Chrysanthemum indicum Linne flower (CF) and Cinnamomum cassia (L.) J. Presl bark (CB) extracts ha... more Chrysanthemum indicum Linne flower (CF) and Cinnamomum cassia (L.) J. Presl bark (CB) extracts have been used as the main ingredients in several prescriptions to treat the hyperuricemia and gout in traditional medicine. In the present study, we investigated the antihyperuricemic effects of DKB114, a CF, and CB mixture, and the underlying mechanisms in vitro and in vivo. DKB114 markedly reduced serum uric acid levels in normal rats and rats with PO-induced hyperuricemia, while increasing renal uric acid excretion. Furthermore, it inhibited the activity of xanthine oxidase (XOD) in vitro and in the liver in addition to reducing hepatic uric acid production. DKB114 decreased cellular uric acid uptake in oocytes and HEK293 cells expressing human urate transporter (hURAT)1 and decreased the protein expression levels of urate transporters, URAT1, and glucose transporter, GLUT9, associated with the reabsorption of uric acid in the kidney. However, data on the mechanism of torilin action against inflammation is limited. This study aimed at determining the anti-inflammatory property of torilin in LPS-induced inflammation using in vitro model of inflammation. We examined torilin's effect on expression levels of inflammatory mediators and cytokines in LPS-stimulated RAW 264.7 macrophages. The involvement of NF-kB and AP-1, MAP kinases, and adaptor proteins were assessed. Torilin strongly inhibited LPS-induced NO release, iNOS, PGE2, COX-2, NF-α, IL-1β, IL-6, and GM-CSF gene and protein expressions. In addition, MAPKs were also suppressed by torilin pretreatment. In this study, we investigated the effect of bamboo, Phyllostachys pubescens, leaves extract (PPE) on human 5α-reductase type 2 (SRD5A2) gene promoter activity in human prostate cell lines and the protective effect of PPE on a testosterone-induced BPH rat model. PPE repressed human SRD5A2 promoter activity and its mRNA expression. The rats treated with PPE for 4 weeks showed a significantly attenuated prostate weight compared to vehicle control. PPE-treated rats also showed reduced serum dihydrotestosterone, testosterone, prostate-specific antigen, and SRD5A2 levels by testosterone injection. The variety in human skin hues is ascribed to the characteristic color called Melanin. However, Melanin overproduction due to UVB beams promotes skin staining and tumorigenesis, a process called photo aging, which damages skin quality. To assess the effects of Korean Red Ginseng Oil (KGO) on photo aging, the murine melanoma cell lines B16/F10 were used in vitro and HRM-2 hairless mice exposed to UVB were studied in vivo. However, its effects on osteoarthritis (OA) and its regulatory mechanisms have not been investigated by network analysis. Here, we investigated the pharmacological effects of Peucedanum japonicum extract (PJE) on OA, by combining in vivo effective verification and network pharmacology prediction. Rats in which OA was induced by monosodium iodoacetate (MIA) were treated with PJE (200 mg/kg), and histopathological parameters, weight bearing distribution and inflammatory factors in serum and joint tissue were measured after 28 days of treatment. Additionally, in silico network analysis was used to predict holistic OA regulatory mechanisms of PJE. The results showed that PJE exerted potential protective effects by recovering hind paw weight bearing distribution, alleviating histopathological features of cartilage and inhibiting inflammatory mediator levels in the OA rat model. Furthermore, network analysis identified caspase-3 (CASP3), caspase-7 (CASP7), and cytochrome P450 2D6 (CYP2D6) as potential target genes; in addition, the TNF (Tumor necrosis factor) signaling pathway was linked to OA therapeutic action. Our combined animal OA model and network analysis confirmed the therapeutic effects of PJE against OA and identified intracellular signaling pathways, active compounds and target genes linked to its therapeutic action.

Objectives : Anti-obesity drugs that have been developed so far have limited efficacies and consi... more Objectives : Anti-obesity drugs that have been developed so far have limited efficacies and considerable adverse effects affecting tolerability and safety. Therefore, most anti-obesity durgs have been withdrawn. We tried to develop anti-obesity agent by combinations from herbs that are used in food ingredients as well as in traditional medicines. Methods : The 80% (v/v) ethanol extracts from Bamboo (Phyllostachys pubescence) leaf (BL) and Scutellaria baicalensis (SB) and their 1:1 combination (BLSB) was evaluated on high fat diet induced obese mice compared to Omega-3 as a positive control. The mice were divided into six groups (n=5), one group fed a normal diet (ND), and the others fed a high fat diets for eight weeks. Two weeks after starting feeding the diets, the high fat diet groups were orally administered vehicle and Omega-3, BL, SB, and BLSB at dosage of 200 mg/kg/day for six weeks. All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : BLSB group showed significant reductions in body weight gain and fat weights of liver and epididymal adipose tissue compared to BL or SB alone as well as control. Total-cholesterol and LDL-cholesterol levels significantly decreased, and HDL-cholesterol level increased. In liver tissue, macrovesicular steaotisis was remarkably improved and its fat cell size was also significantly decreased. Conclusions : These results suggested that a combination preparation of bamboo leaf and S. baicalensis has anti-obesity effect and have synergistic effect compared to bamboo leaf or S. baicalesis.

In the current study, we investigated whether Brassica campestris var narinosa, Canavalia gladiata DC semen and their combinational prescription is related to hemato-potentiating function using Sca-1 + hematopoietic stem cells (Sca-1 + HSCs) as a testing system. Methods : Sca-1 + HSCs isolated from femur in C57bl/6 mice with leukopenia and thrombocytopenia induced by cyclophosphamide (CTX). Then, Real-time PCR was performed to measure the mRNA expression, ELISA and haematopoiesis-related gene (EPO, TPO, IL-3, SCF, c-kit, GM-CSF), the phosphorylation of JAK2, GATA-1 and STAT-5a/b were observed by western blot, and the numbers of CD117 + /Sca-1 + cell and the number of granulocyte erythrocyte monocyte macrophage colony-forming units (CFU-GEMM) and erythroid burst forming units (BFU-E), semisolid clonogenic assay was performed. Result : When Sca-1 + HSCs were treated with Brassica campestris var narinosa, Canavalia gladiata DC semen and their combinational prescription with rIL-3/rSCF, the expression of haematopoiesis-related (EPO, TPO, IL-3, SCF, c-kit, and GM-CSF) were significantly increased at the levels of mRNA as well as production in Sca-1 + HSCs. The present study was designed to test the hypothesis that low doses of two different V. mandshurica extracts have anti-obesity effects. We evaluated the effects of ethanol extract (VME) and aqueous extract (VMA) from V. mandshurica on high-fat diet (HFD)-induced obese mice as well as the acute oral toxicities and chemical compositions of both extracts. Oral administration of VME or VMA (50, 100, or 200 mg/kg) decreased body weight gain, liver and adipose tissue mass, adipocyte size, and serum lipid levels. Both extracts increased adiponectin serum concentrations and mRNA expression in epididymal adipose tissue. Oral treatment of AFB in obese mice reduced body weight, lipid accumulation in liver and adipose tissue as well as adipocyte size, compared to the high fat diet control mice. AFB also decreased serum triacylglycerol, glucose, and insulin concentrations, with increased HDL-cholesterol and adiponectin levels. Furthermore, 5% AFB markedly increased mRNA expression of peroxisome proliferators activated receptor-c, uncoupling protein-2 as well as b3-adrenoreceptor in the visceral adipose tissue. These results demonstrate that the cereal bar with A. fistulosum extract may potentially serve as nutraceutical for improvement of obesity and metabolic disorders.

However, its effects on osteoarthritis (OA) and its regulatory mechanisms have not been investigated by network analysis. Here, we investigated the pharmacological effects of Peucedanum japonicum extract (PJE) on OA, by combining in vivo effective verification and network pharmacology prediction. Rats in which OA was induced by monosodium iodoacetate (MIA) were treated with PJE (200 mg/kg), and histopathological parameters, weight bearing distribution and inflammatory factors in serum and joint tissue were measured after 28 days of treatment. Additionally, in silico network analysis was used to predict holistic OA regulatory mechanisms of PJE. The results showed that PJE exerted potential protective effects by recovering hind paw weight bearing distribution, alleviating histopathological features of cartilage and inhibiting inflammatory mediator levels in the OA rat model. Furthermore, network analysis identified caspase-3 (CASP3), caspase-7 (CASP7), and cytochrome P450 2D6 (CYP2D6) as potential target genes; in addition, the TNF (Tumor necrosis factor) signaling pathway was linked to OA therapeutic action. Our combined animal OA model and network analysis confirmed the therapeutic effects of PJE against OA and identified intracellular signaling pathways, active compounds and target genes linked to its therapeutic action.

Objectives : Anti-obesity drugs that have been developed so far have limited efficacies and consi... more Objectives : Anti-obesity drugs that have been developed so far have limited efficacies and considerable adverse effects affecting tolerability and safety. Therefore, most anti-obesity durgs have been withdrawn. We tried to develop anti-obesity agent by combinations from herbs that are used in food ingredients as well as in traditional medicines. Methods : The 80% (v/v) ethanol extracts from Bamboo (Phyllostachys pubescence) leaf (BL) and Scutellaria baicalensis (SB) and their 1:1 combination (BLSB) was evaluated on high fat diet induced obese mice compared to Omega-3 as a positive control. The mice were divided into six groups (n=5), one group fed a normal diet (ND), and the others fed a high fat diets for eight weeks. Two weeks after starting feeding the diets, the high fat diet groups were orally administered vehicle and Omega-3, BL, SB, and BLSB at dosage of 200 mg/kg/day for six weeks. All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : BLSB group showed significant reductions in body weight gain and fat weights of liver and epididymal adipose tissue compared to BL or SB alone as well as control. Total-cholesterol and LDL-cholesterol levels significantly decreased, and HDL-cholesterol level increased. In liver tissue, macrovesicular steaotisis was remarkably improved and its fat cell size was also significantly decreased. Conclusions : These results suggested that a combination preparation of bamboo leaf and S. baicalensis has anti-obesity effect and have synergistic effect compared to bamboo leaf or S. baicalesis.

The nutritional composition and anti-obesity activities of cereal bars containing Allium fistulos... more The nutritional composition and anti-obesity activities of cereal bars containing Allium fistulosum extract (AFB) in high-fat diet-induced obese mice was determined. AFB is rich in vitamins C, B 2 , and B 3 , and B 9 with high protein. Oral treatment of AFB in obese mice reduced body weight, lipid accumulation in liver and adipose tissue as well as adipocyte size, compared to the high fat diet control mice. AFB also decreased serum triacylglycerol, glucose, and insulin concentrations, with increased HDL-cholesterol and adiponectin levels. Furthermore, 5% AFB markedly increased mRNA expression of peroxisome proliferators activated receptor-c, uncoupling protein-2 as well as b3-adrenoreceptor in the visceral adipose tissue. These results demonstrate that the cereal bar with A. fistulosum extract may potentially serve as nutraceutical for improvement of obesity and metabolic disorders.
