Exploration of probiotics candidate have been accelerated from unique and novel source, one of wh... more Exploration of probiotics candidate have been accelerated from unique and novel source, one of which is from black soldier fly (Hermetia illucens). The black soldier flies are known as omnivores which are commonly reared, harvested and utilized to convert organic wastes into biomass yet reducing the amount of abundant waste in nature. Lactic acid bacteria (LAB) is one of beneficial gut symbionts which can be found across animal taxa, including insect, however, limited information is currently reported from H. illucens as a potential harboring insect. This study aimed to determine the probiotics candidate from the gut of H. illucens fed with different feeding substrates to improve the gut microbiota. The larvae were nourished with chicken manure, tofu solid wastes and vegetable wastes prior isolation of LAB. The LAB isolates were tested for their antagonistic activity against enteric pathogens, pH and bile salt tolerance, and adherence to chicken intestine (ileum). Based on the growth performance under physiological stresses and multivariate analysis using principal component analysis (PCA), four LAB isolates were designated as putative probiotics. The four isolates were Lactiplantibacillus. pentosus 5P2i1, Lacticaseibacillus. paracasei 5P2i5, Lactiplantibacillus. pentosus 5P2i9, and Lactiplantibacillus. plantarum 6P1i1 was then declared as novel strains that could be developed further as poultry probiotics.

In dealing with this problem, studies were carried out, one of which was by utilizing natural medicinal ingredients such as bawang sabrang (Eleutherine palmifolia Merr.) of the Iridaceae family. This study aims to determine the antibacterial activity of the residual fraction of the ethanolic extract of bawang sabrang bulbs (Eleutherine palmifolia Merr.) against gram-positive and gram-negative bacteria and to determine the class of chemical compounds. Extraction was carried out by maceration using ethanol as a solvent and continued by successive fractionation using n-hexane and ethylacetate solvents, so that the remaining fraction was obtained. Antibacterial activity testing was carried out in vitro using the agar diffusion method using metal rings at concentrations of 20 mg/ml, 10 mg/ml and 5 mg/ml against gram-positive bacteria (Bacillus pumilus ATCC 7061, Bacillus subtilis ATCC 6633, Methicillin-resistant Staphylococcus aureus (MRSA) ATCC 25923, Micrococcus luteus ATCC 9341, Propionibacterium acne ATCC 6918, Staphylococcus aureus ATCC 29737, Staphylococcus epidermidis ATCC 12228) and gram-negative bacteria (Klebsiella pneumoniae ATCC 10031, Escherichia coli ATCC 10536, Salmonella thypi ATCC 29213). The analysis of the chemical compound group of the residual fraction was carried out using a phytochemical screening technique. The residual fraction of the ethanolic extract of bawang sabrang bulbs had antibacterial activity on all the tested bacteria. The chemical compounds contained in the remaining fraction of the ethanolic extract of bawang sabrang bulbs are alkaloids, flavonoids, glycosides, saponins, anthraquinone glycosides, tannins, triterpenoids, and steroids.

Bioassay of these isolates to larva of Heliothis armigera, Plutella xylostella, Aedes aegypti, and Culex sp. showed that the isolates have different spectrum and ability in controlling insect larvae. Interestingly, TU1 has similar ability to that of Dipel (B. thuringiensis var kurstaki strain HD-7) in controlling Dipteran (A. aegypti and Culex sp.) and Lepidopteran (H. armigera and P. xylostella). The others were not effective to Dipteran, but have various abilities to kill Lepidopteran. This result indicated that local isolates of B. thuringiensis of North Sumatra were varied in ability to kill different larvae.

Examination of antimicrobial activities to the microbes were done by agar diffusion method, while test for toxicity was done by exposing 2 days-old larvae to certain concentration of the extract. Preliminary tests on phytochemical compounds in leaf were done by Mayer’s, FeCl3, Mg-HCl, foam, and 10% NaOH in methanol tests. The results showed that methanol leaf extract of tabar-tabar in general inhibited more in the growth of gram-positive Bacillus sp. and S. aureus rather than to other tested microbes. Wastewater of textile industry is considered as toxic waste containing various dyes and chemicals... more Wastewater of textile industry is considered as toxic waste containing various dyes and chemicals that makes it recalcitrants. Decolorization of wastewater of the industry was conducted in our laboratorium using fungi isolated from a local forest of North Sumatera. Six potential isolates, previously screened for the ability to grow in plate media containing 100% of wastewater, were used for the study in a liquid culture condition. The results show that all tested isolates grew. The growth response of isolates cultured in a media supplemeted with 1% glucose was about two to three times higher than that of cultures without glucose. In the first culture, the fungi grew on the surface of the media however in the latter experiment, it concentrated on the bottom of culture flask for all of tested isolates. Decolorization analysis shows that all isolates were able to decolorize the wastewater. The cultures supplemented with glucose decolorized wastewater from 20% to 46%, while cultures without glucose did much higher about 56% to 92%. Furthermore, a microscopic analysis shows that dyes molecules in the wastewater were absorbed to fungal mycelia. The findings show that extraceluler molecules existed on the surface of fungal mycelia play an important role in the absorption of dye molecules.

Triglycerides and diglycerides do not show antibacterial activity, but free fatty acids and monog... more Triglycerides and diglycerides do not show antibacterial activity, but free fatty acids and monoglycerides, especially lauric acid and monolaurin do. The purpose of this study was to determine the antibacterial activity of the enzymatic hydrolyzed of virgin coconut oil (VCO) and palm kernel oil (PKO), which produce a combination of lauric acid and monolaurin. Hydrolysis process was done by the enzyme lipase (Lipozym TL IM) which was active at 1.3 position. The hydrolyzed oil then tested for antibacterial activity against Staphylococcus aureus, Escherichia coli and Salmonella thypi using agar diffusion method with paper disc with diameter of 6 mm (Oxoid, England). Antibacterial activity test carried out on VCO and PKO, hydrolyzed VCO (VCOH) and PKO (PKOH) each at 25% concentration, 50%, 75% and 100%. Antibacterial activity of the test material compared with chloramphenicol (30 µg) and tetracycline (30 µg). The results showed that the optimum hydrolysis time is 14 hours. There is low and similar antibacterial activity of VCO and PKO. VCOH and PKOH show higher antibacterial activity than VCO and PKO. There is an increase of antibacterial activity by increased levels of VCOH and PKOH. Antibacterial activity against S. aureus (Gram positive) was higher than S. thypi and E. coli (Gram negative). Antibacterial activity of the test material is lower than the standard chloramphenicol and tetracycline .

Many species of the wild herbs are well known used as traditional medicine not only by local people but also by people out of the area. The methanol extract of the medicinal wild herbs in Tangkahan Conservation Forest, Karo County to relief skin diseases caused by bacteria and fungi never been studied medically. The antimicrobial activity leave extract of the medicinal herbs to pathogenic microorganisms are studied. The leaves extract of kembu-kembu (Callicarpa candicans), rintih bulung (Piper muricatum), cep-cepan (Castanopsis costata), and sereh kayu (Eugenia grandis), has antimicrobial to bacteria (Bacillus sp., Escherichia coli, Serratia marcescens, Staphylococcus aureus) and yeast (Candida albicans). Toxicity assay of these plants by brine shrimp method using Artemia salina indicates that cep-cepan dan sereh kayu have lethal concentration higher than kembu-kembu and rintih bulung.

Isolation of gambir endophytic bacteria was done in nutrient agar incubated at ambient temperature for 48 hours. Samples of gambir leaves and root was used for source of endophytic bacteria. Bacterial isolate was propagated using Mueller Hinton Agar. Bacterial culture was extracted using methanol as solvent. To test methanol extract activity of bacterial isolates, concentration of 20, 40, 60, 80, and 100% (v/v) were used in paper disc. Keratinase is a proteolytic enzyme capable on degrading the hardy polymeric biomolecule or kerati... more Keratinase is a proteolytic enzyme capable on degrading the hardy polymeric biomolecule or keratin. In recent days, the utilization of keratinolytic microorganisms is seen as a promising way in recycling the keratin wastes from the avian and mammalians into valuable derived products. Previous study has reported the presence of a keratinolytic bacterium, later identified as Azotobacter chroococcum B4 obtained from dump soils. The present study investigates the enzyme characteristics of keratinase produced by this strain based on the physical appearance of final degraded product using SEM, the molecular weight of keratinase using SDS-PAGE, the effects of nutrition (C/N-source) on strain production of keratinase and the enzyme stability in metal ions solution. The molecular weight of keratinase produced by A. chroococcum B4 was about 30 kDa. Both sucrose and tryptone supplementation increase the keratinase activity by 71.7 and 97.8 U/mL after 96 h of cultivation. Metal ions, Ca 2+ , Mg 2+ , Mn 2+ , Na + , and K + are regarded as activators by increasing the relative activity of keratinase by 117, 166, 111, 113, and 112% respectively, while phenylmethylsulfonyl flouride (PMSF) is regarded as inhibitor by decreasing the relative activity down to 31%. Based on the metal ion characteristics, this strain produced a serine-protease type of keratinase which may further studied for its application in the field.

Dimethyl sulfoxide was used as negative control, and standard antibiotic Kalmicetine (chloramphenicol) was used as positive control. Result: The ethyl acetate fraction showed the highest antibacterial activity against S. aureus and P. aeruginosa. Conclusion: Karamunting leaf can be used to treat the bacterial infection diseases especially infection from S. aureus and P. aeruginosa.

Exploration of probiotics candidate have been accelerated from unique and novel source, one of wh... more Exploration of probiotics candidate have been accelerated from unique and novel source, one of which is from black soldier fly (Hermetia illucens). The black soldier flies are known as omnivores which are commonly reared, harvested and utilized to convert organic wastes into biomass yet reducing the amount of abundant waste in nature. Lactic acid bacteria (LAB) is one of beneficial gut symbionts which can be found across animal taxa, including insect, however, limited information is currently reported from H. illucens as a potential harboring insect. This study aimed to determine the probiotics candidate from the gut of H. illucens fed with different feeding substrates to improve the gut microbiota. The larvae were nourished with chicken manure, tofu solid wastes and vegetable wastes prior isolation of LAB. The LAB isolates were tested for their antagonistic activity against enteric pathogens, pH and bile salt tolerance, and adherence to chicken intestine (ileum). Based on the growth performance under physiological stresses and multivariate analysis using principal component analysis (PCA), four LAB isolates were designated as putative probiotics. The four isolates were Lactiplantibacillus. pentosus 5P2i1, Lacticaseibacillus. paracasei 5P2i5, Lactiplantibacillus. pentosus 5P2i9, and Lactiplantibacillus. plantarum 6P1i1 was then declared as novel strains that could be developed further as poultry probiotics.

In dealing with this problem, studies were carried out, one of which was by utilizing natural medicinal ingredients such as bawang sabrang (Eleutherine palmifolia Merr.) of the Iridaceae family. This study aims to determine the antibacterial activity of the residual fraction of the ethanolic extract of bawang sabrang bulbs (Eleutherine palmifolia Merr.) against gram-positive and gram-negative bacteria and to determine the class of chemical compounds. Extraction was carried out by maceration using ethanol as a solvent and continued by successive fractionation using n-hexane and ethylacetate solvents, so that the remaining fraction was obtained. Antibacterial activity testing was carried out in vitro using the agar diffusion method using metal rings at concentrations of 20 mg/ml, 10 mg/ml and 5 mg/ml against gram-positive bacteria (Bacillus pumilus ATCC 7061, Bacillus subtilis ATCC 6633, Methicillin-resistant Staphylococcus aureus (MRSA) ATCC 25923, Micrococcus luteus ATCC 9341, Propionibacterium acne ATCC 6918, Staphylococcus aureus ATCC 29737, Staphylococcus epidermidis ATCC 12228) and gram-negative bacteria (Klebsiella pneumoniae ATCC 10031, Escherichia coli ATCC 10536, Salmonella thypi ATCC 29213). The analysis of the chemical compound group of the residual fraction was carried out using a phytochemical screening technique. The residual fraction of the ethanolic extract of bawang sabrang bulbs had antibacterial activity on all the tested bacteria. The chemical compounds contained in the remaining fraction of the ethanolic extract of bawang sabrang bulbs are alkaloids, flavonoids, glycosides, saponins, anthraquinone glycosides, tannins, triterpenoids, and steroids.

Bioassay of these isolates to larva of Heliothis armigera, Plutella xylostella, Aedes aegypti, and Culex sp. showed that the isolates have different spectrum and ability in controlling insect larvae. Interestingly, TU1 has similar ability to that of Dipel (B. thuringiensis var kurstaki strain HD-7) in controlling Dipteran (A. aegypti and Culex sp.) and Lepidopteran (H. armigera and P. xylostella). The others were not effective to Dipteran, but have various abilities to kill Lepidopteran. This result indicated that local isolates of B. thuringiensis of North Sumatra were varied in ability to kill different larvae.

Examination of antimicrobial activities to the microbes were done by agar diffusion method, while test for toxicity was done by exposing 2 days-old larvae to certain concentration of the extract. Preliminary tests on phytochemical compounds in leaf were done by Mayer’s, FeCl3, Mg-HCl, foam, and 10% NaOH in methanol tests. Wastewater of textile industry is considered as toxic waste containing various dyes and chemicals... more Wastewater of textile industry is considered as toxic waste containing various dyes and chemicals that makes it recalcitrants. Decolorization of wastewater of the industry was conducted in our laboratorium using fungi isolated from a local forest of North Sumatera. Six potential isolates, previously screened for the ability to grow in plate media containing 100% of wastewater, were used for the study in a liquid culture condition. The results show that all tested isolates grew. The growth response of isolates cultured in a media supplemeted with 1% glucose was about two to three times higher than that of cultures without glucose. In the first culture, the fungi grew on the surface of the media however in the latter experiment, it concentrated on the bottom of culture flask for all of tested isolates. Decolorization analysis shows that all isolates were able to decolorize the wastewater. The cultures supplemented with glucose decolorized wastewater from 20% to 46%, while cultures without glucose did much higher about 56% to 92%. Furthermore, a microscopic analysis shows that dyes molecules in the wastewater were absorbed to fungal mycelia. The findings show that extraceluler molecules existed on the surface of fungal mycelia play an important role in the absorption of dye molecules.

The purpose of this study was to determine the antibacterial activity of the enzymatic hydrolyzed of virgin coconut oil (VCO) and palm kernel oil (PKO), which produce a combination of lauric acid and monolaurin. Hydrolysis process was done by the enzyme lipase (Lipozym TL IM) which was active at 1.3 position. The hydrolyzed oil then tested for antibacterial activity against Staphylococcus aureus, Escherichia coli and Salmonella thypi using agar diffusion method with paper disc with diameter of 6 mm (Oxoid, England). Antibacterial activity test carried out on VCO and PKO, hydrolyzed VCO (VCOH) and PKO (PKOH) each at 25% concentration, 50%, 75% and 100%. Antibacterial activity of the test material compared with chloramphenicol (30 µg) and tetracycline (30 µg). The results showed that the optimum hydrolysis time is 14 hours. There is low and similar antibacterial activity of VCO and PKO. VCOH and PKOH show higher antibacterial activity than VCO and PKO. There is an increase of antibacterial activity by increased levels of VCOH and PKOH. Antibacterial activity against S. aureus (Gram positive) was higher than S. thypi and E. coli (Gram negative). Antibacterial activity of the test material is lower than the standard chloramphenicol and tetracycline .

The methanol extract of the medicinal wild herbs in Tangkahan Conservation Forest, Karo County to relief skin diseases caused by bacteria and fungi never been studied medically. The antimicrobial activity leave extract of the medicinal herbs to pathogenic microorganisms are studied. The leaves extract of kembu-kembu (Callicarpa candicans), rintih bulung (Piper muricatum), cep-cepan (Castanopsis costata), and sereh kayu (Eugenia grandis), has antimicrobial to bacteria (Bacillus sp., Escherichia coli, Serratia marcescens, Staphylococcus aureus) and yeast (Candida albicans). Toxicity assay of these plants by brine shrimp method using Artemia salina indicates that cep-cepan dan sereh kayu have lethal concentration higher than kembu-kembu and rintih bulung.

In recent days, the utilization of keratinolytic microorganisms is seen as a promising way in recycling the keratin wastes from the avian and mammalians into valuable derived products. Previous study has reported the presence of a keratinolytic bacterium, later identified as Azotobacter chroococcum B4 obtained from dump soils. The present study investigates the enzyme characteristics of keratinase produced by this strain based on the physical appearance of final degraded product using SEM, the molecular weight of keratinase using SDS-PAGE, the effects of nutrition (C/N-source) on strain production of keratinase and the enzyme stability in metal ions solution. The molecular weight of keratinase produced by A. chroococcum B4 was about 30 kDa. Both sucrose and tryptone supplementation increase the keratinase activity by 71.7 and 97.8 U/mL after 96 h of cultivation. Metal ions, Ca 2+ , Mg 2+ , Mn 2+ , Na + , and K + are regarded as activators by increasing the relative activity of keratinase by 117, 166, 111, 113, and 112% respectively, while phenylmethylsulfonyl flouride (PMSF) is regarded as inhibitor by decreasing the relative activity down to 31%. Based on the metal ion characteristics, this strain produced a serine-protease type of keratinase which may further studied for its application in the field.
