This study analyzed nanoencapsulation of a gel combination of Centella asiatica (CA) transfersomes and rosemary essential oil (REO) nanoemulsion with lipid-based nanocarriers for the ability of both biological compounds to synergistically prevent UVB radiation, along with ameliorative and anti-aging effects. Methods: To ensure the quality, lipid-based nanocarriers of transfersomes and nanoemulsion were characterized based on physicochemical properties such as particle size distribution, polydispersity index, zeta potential. Platelet Rich Fibrin (PRF) is rich for growth factors which can improve the Gingival Medicinal Si... more Platelet Rich Fibrin (PRF) is rich for growth factors which can improve the Gingival Medicinal Signaling Cells' (GMSCs) osteogenic differentiation. Aggrecan is chondrogenic differentiation marker which has a significant role in the early stage of GMSCs' osteogenic differentiation. This study aimed to analyze the expression of Aggrecan post PRF administration on the osteogenic differentiation in vitro. This research is a true experimental study using the post-test only control group design with a simple random sampling. GMSCs were isolated from the lower gingival tissue of healthy male Wistar rats (Rattus novergicus) (n=4), weighted around 250 g, a month old, then cultured for 2 weeks and passaged for 4-5 days. GMSCs in the passage 3-5 were cultured in five M24 plates (N=54; n=6/group) for 7 days, 14 days, and 21 days in three different culture mediums, they were negative control group which included α Modified Eagle Medium; positive control group which contained High Glucose-Dulbecco's Modified Eagle Medium (DMEM-HG) combined with osteogenic medium; and at last, treatment group which were DMEM-HG combined with both osteogenic medium and PRF. A one-way Analysis of Variance (ANOVA) test (P<0.05) was performed. The treatment group showed the highest Aggrecan expression of 16.15±2.15 on the 7 th day. The lowest Aggrecan expression with a value of 3.67±0.76 on the 21 th day occurred in the negative control group. There was a significant difference of Aggrecan expression between groups (P<0.05). PRF administration unexpectedly stimulates Aggrecan expression of GMSCs during the osteogenic differentiation that useful to accelerate the bone remodeling or neo-cartilage formation.

Several experimental models have been established to mimic different pathogenesis characteristics of TBI. The purpose of this study was to determine whether there is evidence of hystopathological lesions in the brain tissue after Marmorou TBI models. This study uses Rattus norvegicus Sprague Dawley strain. Macroscopic and microscopic observations on the brain tissue were done. Macroscopic lesions were observed in the brain. Microscopic observation was performed with Haematoxylin-Eosin (HE) staining and immunohistochemistry on the distribution of microglia cells and pyramidal cells in the cortex. Meanwhile, the distribution of NRG-1/ErbB, proliferation, and apoptosis were observed in the hippocampus. Traumatic brain injury (TBI) is a major health problem because of its high mortality and long-ter... more Traumatic brain injury (TBI) is a major health problem because of its high mortality and long-term disability worldwide. Neural progenitor/stem cells (NPSCs) that survive in certain parts of the brain, enable brain to produce new neurons and glia. ACTH 4-10 Pro 8-Gly 9-Pro 10 has a modulation effect on the expression and activation of the BDNF/TrkB system in the hippocampus area. The BDNF/TrkB pathway system is a potential therapeutic target toward NPSCs proliferation after TBI. Thirty male Sprague-Dawley rats were divided into three groups, i.e A=sham-operated controls; B=TBI; C=TBI+intranasal ACTH 4-10 Pro 8-Gly 9-Pro 10 administration. After 24 h, rats' brains were immunohistochemically processed, to observe the number of cells expressing mBDNF, TrkB, and SOX2 in the subgranular zone(SGZ) of the hippocampus dentate gyrus(DG). Data were analyzed with SPSS 17, ANOVA, Post Hoc Tukey HSD test, with p value < 0,05. Mean expression of BDNF group C=16.33 ± 2.83 increased significantly compared to group A=8.33 ± 1.32(p=0.0001) and group B=5.89 ±1.69(p=0.0001). Mean expression of TrkB group C=17.00 ± 1.58 increased significantly compared to group A=4.33 ± 1.73(p=0.0001) and group B=5.89 ± 2.47(p=0.0001), TrkB expression in group B increased insignificantly compared to group A (p= 0.234). Mean expression of SOX2 in group C=12.56 ± 2.07 increased significantly compared to group B = 8.89 ±2.318(p=0.0001) and group A=4.89 ± 2.42(p=0.0001). ACTH 4-10 Pro 8-Gly 9-Pro 10 can increase the expression of BDNF and TrkB, and the proliferation of NPSCs in the subgranular zone (SGZ) of the hippocampus dentate gyrus (DG).

AIM: This study aimed to investigate the effect of curcumin toward heat shock protein 70 (HSP 70) expression against the expression apoptosis marker (apoptosis-inducing factor [AIF], caspase-3, and TUNEL assay) in brain tissue after TBI. METHODS: Thirty-three Sprague Dawley rats were randomized into three treatment groups, that is, sham-operated controls, closed head trauma (CHT), and CHT with curcumin extract (treatment group). Dendritic cells (DC) are responsible for driving the T lymphocyte responses to a variety of exogenous stimuli, but the mechanisms are not completely understood. Several experimental models have been established to mimic different pathogenesis characteristics of TBI. The purpose of this study was to determine whether there is evidence of hystopathological lesions in the brain tissue after Marmorou TBI models. This study uses Rattus norvegicus Sprague Dawley strain. Macroscopic and microscopic observations on the brain tissue were done. Macroscopic lesions were observed in the brain. Microscopic observation was performed with Haematoxylin-Eosin (HE) staining and immunohistochemistry on the distribution of microglia cells and pyramidal cells in the cortex. Meanwhile, the distribution of NRG-1/ErbB, proliferation, and apoptosis were observed in the hippocampus. Neural progenitor/stem cells (NPSCs) that survive in certain parts of the brain, enable brain to produce new neurons and glia. ACTH 4-10 Pro 8-Gly 9-Pro 10 has a modulation effect on the expression and activation of the BDNF/TrkB system in the hippocampus area. The BDNF/TrkB pathway system is a potential therapeutic target toward NPSCs proliferation after TBI. Thirty male Sprague-Dawley rats were divided into three groups, i.e A=sham-operated controls; B=TBI; C=TBI+intranasal ACTH 4-10 Pro 8-Gly 9-Pro 10 administration. After 24 h, rats' brains were immunohistochemically processed, to observe the number of cells expressing mBDNF, TrkB, and SOX2 in the subgranular zone(SGZ) of the hippocampus dentate gyrus(DG). Data were analyzed with SPSS 17, ANOVA, Post Hoc Tukey HSD test, with p value < 0,05. Mean expression of BDNF group C=16.33 ± 2.83 increased significantly compared to group A=8.33 ± 1.32(p=0.0001) and group B=5.89 ±1.69(p=0.0001). Mean expression of TrkB group C=17.00 ± 1.58 increased significantly compared to group A=4.33 ± 1.73(p=0.0001) and group B=5.89 ± 2.47(p=0.0001), TrkB expression in group B increased insignificantly compared to group A (p= 0.234). Mean expression of SOX2 in group C=12.56 ± 2.07 increased significantly compared to group B = 8.89 ±2.318(p=0.0001) and group A=4.89 ± 2.42(p=0.0001). ACTH 4-10 Pro 8-Gly 9-Pro 10 can increase the expression of BDNF and TrkB, and the proliferation of NPSCs in the subgranular zone (SGZ) of the hippocampus dentate gyrus (DG).

AIM: This study aimed to investigate the effect of curcumin toward heat shock protein 70 (HSP 70) expression against the expression apoptosis marker (apoptosis-inducing factor [AIF], caspase-3, and TUNEL assay) in brain tissue after TBI. METHODS: Thirty-three Sprague Dawley rats were randomized into three treatment groups, that is, sham-operated controls, closed head trauma (CHT), and CHT with curcumin extract (treatment group).
