Lumpy skin disease (LSD), a notifiable disease listed by the World Organization for Animal Health... more Lumpy skin disease (LSD), a notifiable disease listed by the World Organization for Animal Health and a fast fast-moving transboundary viral disease infecting cattle and buffaloes, was reported in India in 2019 and has since rapidly spread across the country. This study reports the first complete genome sequence and analysis of a pathogenic LSD virus (LSDV) from India (LSDV/208/PVNRTVU/2020) obtained by direct sequencing of a suspected clinical sample using Illumina and Nanopore sequencing technologies. The complete genome sequence of LSDV/208/PVNRTVU/2020 is 150445 bp long, codes for 156 putative genes and carries identical 2254 bp inverted terminal repeats at either ends. In this study, free vibration analysis is carried out for a glass-fiber reinforced plastic (GFRP) sandwich composite plate. The natural frequencies are obtained by taking into account various uncertainties involved in preparing the sandwich composite plate. The Kolmogorov–Smirnov (KS) and Anderson–Darling (AD) goodness-of-fit (GOF) tests are performed for the quantification of the uncertainty of the natural frequencies. However, despite their fascinating structure and other necessary impetus, only a few synthetic studies of these molecules have been reported so far. We have now devised a concise total synthesis of [3]-ladderanol, a component of ladderane phospholipids, using an organocatalytic enantioselective desymmetrizing formal C(sp2)-H alkylation. Our synthetic strategy rests on the late-stage introduction of chirality, thus allowing facile access to both enantiomers of [3]-ladderanol as well as an analogue. This is the first time a desymmetrization strategy is applied to the synthesis of [3]-ladderanol. The scope of this desymmetrizing C(sp2)-H alkylation of meso-cyclobutane-fused cyclohexenediones is also presented.

The lack of control in the manufacturing of the sandwich composites may lead to non-uniform material properties, and thus variation in its behavior. An investigation has been carried out for identification of both in-plane and out-of-plane elasti... more An investigation has been carried out for identification of both in-plane and out-of-plane elastic parameters of orthotropic composite structures from dynamic responses. A gradient based finite element model updating approach is used to implement the same. The methodology is based on the comparison of modal parameters of composite plates and beams, as obtained from finite element analysis as well as from modal testing. A three-step identification methodology is proposed in which first the in-plane elastic moduli are estimated from observed modes of plate specimens, followed by estimation of out-of-plane shear moduli from separate modal testing of two beam specimens which are oriented along the longer and shorted axes of the rectangular plate. Experimental modal testing data are numerically simulated for the present investigation. Since the in-plane and the out-of-plane elastic parameters have varied order of sensitivities associated with different sets of modes, it is virtually impossible to update all the in-plane and out-of-plane parameters together from a single orthotropic plate type of structures and use of beam specimens oriented in appropriate directions are mandatory. The present method recognizes this difficulty of parameter estimation algorithms to determine material properties of orthotropic plates and uses the multi-step estimation process successfully. The results show rapid conver-gences from varied ranges of initial guesses of parameters and the methodology is found to be robust.

However, it is difficult to identify the accurate damping matrix considering the modal sparsity and coordinate sparsity. A nonproportional viscous damping parameter identification is formulated as an unconstrained optimization problem in the present study. The damping coefficient of each element is considered as the design variable for the optimization problem. The objective function is defined using the incomplete complex eigenvectors, which are generated because of the presence of external damping devices in the structure. This objective function is then minimized using standard particle swarm optimization to identify the damping coefficient of the damping matrix. The accuracy and efficiency of the particle swarm optimization are investigated by solving a few numerical problems with simulated measured data. We assume that the leader has two sets of strategies. He announces only one set of strategies and the other set strategies are not announced but are constrained. We formulate the necessary conditions for the existence of Stackelberg equilibrium for this class of games. In particular, we show under a few assumptions, the necessary conditions can be recasted as a single large scale complementarity problem. We also provide sufficient conditions under which the solution of this complementarity problem is a Stackelbreg equilibrium.

BACKGROUND The physiological significance of a large family of heat-shock proteins (HSPs), compri... more BACKGROUND The physiological significance of a large family of heat-shock proteins (HSPs), comprised of the cytosolic HSP90A and the endoplasmic reticulum component of HSPB, is evident in prokaryotes and eukaryotes. The HSP90A is believed to play critical roles in diverse physiological functions of cell viability and chromosomal stability including stress management. Heightened abundance of hsp90β transcript was documented in Channa striatus, a freshwater fish, which is capable of surviving within an extremely hypoxic environment. METHODS AND RESULTS To better understand the mechanism of hsp90β gene expression, we investigated its genomic organization. Eleven exons were identified, including a long upstream intron with a remarkable similarity with human, but not with chicken counterpart. Dual-luciferase assays identified promoter activity in a 1366 bp 5'-flanking segment beyond the transcription initiation site. Examination detected a minimal promoter of 754 bp containing a TATA-box, CAAT-enhancer in addition to providing clues regarding other enhancer and repressor elements. The driving capability of this minimal promoter was further validated by its binding ability with TATA-box binding protein and the generation of GFP expressing transgenic zebrafish (F2). Further, deletion of an inverted HIF (hypoxia inducible factor) motif RCGTG (upstream of the TATA-box) dramatically reduced luciferase expression in a hypoxic environment (CoCl2 treated cultivable cells) and was identified as a cis-acting HIF responsive element, necessary for the hypoxia-induced expression. CONCLUSIONS The results obtained herein provide an insight regarding how hsp90β gene expression is controlled by HIF responsive element in teleost both during hypoxia stress management and normal physiological functions, and suggested that the hsp90β gene promoter could be used as a potential candidate for generating ornamental and food-fish transgenics.

Abstract The promyelocytic leukemia zinc finger (Plzf) gene, being a transcriptional repressor, p... more Abstract The promyelocytic leukemia zinc finger (Plzf) gene, being a transcriptional repressor, plays a crucial role in self-renewal and maintenance of spermatogonial stem cells (SSCs) across mammalian and non-mammalian species. It is now being considered as a candidate marker of proliferating SSC in teleost. The knowledge on its mechanism of expression is limited. In this study, we intended to clone and characterize Plzf gene promoter of Clarias batrachus, a commercially valuable farmed catfish (also known as magur). A comparative cDNA sequence analysis between Labeo rohita carp and C. batrachus catfish confirmed about perfect homology. We have isolated, sequenced and characterized the regulatory elements of Plzf by genome walking. About 3.6 kb sequence of 5′ upstream region relative to ATG start codon was sequenced. Alignment of genome walking and cDNA sequence data identified transcriptional start site (TSS). The 5′ UTR comprises of non-coding exons (EI and EII) containing a long stretch of intron. Computational analysis revealed several important putative elements like vertebrate TATA binding protein factor (VTBP), ecotropic viral integration site (Evi-1), myoblast determining factors (MyoD), myocyte-specific enhancer binding factor (MEF2), general transcription factor IID initiator (TFIID), etc.; positioned in the upstream sequence above transcriptional start site; indicating as potential promoter/regulatory elements. Dual luciferase reporter assay was performed for various deleted constructs designed to measure their promoter activity in vivo. Its full promoter activity was dependent on upstream region containing a putative Evi-1-like element as enhancer. We also identified the region possibly acting as a transcriptional repressor (Gfi-1). This is the first report of identifying Plzf gene being expressed in spermatogonial cells of C. batrachus. Even though the Plzf cDNA sequence is conserved across species, its regulatory elements are not conserved among human, carps and catfishes.

The current literature of determination of non-proportional damping matrix demands measurement of a large number of complex modes which is extremely difficult in practice. A gradient based finite element model updating algorithm implementing inverse eigensensitivity method has been presented through a series of numerically simulated cantilever beams. Ladderane phospholipids, with their unusual ladder-like arrangement of fused cyclobutane rings, r... more Ladderane phospholipids, with their unusual ladder-like arrangement of fused cyclobutane rings, represent an architecturally unique class of natural products. However, despite their fascinating structure and other necessary impetus, only a few synthetic studies of these molecules have been reported so far. We have now devised a concise synthesis of [3]-ladderanol, a component of ladderane phospholipids, using an organocatalytic enantioselective desymmetrizing formal C(sp2)‒H alkylation. Our synthetic strategy rests on a late-stage introduction of chirality, thus allowing facile access to both the enantiomers of [3] ladderanol as well as its analog. This is the first time a desymmetrization strategy is applied to the synthesis of [3]-ladderanol. The scope of this desymmetrizing C(sp2)‒H alkylation of meso cyclobutane fused cyclohexenediones is also presented.

However, it is difficult to identify the accurate damping matrix considering the modal sparsity and coordinate sparsity. A nonproportional viscous damping parameter identification is formulated as an unconstrained optimization problem in the present study. The damping coefficient of each element is considered as the design variable for the optimization problem. The objective function is defined using the incomplete complex eigenvectors, which are generated because of the presence of external damping devices in the structure. This objective function is then minimized using standard particle swarm optimization to identify the damping coefficient of the damping matrix. The accuracy and efficiency of the particle swarm optimization are investigated by solving a few numerical problems with simulated measured data. We assume that the leader has two sets of strategies. He announces only one set of strategies and the other set strategies are not announced but are constrained. We formulate the necessary conditions for the existence of Stackelberg equilibrium for this class of games. In particular, we show under a few assumptions, the necessary conditions can be recasted as a single large scale complementarity problem. We also provide sufficient conditions under which the solution of this complementarity problem is a Stackelbreg equilibrium.

BACKGROUND The physiological significance of a large family of heat-shock proteins (HSPs), compri... more BACKGROUND The physiological significance of a large family of heat-shock proteins (HSPs), comprised of the cytosolic HSP90A and the endoplasmic reticulum component of HSPB, is evident in prokaryotes and eukaryotes. The HSP90A is believed to play critical roles in diverse physiological functions of cell viability and chromosomal stability including stress management. Heightened abundance of hsp90β transcript was documented in Channa striatus, a freshwater fish, which is capable of surviving within an extremely hypoxic environment. METHODS AND RESULTS To better understand the mechanism of hsp90β gene expression, we investigated its genomic organization. Eleven exons were identified, including a long upstream intron with a remarkable similarity with human, but not with chicken counterpart. Dual-luciferase assays identified promoter activity in a 1366 bp 5'-flanking segment beyond the transcription initiation site. Examination detected a minimal promoter of 754 bp containing a TATA-box, CAAT-enhancer in addition to providing clues regarding other enhancer and repressor elements. The driving capability of this minimal promoter was further validated by its binding ability with TATA-box binding protein and the generation of GFP expressing transgenic zebrafish (F2). Further, deletion of an inverted HIF (hypoxia inducible factor) motif RCGTG (upstream of the TATA-box) dramatically reduced luciferase expression in a hypoxic environment (CoCl2 treated cultivable cells) and was identified as a cis-acting HIF responsive element, necessary for the hypoxia-induced expression. CONCLUSIONS The results obtained herein provide an insight regarding how hsp90β gene expression is controlled by HIF responsive element in teleost both during hypoxia stress management and normal physiological functions, and suggested that the hsp90β gene promoter could be used as a potential candidate for generating ornamental and food-fish transgenics.

Abstract The promyelocytic leukemia zinc finger (Plzf) gene, being a transcriptional repressor, p... more Abstract The promyelocytic leukemia zinc finger (Plzf) gene, being a transcriptional repressor, plays a crucial role in self-renewal and maintenance of spermatogonial stem cells (SSCs) across mammalian and non-mammalian species. It is now being considered as a candidate marker of proliferating SSC in teleost. The knowledge on its mechanism of expression is limited. In this study, we intended to clone and characterize Plzf gene promoter of Clarias batrachus, a commercially valuable farmed catfish (also known as magur). A comparative cDNA sequence analysis between Labeo rohita carp and C. batrachus catfish confirmed about perfect homology. We have isolated, sequenced and characterized the regulatory elements of Plzf by genome walking. About 3.6 kb sequence of 5′ upstream region relative to ATG start codon was sequenced. Alignment of genome walking and cDNA sequence data identified transcriptional start site (TSS). The 5′ UTR comprises of non-coding exons (EI and EII) containing a long stretch of intron. Computational analysis revealed several important putative elements like vertebrate TATA binding protein factor (VTBP), ecotropic viral integration site (Evi-1), myoblast determining factors (MyoD), myocyte-specific enhancer binding factor (MEF2), general transcription factor IID initiator (TFIID), etc.; positioned in the upstream sequence above transcriptional start site; indicating as potential promoter/regulatory elements. Dual luciferase reporter assay was performed for various deleted constructs designed to measure their promoter activity in vivo. Its full promoter activity was dependent on upstream region containing a putative Evi-1-like element as enhancer. We also identified the region possibly acting as a transcriptional repressor (Gfi-1). This is the first report of identifying Plzf gene being expressed in spermatogonial cells of C. batrachus. Even though the Plzf cDNA sequence is conserved across species, its regulatory elements are not conserved among human, carps and catfishes.
