In this study, we analyzed by Northern blotting with a panel of probes the nuclear and cytoplasmic fractions derived from several rodent cell lines. For most of the genes under study, mature mRNAs could easily be detected in the nuclear fraction and accumulated to higher levels than the corresponding precursors. In addition, significant differences in the nucleocytoplasmic partition of mature mRNAs were observed between genes as well as between cell types (NIH 3T3, CTLL-2, D3-ES, PC-12), indicating that this nuclear accumulation of mRNA is regulated. Thus, while it is usually considered that splicing is the limiting step of pre-mRNA processing, these results point towards transport or nuclear retention of mRNA as a key determinant of nuclear mRNA metabolism.

In addition, we have previously shown that src can regulate pre-mRNA processing. To investigate which functional domains of src are involved in the regulation of splicing and transport of Lymphotoxin a (LTa) transcripts, we have used src mutants in the catalytic, SH2 and SH3 domains in association with the Y527F or the E378G activating mutation. Our results establish that the regulation of pre-mRNA processing and transcription can occur independently of each other. The splicing and transport phenotypes require an intact tyrosine kinase domain and both are insensitive to the deletion of the SH3 domain. Therefore these phenotypes do not depend upon the recruitment through the SH3 domain of src of RNA binding proteins (Sam 68, hnRNP K). By contrast, deletions in the SH2 domain have no eect on splicing but either abolish or exacerbate the transport phenotype depending upon the activating mutation (Y527F or E378G). These divergent responses are associated with speci®c changes in the pattern of tyrosine phosphorylated proteins. Thus, the regulation of transcription, splicing and mRNA transport implicate dierent eector pathways of src. Furthermore, analysis of the transport phenotype reveals the interplay between the SH2 and catalytic domain of the protein.

Cette kinase interagit par son domaine sh3 avec la proteine sam68 qui possede des domaines de fixation a l'arn, ce qui suggere l'existence d'un lien direct entre src et le metabolisme des arn. Dans notre systeme, il s'avere que le domaine sh3 n'est pas necessaire a l'obtention du phenotype. En revanche, nos donnees montrent que, avec le domaine catalytique, le domaine sh2 est necessaire a l'expression d'un phenotype complet. Par ailleurs, nous avons cherche a etablir quelles etapes post-transcriptionnelles etaient limitantes dans le noyau pour l'expression d'une serie de genes. Les proportions entre arn premessagers et arn matures dans le noyau de plusieurs lignees de cellules en culture ont donc ete analysees. Nos resultats donnent une image inattendue du contenu en arn du noyau : loin d'etre les especes predominantes comme il est parfois admis, les arn premessagers s'accumulent peu. En revanche, l'existence d'une population d'arn matures dans la fraction nucleaire montre que, pour un certain nombre de genes, comme beta-actine, le transport de l'arnm vers le cytoplasme est l'etape nucleaire limitante. De plus, cette etape est sensible a la mise en quiescence des cellules, suggerant l'existence d'un lien entre efficacite du transport nucleo-cytoplasmique et proliferation cellulaire.

Abstract : Within the framework of Charles S. Peirce’s theory of perception and Firstness, Second... more Abstract : Within the framework of Charles S. Peirce’s theory of perception and Firstness, Secondness, and Thirdness categories, a model of phenomenal experience is put foward, based on René Thom’s predation lace and his theory of salience and pregnance inspired by Jakob von Uexküll. This topological model suggests that the firstness nature of qualia results from a projective process of self-delusion along a retroactive loop: a retrogression of thirdness due to habit. We will show that its theoretical lineage can be traced back to the projective mechanism of Hinausverlegung (“outside transposition”) theorized by Jakob von Uexküll. Semiotic and biological interpretations of the model are presented. We shall see that this projection of qualia hints at a solution to the “illusion problem” (Frankish, 2016).
Résumé : Dans le cadre de la théorie de la perception de Charles S. Peirce et de sa trichotomie fondamentale (priméité, secondéité, tiercéité), un modèle de l’expérience phénoménale est proposé, fondé sur le lacet de prédation de René Thom et sa théorie des prégnances. Ce modèle morphodynamique suggère que le caractère de priméité des qualia résulte d’un processus projectif d’auto-illusion le long d’une boucle rétroactive : une rétrogression de la tiercéité par habitude. On montrera que sa filiation théorique peut être retracée jusqu’au mécanisme projectif d’Hinausverlegung (« outside transposition ») qu’avait théorisé Jakob von Uexküll. Des interprétations sémiotique et biologique du modèle sont présentées et nous verrons en quoi cette projection des qualia offre une solution au « problème de l’illusion » (Frankish, 2016).
