Detección de transgenes en Organismos Genéticamente Modificados (OGM) y sus subproductos Detectio... more Detección de transgenes en Organismos Genéticamente Modificados (OGM) y sus subproductos Detection of transgenes in genetically modified organisms and their subproducts RESUMEN La ingeniería genética de plantas permite la manipulación directa del ácido desoxirribonucleico (DNA, por sus siglas en inglés), posibilitando la creación de nuevos genes (transgenes), usando elementos provenientes de varios organismos y la introducción de éstos en plantas (plantas u Organismos Genéticamente Modificados [OGM]) para conferirles nuevas características. Los principales cultivos OGM son la soya, el maíz y el algodón. México está expuesto a liberaciones no controladas de OGM. La relevancia de esta liberación aumentará cuando se trate de especies cuyo centro de origen o diversidad se encuentre en nuestro país. Por lo tanto, es necesario diseñar estrategias que permitan detectar la presencia de OGM. Este trabajo tiene como objeto desarrollar y validar metodologías para la detección de OGM y sus subproductos, utilizando la técnica de reacción en cadena de la polimerasa (PCR, por sus siglas en inglés). La estandarización incluye procesos que incrementen la reproducibilidad y el uso de reactivos fácilmente accesibles para permitir su implementación en diferentes laboratorios.

ToChLPV and PepGMV are Begomoviruses that have adapted to a wide host range and are able to cause... more ToChLPV and PepGMV are Begomoviruses that have adapted to a wide host range and are able to cause major diseases in agronomic crops. We analyzed the efficacy of induced resistance to PepGMV in Nicotiana benthamiana plants with two constructs: one construct with homologous sequences derived from PepGMV, and the other construct with heterologous sequences derived from ToChLPV. Plants protected with the heterologous construct showed an efficacy to decrease the severity of symptoms of 45%, while plants protected with the homologous construct showed an efficacy of 80%. Plants protected with the heterologous construct showed a reduction of incidence of 42.86%, while the reduction of incidence in plants protected with the homologous construct was 57.15%. The efficacy to decrease viral load was 95.6% in plants protected with the heterologous construct, and 99.56% in plants protected with the homologous construct. Single and mixed infections of Strawberry mottle virus (SMoV) and Strawberry crinkle virus (SCV) were originally reported in the area (3), and subsequently, Strawberry latent ringspot virus (SLRSV) was also found (4). Samples of strawberry plants showing viral symptoms: stunting, mild chlorosis and reddening, occasional wrinkled, curled, and deformed leaves that may exhibit mottling, and chlorotic spots, forming a putative virus complex were collected in April and December 2007 and July and December 2008. The spiny lobster Panulirus argus (Latreille, 1804) is currently affected by an unenveloped, icos... more The spiny lobster Panulirus argus (Latreille, 1804) is currently affected by an unenveloped, icosahedral, DNA virus termed Panulirus argus virus 1 (PaV1), a virulent and pathogenic virus that produces a long-lasting infection that alters the physiology and behaviour of heavily infected lobsters. Gut-associated microbiota is crucial for lobster homeostasis and well-being, but pathogens could change microbiota composition affecting its function. In PaV1 infection, the changes of gut-associated microbiota are yet to be elucidated. In the present study, we used high-throughput 16S rRNA sequencing technology to compare the bacterial microbiota in intestines of healthy and heavily PaV1-infected male and female juveniles of spiny lobsters P. argus captured in Puerto Morelos Reef lagoon, Quintana Roo, Mexico. We found that basal gut-associated microbiota composition showed a sex-dependent bias, with females being enriched in amplicon sequence variants (ASVs) assigned to Sphingomonas, while males were enriched in the genus Candidatus Hepatoplasma and Aliiroseovarius genera. Moreover, the alpha diversity of microbiota decreased in PaV1-infected lobsters. A significant increase of the genus Candidatus Bacilloplasma was observed in infected lobsters, as well as a significant decrease in Nesterenkonia, Caldalkalibacillus, Pseudomonas, Cetobacterium and Phyllobacterium. We also observed an alteration in the abundances of Vibrio species. Results from this study suggest that PaV1 infection impacts intestinal microbiota composition in Panulirus argus in a sex-dependent manner.

D... more A survey for citrus viroids was carried out on Tahiti lime in several citrus regions of México. Detection of citrus viroids was conducted by reverse transcription-polymerase chain reaction (RT-PCR) and sequential polyacrylamide gel electrophoresis (sPAGE). Both citrus exocortis (CEVd) and citrus cachexia (CCaVd) viroids were consistently detected in 33 out of 35 samples analyzed from Tamaulipas, Veracruz, Tabasco, and Yucatán citrus groves. There was no evidence of infection with other citrus viroids in the samples analyzed. Disease symptoms present in the groves included in the survey were severe bark cracking on main branches and trunk. These symptoms were frequently associated with differing degrees of tree deterioration and poor growth. In addition, infected trees which were grafted on Alemow rootstock had reduced growth in trunk diameter below the bud union along with brownish lesions on the wood. The consistent presence of both CEVd and CCaVd in Tahiti lime groves from diverse geographical origins in México indicates the continuing use of infected propagative budwood sources.

Nucleic acid preparations from shoot samples were analyzed by sequential polyacrylamide gel electrophoresis. All source from both California and Spain contained one to four viroids with distinct physical and biological properties. The size range was estimated from 371 nucleotides for the citrus exocortis viroid (CEV) to 275 for the smallest viroid. The recovery of single viroids suggested a relationship between the distinct viroids and the symptom reaction expressed in citron.

In some geminivirus–host interactions, infected plants show recovery, a phenomenon characterized by symptom disappearance in newly emerging leaves. In pepper–Pepper golden mosaic virus (PepGMV) interaction, the host recovery process involves a silencing mechanism that includes both post-transcriptional (PTGS) and transcriptional (TGS) gene silencing pathways. Under field conditions, PepGMV is frequently found in mixed infections with Pepper huasteco yellow vein virus (PHYVV), another bipartite begomovirus. Mixed infected plants generally show a synergetic phenomenon and do not present recovery. Little is known about the molecular mechanism of this interaction. Geminiviruses are transmitted by various types of insect (whiteflies, leafhoppers, treehoppers and aphids). Members of the genus Begomovirus are transmitted by whiteflies, those in the genera Becurtovirus, Curtovirus, Grablovirus, Mastrevirus and Turncurtovirus are transmitted by specific leafhoppers, the single member of the genus Topocuvirus is transmitted by a treehopper and one member of the genus Capulavirus is transmitted by an aphid. Geminiviruses are transmitted by various types of insect (whiteflies, leafhoppers, treehoppers and aphids). Members of the genus Begomovirus are transmitted by whiteflies, those in the genera Becurtovirus, Curtovirus, Grablovirus, Mastrevirus and Turncurtovirus are transmitted by specific leafhoppers, the single member of the genus Topocuvirus is transmitted by a treehopper and one member of the genus Capulavirus is transmitted by an aphid. Geminiviruses are plant pathogens causing economically important diseases in most tropical and subtropical regions of the world.
