Numerous studies in different species of mammals show the presence of seminal plasma proteins directly linked to fertility. Thus, the objective of the present study was to develop a protocol for purification of two proteins of the Binder Sperm protein (BSP) family, named Ram Seminal Vesicle Proteins with 14 kDa (RSVP14) and 22 kDa (RSVP22). Seminal samples were collected from 16 adult Morada Nova rams by eletroejaculation. Andrological evaluation confirmed that all animals were healthy and reproductively sound. Seminal plasma was separated from sperm by two centrifugations (700g 4oC 15 min and 5000g 4oC 60 min). A pool was made with the seminal plasma from all 16 animals and stored at -20°C. Protein concentration was determined by the Bradford method. Fast Protein Liquid Chromatography (FPLC) was used to separate proteins (10 mg total) from the seminal plasma samples using a Gelatin-SepharoseTM column and binding buffer consisting of 40mM Tris and 2mM CaCl2, pH 7.0. In addition, elution buffer comprised of 40mM Tris, 8M Urea and 2mM CaCl2, pH 7.0 was also used. Proteins were eluted in three chromatographic peaks. The first two peaks contained proteins with low affinity for gelatin and the third peak included proteins with high affinity for gelatin. This last group of proteins probably contains fibronectin type II domains in their structure. All three peaks of the eluted content were concentrated using a 10kDa filter (MWCO Vivaspin 10,000 Da, GE) and the material retained by the filter was quantified by Bradford’s method. An aliquot of 20 µg was removed from each peak and subjected to 12.5% SDS-PAGE. To confirm the presence of the BSP proteins in the seminal plasma samples, Western blots were carried out using antiram BSP as primary antibody (1: 6,000) and Dnk to pAb Rb IgG anti-rabbit (1: 5,000) as secondary antibody (Abcam, Cambridge, UK). The molecular weights of the proteins separated by electrophoresis were confirmed using Quantity One software (Bio-Rad Laboratories, USA). The bands detected in the gelatin affinity peak were between 12 and 52kDa. Immunodetection by Western Blot showed specific staining for BSPs in all peaks analyzed, with higher staining in the gelatin affinity peak. The gelatin affinity peak was subjected to affinity chromatography on HiTrap ™ Heparin HP column with binding buffer consisting of Tris 40 mM and ClCa2 2mM, pH 7.0 and elution buffer consisting of Tris 40 mM, 2 mM CaCl2 and NaCl2 1mM, pH 7.0. Two chromatographic peaks were well formed and defined. The first peak represented the non-ligated fraction with a maximum amplitude of 120 mAu; and the second peak representing the heparin affinity with 80 mAu for maximum amplitude. The two peaks were centrifuged twice at 5000g for 60 min at 4°C using the 10kDa filter used before. All the material retained on the filter was again quantified by Bradford’s method. The proteins in the two peaks were detected by SDS-PAGE and Western Blots as described above. By Quantity One analyses, the first peak showed 5 bands of 14, 15, 22, 24 and 40 kDa, while the second peak showed 6 bands of 12, 16, 21, 22, 170 and 198 kDa. The Western blot testing confirmed the presence of BSP proteins of 14 kDa for the first peak and 22-kDa in the both peaks Therefore, it was observed in the present study that quantitative technique using two affinity columns identified two proteins of interest, but RSVP´s couldn’t be completely isolated. Soon, it is necessary to use a third chromatographic column to separate completely the two protein bands and use them in functional tests.

Plastids of higher plants are generally semiautonomous with a ~120–150 kb genome. Chloroplast transformation has become an attractive alternative to nuclear gene transformation due to its advantages, high protein levels, the feasibility of expressing multiple proteins from polycistronic mRNAs, and gene containment through the lack of pollen transmission. The review presents the recent trends and methods for plastid genome engineering and transgene expression and summarizes the potential of plastid transformation in various fields of biotechnology and also as a source of therapeutic proteins.

The present overview is based on recent available information on nasopharyngeal carcinoma (NPC) f... more The present overview is based on recent available information on nasopharyngeal carcinoma (NPC) from various parts of the world including India. The available data suggests that NPC is a rare tumour in most parts of the world. But the incidence is higher in China and South East Asia and also among the Chinese wherever they have migrated. NPC is also relatively higher especially among the mongoloid group of the people in the North Eastern Region of India as compared to other parts of the country. The distinct geographical and ethnic distribution of NPC have stimulated much research to find out its etiology. The results suggest that Epstein Barr Virus (EBV) infection and genetic susceptibility are the constant aetiological factors for the higher incidence of NPC among various ethnic groups while other factors such as ingestants and inhalants may depend on the distinct dietary practices and living environment adopted by various ethnic groups in different geographical region of the world.

Mathematical simulation is widely used for investigating and designing the compressors. Investigation of the processes of reciprocating compressors using mathematical models is an effective tool by high development of computing technique, which enables complicated problems to be solved with a minimal number of simplifying assumptions. A considerable number of previous works has been done on the mathematical modeling and simulation. This paper presents a simplified and effective mathematical model for the estimation of reciprocating compressor performance using personal computers that can be easily handled. The developed model has been validated using the experimental results from the compressor with reed vale in the delivery side and ring valve in the suction side. The compressor is tested for different delivery pressures and different shaft speeds. The effect of parameters speed, discharge pressure on thermodynamic behaviour of compressor in working condition has been analyzed. The model can predict cylinder pressure, cylinder volume, cylinder temperature, valve lift and resultant torque at different crank angles and free air delivered and indicated power of the compressor. The predicted results using the developed mathematical model are very much comparable with the experimental results.

Ceramic-metal functionally graded materials (FGMs) have been attracting a great deal of attention... more Ceramic-metal functionally graded materials (FGMs) have been attracting a great deal of attention as environmental barrier coatings (EBCs) for aerospace structures and turbine blades working under elevated temperatures and chemically harsh environment. A relatively new, simple and low cost manufacturing method of environmental barrier coatings based on the slurry spray technique (SST) has been developed. In this research, it is desired to develop this technique to achieve the coating quality comparable to the existing manufacturing methods, which are often expensive. This paper describes the development and optimization of mullite-nickel based coating using Slurry Spray Technique analysing the effect of MgO as liquid phase sintering (LPS) additive. Taguchi's experimental design based on L9 orthogonal array was employed for optimizing the tech- nique and analyzing the effect of selected process parameters (sintering temperature, sintering time and percentage of additives). The response parameter of the coating so produced is measured in terms of coating interface adhesion strength. The test results have been used to downselect the significant process parameters and help demonstrate the feasibility of coatings produced by slurry spray technique. The test results demonstrate satisfactory adhesion strength of the developed coating, which is comparable with that pro- duced from traditional techniques such as flame spray method.

An automatic weather station (AWS) or data collection platform (DCP) is a rugged, environmentally... more An automatic weather station (AWS) or data collection platform (DCP) is a rugged, environmentally qualified system of sensors, computers and communication equipment, which automatically and continuously measures real time surface weather conditions and three times hourly sends the observed meteorological parameters to the central station through a satellite link in a self-timed pseudo random manner in its prescribed 10 minute time slot within the 60 minutes before the next observation takes place. Loss of AWS data is due to the collision of data bursts transmitted simultaneously by any two or more AWS. Generally, the collision of AWS data bursts depends upon the transmission time or transmission baud rate, number of AWS in a network, and total number of bits in the AWS data burst. This paper describes the influence of data burst collisions on pseudo random burst sequence (PRBS) transmission through satellite.

The synthesized nanostructures are characterized by scanning electron microscope and X-ray diffraction. The XRD analysis confirmed the formation of nanowires of copper telluride. The photoluminescence emission spectrum was obtained at 457.56 nm for 100 nm copper telluride nanowires.

A skeleton based strategy is adopted to extract the features from a video sequence representing any human gesture. In our binary tree implementation of SVM, the number of binary classifiers required is reduced since, instead of grouping different classes together in order to train a global classifier, we select two classes for training at every node of the tree and use probability theory to classify the remaining points based on their similarities and differences to the two classes used for training. This process is carried on, randomly selecting two classes for training at a node, thus creating two child nodes and subsequently assigning the classes to the nodes derived. In the classification phase, we start out at the root node. At each node of the tree, a binary decision is made regarding the assignment of the input data point to either of the group represented by the left and right sub-tree of the node which may contain multiple classes. This is repeated recursively downward until we reach a leaf node that represents the class to which the input data point belonging. Finally, the proposed framework is tested on various data sets to check its efficiency. Encouraging results are achieved in terms of classification accuracy.

Ductal carcinoma in situ (DCIS) is a heterogeneous and complex disorder that has changed greatly ... more Ductal carcinoma in situ (DCIS) is a heterogeneous and complex disorder that has changed greatly as an entity over the past few decades. Despite an overall favorable prognosis and evidence-based recommendations from randomized trials, the clinical management of DCIS varies considerably among surgeons. While the goal is to achieve maximal local control with minimal morbidity, surgeons at times both over- and undertreat these patients. Currently, the rates of mastectomy and contralateral prophylactic mastectomy have been dramatically rising without clear evidence of better control. Radiotherapy is omitted in about one- half of patients undergoing lumpectomy. Tools to help stratify patients at highest risk for DCIS or invasive recurrence may assist in determining appropriate therapy. This article reviews some of the areas of controversy in the surgical approach to DCIS, including breast conservation versus mastectomy, the use of radiotherapy, the appropriate extent of margins, and the criteria for including sentinel nodal biopsy.

The present/current diagnostic kits and methods to diagnose BTV are laborious, time consuming and expensive. In the present study, we have attempted to develop a novel approach to detect BTV antibodies in sera that in future can be harnessed for developing a pen side diagnostic test. Briefly, we identified the immunodominant regions of the VP7 protein of BTV and synthesized them in the multiple antigenic peptide (MAP) format with cysteine at C-terminal of the lysine mosaic, which elicited highly ordered conformation as well as ELISA reactivity. Finally, we coated the MAP peptides on the gold nanoparticles that can be used to detect BTV specific antibodies in the sera using a spot test.

The results suggest that Epstein Barr Virus (EBV) infection and genetic susceptibility are the constant aetiological factors for the higher incidence of NPC among various ethnic groups while other factors such as ingestants and inhalants may depend on the distinct dietary practices and living environment adopted by various ethnic groups in different geographical region of the world.

Mathematical simulation is widely used for investigating and designing the compressors. Investigation of the processes of reciprocating compressors using mathematical models is an effective tool by high development of computing technique, which enables complicated problems to be solved with a minimal number of simplifying assumptions. A considerable number of previous works has been done on the mathematical modeling and simulation. This paper presents a simplified and effective mathematical model for the estimation of reciprocating compressor performance using personal computers that can be easily handled. The developed model has been validated using the experimental results from the compressor with reed vale in the delivery side and ring valve in the suction side. The compressor is tested for different delivery pressures and different shaft speeds. The effect of parameters speed, discharge pressure on thermodynamic behaviour of compressor in working condition has been analyzed. The model can predict cylinder pressure, cylinder volume, cylinder temperature, valve lift and resultant torque at different crank angles and free air delivered and indicated power of the compressor. The predicted results using the developed mathematical model are very much comparable with the experimental results.

A relatively new, simple and low cost manufacturing method of environmental barrier coatings based on the slurry spray technique (SST) has been developed. In this research, it is desired to develop this technique to achieve the coating quality comparable to the existing manufacturing methods, which are often expensive. This paper describes the development and optimization of mullite-nickel based coating using Slurry Spray Technique analysing the effect of MgO as liquid phase sintering (LPS) additive. Taguchi's experimental design based on L9 orthogonal array was employed for optimizing the tech- nique and analyzing the effect of selected process parameters (sintering temperature, sintering time and percentage of additives). The response parameter of the coating so produced is measured in terms of coating interface adhesion strength. The test results have been used to downselect the significant process parameters and help demonstrate the feasibility of coatings produced by slurry spray technique. The test results demonstrate satisfactory adhesion strength of the developed coating, which is comparable with that pro- duced from traditional techniques such as flame spray method.

Loss of AWS data is due to the collision of data bursts transmitted simultaneously by any two or more AWS. Generally, the collision of AWS data bursts depends upon the transmission time or transmission baud rate, number of AWS in a network, and total number of bits in the AWS data burst. This paper describes the influence of data burst collisions on pseudo random burst sequence (PRBS) transmission through satellite.

The synthesized nanostructures are characterized by scanning electron microscope and X-ray diffraction. The XRD analysis confirmed the formation of nanowires of copper telluride. The photoluminescence emission spectrum was obtained at 457.56 nm for 100 nm copper telluride nanowires.

A skeleton based strategy is adopted to extract the features from a video sequence representing any human gesture. In our binary tree implementation of SVM, the number of binary classifiers required is reduced since, instead of grouping different classes together in order to train a global classifier, we select two classes for training at every node of the tree and use probability theory to classify the remaining points based on their similarities and differences to the two classes used for training. This process is carried on, randomly selecting two classes for training at a node, thus creating two child nodes and subsequently assigning the classes to the nodes derived. In the classification phase, we start out at the root node. At each node of the tree, a binary decision is made regarding the assignment of the input data point to either of the group represented by the left and right sub-tree of the node which may contain multiple classes. This is repeated recursively downward until we reach a leaf node that represents the class to which the input data point belonging. Finally, the proposed framework is tested on various data sets to check its efficiency. Encouraging results are achieved in terms of classification accuracy.

Despite an overall favorable prognosis and evidence-based recommendations from randomized trials, the clinical management of DCIS varies considerably among surgeons. While the goal is to achieve maximal local control with minimal morbidity, surgeons at times both over- and undertreat these patients. Currently, the rates of mastectomy and contralateral prophylactic mastectomy have been dramatically rising without clear evidence of better control. Radiotherapy is omitted in about one- half of patients undergoing lumpectomy. Tools to help stratify patients at highest risk for DCIS or invasive recurrence may assist in determining appropriate therapy. This article reviews some of the areas of controversy in the surgical approach to DCIS, including breast conservation versus mastectomy, the use of radiotherapy, the appropriate extent of margins, and the criteria for including sentinel nodal biopsy.

The present/current diagnostic kits and methods to diagnose BTV are laborious, time consuming and expensive. In the present study, we have attempted to develop a novel approach to detect BTV antibodies in sera that in future can be harnessed for developing a pen side diagnostic test. Briefly, we identified the immunodominant regions of the VP7 protein of BTV and synthesized them in the multiple antigenic peptide (MAP) format with cysteine at C-terminal of the lysine mosaic, which elicited highly ordered conformation as well as ELISA reactivity. Finally, we coated the MAP peptides on the gold nanoparticles that can be used to detect BTV specific antibodies in the sera using a spot test.
