Background and Aim: Bali-polled bulls, known for their favorable traits, require advanced reprodu... more Background and Aim: Bali-polled bulls, known for their favorable traits, require advanced reproductive performance analysis to optimize breeding programs. This study aimed to evaluate sperm kinematics and seminal plasma proteomic profiles as biomarkers for sperm motility and fertility in Bali-polled bulls.

Materials and Methods: Semen from five Bali-polled bulls was collected biweekly over five batches using artificial vaginas. Sperm kinematics were assessed using computer-assisted semen analysis (CASA). Fertility was evaluated through service per conception (S/C) in artificial insemination trials. Seminal plasma proteins were analyzed through liquid chromatography–tandem mass spectrometry (LC-MS/MS) and annotated using the UniProt database, PANTHER for gene ontology, and STRING database for protein interactions.

Results: Post-thaw sperm kinematics showed satisfactory results, with a mean S/C of 1.52. Proteomic analysis identified 138 proteins, including six (TEX101, BSP1, PRSS55, BSP3, SPADH2, and TPPP2) linked to sperm motility. These proteins were involved in key biological processes such as sperm capacitation, motility regulation, and sperm-oocyte interaction.

Conclusion: Sperm kinematics and seminal plasma proteomics provide insights into Bali-polled bull fertility. Identified proteins can serve as fertility biomarkers, aiding in superior local breed development and reproductive efficiency improvement.

Telah dilakukan suatu penelitian untuk mengidentifikasi variasi gen myostatin dan mengevaluasi ke... more Telah dilakukan suatu penelitian untuk mengidentifikasi variasi gen myostatin dan mengevaluasi keberadaan marker genetik untuk sifat double muscling (delesi 11 basa pada ekson 3 gen myostatin) pada generasi pertama (F1) hasil persilangan dengan sapi Belgian Blue pertama di Indonesia. Sebanyak 8 sampel DNA sapi yang dipelihara di PT. Karya Anugerah Rumpin (KAR) digunakan dalam penelitian ini. Analisa Single Strand Conformation Polymorphism (SSCP) dan sekuensing digunakan untuk mengidentifikasi keragaman dari gen myostatin. Terdapat 3 tipe alel gen myostatin berdasarkan hasil analisa SSCP. Pejantan Belgian Blue memiliki tipe alel A. Sapi Simmental, Wagyu, SO x BX, Charolais, dan sapi PO memiliki tipe alel B. Sementara itu anak sapi hasil perkawinan Belgian Blue x FH dan Belgian Blue x SO memiliki tipe alel C (heterozigot). Berdasarkan hasil analisa sekuen gen myostatin, ditemukan adanya delesi 11 basa pada ekson 3 gen myostatin pejantan Belgian Blue. Hasil analisa sekuensing juga memberikan informasi bahwa generasi F1 (Belgian Blue x FH dan Belgian Blue x SO) memiliki gen myostatin yang berada dalam kondisi heterozigot. Hasil penelitian ini memberikan bukti ilmiah bahwa delesi 11 basa pada ekson 3 gen myostatin yang merupakan marker untuk sifat double muscling pada sapi Belgian Blue juga ada (diwariskan) pada anak-anak sapi generasi F1 (jantan dan betina).

Penelitian ini bertujuan untuk menentukan angka kebuntingan dan kelahiran sapi Bali sesudah sinkr... more Penelitian ini bertujuan untuk menentukan angka kebuntingan dan kelahiran sapi Bali sesudah sinkronisasi estrus dan inseminasi buatan (IB) dengan semen beku sexing serta kesesuaian jenis kelamin anak. Penelitian dilakukan pada dua lokasi peternakan di Kabupaten Lombok Barat. Sinkronisasi estrus dilakukan melalui pemberian satu suntikan 5 cc Lutalyse ® yang mengandung 25 mg PGF 2α (Lutalyse, Sterile Solution, Pharmacia Animal Health) secara intra muscular. Pengamatan dilakukan pada sapi yang memberikan respon dan diinseminasi pada saat estrus. IB juga langsung dilakukan pada sapi-sapi yang menunjukkan birahi alami. Diagnosa kebuntingan dilakukan melalui palpasi rektal pada hari ke-90. Pada Peternakan 1, angka kebuntingan sapi-sapi yang mendapat perlakuan dan yang tidak mendapat perlakuan PGF 2α masing-masing sebesar 29/63 (46,03%) dan 71/75 (94,66%). Pada Peternakan 2, nilainilai yang bersesuaian masing-masing adalah 38/89 (42,70%) dan 10/23 (43,48%). Angka kelahiran anak pada kedua farm diperoleh 100%. Angka kesesuaian jenis kelamin untuk anak betina yang lahir di Peternakan 1 adalah 94.00%, sedangkan di Peternakan 2, untuk anak sapi yang terlahir jantan adalah 93.75%. Dibawah kondisi penelitian ini, sinkronisasi estrus yang diikuti dengan IB semen beku hasil sexing tampaknya dapat diaplikasikan di lapang sehingga menghasilkan angka kebuntingan dan kelahiran anak yang pantas diterima, serta angka kesesuaian di atas 90% untuk masing-masing jenis kelamin betina maupun jantan. The presented study was aimed at determining the association of growth hormone (GH) gene polymorp... more The presented study was aimed at determining the association of growth hormone (GH) gene polymorphism with the traits that characterise carcass and meat quality and meat basic chemical composition in porkers with genotype CC with respect to RYR1 locus. The study was carried out on 126 PIC porkers originating after Camborough 22 sows and PIC 337 boars. The polymorphism of GH gene was identified respectively with restrictive enzymes MspI, HaeII, CfoI and ApaI. The allele frequencies were as follows: GH/MspI A = 0.78 and B = 0.22; GH/HaeII A = 0.86 and B = 0.14; GH/CfoI C1 = 0.09 and C2 = 0.03 and C3 = 0.18 and C4 = 0.70; GH/ApaI A1 = 0.28 and A2 = 0.72. The analysis of association of GH genotypes identified with four restrictive enzymes did not show any relationship between the GH gene polymorphism and the carcass slaughter traits determined with meatiness measuring apparatus CGM and the meat quality and meat basic chemical composition in PIC hybrid porkers. This research had two steps experimental, the first steps experiment to determine the nutrient content and digestibility on wafer of feed supplement. The composition wafer of feed supplement i.e. T1 = wafer containing lamtoro leaf, T2 = wafer containing lamtoro and papaya leaf, T3 = wafer containing moringa leaf, T4 = wafer containing gamal leaf, T5= wafer containing corn leaf and corn. Nutrient content and digestibility of T1 had highest compared among the others, i.e. crude protein (32.34%), ash (7.24%), crude fiber (16.85%), crude fat (4.52%), NFE (39.06 cal/g), digestibility of dry matter 82.87%, digestibility of organic matter 81.78%, NH3 9.33% and VFA 164.55%. A performance test was conducted using 25 males and 25 females of Sumba Ongole (SO) cattle. The performance test were conducted in three periods (2014, 2015 and 2016) for about 535 days. Descriptive statistics ws used to analyze data of body weight. Cattle with age between 300 to 600 days were used for evaluation. Result showed that heritability value of yearling weight (YW365) was 0.77+0.68 as a high category. The highest breeding value of YW365 were 66.05 kg (male) and 41.89 kg (female). The average of corrected final weight (CFW) weight were 172.55+34.22 kg (male) and 159.80+37.73 kg (female). The performance test in this study obtained six A class (1 bull and 5 cows) based on the standard minimum of body measurements for SO cattle.

The Thyroglobulin (TG) and Leptin (LEP) genes are two candidate genes that widely used for molecu... more The Thyroglobulin (TG) and Leptin (LEP) genes are two candidate genes that widely used for molecular selection to improve carcass traits in beef cattle. This research was carried out to identify the genetic characterization of TG and LEP genes from 47 heads of Pasundan cows at West Java using PCR-RFLP method. Research shows that TG gene of Pasundan cattle is monomorphic with C allele as the dominant allele (1.00). However, LEP gene of Pasundan cattle is polymorphic with C allele as the dominant allele (0.98) and T as the rare allele (0.02). The polymorphic informative content (PIC) and numberof effective allele (ne) values in the LEP gene in the animal studied were 0.04 and 1.04 respectively. It was concluded that TG/BstYI and LEP/Sau3AI gene in the present study can not be used as molecular selection in Pasundan cattle. These results are important as the basic information for preparing the molecular selection program in the future.

However, the standard of desirable birth weight in cattle during the selection program is important to prevent dystocia incident risk. This study was aimed to select Bali cattle (Bos javanicus) based on Estimated Breeding Value of birth weight (EBVBW) and Most Probable Producing Ability of birth weight (MPPABW) and calving interval (MPPACI). Total of 758 records data of BW were collected from Lombok and Sumbawa islands, West Nusa Tenggara Province. Research showed the average of BW in Bali calves were 15.69±1.70 kg (Lombok) and 13.49±1.89 (Sumbawa). The average of CI in Bali cows at both islands were about 385 days. In addition, the heritability (h2) values of BW in both islands was about 0.90. The repeatability (r) values of BW in both islands were about 0.30. Hasil analisa sekuensing juga memberikan informasi bahwa generasi F1 (Belgian Blue x FH dan Belgian Blue x SO) memiliki gen myostatin yang berada dalam kondisi heterozigot. Hasil penelitian ini memberikan bukti ilmiah bahwa delesi 11 basa pada ekson 3 gen myostatin yang merupakan marker untuk sifat double muscling pada sapi Belgian Blue juga ada (diwariskan) pada anak-anak sapi generasi F1 (jantan dan betina).

Penelitian ini bertujuan untuk menentukan angka kebuntingan dan kelahiran sapi Bali sesudah sinkr... more Penelitian ini bertujuan untuk menentukan angka kebuntingan dan kelahiran sapi Bali sesudah sinkronisasi estrus dan inseminasi buatan (IB) dengan semen beku sexing serta kesesuaian jenis kelamin anak. Penelitian dilakukan pada dua lokasi peternakan di Kabupaten Lombok Barat. Sinkronisasi estrus dilakukan melalui pemberian satu suntikan 5 cc Lutalyse ® yang mengandung 25 mg PGF 2α (Lutalyse, Sterile Solution, Pharmacia Animal Health) secara intra muscular. Pengamatan dilakukan pada sapi yang memberikan respon dan diinseminasi pada saat estrus. IB juga langsung dilakukan pada sapi-sapi yang menunjukkan birahi alami. Diagnosa kebuntingan dilakukan melalui palpasi rektal pada hari ke-90. Pada Peternakan 1, angka kebuntingan sapi-sapi yang mendapat perlakuan dan yang tidak mendapat perlakuan PGF 2α masing-masing sebesar 29/63 (46,03%) dan 71/75 (94,66%). Pada Peternakan 2, nilainilai yang bersesuaian masing-masing adalah 38/89 (42,70%) dan 10/23 (43,48%). Angka kelahiran anak pada kedua farm diperoleh 100%. Angka kesesuaian jenis kelamin untuk anak betina yang lahir di Peternakan 1 adalah 94.00%, sedangkan di Peternakan 2, untuk anak sapi yang terlahir jantan adalah 93.75%. Dibawah kondisi penelitian ini, sinkronisasi estrus yang diikuti dengan IB semen beku hasil sexing tampaknya dapat diaplikasikan di lapang sehingga menghasilkan angka kebuntingan dan kelahiran anak yang pantas diterima, serta angka kesesuaian di atas 90% untuk masing-masing jenis kelamin betina maupun jantan. The study was carried out on 126 PIC porkers originating after Camborough 22 sows and PIC 337 boars. The polymorphism of GH gene was identified respectively with restrictive enzymes MspI, HaeII, CfoI and ApaI. The allele frequencies were as follows: GH/MspI A = 0.78 and B = 0.22; GH/HaeII A = 0.86 and B = 0.14; GH/CfoI C1 = 0.09 and C2 = 0.03 and C3 = 0.18 and C4 = 0.70; GH/ApaI A1 = 0.28 and A2 = 0.72. This research had two steps experimental, the first steps experiment to determine the nutrient content and digestibility on wafer of feed supplement. The composition wafer of feed supplement i.e. T1 = wafer containing lamtoro leaf, T2 = wafer containing lamtoro and papaya leaf, T3 = wafer containing moringa leaf, T4 = wafer containing gamal leaf, T5= wafer containing corn leaf and corn. Nutrient content and digestibility of T1 had highest compared among the others, i.e. crude protein (32.34%), ash (7.24%), crude fiber (16.85%), crude fat (4.52%), NFE (39.06 cal/g), digestibility of dry matter 82.87%, digestibility of organic matter 81.78%, NH3 9.33% and VFA 164.55%. A performance test was conducted using 25 males and 25 females of Sumba Ongole (SO) cattle. The performance test were conducted in three periods (2014, 2015 and 2016) for about 535 days. Descriptive statistics ws used to analyze data of body weight. Cattle with age between 300 to 600 days were used for evaluation. Result showed that heritability value of yearling weight (YW365) was 0.77+0.68 as a high category. The highest breeding value of YW365 were 66.05 kg (male) and 41.89 kg (female). The average of corrected final weight (CFW) weight were 172.55+34.22 kg (male) and 159.80+37.73 kg (female). The performance test in this study obtained six A class (1 bull and 5 cows) based on the standard minimum of body measurements for SO cattle.

This research was carried out to identify the genetic characterization of TG and LEP genes from 47 heads of Pasundan cows at West Java using PCR-RFLP method. Research shows that TG gene of Pasundan cattle is monomorphic with C allele as the dominant allele (1.00). However, LEP gene of Pasundan cattle is polymorphic with C allele as the dominant allele (0.98) and T as the rare allele (0.02). The polymorphic informative content (PIC) and numberof effective allele (ne) values in the LEP gene in the animal studied were 0.04 and 1.04 respectively. It was concluded that TG/BstYI and LEP/Sau3AI gene in the present study can not be used as molecular selection in Pasundan cattle. These results are important as the basic information for preparing the molecular selection program in the future.

However, the standard of desirable birth weight in cattle during the selection program is important to prevent dystocia incident risk. This study was aimed to select Bali cattle (Bos javanicus) based on Estimated Breeding Value of birth weight (EBVBW) and Most Probable Producing Ability of birth weight (MPPABW) and calving interval (MPPACI). Total of 758 records data of BW were collected from Lombok and Sumbawa islands, West Nusa Tenggara Province. Research showed the average of BW in Bali calves were 15.69±1.70 kg (Lombok) and 13.49±1.89 (Sumbawa). The average of CI in Bali cows at both islands were about 385 days. In addition, the heritability (h2) values of BW in both islands was about 0.90. The repeatability (r) values of BW in both islands were about 0.30.
