). Vo2 was 3.8, 74 and 128 ml-min-l.kg-1, at rest and at 10 and 14 m/sec, and did not vary with FIo2 at any speed. At 14 m/sec only, when Fio2 was lowered to 0.16, Cao2 fell (to 14.7 from 20 ml/dl on air), Q increased (to 0.86 from 0.66 L'min-l.kg-I on air), and stroke volume increased (to 4.1 from 3.2 ml.kg-1 on air). Slopes and intercepts of Ppa-Q curves did not vary with FIo2. We conclude that HPV does not contribute to the high Ppa of exercising horses breathing air near sea level.

The determination of the arterial blood pressure was done on 12 healthy mixed breed dogs in both ... more The determination of the arterial blood pressure was done on 12 healthy mixed breed dogs in both the anesthetized and the conscious state, to evaluate two instruments (Doppler flow detector and infrasonde D4000), in their ability to indirectly determine arterial blood pressure. The coefficients of variation were higher with indirect methods when compared with the results obtained by cannulation. These coefficients were lower with the Doppler flow detector. The correlation study showed that both apparatuses were reliable in most situations. The infrasonde D4000 was more accurate than the Doppler in the conscious animals. However the results showed a lack of precision in hypertensive conscious dogs. The diastolic arterial blood pressure was particularly precise in the case of the anesthetized hypotensive dogs. Several strategies are proposed for its improvement. First, light collection in conventional microarrays scanners is quite limited. It was recently shown that almost full collection can be achieved in an integrated lens-free biosensor, with labelled species hybridizing practically on the surface of a sensitive silicon detector [L. Martinelli et al. Appl. Phys. Lett. 91, 083901 (2007)]. However, even with such an improvement, the ultimate goal of real-time measurements during hybridization is challenging: the detector is dazzled by the large fluorescence of labelled species in the solution. In the present paper we show that this unwanted signal can effectively be reduced if the excitation light is confined in a waveguide. Moreover, the concentration of excitation light in a waveguide results in a huge signal gain. In our experiment we realized a structure consisting of a high index sol-gel waveguide deposited on a low-index substrate. The fluorescent molecules deposited on the surface of the waveguide were excited by the evanescent part of a wave travelling in the guide. The comparison with free-space excitation schemes confirms a huge gain (by several orders of magnitude) in favour of waveguide-based excitation. An optical guide deposited onto an integrated biosensor thus combines both advantages of ideal light collection and enhanced surface localized excitation without compromising the imaging properties. Modelling predicts a negligible penalty from spatial cross-talk in practical applications. We believe that such a system would bring microarrays to hitherto unattained sensitivities.

Large amounts of organic carbon are stored in permafrost peatlands in northwestern Canada. Their possible degradation into permafrost-free wetlands including thermokarst bogs may affect carbon (C) stocks, but the direction and magnitude of change are uncertain. Using peat core reconstructions, we characterized the temporal and spatial variability in vegetation macrofossil, testate amoebae, C content, and peat decomposition along a permafrost thaw chronosequence in the southern portion of the Scotty Creek watershed near Fort Simpson, Northwest Territories. The accumulation of limnic and minerotrophic peat prevailed at the site until permafrost formed around 5000 cal. yr BP. La proteomique ouvre de nouvelles perspectives dans la caracterisation des facteurs de virulence ... more La proteomique ouvre de nouvelles perspectives dans la caracterisation des facteurs de virulence des bacteries associees au bio-terrorisme. L’electrophorese en 2 dimensions (2-DE) est une des techniques les plus couramment utilisee permettant une separation tres resolutive des proteines. Cependant au cours de mes travaux de these nous avons developpe au laboratoire une nouvelle technique plus performante, la 2D-DIGE (Two-Dimensional Differential Gel Electrophoresis) afin d’etudier plus precisement les proteines synthetisees par ces agents du bio-terrorisme. En effet, cette technique permet de comparer plusieurs conditions dans un meme gel avec une meilleure sensibilite, reproductibilite et efficacite que la 2-DE. L’utilisation de cette technique pour la comparaison de souches virulentes et avirulentes chez Francisella tularensis et Rickettsia prowazekii est une premiere, et a permis d’identifier de nouvelles proteines presentant des niveaux d’expressions differents ainsi que confirmer des resultats obtenus avec d’autres approches (genomique, transcriptomique). D’autre part l’etude de l’immunogenicite de F. Tularensis, avec l’utilisation de la 2-DE associee aux Western blots, a permis d’identifier de nouvelles proteines immunoreactives et de comprendre l’origine de la reaction croisee avec Mimivirus. De plus, l’etude proteique presente un avantage considerable par rapport aux approches geniques ou transcriptionnelles, qui est de pouvoir apprecier les modifications post-traductionnelles. Nous avons donc decide d’etudier les methylations lors de l’analyse du proteome de R. Prowazekii. L’approche proteomique de ces bacteries potentielles du bio-terrorisme est donc essentielle pour une meilleure comprehension des mecanismes responsables de la virulence et de la maladie. Ces nouvelles pistes ouvrent des perspectives interessantes dans le developpement de nouveaux outils diagnostics et therapeutiques. Des etudes plus precises sur les modifications posttraductionnelles mais aussi sur des fractions enrichies en proteines membranaires pourraient permettre de mieux comprendre ces bacteries et leurs interactions avec l’hote

The mechanisms are unknown. To see if hypoxic pulmonary vasoconstriction (HPV) plays a role, we compared pulmonary artery pressure-flow (Ppa-Q) curves when inspired 02 fraction (FIo2) was 0.16, 0.21, and 0.30, in 5 normal Thoroughbred horses standing quietly and while galloping at 10 and 14 m/sec on a level treadmill. We calculated 02 consumption (Xto2) from measurements of respired gas composition and flow, and calculated 0 from ~Zo: and measurements of oxygen content in arterial and mixed venous blood (Cao2 and Crlo2). Vo2 was 3.8, 74 and 128 ml-min-l.kg-1, at rest and at 10 and 14 m/sec, and did not vary with FIo2 at any speed. At 14 m/sec only, when Fio2 was lowered to 0.16, Cao2 fell (to 14.7 from 20 ml/dl on air), Q increased (to 0.86 from 0.66 L'min-l.kg-I on air), and stroke volume increased (to 4.1 from 3.2 ml.kg-1 on air). Slopes and intercepts of Ppa-Q curves did not vary with FIo2. We conclude that HPV does not contribute to the high Ppa of exercising horses breathing air near sea level.

The determination of the arterial blood pressure was done on 12 healthy mixed breed dogs in both ... more The determination of the arterial blood pressure was done on 12 healthy mixed breed dogs in both the anesthetized and the conscious state, to evaluate two instruments (Doppler flow detector and infrasonde D4000), in their ability to indirectly determine arterial blood pressure. The coefficients of variation were higher with indirect methods when compared with the results obtained by cannulation. These coefficients were lower with the Doppler flow detector. The correlation study showed that both apparatuses were reliable in most situations. The infrasonde D4000 was more accurate than the Doppler in the conscious animals. However the results showed a lack of precision in hypertensive conscious dogs. The diastolic arterial blood pressure was particularly precise in the case of the anesthetized hypotensive dogs. Several strategies are proposed for its improvement. First, light collection in conventional microarrays scanners is quite limited. It was recently shown that almost full collection can be achieved in an integrated lens-free biosensor, with labelled species hybridizing practically on the surface of a sensitive silicon detector [L. Martinelli et al. Appl. Phys. Lett. 91, 083901 (2007)]. However, even with such an improvement, the ultimate goal of real-time measurements during hybridization is challenging: the detector is dazzled by the large fluorescence of labelled species in the solution. In the present paper we show that this unwanted signal can effectively be reduced if the excitation light is confined in a waveguide. Moreover, the concentration of excitation light in a waveguide results in a huge signal gain. In our experiment we realized a structure consisting of a high index sol-gel waveguide deposited on a low-index substrate. The fluorescent molecules deposited on the surface of the waveguide were excited by the evanescent part of a wave travelling in the guide. The comparison with free-space excitation schemes confirms a huge gain (by several orders of magnitude) in favour of waveguide-based excitation. An optical guide deposited onto an integrated biosensor thus combines both advantages of ideal light collection and enhanced surface localized excitation without compromising the imaging properties. Modelling predicts a negligible penalty from spatial cross-talk in practical applications. We believe that such a system would bring microarrays to hitherto unattained sensitivities.
