The effects of pinealectomy on the dynamics of morphometric changes in interdigitating cells(IDC)... more The effects of pinealectomy on the dynamics of morphometric changes in interdigitating cells(IDC) of rat thymus were investigated. The experimental animals were divided into two groups: an experimental one and the control group. Animals from the first group were subjected to pinealectomy while the second group were treated in the same manner, but without the removal of the pineal gland. All the animals were sacrificed 60. days following the surgery. The thymus tissue were fixed by means of immersion and prepared for transmission electron microscopy. Using Weibl's multipurpose test system and multilavel sampling technique on electron micrographs, the nuclear volume density(Vvn) and cytoplasmic volume density(Vvc) were calculated. Different physiological states have been compared and tahat pre- and postovulatory. Volume density (VV), surface density (SV) and specific surface density (SV/VV) of nuclei and epithelial cells cytoplasm have been stereologically examined and compared. Postovulatory, the volume density (VV) and surface density (SV) of nuclei as well as specific surface density (SV/VV) of epithelial cells cytoplasm are much less, while a volume density (VV) of epithelial cells cytoplasm is greater.

The mammary glands of primigravid Wistar rats were investigated by stereological analysis in cond... more The mammary glands of primigravid Wistar rats were investigated by stereological analysis in conditions of lead acetate administration via drinking water. The experiment was made in early summer period when absorbing ability of rat intestinum for lead is highest. Morphological state of glands was observed on 7th, 14th and 21st day of pregnancy. Groups of control animals were drinking deionized water but groups of experimental animals were drinking deionized water with added lead acetate. On paraffin sections coloured by HE method, volume density (VV) of glands structural elements were determined by multipurpose test system M42. Stereological analysis shows that lead diminishes the proliferative capacity of the mammary parenchyma and changes the morphofunctional differentiation of the overall structure in mammary gland during pregnancy. This article presents results of stereological ultrastructure parameters of thymuscortical and medullar epithelial cells of male and female rats two month after pinealectomy. Methods: The study was retrospective, comparative, and data were obtained at the Clinic of Obstetrics and Gynecology of the University Clinical Centre Sarajevo for the period from October 2013 through February 2014. The sample was divided into two groups based on the timing of delivery: preterm delivery group (before 37th week of pregnancy) and term delivery group (after 37th week of pregnancy). The standard methods of descriptive statistics and the univariate analysis were performed. Nuclear and cytoplasm surface of Leydig cells in sham pinealectomized and pinealectomized adult Wistar rats was determined after their total irradiation with 8 Gy of gama rays produced by 60 Co and with previously performed 14 days treatment by injecting of melatonin solution in a daily dose of 0.2 mg. Rats testes were histologically prepared 17 days after irradiation. Total melatonin administration significantly decreased surface of Leydig cells including both decrease of their nuclear and cytoplasm surface in irradiated sham pinealectomized and pinealectomized rats. The results suggested about direct effects of exogenous melatonin on Leydig cells in irradiation stress.

It competitively binds to estrogen receptors and generates oxidative stress. Ultrastructural characteristics of ductular parenchyma epithelial and myoepithelial cells were an... more Ultrastructural characteristics of ductular parenchyma epithelial and myoepithelial cells were analysed in 18 cases of normal female breast tissue. Different physiological states have been compared and that pre- and postmenopausal. Nuclei and cytoplasm of epithelial and myoepithelial cells have been examined and compared. There were obvious signs of secretion and that apical protein granules, "dense-core" granules and light vesicles and lipid vacuoles and glycogen granules were accumulated in the deeper parts of premenopausal breast epithelial cells cytoplasm. Morphologic equivalents of secretion and accumulated glycogen were presented in the postmenopausal breast epithelial cells. Ultrastructural morphologic characteristics of postmenopausal myoepithelial cells were not different from those in premenopausal breasts.

Retinal melatonin levels show a marked daily variation, with high levels at night and low levels during the light period. Melatonin rhythm generating system includes photodetector, circadian clock and melatonin synthesis machinery and they are located in pincal organ and retina. In both, light regulates daily variations in melatonin secretion by controlling the activity of arylalkylamine N-acetyltransferase (AANAT). In mammals, circadian system is comprised of three major components: the eyes, the hypothalamic suprachiasmatic nucleus (SCN) and the pincal gland. The SCN harbours the endogenous oscillator that is entrained every day to the ambient lighting conditions via retinal input. The aim of our study was to quantitatively analyse morphological parameters of the rat telencephalon during embryonic development. The investigation was carried out on semithin serial sections of rat brain from embryonic days 12 to 15. The volume densities (VV) of the lateral ventricles, the telencephalic neuroepithelium and the surrounding mesenchyme have been analysed stereologically and compared in examined embryonic stages. The neuroepithelial volume density was the smallest (28%) at E13 and the biggest (44%) at E15 (p<0.0005). The mesenchymal volume density was the smallest (32%) at E13 and the biggest (48%) at E14 (p<0.0005). Objectives: The aim of this study was to investigate the effects of pinealectomy and melatonin tr... more Objectives: The aim of this study was to investigate the effects of pinealectomy and melatonin treatment on the rat thymus gland characteristics, taking into consideration possible gender differences. Materials and methods: Thirty adult Wistar rats of both sexes were divided into three groups. Group C and group PX served as control groups and included sham-pinealectomized and pinealectomized animals that were treated with 10% ethanol solution (0,1ml/ daily, subcutaneous). Animals from third group (group PXM) underwent pinealectomy and seven days after surgery started receiving melatonin dissolved in 10% ethanol solution (3mg/ kg/daily, subcutaneous). All animals were treated for 4 weeks. Results: Volume density of the thymus cortex showed statistically significant (p<0,05) decrease while the volume density of the thymus medulla was increased in the pinealectomized compared to the sham-pinealectomized female rats. Numerical density of macrophages as well as the distribution of blood vessels showed no gender differences. The numerical density of lymphocytes was statistically significantly decreased in female in comparison to the male pinealectomized rats. Melatonin treatment was proved to cause reverse effects in the sense that the results from the melatonin treated group corresponded to the results obtained from the control group of animals. Conclusion: The results of this study suggest that the pinealectomy causes gender-related changes in the rat thymus. Short-term melatonin treatment showed reverse effect, equally in both sexes.

Introduction: Hydatidiform moles (HM), presenting as complete (CHM) and partial (PHM) form, are r... more Introduction: Hydatidiform moles (HM), presenting as complete (CHM) and partial (PHM) form, are rare pregnancy disorder. Diagnosis is based on clinical presentation, ultrasound imaging fi ndings and pathological examination of products of conception. Protein p57, encoded by CKDN1C gene, is paternally imprinted and maternally expressed gene and provides quick insight in genetic basis of HM and allows distinction of CHM from all other conceptions. compare the preevacuational and pathohistological diagnosis with outcome of p57 immunostaining. Material and methods: All cases of HM diagnosed between January 2011 and December 2015 were included in this research. Maternal age, gestational age and input diagnosis data were recored. p57 immunostaining was performed in order to evaluate the diagnosis based on tissue slides examination. Results: There were 198 cases of histologically confi rmed HM, 185 PHM, 12 CHM and one case of undefi ned HM. Mean maternal age in the CHM group was 24,7 and in the PHM group 26,9 years, with no signifi cant diff erences among these two groups (p=0,27). For CHM mean gestational age was estimated at eight and for PHM 9,2 gestational weeks. Pregnant woman older than 40 years present signifi cant earlier compared with younger woman (p<0,01), and those younger than 20 years tend to present at the beginning of the second trimester more often than older women (pα0,05). In the CHM group, 9 (75%) input diagnoses were mola in obs, and 3 (25%) of them were signed as abortion, unlike the PHM where 126 (67%) were qualifi ed as abortion, 35 (19%) as blighted ovum, and 26 (14%) were suggestive for molar pregnancy. p57 immunostaining results confi rmed all pathohistological diagnosis of CHM whereas 8% of PHM demonstrated divergent p57 expression. Conclusion: PHM, compared with CHM, represent a greater diagnostic challenge for both gynecologist and pathologist even when presenting in more advanced pregnancies.

Objectives: Two main and the most common types of congenital abdominal wall defects are omphaloce... more Objectives: Two main and the most common types of congenital abdominal wall defects are omphalocele and gastroschisis. According to the literature, the frequency of omphalocele is generally stable, the rate of gastroschisis has increased lately, and cloacal exstrophy is a very rare birth defect. Methods: A retrospective study was performed on the basis of clinical records for the period January 2000. to December 2008. and the standard methods of descriptive statistics were performed for the data analysis. Investigation was carried out to obtain the frequency and gender distribution of congenital abdominal wall defects among patients surgically treated in the Clinic for Children's Surgery, Clinical Center University of Sarajevo, Bosnia and Herzegovina. Results: A register-based data show that total of 26 abdominal wall defect cases were surgically treated in the investigated period. Out of that number 15 (60%) were male patients, while 11 (40%) were female patients; sex ratio-1.5:1. Twenty patients had gastroschisis, five omphalocele and one patient were with cloacal exstrophy. Anomalies associated with abdominal wall defects were present in total of six patients (23%), four male (15%) and two female (8%) patients. Conclusion: The study allows the conclusion that abdominal wall defects are generally rare congenital anomalies of which cloacal exstrophy is the rarest, the gastroschisis is on the rising trend and omphalocele is generally stable.

Seven days after the application of 2% lidocaine (4 ml doze, the speed of application 3 ml/min, m... more Seven days after the application of 2% lidocaine (4 ml doze, the speed of application 3 ml/min, mean application pressure 15 +/- 2.47 kPa) into mesoneurium of adult rabbit sciatic nerve, a nerve segment was intravitally excided from the injection site with a length of 1 cm and qualitatively and quantitatively analyzed. Histological changes at the level of total nerve structures were found, especially of its sheaths. Epi- and interfascicular epineurium were thickened with a hipercellularity of a mononuclear inflammatory process type. On the side of application, occasional delamination of perineurium was registered. Myelinated fibers were sporadically demyelinated, followed by axon degeneration which, in thick fibers, reached a full disintegration. In general, the damages were more severe at the side of application although some zonal differences in the degree of damage (above, below and at the site of application) within individual fasciculi were expressed. A significantly higher volume density of epifascicular and interfascicular connective tissue in comparison to the control, untreted nerves, observed by the stereological analysis, may be considered as an inflammatory-repairing reaction which we identified microscopically. At the same time, we found a significantly higher volume density of endoneurium in lidocaine treated nerves.

Background: the most widely used tumor marker in ovarian cancer, often considered the 'gold stand... more Background: the most widely used tumor marker in ovarian cancer, often considered the 'gold standard' is CA125 but reliable clinical evidence demonstrates that human epididymis protein (HE4), used alone or in combination with CA125, substantially improves the accuracy of screening and/or disease monitoring. Aim: to evaluate the reliability of the determination a tumor marker HE4 in comparison with CA125 on the Elecsys analyzer 2010 in epithelial ovarian cancer, benign ovarian cyst and healthy controls. Methods: we prospectively determined CA125 and HE4 serum levels in the Biochemical-Immunological-Haematological "Medical Laboratory" Ilidza, Sarajevo, B&H between June 1 st and December 31 st 2011. Electro-chemiluminescence immunoassay (ECLIA) methods for quantitative determination in vitro were performed on the Roche/Hitachi Elecsys 2010 Immunoassay Analyzer. Standard methods of descriptive statistics were performed for the data analysis. Results: univariate statistical analyze of tumor marker control serum revealed a high reliability for both CA125 and HE4 determination (p>0.05). Levey-Jennings charts of quality control data show that the target and the obtained values of both markers control sera do not differ significantly in relation to the ideal value. In a total number of 60 patients compared values of tumor markers show a high correlation (r=0.85). This study confirmed higher sensitivity and specificity of HE4 tumor marker compared with CA125. ROC-AUC values show that the diagnostic performance of HE4 was significantly higher compared with CA125. Conclusion: We concluded that HE4 was better than CA125 as a single tumor marker.

In Bosnia and Herzegovina there is no existent unique evidence of congenital anomalies and registries. The aim of this study was to obtain the frequency of different urinary tract anomalies types and their sex distribution among cases hospitalized in the Department of Pediatric Surgery of the University of Sarajevo Clinics Centre, Bosnia and Herzegovina, during the period from January 2002 to December 2006. Retrospective study was carried out on the basis of clinical records. Standard methods of descriptive statistics were performed for the data analysis. Among 289 patients that were surgically treated 62,37% of the patients were male patients, while 37,63% were female patients. The prevalence of these anomalies shows considerable geographical variation and female predominance. The aim of this work was to obtain the frequency of different CNS congenital anomalies types and their sex distribution among cases hospitalized in a Department of Neurosurgery, University of Sarajevo Clinics Center, Bosnia and Herzegovina, during the period January 2001 to December 2004. Retrospective study was carried out on the basis of the clinical records. Standard methods of descriptive statistics were performed for the data analysis. A total of 103 cases were surgically treated in the period from 2001 through 2004. Out of that number 56 (54.4%) were female patients, while 47 (46.6%) were male patients. Seven different CNS birth defect types were found in this investigation. The experimental animals were divided into two groups: an experimental one and the control group. The experiment was made in early summer period when absorbing ability of rat intestinum for lead is highest. Morphological state of glands was observed on 7th, 14th and 21st day of pregnancy. Groups of control animals were drinking deionized water but groups of experimental animals were drinking deionized water with added lead acetate. On paraffin sections coloured by HE method, volume density (VV) of glands structural elements were determined by multipurpose test system M42. Stereological analysis shows that lead diminishes the proliferative capacity of the mammary parenchyma and changes the morphofunctional differentiation of the overall structure in mammary gland during pregnancy. Nuclear and cytoplasm surface of Leydig cells in sham pinealectomized and pinealectomized adult Wistar rats was determined after their total irradiation with 8 Gy of gama rays produced by 60 Co and with previously performed 14 days treatment by injecting of melatonin solution in a daily dose of 0.2 mg. Rats testes were histologically prepared 17 days after irradiation. Total melatonin administration significantly decreased surface of Leydig cells including both decrease of their nuclear and cytoplasm surface in irradiated sham pinealectomized and pinealectomized rats. The results suggested about direct effects of exogenous melatonin on Leydig cells in irradiation stress.

Different physiological states have been compared and that pre- and postmenopausal. Nuclei and cytoplasm of epithelial and myoepithelial cells have been examined and compared. There were obvious signs of secretion and that apical protein granules, "dense-core" granules and light vesicles and lipid vacuoles and glycogen granules were accumulated in the deeper parts of premenopausal breast epithelial cells cytoplasm. Morphologic equivalents of secretion and accumulated glycogen were presented in the postmenopausal breast epithelial cells. Ultrastructural morphologic characteristics of postmenopausal myoepithelial cells were not different from those in premenopausal breasts.

In both, light regulates daily variations in melatonin secretion by controlling the activity of arylalkylamine N-acetyltransferase (AANAT). The aim of our study was to quantitatively analyse morphological parameters of the rat telencephalon during embryonic development. The investigation was carried out on semithin serial sections of rat brain from embryonic days 12 to 15. The volume densities (VV) of the lateral ventricles, the telencephalic neuroepithelium and the surrounding mesenchyme have been analysed stereologically and compared in examined embryonic stages. The neuroepithelial volume density was the smallest (28%) at E13 and the biggest (44%) at E15 (p<0.0005). Objectives: The aim of this study was to investigate the effects of pinealectomy and melatonin tr... more Objectives: The aim of this study was to investigate the effects of pinealectomy and melatonin treatment on the rat thymus gland characteristics, taking into consideration possible gender differences. Materials and methods: Thirty adult Wistar rats of both sexes were divided into three groups. Group C and group PX served as control groups and included sham-pinealectomized and pinealectomized animals that were treated with 10% ethanol solution (0,1ml/ daily, subcutaneous). Animals from third group (group PXM) underwent pinealectomy and seven days after surgery started receiving melatonin dissolved in 10% ethanol solution (3mg/ kg/daily, subcutaneous). All animals were treated for 4 weeks. Results: Volume density of the thymus cortex showed statistically significant (p<0,05) decrease while the volume density of the thymus medulla was increased in the pinealectomized compared to the sham-pinealectomized female rats. Numerical density of macrophages as well as the distribution of blood vessels showed no gender differences. The numerical density of lymphocytes was statistically significantly decreased in female in comparison to the male pinealectomized rats. Melatonin treatment was proved to cause reverse effects in the sense that the results from the melatonin treated group corresponded to the results obtained from the control group of animals. Conclusion: The results of this study suggest that the pinealectomy causes gender-related changes in the rat thymus. Short-term melatonin treatment showed reverse effect, equally in both sexes.

Introduction: Hydatidiform moles (HM), presenting as complete (CHM) and partial (PHM) form, are r... more Introduction: Hydatidiform moles (HM), presenting as complete (CHM) and partial (PHM) form, are rare pregnancy disorder. Diagnosis is based on clinical presentation, ultrasound imaging fi ndings and pathological examination of products of conception. Protein p57, encoded by CKDN1C gene, is paternally imprinted and maternally expressed gene and provides quick insight in genetic basis of HM and allows distinction of CHM from all other conceptions. compare the preevacuational and pathohistological diagnosis with outcome of p57 immunostaining. Material and methods: All cases of HM diagnosed between January 2011 and December 2015 were included in this research. Maternal age, gestational age and input diagnosis data were recored. p57 immunostaining was performed in order to evaluate the diagnosis based on tissue slides examination. Results: There were 198 cases of histologically confi rmed HM, 185 PHM, 12 CHM and one case of undefi ned HM. Mean maternal age in the CHM group was 24,7 and in the PHM group 26,9 years, with no signifi cant diff erences among these two groups (p=0,27). For CHM mean gestational age was estimated at eight and for PHM 9,2 gestational weeks. Pregnant woman older than 40 years present signifi cant earlier compared with younger woman (p<0,01), and those younger than 20 years tend to present at the beginning of the second trimester more often than older women (pα0,05). In the CHM group, 9 (75%) input diagnoses were mola in obs, and 3 (25%) of them were signed as abortion, unlike the PHM where 126 (67%) were qualifi ed as abortion, 35 (19%) as blighted ovum, and 26 (14%) were suggestive for molar pregnancy. p57 immunostaining results confi rmed all pathohistological diagnosis of CHM whereas 8% of PHM demonstrated divergent p57 expression. Conclusion: PHM, compared with CHM, represent a greater diagnostic challenge for both gynecologist and pathologist even when presenting in more advanced pregnancies.

According to the literature, the frequency of omphalocele is generally stable, the rate of gastroschisis has increased lately, and cloacal exstrophy is a very rare birth defect. Methods: A retrospective study was performed on the basis of clinical records for the period January 2000. to December 2008. and the standard methods of descriptive statistics were performed for the data analysis. Investigation was carried out to obtain the frequency and gender distribution of congenital abdominal wall defects among patients surgically treated in the Clinic for Children's Surgery, Clinical Center University of Sarajevo, Bosnia and Herzegovina. Results: A register-based data show that total of 26 abdominal wall defect cases were surgically treated in the investigated period. Out of that number 15 (60%) were male patients, while 11 (40%) were female patients; sex ratio-1.5:1. Twenty patients had gastroschisis, five omphalocele and one patient were with cloacal exstrophy. Anomalies associated with abdominal wall defects were present in total of six patients (23%), four male (15%) and two female (8%) patients. Conclusion: The study allows the conclusion that abdominal wall defects are generally rare congenital anomalies of which cloacal exstrophy is the rarest, the gastroschisis is on the rising trend and omphalocele is generally stable.

Seven days after the application of 2% lidocaine (4 ml doze, the speed of application 3 ml/min, m... more Seven days after the application of 2% lidocaine (4 ml doze, the speed of application 3 ml/min, mean application pressure 15 +/- 2.47 kPa) into mesoneurium of adult rabbit sciatic nerve, a nerve segment was intravitally excided from the injection site with a length of 1 cm and qualitatively and quantitatively analyzed. Histological changes at the level of total nerve structures were found, especially of its sheaths. Epi- and interfascicular epineurium were thickened with a hipercellularity of a mononuclear inflammatory process type. On the side of application, occasional delamination of perineurium was registered. Myelinated fibers were sporadically demyelinated, followed by axon degeneration which, in thick fibers, reached a full disintegration. In general, the damages were more severe at the side of application although some zonal differences in the degree of damage (above, below and at the site of application) within individual fasciculi were expressed. A significantly higher volume density of epifascicular and interfascicular connective tissue in comparison to the control, untreted nerves, observed by the stereological analysis, may be considered as an inflammatory-repairing reaction which we identified microscopically. At the same time, we found a significantly higher volume density of endoneurium in lidocaine treated nerves.

Background: the most widely used tumor marker in ovarian cancer, often considered the 'gold stand... more Background: the most widely used tumor marker in ovarian cancer, often considered the 'gold standard' is CA125 but reliable clinical evidence demonstrates that human epididymis protein (HE4), used alone or in combination with CA125, substantially improves the accuracy of screening and/or disease monitoring. Aim: to evaluate the reliability of the determination a tumor marker HE4 in comparison with CA125 on the Elecsys analyzer 2010 in epithelial ovarian cancer, benign ovarian cyst and healthy controls. Methods: we prospectively determined CA125 and HE4 serum levels in the Biochemical-Immunological-Haematological "Medical Laboratory" Ilidza, Sarajevo, B&H between June 1 st and December 31 st 2011. Electro-chemiluminescence immunoassay (ECLIA) methods for quantitative determination in vitro were performed on the Roche/Hitachi Elecsys 2010 Immunoassay Analyzer. Standard methods of descriptive statistics were performed for the data analysis. Results: univariate statistical analyze of tumor marker control serum revealed a high reliability for both CA125 and HE4 determination (p>0.05). Levey-Jennings charts of quality control data show that the target and the obtained values of both markers control sera do not differ significantly in relation to the ideal value. In a total number of 60 patients compared values of tumor markers show a high correlation (r=0.85). This study confirmed higher sensitivity and specificity of HE4 tumor marker compared with CA125. ROC-AUC values show that the diagnostic performance of HE4 was significantly higher compared with CA125. Conclusion: We concluded that HE4 was better than CA125 as a single tumor marker.

A total of 103 cases were surgically treated in the period from 2001 through 2004. Out of that number 56 (54.4%) were female patients, while 47 (46.6%) were male patients.
