Bergia suffruticosa (Elatinaceae family) is an important Indian medicinal plant hitherto unexplor... more Bergia suffruticosa (Elatinaceae family) is an important Indian medicinal plant hitherto unexplored for its chemical constituents and its pharmacological activity. In the present paper we report our work on isolation of gallicin, gallic acid, lupeol and β-sitosterol from the plant. To the best of our knowledge, this is the first report of these four compounds from this plant. Further, these four marker compounds were quantified by thin layer chromatography densitometric methods using high performance thin layer chromatography. The thin layer chromatography densitometric methods were found to be precise with RSD for intra-day in the range of 0.61–1.83, 1.14–1.57, 0.38–0.52 and 0.15–0.52 and for inter-day in the range of 0.97–1.45, 0.58–1.27, 0.42–0.50 and 0.26–0.61 for different concentrations of gallicin, gallic acid, lupeol and β-sitosterol. Instrumental precision was 1.05, 1.20, 0.65 and 0.85 (% RSD) for gallicin, gallic acid, lupeol and β-sitosterol. Accuracy of the method was checked by conducting recovery studies at three different levels for the four compounds and the average percentage recoveries obtained were 99.89, 100.58, 99.79 and 100.11%. B. suffruticosa sample was found to contain 0.34% w/w of gallicin, 0.288% w/w of gallic acid, 0.064% w/w of lupeol and 0.034% w/w of β-sitosterol.

Leaf of Adhatoda vasica (Vasaka) is an important drug of Ayurveda, prescribed as an expectorant. ... more Leaf of Adhatoda vasica (Vasaka) is an important drug of Ayurveda, prescribed as an expectorant. Quinazoline alkaloids present in the leaves are established as active principles. In Ayurveda, its leaf juice (Vasa swarasa) is incorporated in many formulations. Classical method for extracting the juice (swarasa) from the leaf is an elaborate process, which involves subjecting a bolus of crushed fresh leaf to heat followed by squeezing out the juice. Commercially, to prepare the juice of Vasaka, manufacturers have been adopting different methods other than the traditional method. In an effort to evaluate these modified processes phytochemically to identify the process which gives juice of the quality that is obtained by traditional method, in terms of its alkaloid content, we prepared the leaf juice by traditional Ayurvedic method, its modification by steaming of leaf to simulate the traditional method and other methods adopted by some manufacturers. These juice samples were evaluated for the total alkaloid content by spectrophotometric method and vasicine content by thin layer chromatography densitometric method using high performance thin layer chromatography. The high performance thin layer chromatography method was validated for precision, repeatability and accuracy. The total alkaloid content varied from 0.3 mg/ml to 5.93 mg/ml and that of vasicine content varied from 0.2 mg/ml to 5.64 mg/ml in the juice samples prepared by different methods. The present study revealed that steaming of fresh leaves under 15 lb pressure yielded same quantity of juice as the traditional bolus method (25 ml/100 g leaf) and its total alkaloid content and vasicine content (4.05+/-0.12 and 3.46+/-0.06 mg/ml, respectively) were very high when compared to the other methods, though the traditional method was found to give the best quality juice with highest amount of total alkaloids (5.93+/-0.55 mg/ml) and vasicine (5.64+/-0.10 mg/ml) content.

Bergia suffruticosa (Family Elatinaceae) is an important Indian medicinal plant, traditionally us... more Bergia suffruticosa (Family Elatinaceae) is an important Indian medicinal plant, traditionally used to repair bones and applied on sores. The plant has not been studied much. Preliminary phytochemical screening of the plant showed the presence of high amount of phenolics, tannins and flavonoids. Subsequent quantification showed the presence of 8.8% w/w phenolics (calculated as gallic acid), and 7.5% w/w tannins. This high amount of tannins and phenolics prompted us to evaluate its antioxidant activity. We studied the antioxidant activity of B. suffruticosa whole plant in three in vitro models: DPPH radical scavenging activity, superoxide scavenging activity and measurement of reducing power. Methanolic extract of whole plant of B. suffruticosa showed a very good DPPH radical scavenging activity (EC50 of 13.1 μg) and superoxide scavenging activity (EC50 of 139.4 μg) in a dose-dependent manner. The extract showed dose-dependent reduction ability (Fe3+ to Fe2+ transformation) with a maximum absorbance of 1.195 at a concentration of 300 μg of the extract.

The oral bioavailability of berberine from Rasont (aqueous extract of Berberis aristata DC., Fami... more The oral bioavailability of berberine from Rasont (aqueous extract of Berberis aristata DC., Family: Berberidaceae) was investigated in rats. The in vivo bioavailability enhancement potential of lysergol was also evaluated for berberine from Rasont. One animal group was dosed with freshly prepared lyophilized Rasont (Dose: 1.07 g/kg bodyweight equivalent to 200 mg of berberine) and the other with both freshly prepared lyophilized Rasont (same dose as in 1st group) and lysergol (20 mg/kg bodyweight). The rat pharmacokinetic profiles of berberine upon oral administration Rasont alone and in combination with lysergol were compared. Results of the study inferred that lysergol improved the oral bioavailabilty of berberine from Rasont.

In the present work we developed and validated densitometric methods for quantification of glycyrrhizin, glycyrrhetinic acid, apigenin, kaempferol and quercetin using HPTLC. The developed methods were found to be precise and accurate. The amount of glycyrrhizin, glycyrrhetinic acid, and quercetin was found to be 1.070, 0.84 and 0.271% w/w, respectively. Apigenin and kaempferol were quantified in free (0.007 and 0.033% w/w) as well as bound (0.021 and 0.074% w/w) forms. This is the first report of simultaneous quantification of glycyrrhetinic acid and apigenin as well as kaempferol and quercetin from G. glabra. Furthermore, no TLC densitometric methods have been reported for the quantification of apigenin, kaempferol and quercetin from G. glabra.

Acanthaceae) with that of the pure vasicine and vasicinone (chief marker compounds of A. vasica) upon oral administration of Vasa Swaras in rats, and also in different animal groups. Significant difference with p < 0.05 was found in the oral bioavailability of vasicine and vasicinone when administered as Vasa Swaras and as single vasicine and/or vasicinone. Vasicine and vasicinone were found to be more bioavailable from Vasa Swaras than pure vasicine and/or vasicinone.
