The effect of tryptophan on catharanthine production was studied in aggregate cells of Catharanthus roseus (L.) G Don. Aggregate cells of C. roseus were cultured in Murashige and Skoog (MS) medium supplemented with 2 ppm NAA and 0.ppm kinetin. Aggregate cells culture were devided into control and treatment groups and were fed by 100, 125, 150, 175, 200, 225 mg/L tryptophan, respectively. The growth of cells and the catharanthine production in the aggregate cells as well as in medium were observed on the 1st and 10th days. Qualitative and quantitative analysis were conducted using High Performance Liquid Chromatography (HPLC) in VP-ODS C-18 column, eluted using methanol : acetonitrile : diamonium hydrogen phosphate (3 : 4 : 3), rate 1 ml/minute. The results showed that catharanthine was detected in the aggregate cells and culture medium. The highest concentration of catharanthine in the aggregate cell and the medium were obtained on the addition of 175 mg/L tryptophan, i.e. 1650.48 ug/g and 10329.75 ug/L respectively. Tryptophan feeding enhanced catharanthine content in aggregate cell by 171.02%, as well as its secretion into medium by 329.34%.

Salah satu cara untuk meningkatkan kandungan metabolit sekunder dalam kultur jaringan tumbuhan ad... more Salah satu cara untuk meningkatkan kandungan metabolit sekunder dalam kultur jaringan tumbuhan adalah dengan penambahan prazat (prekursor). Oleh karena itu telah dilakukan penelitian hubungan antara kandungan IAA dengan pertumbuhan dan kandungan kultur agreget sel tapak darah yang diberi perlakuan triptofan dalam erlenmayer. Penelitian ini bertujuan untuk meningkatkan kandungan katarantin dalam kultur agregat sel Chatharantus roseus (l) eksperimental di laboratorium. Konsentrasi prekursor triptofan yang digunakan ialah 0-250 mg/L. Pertumbuhan ditentukan dengan penimbangan berat basah dan berat kering serta pengamatan perubahan secara morfologis. Analisis kandungan katarantin dilakukan dilakukan dengan Kromatografi Cair Kenerja Tinggi (KCKT) menggunakan kolom VP-ODS c-18. Hasil penelitian menunjukkan bahwa kandungan IAA meningkat dengan bertambahnya kosentrasi triptofan sampai 150mg/L. Kansungan tertinggi sebesar (214, 79+0.90 ug/g.bk) diamati pada perlakuan 150mg/L triptofan dengan peningkatan 50,60%. Peningkatan IAA berhubungan erat dengan pertumbuhan agregrat sel dengan r= 0,931. Hubungan IAA dan Katarantin yang diberi perlakuan triptofan juga positif dengan r= 0,83. Pertumbuhan yang optimum pada kandungan katarantin dan IAA yang lebih tinggi dari kontrol merupakan dasar untuk kultur dalam skala besar yang dapat berkelanjutan dalam bioreaktor

This study aims to determine the types of plants used for Bakera in Sea Mitra Village, Pineleng District. This study used a survey method with an interview data collection technique. The sampling used was 50 people. Furthermore, randomly interviewed mothers at Posyandu, and people who were willing to be interviewed were partly determined by purposive sampling who had knowledge of traditional medicine. Variables observed in the study are the characteristics of respondents, knowledge about Bakera, ways of use, and uses of Bakera, types, and parts of plants used, and perceived characteristics. Penelitian ini bertujuan untuk mencri komposisi media yang optimum untuk pembentukan kalus dan menganalisis alkaloid yang terdapat pada kalus Chataranthus roseus yang terbentuk Manfaat penelitian adalah untuk memproduksi atkaloid sebagai obat kanker. Kalus diinduksi pada media Murashige and Skoog (MS) dengan perlakuan sebagai berikut: D[a (tanpa 2,4-D & Kinetin), DKI (1 ppm 2,4-D and 0,1 ppm kinetin), DK2 (1,5 ppm2,4-D and 0,15 ppm kinetin), DK3 Qppm2,4- D and 0,2 ppm kinstin), DK4 (2 ppm 2,4-D). Analisis pertumbuhm kalus dilakukan melalui pe,nimbangan berat basah kalus. Kalus dipanen setelah melewati fase linier pertumbuhar kalus. Kalus diekstraksi dalam metanol yang mengandung 0,25 o/o HCI 1 N. selarna 48 jarn. Analisis kualitatif dilakrftan dengan teknik kromatografi lapis tipis, yang me,nggunakan UV pada panjang gelombang 254 nm untuk menampakkan noda dan untuk penampakan alkaloid disemprot dengan Dragendorf ke,mudian ditentukan RF-nya. Analisis kuantitatif dilakukan dengan melewatkan lromatogram pada spektrofotodensitometer pada panjang gelombang 300 nm. Hasil penelitian menunjulrkan bahwa kalus terbentuk pada semua media (perlakuan) yang diberi zat pergatur tumbuh (zpt). Kalus terbentuk mulai hari kedelapan setelah kultur. Pertumbutran kalus yang terbaik terjadi pada media DK2 (1,5 ppm 2,4-D and 0,15 ppm kinetin) dengan sifa kalus kompalq embriogenik dan kalumya tsrbentuk lebih cepat. Psrtumbuhan liniernya berakhir pada hari ke 21, sehingga dilalrukan panen kalus pada hari ke-21 setelah sub kultur. Alkaloid terdeteksi pada semua sampel, tetapi kadamya tidak selalu meningkat dengan meningkatnya zpt. Kandungan alkaloid paling tingg terjadi pada Df; dengan rata-rafa 0, 1289 %. Kandungan tersebut sargal berbeda nyata pada LSR 1% dengan kandungan alkaloid antikanker pada kalus lainnya.

Immobilization method is frequently used as one of the reliable techniques to increase in vitro s... more Immobilization method is frequently used as one of the reliable techniques to increase in vitro secondary metabolite production. Research has been conducted to evaluate the influence of cell immobilization on cell growth and catharanthine production in C.roseus culture. Callus was first initiated in MS medium with 1,5 mg/L 2,4-D and 0,15 mg/L kinetin. The cell agregate derived from callus of Catharanthus roseus was cultured in MS medium with 2mg/L NAA and 0,2 mg/L kinetin, and subcultured every 10 days for 4 times before immobilization. Immobilization was conducted by using 2,5% Na-alginate. The cell growth and production pattern of catharanthine in the cells agregate were observed every 3 days for 27 days. Qualitative and quantitative analyses were conducted by using HPLC, eluted by using methanol : acetonitrile : diamoniumhydrogenphosphate (3:4:3), rate: 1ml/minute. The results showed that catharanthine was detected in the cells agregate and culture medium. The highest concentration of catharanthine in the control cell was detected on the 9th day of culture, i.e. 39,1843 µg/g, while in medium was obtained on the 12th day of culture, i.e. 5,6892 µg/L. Analysis on the immobilized cells showed that optimal content on the cells agregate was obtained on the 9th day (136,4230 µg/g), while in the medium was on the12th day of culture, i.e. 17,6306 µg/L. The average concentration of catharanthine in immobilized agregate cell culture increased 248 % compared to the control. The result also showed that catharanthine was still retained in the alginate matrix, and reached 3,6974 µg/L on the 18th day. Therefore further experiment is still needed to optimize the secretion of catharanthine into the medium.

This plant is known to contain flavonoids, terpenes, sesquiterpenes, pigmol, xylosides, coumarin and essential oils. Pasote plants are also known to have biological activities such as antimicrobial, cytoxicity, antioxidants, larvacides, antidiabetic, antiparasitic, antiviral and molluscidal. In this study, the extraction of pasote leaves with the maceration method using water and acetone solvents was carried out in order to determine the best type of solvent that can produce pasote leaf extract with the highest antioxidant activity. Determination of antioxidant activity was carried out using DPPH on a UV Vis Spectrophotometer at a wavelength of 517 nm. Commonly found heavy metals such as zinc (Zn), mercury (Hg), and copper (Cu). To reduce or remove heavy metals in the waters, phytoremediation techniques are used. The purpose of this study was to determine the ability of water hyacinth plants (E. crassipes) in remediating Zn2+ metal in contaminated Lake Tempe water. The research method used includes the sampling process, acclimatization, physicochemical analysis, Zn analysis, Bio-Concentration Factor (BCF), and infrared (IR) analysis. The Zn concentration in Lake Tempe water decreased after 30 days of the phytoremediation process. And the highest adsorbed Zn (II) metal ions on water hyacinth plants was 77,257 ppm on the 30th day. The purpose of this study was to obtain research information about the anticancer and antioxidant... more The purpose of this study was to obtain research information about the anticancer and antioxidant potential of Pasote leaf powder extract. Anticancer testing was carried out by the MTT test method on P388 leukemia cells. The antioxidant test was carried out by the DPPH method and determined by UV-Vis Spectro at a wavelength of 517 nm. Data analysis with the Origin Lab program for anticancer, and Excel programs for antioxidant data analysis. Pasote water extract has anticancer activity potential category as an anticancer with IC50 of 0.105 μg / mL. The results of the antioxidant testing of Pasote water included in the medium category with an IC50 value of 50.736 µg/mL. This shows that Pasote leaf water has the potential to be made antioxidant and is not different from antioxidant vitamin C. The conclusion is Pasote leaf water has the potential to be used as an anticancer and antioxidant.

The purpose of this study was to determine the morphological and molecular characters of nutmeg (Myristicasp) with matK markers from West Halmahera. The method used is in three stages, namely a field survey for observations, data collection, analysis of morphological diversity and molecular analysis using the matK gene DNA marker. Penelitian ini bertujuan menganalisis keanekaragaman dan pemanfaatan tanaman pekarangan di Desa Taripa, Kecamatan Pamona Timur, Kabupaten Poso, Sulawesi Tengah. Pengambilan sampel berdasarkan luas pekarangan yaitu kategori sempit (100-200 m2), sedang (300-400 m2) dan luas (500-600 m2). Pada setiap kategori diambil 10 sampel pekarangan dan dicatat seluruh spesies tanaman yang terdapat pada pekarangan tersebut. Pemanfaatan tanaman ditentukan berdasarkan alasan penanaman dengan wawancara langsung dengan pemilik pekarangan. Analisis data meliputi kelimpahan, indek kekayaan, indek keanekaragaman dan indek kemerataan spesies tanaman. Hasil didapatkan sebanyak 64 famili yang terdiri dari 155 spesies dan 1265 individu. Famili yang banyak ditemukan jumlah spesiesnya adalah Araceae. Spesies yang memiliki kelimpahan tertinggi yaitu Garcinia mangostana dan Curcuma longa. Kelimpahan spesies tertinggi pada lahan pekarangan katagori luas. Keanekaragaman dan kekayaan spesies tanaman tertinggi pada lahan pekarangan kategori sempit, sedangkan kemerataan pada lahan kategori sedang. Habitus tanaman yang banyak dimanfaatkan adalah herba. Organ tanaman yang dimanfaatkan umumnya daun, sedangkan pemanfaatan tanaman pekarangan banyak digunakan sebagai sumber pangan. Kata kunci: Keanekaragaman; Garcinia mangostoma; herba; daun. ABSTRACTYard is a plot of land on which there are residential buildings and various plant species can be cultivated. This research aims to analyze the diversity and utilization of garden plants in Taripa Village, East Pamona District, Poso Regency, Central Sulawesi. Sampling was based on the area of the yard, namely the narrow (100-200 m2), medium (300-400 m2) and broad (500-600 m2). In each category, 10 samples of the yard were taken and recorded all plant species found in the yard. The use of plants is determined based on the reasons for planting by direct interviews with the owners of the yards. Data analysis includes abundance, Richness index, diversity index and index evenness of plant species. The results obtained were 64 families consisting of 155 species and 1265 individuals. The family with the most number of species found is Araceae. The species that had the highest abundance were Garcinia mangostana and Curcuma longa. The highest species abundance was in the large yard area. The highest diversity and richness of plant species was in the narrow category land, while evenness was in the medium category. Plant habitus that is widely used is herbaceous. The plant organs that are used are generally leaves, while the use of garden plants is widely used as a food source. Oleh karena itu telah dilakukan penelitian hubungan antara kandungan IAA dengan pertumbuhan dan kandungan kultur agreget sel tapak darah yang diberi perlakuan triptofan dalam erlenmayer. Penelitian ini bertujuan untuk meningkatkan kandungan katarantin dalam kultur agregat sel Chatharantus roseus (l) eksperimental di laboratorium. Konsentrasi prekursor triptofan yang digunakan ialah 0-250 mg/L. Pertumbuhan ditentukan dengan penimbangan berat basah dan berat kering serta pengamatan perubahan secara morfologis. Analisis kandungan katarantin dilakukan dilakukan dengan Kromatografi Cair Kenerja Tinggi (KCKT) menggunakan kolom VP-ODS c-18. Hasil penelitian menunjukkan bahwa kandungan IAA meningkat dengan bertambahnya kosentrasi triptofan sampai 150mg/L. Kansungan tertinggi sebesar (214, 79+0.90 ug/g.bk) diamati pada perlakuan 150mg/L triptofan dengan peningkatan 50,60%. Peningkatan IAA berhubungan erat dengan pertumbuhan agregrat sel dengan r= 0,931. Hubungan IAA dan Katarantin yang diberi perlakuan triptofan juga positif dengan r= 0,83. Pertumbuhan yang optimum pada kandungan katarantin dan IAA yang lebih tinggi dari kontrol merupakan dasar untuk kultur dalam skala besar yang dapat berkelanjutan dalam bioreaktor

Furthermore, randomly interviewed mothers at Posyandu, and people who were willing to be interviewed were partly determined by purposive sampling who had knowledge of traditional medicine. Penelitian ini bertujuan untuk mencri komposisi media yang optimum untuk pembentukan kalus dan menganalisis alkaloid yang terdapat pada kalus Chataranthus roseus yang terbentuk Manfaat penelitian adalah untuk memproduksi atkaloid sebagai obat kanker. Kalus diinduksi pada media Murashige and Skoog (MS) dengan perlakuan sebagai berikut: D[a (tanpa 2,4-D & Kinetin), DKI (1 ppm 2,4-D and 0,1 ppm kinetin), DK2 (1,5 ppm2,4-D and 0,15 ppm kinetin), DK3 Qppm2,4- D and 0,2 ppm kinstin), DK4 (2 ppm 2,4-D). Analisis pertumbuhm kalus dilakukan melalui pe,nimbangan berat basah kalus. Kalus dipanen setelah melewati fase linier pertumbuhar kalus. Kalus diekstraksi dalam metanol yang mengandung 0,25 o/o HCI 1 N. selarna 48 jarn. Analisis kualitatif dilakrftan dengan teknik kromatografi lapis tipis, yang me,nggunakan UV pada panjang gelombang 254 nm untuk menampakkan noda dan untuk penampakan alkaloid disemprot dengan Dragendorf ke,mudian ditentukan RF-nya. Analisis kuantitatif dilakukan dengan melewatkan lromatogram pada spektrofotodensitometer pada panjang gelombang 300 nm. Hasil penelitian menunjulrkan bahwa kalus terbentuk pada semua media (perlakuan) yang diberi zat pergatur tumbuh (zpt). Kalus terbentuk mulai hari kedelapan setelah kultur. Pertumbutran kalus yang terbaik terjadi pada media DK2 (1,5 ppm 2,4-D and 0,15 ppm kinetin) dengan sifa kalus kompalq embriogenik dan kalumya tsrbentuk lebih cepat. Psrtumbuhan liniernya berakhir pada hari ke 21, sehingga dilalrukan panen kalus pada hari ke-21 setelah sub kultur. Alkaloid terdeteksi pada semua sampel, tetapi kadamya tidak selalu meningkat dengan meningkatnya zpt. Kandungan alkaloid paling tingg terjadi pada Df; dengan rata-rafa 0, 1289 %. Kandungan tersebut sargal berbeda nyata pada LSR 1% dengan kandungan alkaloid antikanker pada kalus lainnya.

Immobilization method is frequently used as one of the reliable techniques to increase in vitro s... more Immobilization method is frequently used as one of the reliable techniques to increase in vitro secondary metabolite production. Research has been conducted to evaluate the influence of cell immobilization on cell growth and catharanthine production in C.roseus culture. Callus was first initiated in MS medium with 1,5 mg/L 2,4-D and 0,15 mg/L kinetin. The cell agregate derived from callus of Catharanthus roseus was cultured in MS medium with 2mg/L NAA and 0,2 mg/L kinetin, and subcultured every 10 days for 4 times before immobilization. Immobilization was conducted by using 2,5% Na-alginate. The cell growth and production pattern of catharanthine in the cells agregate were observed every 3 days for 27 days. Qualitative and quantitative analyses were conducted by using HPLC, eluted by using methanol : acetonitrile : diamoniumhydrogenphosphate (3:4:3), rate: 1ml/minute. The results showed that catharanthine was detected in the cells agregate and culture medium. The highest concentration of catharanthine in the control cell was detected on the 9th day of culture, i.e. 39,1843 µg/g, while in medium was obtained on the 12th day of culture, i.e. 5,6892 µg/L. Analysis on the immobilized cells showed that optimal content on the cells agregate was obtained on the 9th day (136,4230 µg/g), while in the medium was on the12th day of culture, i.e. 17,6306 µg/L. The average concentration of catharanthine in immobilized agregate cell culture increased 248 % compared to the control. The result also showed that catharanthine was still retained in the alginate matrix, and reached 3,6974 µg/L on the 18th day. Therefore further experiment is still needed to optimize the secretion of catharanthine into the medium.

Pasote plants are also known to have biological activities such as antimicrobial, cytoxicity, antioxidants, larvacides, antidiabetic, antiparasitic, antiviral and molluscidal. In this study, the extraction of pasote leaves with the maceration method using water and acetone solvents was carried out in order to determine the best type of solvent that can produce pasote leaf extract with the highest antioxidant activity. Determination of antioxidant activity was carried out using DPPH on a UV Vis Spectrophotometer at a wavelength of 517 nm. The purpose of this study was to determine the ability of water hyacinth plants (E. crassipes) in remediating Zn2+ metal in contaminated Lake Tempe water. The research method used includes the sampling process, acclimatization, physicochemical analysis, Zn analysis, Bio-Concentration Factor (BCF), and infrared (IR) analysis. The Zn concentration in Lake Tempe water decreased after 30 days of the phytoremediation process. And the highest adsorbed Zn (II) metal ions on water hyacinth plants was 77,257 ppm on the 30th day. Anticancer testing was carried out by the MTT test method on P388 leukemia cells. The antioxidant test was carried out by the DPPH method and determined by UV-Vis Spectro at a wavelength of 517 nm. Data analysis with the Origin Lab program for anticancer, and Excel programs for antioxidant data analysis. Pasote water extract has anticancer activity potential category as an anticancer with IC50 of 0.105 μg / mL. The results of the antioxidant testing of Pasote water included in the medium category with an IC50 value of 50.736 µg/mL. This shows that Pasote leaf water has the potential to be made antioxidant and is not different from antioxidant vitamin C. The conclusion is Pasote leaf water has the potential to be used as an anticancer and antioxidant.

Penelitian ini bertujuan menganalisis keanekaragaman dan pemanfaatan tanaman pekarangan di Desa Taripa, Kecamatan Pamona Timur, Kabupaten Poso, Sulawesi Tengah. Pengambilan sampel berdasarkan luas pekarangan yaitu kategori sempit (100-200 m2), sedang (300-400 m2) dan luas (500-600 m2). Pada setiap kategori diambil 10 sampel pekarangan dan dicatat seluruh spesies tanaman yang terdapat pada pekarangan tersebut. Pemanfaatan tanaman ditentukan berdasarkan alasan penanaman dengan wawancara langsung dengan pemilik pekarangan. Analisis data meliputi kelimpahan, indek kekayaan, indek keanekaragaman dan indek kemerataan spesies tanaman. Hasil didapatkan sebanyak 64 famili yang terdiri dari 155 spesies dan 1265 individu. Famili yang banyak ditemukan jumlah spesiesnya adalah Araceae. Spesies yang memiliki kelimpahan tertinggi yaitu Garcinia mangostana dan Curcuma longa. Kelimpahan spesies tertinggi pada lahan pekarangan katagori luas. Keanekaragaman dan kekayaan spesies tanaman tertinggi pada lahan pekarangan kategori sempit, sedangkan kemerataan pada lahan kategori sedang. Habitus tanaman yang banyak dimanfaatkan adalah herba. Organ tanaman yang dimanfaatkan umumnya daun, sedangkan pemanfaatan tanaman pekarangan banyak digunakan sebagai sumber pangan. Kata kunci: Keanekaragaman; Garcinia mangostoma; herba; daun. This research aims to analyze the diversity and utilization of garden plants in Taripa Village, East Pamona District, Poso Regency, Central Sulawesi. Sampling was based on the area of the yard, namely the narrow (100-200 m2), medium (300-400 m2) and broad (500-600 m2). In each category, 10 samples of the yard were taken and recorded all plant species found in the yard. The use of plants is determined based on the reasons for planting by direct interviews with the owners of the yards. Data analysis includes abundance, Richness index, diversity index and index evenness of plant species. The results obtained were 64 families consisting of 155 species and 1265 individuals. The family with the most number of species found is Araceae. The species that had the highest abundance were Garcinia mangostana and Curcuma longa. The highest species abundance was in the large yard area. The highest diversity and richness of plant species was in the narrow category land, while evenness was in the medium category. Plant habitus that is widely used is herbaceous. The plant organs that are used are generally leaves, while the use of garden plants is widely used as a food source.
