The endoplasmic reticulum (ER) is a multi-layered organelle that is essential for the synthesis, ... more The endoplasmic reticulum (ER) is a multi-layered organelle that is essential for the synthesis, folding, and structural maturation of almost one-third of the cellular proteome. It houses several resident proteins for these functions including the 21 members of the protein disulfide isomerase (PDI) family. The signature of proteins belonging to this family is the presence of the thioredoxin domain which mediates the formation, and rearrangement of disulfide bonds of substrate proteins in the ER. This process is crucial not only for the proper folding of ER substrates but also for maintaining a balanced ER proteostasis. The inclusion of new PDI members with a wide variety of structural determinants, size and enzymatic activity has brought additional epitomes of how PDI functions. Notably, some of them do not carry the thioredoxin domain and others have roles outside the ER. This also reflects that PDIs may have specialized functions and their functions are not limited within the ER. ...

Management of PGK patients undergoing hemodialysis one of which is diet, adherence in the diet is... more Management of PGK patients undergoing hemodialysis one of which is diet, adherence in the diet is influenced by knowledge and family support. The purpose of this study was to determine the relationship of knowledge and family support with adherence to implement the diet in patients with Chronic Kidney Disease (PGK) who underwent hemodialysis in RSUP DR. M. Djamil Padang in 2017. This type of research is an analytic survey with cross sectional design. The sampling technique was accurate sampling with 41 samples. The data collection used primary data using knowledge and family support questionnaires, while for dietary compliance using secondary data looked at references from nurse or medical records. Univariate analysis is shown in frequency distribution and bivariate table by using chi-square test with 95% confidence α = 0.05. The results showed that there was a correlation between knowledge and adherence in the implementation of diet in patients with Chronic Kidney Disease (PGK) who underwent hemodialysis at RSUP.DR.M.Djamil Padang in 2017 (P value = 0.048) and there was a relationship between family support with Obedience in carrying out the diet in patients with Chronic Kidney Disease (PGK) undergoing hemodialysis at RSUP.DR.M.Djamil Padang in 2017 (P value = 0.042). It is suggested to the head of the hemodialysis room to educate the patient about the importance of carrying out the diet regularly and provide direction to the families who accompany the patient in order to pay more attention to the patient's compliance in following the recommended diet.

Protein-protein interactions are fundamental to various aspects of cell biology with many protein... more Protein-protein interactions are fundamental to various aspects of cell biology with many protein complexes participating in numerous fundamental biological processes such as transcription, translation and cell cycle. MS-based proteomics techniques are routinely applied for characterising the interactome, such as affinity purification coupled to mass spectrometry that has been used to selectively enrich and identify interacting partners of a bait protein. In recent years, many orthogonal MS-based techniques and approaches have surfaced including proximity-dependent labelling of neighbouring proteins, chemical cross-linking of two interacting proteins, as well as inferring PPIs from the co-behaviour of proteins such as the co-fractionating profiles and the thermal solubility profiles of proteins. This review discusses the underlying principles, advantages, limitations and experimental considerations of these emerging techniques. In addition, a brief account on how MS-based techniques are used to investigate the structural and functional properties of protein complexes, including their topology, stoichiometry, copy number and dynamics, are discussed. Penulis mengambil studi kasus implementasi kebi- jakan Program Nasional Pemberdayaan Masyarakat (PNPM) Mandiri Pariwisata di Desa Wisata Brayut Kabupaten Sleman. PNPM Mandiri Pariwisata diimplementasikan di desa ini pada tahun 2009, 2010, dan 2011. Untuk mengukur kinerja im- plementasi PNPM Mandiri Pariwisata di Desa Wisata Brayut, penulis menggunakan dua indikator. Pertama, indikator policy output yang terdiri dari indikator cakupan, bias, akses, dan kesesuaian program dengan kebutuhan. Kedua, indikator policy outcomes yang terdiri dari initial outcome, intermediate outcome, dan long-term outcome. Berdasarkan pengukuran, didapat hasil bahwa kinerja implementasi PNPM Mandiri Pariwisata di Desa Wisata Brayut terbilang rendah. Anterior gradient-2 (AGR2) protein mediates the formation, breakage and isomerization of disulphi... more Anterior gradient-2 (AGR2) protein mediates the formation, breakage and isomerization of disulphide bonds during protein maturation in the endoplasmic reticulum (ER) and contributes to the homoeostasis of the secretory pathway. AGR2 promotes tumour development and metastasis and its elevated expression is almost completely restricted to malignant tumours. Interestingly, this supposedly ER-resident protein can be localised to other compartments of cancer cells and can also be secreted into the extracellular milieu. There are emerging evidences that describe the gain-of-function activities of the extracellular AGR2, particularly in cancer development. Here, we reviewed studies detailing the expression, pathological and physiological roles associated with AGR2 and compared the duality of localization, intracellular and extracellular, with special emphasis on the later. We also discussed the possible mechanisms of AGR2 secretion as well as deliberating the functional impacts of AGR2 in cancer settings. Last, we deliberate the current therapeutic strategies and posit the potential use AGR2, as a prognosis and diagnosis marker in cancer.

However, it is easy to degrade by the environment. In this study, the degradation of Gold Tri.E™ was studied and determined. Gold Tri.E™ is a mixture of Vitamin E homologs (tocotrienol and tocopherol) extracted from palm oil (Elais Guineensis). A nanocarrier system has been optimized to encapsulate Gold Tri.E™ from degrading and increasing its stability as a bioactive compound. An oil-in-water (o/w) microemulsion was formulated and optimized as the best carrier to encapsulate Gold Tri.E™ with the mean particle size of 32.60±3.60 nm and 99.99±0.01% encapsulation efficiency (EE). Degradation of the Gold Tri.E™ in o/w microemulsion was significantly reduced as compared to the bare Gold Tri.ETM. This suggested that the system could protect Gold Tri.E™ from thermal and hydrolysis degradation.
