Two peptide nucleic acids (PNAs) containing three adjacent modified chiral monomers (chiral box) ... more Two peptide nucleic acids (PNAs) containing three adjacent modified chiral monomers (chiral box) were synthesized. The chiral monomers contained either a C2- or a C5-modified backbone, synthesized starting from D- and L-arginine, respectively (2D- and 5L-PNA). The C2-modified chiral PNA was synthesized using a submonomeric strategy to avoid epimerization during solid-phase synthesis, whereas for the C5-derivative, the monomers were first obtained and then used in solid-phase synthesis. The melting temperature of these PNA duplexes formed with the full-match or with single-mismatch DNA were measured both by UV and by CD spectroscopy and compared with the unmodified PNA. The 5L-chiral-box-PNA showed the highest T(m) with full-match DNA, whereas the 2D-chiral-box-PNA showed the highest sequence selectivity. The PNA were spotted on microarray slides and then hybridized with Cy5-labeled full match and mismatched oligonucleotides. The results obtained showed a signal intensity in the order achiral >2D-chiral box >5L-chiral box, whereas the full-match/mismatch selectivity was higher for the 2D chiral box PNA.

Celiac disease is an autoimmune enteropathy triggered by gluten proteins, that develops in some g... more Celiac disease is an autoimmune enteropathy triggered by gluten proteins, that develops in some genetically susceptible subjects after gluten consumption. The high proline and glutamine content of gliadins and glutenins make them very resistant to the degradation by digestive proteases and peptides derived from digestion are absorbed into the lamina propria, where they cause immunological reactions that damage intestinal epithelium. Gluten content of wheat is highly variable, depending on the plant genetics and the growing conditions. The prevalence of celiac sprue seems to be promoted by an early exposition to a large amount of gluten, when gluten-containing food are introduced in the children diet. In the present work, an extraction and digestion method for the prolamine fraction of wheat was developed. Pyroglutamic acid is present in many cheese varieties and particularly in high amounts (0.5 g/100... more Pyroglutamic acid is present in many cheese varieties and particularly in high amounts (0.5 g/100 g of cheese) in extensively ripened Italian cheeses (Grana Padano and Parmigiano Reggiano) that are produced with thermophilic lactic acid bacteria as starters. The mechanism of pyroglutamic acid formation in cheese seems to be mostly enzymatic, as demonstrated by the presence of only L-pyroglutamic acid enantiomer. Thermophilic lactobacilli are involved in pyroglutamic acid production, as suggested by the low pyroglutamic acid content found in Bagos, a ripened Italian mountain cheese produced without addition of starter. Because milk pasteurization did not influence the pyroglutamic acid content in the ripened Grana Padano cheese, the formation of pyroglutamic acid mainly depends on the whey starter microflora rather than that of raw milk. Pyroglutamic acid concentration is linearly correlated (R2 = 0.94) with the age of Grana Padano cheese.

Enantiomeric analysis is one of the crucial points for the sensor technology, due to the increasi... more Enantiomeric analysis is one of the crucial points for the sensor technology, due to the increasing importance that enantiomerically pure compounds and drugs have in pharmaceutic and agrochemical applications. Enantiomeric luminescent sensors give different responses by interaction or reaction with chiral molecules, allowing one to assess their optical purity by spectroscopic measurements. Moreover, chemosensors have been developed to perform enantiomeric analysis of both luminescent and non-luminescent organic compounds. In the present chapter we focus on the recent advances in the sensing of chiral molecules by luminescent sensory systems, with the aim of outlining different mechanisms: fluorescence quenching by metal complexes, photoinduced electron transfer (PET) quenching, fluorescence enhancement by PET inhibition, analyte induced sensor conformational changes, modulation of excimer and exciplex formation, and aggregation induced emission enhancement (AIEE). Recent advances in the use of more elaborate techniques such as anisotropy measurements, gated detection, circularly polarized luminescence (CPL) and perspectives in the field are also discussed. Emphasis is given to the methods which have provided high enantioselectivity and which are amenable to fast screening procedures.
