This chapter presents an evaluation of a program called "Student Voices", developed in ... more This chapter presents an evaluation of a program called "Student Voices", developed in 1999 as a joint effort by the Annenberg Public Policy Center and the Pew Charitable Trusts. "Student Voices" was an in-school intervention aimed at engaging high school seniors in the discourse and political activity surrounding the Philadelphia mayoral election. A key component of this program was providing students with access to the Internet and web-based information about politics, candidates, and the issues surrounding the campaign. Working within the school setting, students were encouraged to get to know the candidates, explore the issues, form opinions, find their voices, and ultimately participate in the voting process. A quasi-experiment was designed to evaluate the project. 419 students completed pretest and posttest questionnaires. The "Student Voices" program was successful in achieving many of its goals. The adenovirus (Adv) oncoprotein E1A stimulates cell proliferation and inhibits differentiation. ... more The adenovirus (Adv) oncoprotein E1A stimulates cell proliferation and inhibits differentiation. These activities are primarily linked to the N-terminal region (exon 1) of E1A, which interacts with multiple cellular protein complexes. The C terminus (exon 2) of E1A antagonizes these processes, mediated in part through interaction with C-terminal binding proteins 1 and 2 (CtBP1/2). To identify additional cellular E1A targets that are involved in the modulation of E1A C-terminus-mediated activities, we undertook tandem affinity purification of E1A-associated proteins. Through mass spectrometric analysis, we identified several known E1A-interacting proteins as well as novel E1A targets, such as the forkhead transcription factors, FOXK1/K2. We identified a Ser/Thr-containing sequence motif in E1A that mediated interaction with FOXK1/K2. We demonstrated that the E6 proteins of two beta-human papillomaviruses (HPV14 and HPV21) associated with epidermodysplasia verruciformis also interacted with FOXK1/K2 through a motif similar to that of E1A. The E1A mutants deficient in interaction with FOXK1/K2 induced enhanced cell proliferation and oncogenic transformation. The hypertransforming activity of the mutant E1A was suppressed by HPV21 E6. An E1A-E6 chimeric protein containing the Ser/Thr domain of the E6 protein in E1A interacted efficiently with FOXK1/K2 and inhibited cell transformation. Our results suggest that targeting FOXK1/K2 may be a common mechanism for certain beta-HPVs and Adv5. E1A exon 2 mutants deficient in interaction with the dual-specificity kinases DYRK1A/1B and their cofactor HAN11 also induced increased cell proliferation and transformation. Our results suggest that the E1A C-terminal region may suppress cell proliferation and oncogenic transformation through interaction with three different cellular protein complexes: FOXK1/K2, DYRK(1A/1B)/HAN11, and CtBP1/2.

Purpose: Using long-term gene-expression lentiviral (LV) vectors, we have previously demonstrated... more Purpose: Using long-term gene-expression lentiviral (LV) vectors, we have previously demonstrated that LV-IL-10 gene delivery through the recipient airway can attenuate the process of allograft airway obliteration in an intrapulmonary tracheal transplant model of obliterative bronchiolitis. The objective of this study is to explore the underlying immunological mechanism. Methods and Materials: A mouse intra-pulmonary tracheal transplant model (BALBc¡C57BL6) was used. Recipient mice received LV-huma-nIL-10 or LV-eGFP through the airway at days Ϫ9, Ϫ8, and Ϫ7 and then transplantation was conducted at day 0. Animals were sacrificed at day 28 and 42 (n ϭ 5/group at each time point). Allograft obliteration was morphometrically quantified. Gene transduction and expression of markers for classical and alternative macrophage activation were immunohistochemically examined. Results: Human IL-10 expression in the LV-IL-10 group was confirmed by ELSIA at day 42 (average 11.8 pg/mg lung tissue protein). eGFP transduction was observed mainly in mac-3ϩ macrophages (M⌽, 70% among all eGFP positive cells). Histologically, at post-operative day 28, all the groups showed near-complete obliteration of the lumen (83.0Ϯ6.7 (meanϮSEM) vs 82.7Ϯ7.9%). However, interestingly, humanIL-10 significantly reduced luminal occulusion by day 42 compared with the eGFP group (62.9Ϯ4.9 vs 91.6Ϯ5.3% (eGFP), pϽ0.05). The IL-10 group showed a trend of decreased cells positive for iNOS (a classical M⌽ activation marker, 1.9Ϯ0.4 vs 2.3Ϯ0.4 (eGFP), pϭ0.036) and significantly decreased cells positive for arginase-1 (an alternative M⌽ activation marker, 1.0Ϯ0.1 vs 2.3Ϯ0.5, pϭ0.009) in the graft lumen. Conclusions: LV-IL-10-mediated gene therapy attenuates allograft airway fibrosis. Down-regulation of both classical and alternative M⌽ activation pathways appear to be mechanistically implicated in this process.

In 2 studies, the authors explored preadolescent and adolescent use of personal homepages in rela... more In 2 studies, the authors explored preadolescent and adolescent use of personal homepages in relation to mastery and identity formation. In Study 1, the authors attempted to determine the prevalence of personal homepage and online journal (blog) construction among a random sample (N ϭ 500) of preadolescents and adolescents. Adolescents were more likely to create personal homepages or blogs than preadolescents. Creation was related to feelings of mastery, expressions of identity, and a means to socialize. In Study 2, the authors explored the relationship of homepages to mastery and identity formation by content analysis of a random sample of homepages. Results suggest children use personal homepages to express and explore their forming identities.
