6-Phosphofructokinase purified from honey-bee flight muscle is inhibited by ATP and, unusually, b... more 6-Phosphofructokinase purified from honey-bee flight muscle is inhibited by ATP and, unusually, by glucose 1,6-bisphosphate and fructose 1,6-bisphosphate. The inhibition by either of the bisphosphates is not relieved by AMP, but is relieved by fructose 6-phosphate and especially by fructose 2,6-bisphosphate. Lack of effect by AMP is consistent with a low activity of adenylate kinase in this muscle.

Arginine and creatine kinase activities in different muscles are compared with calculated maximum... more Arginine and creatine kinase activities in different muscles are compared with calculated maximum rates of ATP turnover. The magnitude of the kinase activities decreases in the following order: anaerobic muscles and vertebrate skeletal muscles greater than heart muscle greater than insect flight muscle. The maximum activity of phosphagen kinases (i.e. creatine kinase and arginine kinase), in the direction of phosphagen formation, is lower than the calculated maximum rate of ATP turnover in insect flight muscle or rat heart.

IN October 2006, one of the authors (A. G. Irwin) was contacted by a woman whose nine-year-old co... more IN October 2006, one of the authors (A. G. Irwin) was contacted by a woman whose nine-year-old cocker spaniel had died after apparently consuming fungus fruiting bodies from the garden lawn. The owner had collected a sample of the dog's vomit, which clearly showed fragments of undigested fungus. Fungi collected from the lawn included fool's funnel ( Clitocybe rivulosa ), a species known to cause poisoning in humans. Examination of the ingested fragments under the microscope revealed distinctive spores that matched those of C rivulosa . The time between ingestion and the onset of clinical signs is not known but within 45 minutes of the dog displaying salivation, vomiting and diarrhoea it had collapsed and died. Simple and specific radiochemical asays for adenylate kinase and AMP deaminase activities in crud... more Simple and specific radiochemical asays for adenylate kinase and AMP deaminase activities in crude tissue extracts are described. The radioactive substrate (AMP) is separated from the radioactive product (ADP or IMP) by chromatography on polyethyleneimine-cellulose thin layers. A rapid modification of the adenylate kinase assay is described in which samples of the reaction mixture are transferred directly to the polyethyleneimine-cellulose thin layer.

Simple and specific radiochemical assays for the determination of creatine and arginine kinase ac... more Simple and specific radiochemical assays for the determination of creatine and arginine kinase activities in crude tissue extracts are described. Creatine kinase is assayed by incubation with radioactive creatine and subsequent determination of the radioactivity in creatine phosphate. Creatine and creatine phosphate are separated on DEAE-cellulose ion exchange papers. Arginine kinase is assayed similarly by using radioactive arginine and separating it from arginine phosphate on short ion exchange columns. An assay for creatine kinase in the direction of creatine formation is also described.
