Type 2 diabetes mellitus (T2DM) and metabolic syndrome (MetS) are diseases caused by the interact... more Type 2 diabetes mellitus (T2DM) and metabolic syndrome (MetS) are diseases caused by the interaction of genetic and non-genetic factors. Therefore, the aim of our study was to investigate the association between six common genetic polymorphisms and T2DM and MetS in males. A total of 120 T2DM, 75 MetS, and 120 healthy controls (HC) were included in the study. ACE ID, eNOS 4a/b, ATR1 A1166C, OXTR (A>G), SOD1 +35A/C, CAT-21A/T gene polymorphisms were genotyped by PCR or PCR-RFLP techniques. T2DM was diagnosed at an earlier age compared to MetS (54 vs 55 years old, p=0.0003) and the difference was greater in carriers of the OXTR G allele (54 vs 56 years old, p=0.0002) or both OXTR G and eNOS b alleles (54 vs 56, p=0.00016). The SOD1 AA genotype (O.R.=0.11, p=0.0006) and the presence of both ACE I and OXTR1 A (O.R.=0.39, p=0.0005) alleles revealed to be protective for T2DM. The vitamin D receptor (VDR) might be a candidate gene for this disease. Our study aimed to assess the preferential transmission of VDR polymorphisms from parents to SMA children. We genotyped 261 subjects (87 SMA nuclear families) for VDR FokI, BsmI, ApaI, and TaqI polymorphisms. The transmission of the genetic marker was estimated with Plink and FBAT software. It detected a preferential transmission of the rs731236 and rs7975232 variants to SMA1 patients and of rs1544410 variants to SMA2 patients. The variants of rs2228570 were preferentially transmitted to parents of all SMA patients. Haplotype analysis identified that haplotypes C-C-G-A and T-A-A-G seem to be involved in the booth type of SMA whereas the impact of T-A-A-A seems to be limited only to SMA2. Investigation regarding effects of low doses of DOX on chromosomal aberrations frequency in mice bone marrow. The study explored a possible relationship between diet, follow-up period, and cytostatic doses. Four mice groups received normal or high-fat (pork) diet. At the beginning of the 8th day mice from each group received a single intraperitoneal injection with DOX (dose range: 1.7-7.7 mg/kg b.w.) or isotonic saline solution (0.02 mL/g control mice). In the 10th (groups I, II) and 14th (groups III, IV) days from the beginning of the experiment, the mice were euthanized and chromosome aberrations were tested in bone marrow cells. The body weight of all mice who received DOX decreases, especially at higher doses. In the first three days post-injection, weight loss percentage was significantly influenced by DOX doses and/or type of diet (p<0.003). The interaction between the type of diet and DOX doses was not statistically significant in the follow-up period (p interactions >0.05). DOX doses were able to increase the frequency of chromosomal aberrations. DOX acts as a potent inductor of cytogenetic aberrations in bone marrow cells, regardless of the type of diet."

COVID-19 may remain with symptoms which can persist
for an uncertain period of time.
Aim. The purpose of this study was to investigate
the reasons why patients who passed the acute phase
of COVID-19 presented themselves to the Emergency
Department.
Patients and Methods. We selected 87 patients
admitted to the Emergency Department of the Bucharest
University Emergency Hospital between 01.01.2021-
31.05.2021. Patients had pulmonary fibrosis (11.49%),
pleural effusion (16.09%) or a history of hypertension
(73.56%), type 2 diabetes (42.53%), stroke (24.14%),
malignant diseases (10.34%).
Results. Association between neutrophil levels
and acute stroke and between fibrinogen levels and alveolar
condensation were identified. The percentage of deaths was
significantly higher in the subgroup of subjects that had
maxim 11 days of hospitalization (p=0.004); we observed a
trend of association between the age of more than 51 years
old and admission in the Emergency Unit at less than a
month after the SARS Cov2 infection, the positive result at
the RT-PCR test or a lung damage of over 30% (p<0.05).
Conclusion. A significant percentage of patients
that were admitted to the Emergency Unit post COVID-19
had chronic pathologies and their characteristics were
associated with neutrophilia, high fibrinogen levels or length
of hospitalization.

The study aims to follow the evolution of the patients with acute post-COVID 19 clinical symptoms presented in the Emergency Department. Methods: We analyzed 262 presentations more than 14 days after the initial COVID-19 diagnosis and with at least one acute COVID-19-related symptom. This study included presented between 01.01 and 31.05.2021 in the Emergency Department of Bucharsti- Emergency University Hospital. Data were obtained in real-time, at the presentation, from the patients’ medical charts, and afterward from SUUB’s database. Results. Out of the total of 262 presentations, 40.84% required patient hospitalization. The highest number of presentations with (18.70%) or without (22.90%) hospitalization was recorded in the first month after the COVID-19 diagnosis. In an attempt to standardize the X-ray analysis and to create relevant correlations between it an... more In an attempt to standardize the X-ray analysis and to create relevant correlations between it and the functional results, we present a pilot model of an artificial neural network based on a Bayesian belief network (BNN) designed for the automatic analysis of anteroposterior X-ray of the knee following total knee arthroplasty and more precisely the analysis of the tibial component. A prospective analysis was conducted in which 30 patients were analyzed and two groups resulted: the first was made up of 12 cases considered “normal” by the examiners (optimal positioning and cementation) and the second one, which was considered “pathological” (incorrect positioning or cementation) and included 16 cases. Based on 22 points established on the tibial component several geometric sizes (calculated parameters) were computed. The predisposition for this multifactorial disease is partially known. The aim of this study is to investigate the predisposition for stroke in Romanian population. Material and methods. We selected cases with recent ischemic stroke (n=100) and healthy control subjects (n=100). The AGTR1 A1166C (rs5186) polymorphism was genotyped by restriction of amplicons with Dde I endonuclease. Results. Active cigarette smoking (O.R. =6.92, p=0.0001) or presence of the AT1R C variant (O.R. =6.69, p=0.0006) in overweight or obese women significantly increase the risk for ischemic stroke. The diagnosis of stroke (71.5 vs 68 years old) or T2DM (63.39 vs 60.77 years old) was recorded at an older age in women compared to men (p<0.05). Obesity considered independent (O.R. =4.22, p<0.05) or in association with T2DM (O.R. = 10.16, p=0.0002) confers the highest risk of stroke when compared to women. Conclusions. Obesity in association with T2DM confers the highest risk of stroke for men when compared to women. The aim of this study was to evaluate the association between vitamin D receptor, IL6 polymorphis... more The aim of this study was to evaluate the association between vitamin D receptor, IL6 polymorphisms and DM. DNA was used to genotype the VDR FokI (rs2228570), TaqI (rs731236) and ApaI (rs7975232) polymorphisms by PCR RFLP and IL6 G-174C (rs1800795) by tetra-primer ARMS PCR. The presence of torque teno viruses DNA was assessed with heminested-PCR. For this study T1DM (n = 107) and T2DM (n = 124) patients and matched clinically healthy subjects (n = 200) were recruited. T1DM patients have a tendency to be more frequent carriers of the C allele and TTV infection than controls (OR = 1.9, p = 0.03). VDR tt genotype and VDR “FAt” haplotype are risk factors for T1DM. VDR “fAt” haplotype may increases the risk for T2DM. These associations were not changed after exclusion from statistical analysis of patients with hypertension, myocardial infarction, stroke or breast cancer.

We aimed to assess the geographical distribution and phylogeny of TTV from Romanians. A fragment of TTV UTR B was sequenced in samples from volunteers from all seven development regions of Romania. Additional sequences from Romanian dialyzed patients were added for phylogenetic analysis. More than 80% of Romanian sequences clustered with isolates assigned to TTV1 and TTV3 (former genogroup 1), while the rest clustered with TTV19-SANBAN and TTV22-svi-1 (former genogroup 3) and TTV-Hebei-1. Phylogenetic analysis showed segregation between isolates from North-East and West Romania providing possible evidence of distinct geographical distribution.

The mechanism of mesenteric injury in blunt abdominal trauma involves compression and deceleratio... more The mechanism of mesenteric injury in blunt abdominal trauma involves compression and deceleration forces which result in a spectrum of injuries that range from contusions to tearing of the bowel wall, tearing of the mesentery, and loss of vascular supply. Hemoperitoneum is a major sign of severity in abdominal trauma, which, in most cases, requires emergency surgery. Our patient showed that the diagnosis of mesenteric injury should be considered even in patients who sustain only minor abdominal trauma. In this case the patient’s symptoms were suggestive only for the orthopedic lesion and not for the hemoperitoneum. Trans-scapho-lunate dislocation fracture is a rare lesion that may go unnoticed at initial assessment in a polytraumatized patient. A delay in the diagnosis and treatment of a scaphoid fracture may alter the prognosis after consolidation.

Methods: This was a sub-analysis of the European EURODEM study. Data on patients presenting with dyspnoea were collected prospectively from 66 European emergency departments. Materials and method: The retrospective study was conducted in the Emergency Unit of the University Emergency Hospital Bucharest, Romania, between 01.06.2020 and 30.09.2020, by analyzing the data from the presentation sheets and the hospital database. It followed and compared a total of 55 cases, out of which 29 patients were involved in motorcycle accidents, and 26 patients were involved in electric scooter accidents, by number, age, sex, imaging investigations, severity of injuries, hospitalization days, and the treatment of discharged patients. This study aims to investigate the genetic distribution of di-allelic DNA polymorphisms associated with different diseases in the South Romanian population. We investigated eleven di-allelic DNA polymorphisms in blood samples from 648 healthy Caucasian subjects (age 18-66 years old) from Bucharest and eight South Romanian districts. All markers present no significant deviation from expected Hardy–Weinberg equilibrium even if subjects were stratified according to the birth place, gender or age group. The frequency of allelic variants and genotypes in South Romanian areas situates in the range of values found with other Caucasian populations. When testing the distribution between the tested districts only VDR Fok was found to present a different distribution in samples from Bucharest and in those from Calarasi, Dambovita or Teleorman (p < 0.00008). The aim of this retrospective study was to analyze the results of tests for SARSCoV- 2 performed ... more The aim of this retrospective study was to analyze the results of tests for SARSCoV- 2 performed during 07.04.2020- 20.06.2020 in the Department of the Emergency from Bucharest University Emergency Hospital. We detected 173 men and 133 women that were tested with both RT-PCR and serologic tests. The results were concordant for 287 samples (93,8%) that were collected from subjects for whom the diagnosis of COVID-19 was subsequently confirmed (10) or infirmed (277). We found that the most frequent signs and symptoms of patients with COVID-19 were at the respiratory (e.g. dyspnea), neurological (e.g. vertigo, cephalgia) and gastrointestinal (e.g. abdominal pain, vomiting, high volume of the abdomen) systems. There was no situation with positive RT-PCR and IgG and negative IgM results. In our study the RT-PCR and quick serological tests were concordant in 93,8% of cases. The combination of RT-PCR and serological testing can enhance the accuracy of COVID-19 diagnosis.

Only a rescuer in optimal physical and mental shape can ensure the maximum chances of rescuing a victim, while an injured rescuer represents a new victim who will share the rescue resources with the victim(s) or will cancel any chance of rescue either for the initial victim as well as for himself. Rescuers must be able to perform resuscitation maneuvers and rescue operations anytime and anywhere. The safety of the savior is not always entirely his responsibility.

The aim of this study was to investigate the growth pattern of a lyophilized Candida albicans str... more The aim of this study was to investigate the growth pattern of a lyophilized Candida albicans strain using microcalorimetry. Microcalorimetry is a method that can record the thermal flow released by the basal metabolism of a microorganism. Using a dedicated data acquisition and processing software, the method can provide a growth curve, which is specific to every microorganism. We have obtained the growth pattern of Candida albicans in different conditions with the objective of determining various growth “fingerprints” that we can afterwards use within a diagnostic method. Experiments where undertaken at five different temperatures 26°C, 30°C, 34°C, 37°C and 40°C and at different suspension volumes in the microcalorimetric cell: 200 μl, 400 μl, 600 μl, 800 μl. Our experiments provided real-time, reproducible information about fungal growth, with expected differences due to the various changes in growth conditions.

Results. Active cigarette smoking (O.R. =6.92, p=0.0001) or presence of the AT1R C variant (O.R. =6.69, p=0.0006) in overweight or obese women significantly increase the risk for ischemic stroke. The diagnosis of stroke (71.5 vs 68 years old) or T2DM (63.39 vs 60.77 years old) was recorded at an older age in women compared to men (p<0.05). Obesity considered independent (O.R. =4.22, p<0.05) or in association with T2DM (O.R. = 10.16, p=0.0002) confers the highest risk of stroke when compared to women. Conclusions. Obesity in association with T2DM confers the highest risk of stroke for men when compared to women. The aim of this study was to evaluate the association between vitamin D receptor, IL6 polymorphis... more The aim of this study was to evaluate the association between vitamin D receptor, IL6 polymorphisms and DM. DNA was used to genotype the VDR FokI (rs2228570), TaqI (rs731236) and ApaI (rs7975232) polymorphisms by PCR RFLP and IL6 G-174C (rs1800795) by tetra-primer ARMS PCR. The presence of torque teno viruses DNA was assessed with heminested-PCR. For this study T1DM (n = 107) and T2DM (n = 124) patients and matched clinically healthy subjects (n = 200) were recruited. T1DM patients have a tendency to be more frequent carriers of the C allele and TTV infection than controls (OR = 1.9, p = 0.03). VDR tt genotype and VDR “FAt” haplotype are risk factors for T1DM. VDR “fAt” haplotype may increases the risk for T2DM. These associations were not changed after exclusion from statistical analysis of patients with hypertension, myocardial infarction, stroke or breast cancer.

We aimed to assess the geographical distribution and phylogeny of TTV from Romanians. A fragment of TTV UTR B was sequenced in samples from volunteers from all seven development regions of Romania. Additional sequences from Romanian dialyzed patients were added for phylogenetic analysis. More than 80% of Romanian sequences clustered with isolates assigned to TTV1 and TTV3 (former genogroup 1), while the rest clustered with TTV19-SANBAN and TTV22-svi-1 (former genogroup 3) and TTV-Hebei-1. Phylogenetic analysis showed segregation between isolates from North-East and West Romania providing possible evidence of distinct geographical distribution.

The mechanism of mesenteric injury in blunt abdominal trauma involves compression and deceleratio... more The mechanism of mesenteric injury in blunt abdominal trauma involves compression and deceleration forces which result in a spectrum of injuries that range from contusions to tearing of the bowel wall, tearing of the mesentery, and loss of vascular supply. Hemoperitoneum is a major sign of severity in abdominal trauma, which, in most cases, requires emergency surgery. Our patient showed that the diagnosis of mesenteric injury should be considered even in patients who sustain only minor abdominal trauma. In this case the patient’s symptoms were suggestive only for the orthopedic lesion and not for the hemoperitoneum. Trans-scapho-lunate dislocation fracture is a rare lesion that may go unnoticed at initial assessment in a polytraumatized patient. A delay in the diagnosis and treatment of a scaphoid fracture may alter the prognosis after consolidation.

This study aims to investigate the genetic distribution of di-allelic DNA polymorphisms associated with different diseases in the South Romanian population. We investigated eleven di-allelic DNA polymorphisms in blood samples from 648 healthy Caucasian subjects (age 18-66 years old) from Bucharest and eight South Romanian districts. All markers present no significant deviation from expected Hardy–Weinberg equilibrium even if subjects were stratified according to the birth place, gender or age group. The frequency of allelic variants and genotypes in South Romanian areas situates in the range of values found with other Caucasian populations. When testing the distribution between the tested districts only VDR Fok was found to present a different distribution in samples from Bucharest and in those from Calarasi, Dambovita or Teleorman (p < 0.00008). The aim of this retrospective study was to analyze the results of tests for SARSCoV- 2 performed ... more The aim of this retrospective study was to analyze the results of tests for SARSCoV- 2 performed during 07.04.2020- 20.06.2020 in the Department of the Emergency from Bucharest University Emergency Hospital. We detected 173 men and 133 women that were tested with both RT-PCR and serologic tests. The results were concordant for 287 samples (93,8%) that were collected from subjects for whom the diagnosis of COVID-19 was subsequently confirmed (10) or infirmed (277). We found that the most frequent signs and symptoms of patients with COVID-19 were at the respiratory (e.g. dyspnea), neurological (e.g. vertigo, cephalgia) and gastrointestinal (e.g. abdominal pain, vomiting, high volume of the abdomen) systems. There was no situation with positive RT-PCR and IgG and negative IgM results. In our study the RT-PCR and quick serological tests were concordant in 93,8% of cases. The combination of RT-PCR and serological testing can enhance the accuracy of COVID-19 diagnosis.

Rescuers must be able to perform resuscitation maneuvers and rescue operations anytime and anywhere. The safety of the savior is not always entirely his responsibility.

The aim of this study was to investigate the growth pattern of a lyophilized Candida albicans str... more The aim of this study was to investigate the growth pattern of a lyophilized Candida albicans strain using microcalorimetry. Microcalorimetry is a method that can record the thermal flow released by the basal metabolism of a microorganism. Using a dedicated data acquisition and processing software, the method can provide a growth curve, which is specific to every microorganism. We have obtained the growth pattern of Candida albicans in different conditions with the objective of determining various growth “fingerprints” that we can afterwards use within a diagnostic method. Experiments where undertaken at five different temperatures 26°C, 30°C, 34°C, 37°C and 40°C and at different suspension volumes in the microcalorimetric cell: 200 μl, 400 μl, 600 μl, 800 μl. Our experiments provided real-time, reproducible information about fungal growth, with expected differences due to the various changes in growth conditions.
