Expressed sequence tags (ESTs) and differentially expressed cDNAs from the self-fertilizing fish,... more Expressed sequence tags (ESTs) and differentially expressed cDNAs from the self-fertilizing fish, Kryptolebias marmoratus were mined to develop alternative biomarkers for endocrine-disrupting chemicals (EDCs). 1,577 K. marmoratus cDNA clones were randomly sequenced from the 5'-end. These clones corresponded to 1,518 and 1,519 genes in medaka dbEST and zebrafish dbEST, respectively. Of the matched genes, 197 and 115 genes obtained Unigene IDs in medaka dbEST and zebrafish dbEST, respectively. Many of the annotated genes are potential biomarkers for environmental stresses. In a differential display reverse transcriptase-polymerase chain reaction (DD RT-PCR) study, 56 differential expressed genes were obtained from fish liver exposed to bisphenol A. Of these, 16 genes were identified after BLAST search to GenBank, and the annotated genes were mainly involved in catalytic activity and binding. Deltamethrin, an a-cyano class of pyrethroid insecticide is used in insect pest control and antim... more Deltamethrin, an a-cyano class of pyrethroid insecticide is used in insect pest control and antimalaria programs in several countries including India. Although various toxic manifestations of deltamethrin are reported in mammals, its ecotoxicologic dimensions are not adequately researched in ecologically and commercially important fishes. In this study, we report genotoxic effect of deltamethrin in a biomarker fish Channa punctata (Bloch). Adult fish were exposed to three concentrations of technical grade deltamethrin (0.4, 0.8, and 1.2 lg/L) for 48 and 72 h. Ethyl methane sulfonate was used as a positive control. Fish were analyzed for induction of micronucleus (MN), nuclear abnormalities (NAs), and oxidative stress biomarkers in erythrocytes. Deltamethrin significantly induced MN and NAs accompanied by increased lipid peroxidation. Activity of antioxidant enzyme superoxide dismutase was significantly decreased but an increase was observed in reduced glutathione level after 72 h of exposure. The NAs in exposed fish included blebbed, lobed and notched nuclei, and binucleated erythrocytes. Our findings suggest that oxidative stress may, in part, be contributing to deltamethrin-induced genotoxic damage to erythrocytes. Although MN induction is a nonspecific biomarker, it may provide an indication of pollution load of deltamethrin in the affected fish population when used as part of suite of other biomarkers.

Reactive oxygen species (ROS) and mitochondrial perturbations have been implicated in drug induced hepatotoxicity, although their role in the pathway needs exploration. Study was undertaken to elucidate the effect of Fumaria parviflora Lam. (Fp) on nimesulide induced cell death in primary rat hepatocyte cultures. Fp extract treated cells showed increased viability as compared to nimesulide stressed cells as assessed by MTT assay. LDH leakage increased significantly at 500 lM nimesulide, and the data suggested that apoptosis was the predominant mechanism responsible for cell death. Nimesulide induced apoptosis was further confirmed by DNA fragmentation and chromatin condensation. Nimesulide exposure increased intracellular ROS, translocation of Bax and Bcl2 followed by mitochondrial depolarization and cytochrome c (Cyt c) release along with caspase-9/-3 activity confirming involvement of mitochondria in nimesulide induced apoptosis. Events like membrane depolarization of mitochondria, expression of Bax, Bcl2, externalization of phosphatidyl serine are substantially reversed by the pre-treatment of Fp extract. Thus, the study indicates that Fp extract modulates critical events regulating pro and anti-apoptotic proteins in mitochondria dependent apoptosis induced by nimesulide.

Protein A (PA), a cell wall glycoprotein of Staphylococcus aureus Cowan I, is known to possess im... more Protein A (PA), a cell wall glycoprotein of Staphylococcus aureus Cowan I, is known to possess immunomodulatory and antitumor activities. In the present study the effect of protein A on the anti-sheep erythrocyte primary antibody response in normal mice has been investigated. Intraperitoneal administration of PA resulted in suppression of primary antibody response in mice immunized with sheep red blood cells (SRBC) as assessed by plaque forming cell (PFC) assay. The suppressive effect was dose dependent. Maximum suppression in PFC response was observed at 12 micrograms PA/animal and could be observed as low as 1 microgram PA/animal. The suppression at 6 and 12 micrograms PA/animal was significant when compared with control values (p < 0.02 and p < 0.001, respectively). However, no significant suppression was recorded at 1 microgram dose. The suppression was proportional to the numbers of administrations of PA to the mice and critically dependent on the timing of inoculation of PA. PA produced pronounced suppression when injected prior to the immunization of animals with SRBC. PA reduced splenic localization of 51Cr labelled SRBC to 55%. It is proposed that the altered localization of antigen (SRBC) in spleen may be responsible for reduced plaque forming cells in normal mice.

We have previously shown that PA suppresses the anti-sheep erythrocyte primary antibody response in normal mice. The present investigation evaluates the effect of protein A on the anti-sheep erythrocyte primary antibody response in tumor-bearing mice. The primary antibody response in tumor-bearing mice immunized with sheep red blood cells (SRBC) was suppressed by the intraperitoneal administration of PA in a dose-dependent fashion. The plaque forming cell (PFC) assay was used to assess this response. Maximum suppression of the PFC response was observed at 12 micrograms PA/animal (p < 0.001) and could be observed at doses as low as 1 microgram PA/animal (p < 0.01). The amount of suppression was proportional to the number of PA doses administered. In addition this effect was critically dependent on the timing of PA administration. PA showed no significant effect on PFC when injected after immunization, but it produced pronounced suppression when injected prior to the immunization with SRBC. Maximum suppression of the PFC response was observed when PA was administered one day before the antigen challenge. PA also reduced splenic localization of 51Cr labeled SRBC to 42% (p < 0.01). The altered localization of antigen in spleen may be responsible for reduced PFC response in tumor-bearing mice. Depletion of B-lymphocyte is reported to exhibit tumor inhibition. Therefore, we propose that the suppression of the primary antibody response by PA helps in tumor regression by reducing the soluble immunosuppressive immune complexes.

In the present communication an attempt has been made to evaluate the effects of aflatoxin BI (AFBt) on hepatic microsomal drug metabolizing enzymes in growing rats, The weanling rats were exposed to 60, 300 or 600 ug AFBllkg body weight, per os, on alternate days for 4 weeks, in 0.2 ml com oil. A significant depression in the activities of aryl hydrocarbon hydroxylase (AHH), aniline hydroxylase (AH) and aminopyrene-N-demethylase (AND) was observed at 300 J.lg and 600 J.lg doses of AFBt. However, no significant change was recorded in glutathione-S-transferase (GST) activity and total sulphydryl (SH) content upon AFBt exposure in weanling rats. Thus, AFBt appears to have more pronounced effect on the phase I, rather than phase II, biotransformation enzyme system in weanling rats. The depression of drug metabolizing enzymes together with suppression of immunity by AFBt, as reported earlier by us, may increase the susceptibility of the host to toxic chemicals, drugs and infectious agents, particularly during the post-natal period.

The present investigation was undertaken to assess whether aflatoxin B1 (AFB1) has any modulatory... more The present investigation was undertaken to assess whether aflatoxin B1 (AFB1) has any modulatory effect on the growth and progression of Ehrlich's ascites tumor (EAT) in mice or not. Male Swiss albino mice were treated with 0, 70, 350 and 700 micrograms AFB1/kg body weight in 0.2 ml corn oil on alternate days, orally, for two weeks. Treated animals were challenged with 1 x 10(6) cells of Ehrlich's ascites tumor. Animals were monitored for the appearance of palpable tumor, body weight gain as a measure of tumor burden, mortality profile and tumor cell population. Some parameters of cell mediated immunity (CMI), humoral immunity and non-specific immunity were also studied in aflatoxin treated animals in order to find out the mechanism of action of AFB1 on host immunity. It was observed that AFB1 treatment resulted in an early appearance of tumor, enhanced mortality, appreciable increase in body weight gain and EAT cell population following tumor challenge, in comparison with the control animals. Aflatoxin treatment caused suppression of CMI including an impairment of macrophage function, while humoral immunity was not much affected. It may be concluded that impaired CMI and macrophage function might be contributing to the increased growth of EAT in the AFB1 treated animals. Our findings may be relevant in that when a host is exposed to aflatoxins concomitantly with other carcinogens and cancer causing situations.

Swiss albino mice were treated with zinc acetate (3 mg/kg body weight) in one or two intraperitoneal injections. Various immune function assays were performed in treated animals. Zinc treatment to normal animals caused potentiation of T-lymphocyte and macrophage functions. Zinc treatment was also found to increase host resistance against Candida albicans and Semliki Forest virus infections. Increased resistance against endotoxin shock and Ehrlich's ascites tumor challenge was also observed in zinc treated animals. It can be stated from this study that zinc treatment potentiates the cell mediated immunity and host resistance against infection and tumor challenge.

The weanling rats were subchronically exposed to 60, 300 and 600 Ixg AFBJkg body weight for four weeks on alternate days by oral feeding. Various parameters of cell mediated immunity (CMI) and humoral immunity were assessed in control and treated animals. CMI was evaluated by measuring delayed type of hypersensitivity (DTH) response and humoral by plaque forming cell (PFC) assay. The lymphoproliferative response assay for T-and B-cells was also performed. It was observed that AFB1 selectively suppressed cell mediated immunity in growing rats. AFB1 suppressed CMI at the 300 and 600 p~g dose levels only as measured by DTH response assay. It is concluded that continuous low level exposure of aflatoxin to growing host may enhance its susceptibility to infection and tumorigenesis.

Walnut (Juglans regia L.) bark has been claimed to possess anti-inflammatory, blood purifying, an... more Walnut (Juglans regia L.) bark has been claimed to possess anti-inflammatory, blood purifying, anticancer, depurative, diuretic and laxative activities. It contains several therapeutically active constituents, especially polyphenols. We studied the antioxidant potential of aqueous extract of walnut bark and its modulatory effect on cyclophosphamide (CP)-induced urotoxicity in Swiss albino male mice. Free radical-scavenging activity of extract was assessed in four in vitro assays. The phenolic and flavonolic contents of the extract were also measured. Walnut bark extract treatment (150 mg/kg p.o. × 10 days) resulted in protective restoration of decreased antioxidants in CP-treated (18 mg/kg i.p. × 10 days) animals. CP treatment caused decreases in the activities of catalase (CAT), glutathione peroxidase (GP), glutathione reductase (GR) and glutathione Stransferase (GST) and in the glutathione (GSH) content in urinary bladder and a significant concomitant increase in lipid peroxidation (LPO). Administration of extract restored all the antioxidants significantly and lowered the elevated LPO in the bladder. A correlation between radical scavenging capacities of the extract with phenolic content was observed thus justifying its antioxidant potential against oxidative stress-mediated urotoxicity in mice. Walnut is reported to possess antiproliferative activity. Its protective effect on CP-induced toxicity in bladder is a promising activity, which warrants possible clinical investigations on this medicinal plant.

Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidin-1-oxyl) has powerful antioxidant activity. We investigated its potential nephroprotective effects and the underlying mechanisms that may add further benefits to its clinical usefulness in a CP-induced AKI model. Male Swiss albino mice were divided randomly into four groups: control, CP (20 mg/kg intraperitoneally), tempol (100 mg/kg/day, per os) + CP, and tempol only treatments. Events like membrane depolarization of mitochondria, expression of Bax, Bcl2, externalization of phosphatidyl serine are substantially reversed by the pre-treatment of Fp extract. Thus, the study indicates that Fp extract modulates critical events regulating pro and anti-apoptotic proteins in mitochondria dependent apoptosis induced by nimesulide.

In the present study the effect of protein A on the anti-sheep erythrocyte primary antibody response in normal mice has been investigated. Intraperitoneal administration of PA resulted in suppression of primary antibody response in mice immunized with sheep red blood cells (SRBC) as assessed by plaque forming cell (PFC) assay. The suppressive effect was dose dependent. Maximum suppression in PFC response was observed at 12 micrograms PA/animal and could be observed as low as 1 microgram PA/animal. The suppression at 6 and 12 micrograms PA/animal was significant when compared with control values (p < 0.02 and p < 0.001, respectively). However, no significant suppression was recorded at 1 microgram dose. The suppression was proportional to the numbers of administrations of PA to the mice and critically dependent on the timing of inoculation of PA. PA produced pronounced suppression when injected prior to the immunization of animals with SRBC. PA reduced splenic localization of 51Cr labelled SRBC to 55%. It is proposed that the altered localization of antigen (SRBC) in spleen may be responsible for reduced plaque forming cells in normal mice.

We have previously shown that PA suppresses the anti-sheep erythrocyte primary antibody response in normal mice. The present investigation evaluates the effect of protein A on the anti-sheep erythrocyte primary antibody response in tumor-bearing mice. The primary antibody response in tumor-bearing mice immunized with sheep red blood cells (SRBC) was suppressed by the intraperitoneal administration of PA in a dose-dependent fashion. The plaque forming cell (PFC) assay was used to assess this response. Maximum suppression of the PFC response was observed at 12 micrograms PA/animal (p < 0.001) and could be observed at doses as low as 1 microgram PA/animal (p < 0.01). The amount of suppression was proportional to the number of PA doses administered. In addition this effect was critically dependent on the timing of PA administration. PA showed no significant effect on PFC when injected after immunization, but it produced pronounced suppression when injected prior to the immunization with SRBC. Maximum suppression of the PFC response was observed when PA was administered one day before the antigen challenge. PA also reduced splenic localization of 51Cr labeled SRBC to 42% (p < 0.01). The altered localization of antigen in spleen may be responsible for reduced PFC response in tumor-bearing mice. Depletion of B-lymphocyte is reported to exhibit tumor inhibition. Therefore, we propose that the suppression of the primary antibody response by PA helps in tumor regression by reducing the soluble immunosuppressive immune complexes.

In the present communication an attempt has been made to evaluate the effects of aflatoxin BI (AFBt) on hepatic microsomal drug metabolizing enzymes in growing rats, The weanling rats were exposed to 60, 300 or 600 ug AFBllkg body weight, per os, on alternate days for 4 weeks, in 0.2 ml com oil. A significant depression in the activities of aryl hydrocarbon hydroxylase (AHH), aniline hydroxylase (AH) and aminopyrene-N-demethylase (AND) was observed at 300 J.lg and 600 J.lg doses of AFBt. However, no significant change was recorded in glutathione-S-transferase (GST) activity and total sulphydryl (SH) content upon AFBt exposure in weanling rats. Thus, AFBt appears to have more pronounced effect on the phase I, rather than phase II, biotransformation enzyme system in weanling rats. The depression of drug metabolizing enzymes together with suppression of immunity by AFBt, as reported earlier by us, may increase the susceptibility of the host to toxic chemicals, drugs and infectious agents, particularly during the post-natal period.

Male Swiss albino mice were treated with 0, 70, 350 and 700 micrograms AFB1/kg body weight in 0.2 ml corn oil on alternate days, orally, for two weeks. Treated animals were challenged with 1 x 10(6) cells of Ehrlich's ascites tumor. Animals were monitored for the appearance of palpable tumor, body weight gain as a measure of tumor burden, mortality profile and tumor cell population. Some parameters of cell mediated immunity (CMI), humoral immunity and non-specific immunity were also studied in aflatoxin treated animals in order to find out the mechanism of action of AFB1 on host immunity. It was observed that AFB1 treatment resulted in an early appearance of tumor, enhanced mortality, appreciable increase in body weight gain and EAT cell population following tumor challenge, in comparison with the control animals. Aflatoxin treatment caused suppression of CMI including an impairment of macrophage function, while humoral immunity was not much affected. It may be concluded that impaired CMI and macrophage function might be contributing to the increased growth of EAT in the AFB1 treated animals. Our findings may be relevant in that when a host is exposed to aflatoxins concomitantly with other carcinogens and cancer causing situations.

Swiss albino mice were treated with zinc acetate (3 mg/kg body weight) in one or two intraperitoneal injections. Various immune function assays were performed in treated animals. Zinc treatment to normal animals caused potentiation of T-lymphocyte and macrophage functions. Zinc treatment was also found to increase host resistance against Candida albicans and Semliki Forest virus infections. Increased resistance against endotoxin shock and Ehrlich's ascites tumor challenge was also observed in zinc treated animals. It can be stated from this study that zinc treatment potentiates the cell mediated immunity and host resistance against infection and tumor challenge.

CMI was evaluated by measuring delayed type of hypersensitivity (DTH) response and humoral by plaque forming cell (PFC) assay. The lymphoproliferative response assay for T-and B-cells was also performed. It was observed that AFB1 selectively suppressed cell mediated immunity in growing rats. AFB1 suppressed CMI at the 300 and 600 p~g dose levels only as measured by DTH response assay. It is concluded that continuous low level exposure of aflatoxin to growing host may enhance its susceptibility to infection and tumorigenesis.

It contains several therapeutically active constituents, especially polyphenols. We studied the antioxidant potential of aqueous extract of walnut bark and its modulatory effect on cyclophosphamide (CP)-induced urotoxicity in Swiss albino male mice. Free radical-scavenging activity of extract was assessed in four in vitro assays. The phenolic and flavonolic contents of the extract were also measured. Walnut bark extract treatment (150 mg/kg p.o. × 10 days) resulted in protective restoration of decreased antioxidants in CP-treated (18 mg/kg i.p. × 10 days) animals. CP treatment caused decreases in the activities of catalase (CAT), glutathione peroxidase (GP), glutathione reductase (GR) and glutathione Stransferase (GST) and in the glutathione (GSH) content in urinary bladder and a significant concomitant increase in lipid peroxidation (LPO). Administration of extract restored all the antioxidants significantly and lowered the elevated LPO in the bladder. A correlation between radical scavenging capacities of the extract with phenolic content was observed thus justifying its antioxidant potential against oxidative stress-mediated urotoxicity in mice. Walnut is reported to possess antiproliferative activity. Its protective effect on CP-induced toxicity in bladder is a promising activity, which warrants possible clinical investigations on this medicinal plant.
