Polymethyl mehtacrylates polymers such as Eudragit RL100 have been used in preparation of matrix ... more Polymethyl mehtacrylates polymers such as Eudragit RL100 have been used in preparation of matrix type oral sustained release, coating and in microencapsulation of drug. Embedding of drug within an insoluble matrix is convenient way of controlling the drug release.The prepared microspheres by emulsion solvent evaporation technique was simple and reproducible, possessed good sphericity, smooth surface morphology, uniform and narrow size distribution (170-200 μm), when analyzed by scanning electron microscopy, and optical microscopy. Method of preparation has influenced the particle size and drug loading efficiency. The carrier ethyl cellulose and Eudragit RL100 are easily available and compatible. Drug-polymer compatibility was confirmed by Fourier transform infrared spectroscopy and DSC and X-ray diffraction studies revealed that the drug was dispersed inside the microspheres in the form of an insoluble matrix. To administer drug optimally, knowledge is needed not only of the mechanism of drug absorption, d... more To administer drug optimally, knowledge is needed not only of the mechanism of drug absorption, distribution and elimination but also of the kinetics of these processes, that is, pharmacokinetics. The application of pharmacokinetic principles of the therapeutic management of patients is clinical pharmacokinetics1. Pharmacokinetics and biotransformation of ambroxol chemically trans-4-(2-amino-3, 5-dibromo-benzylamino) cyclohexanol hydrochloride was studied by using increasing the number of rats or groups of rats. Absorption after oral administration was found to be fast and complete. Pharmacokinetic parameters in the blood were estimated by Standard chromatogram of Ambroxol by HPLC with electrochemical detector as 20--25 hrs in rat by statistical analysis. Ambroxol is an active substance with a long history that influences parameters considered to be the basis for the physiological production and the transport of the bronchial mucus. The applicability of gum karaya (GK), obtained from sterculia species was studied for its applica... more The applicability of gum karaya (GK), obtained from sterculia species was studied for its applicability in the design of colon drug delivery systems of 5fluorouracil (5-FU). Colon targeted tablets of 5 FU were prepared with varying drug-GK ratios and evaluated for the drug release in colonic pH environment. The optimum concentration of drug-GK was found to be 1:1.25 w/w ratio for extending the drug release around 24 hours. The effect of channeling agents was studied and observed that soluble channeling agent lactose markedly affected the drug release compared to insoluble channeling agents and nearly 90% drug release was observed in 24 hours. The optimized formulations were evaluated in dissolution medium containing rat caecal content and 100% drug release was observed over 24 hours. The formulations followed first order without channeling agent and in rat caecal content medium where as zero order was observed for tablets with lactose as channeling agent. The alginate/chitosan micro granules were prepared by the cross linking technique with calcium chloride(4%). Microscopy and Digital photography was used for morphology observation, which shows spherical shape but rough surface of the micro particles. Transmission infrared spectra of chitosan powder, 5-fluorouracil, sodium alginate pectin, and prepared microcapsules were acquired to draw information on the molecular state of chitosan and 5-fluorouracil. Differential scanning calorimetry (DSC) studies of 5-Fluorouracil, Chitosan, sodium alginate and pectin, were performed with PerkinElmer Thermal Analysis (Mettler Toledo 821Thermal analyzer) Calibrated with indium as standard.
