CCR7 is involved in the initiation of immune responses and has been recently implicated in the co... more CCR7 is involved in the initiation of immune responses and has been recently implicated in the control of tolerance. To analyze the role of CCR7 in autoimmunity, we backcrossed CCR7 ko/ko mice (in which ko signifies deficient) onto the autoimmune-prone NOD background. Surprisingly, NODCCR7 ko/ko mice never developed diabetes, but showed severe inflammation in multiple tissues including thyroid, lung, stomach, intestine, uterus, and testis. NODCCR7 ko/ko mice had a marked enlargement of the thyroid gland (goiter) that was associated with circulating autoantibodies against thyroglobulin, and development of primary hypothyroidism (decreased levels of serum thyroxin, and augmented levels of thyroid-stimulating hormone in the pituitary gland), features found in Hashimoto's thyroiditis. Cells isolated from diseased thyroids and activated splenocytes from NODCCR7 ko/ko animals induced goiter in NOD.SCID recipients, demonstrating that autoreactive cells were generated in the absence of CCR7. Moreover, thyroid disease could be accelerated in young NODCCR7 ko/ko mice by immunization with thyroglobulin. These results demonstrate the complexity in the generation of multiple autoimmune phenotypes in NOD mice and indicate that CCR7 is a key molecule in their development.

A kinetic spectrophotometric method for determining the residues of herbicide bromofenoxim (BrFX)... more A kinetic spectrophotometric method for determining the residues of herbicide bromofenoxim (BrFX) has been developed and validated. The proposed method is based on the inhibitory effect of BrFX on the oxidation of sulfanilic acid (SA) by hydrogen peroxide in the presence of Cu(II) ion, which was monitored at 370 nm. The variables affecting the rate of the reaction were investigated and the optimum conditions were established. BrFX can be measured in the range of 0.041-0.46 μg/ml and 0.46-13.86 μg/ml. The detection limit of the method with 3σ criteria is 0.0077 μg/ml. The relative standard deviations for five replicate determinations of 0.041, 0.24 and 0.46 μg/ml BrFX are 3.0, 5.32 and 2.85%, respectively. This method can be successfully used to determine BrFX concentration in baby juice samples. The HPLC method is used to verify the results. The results obtained for the same samples by the two methods are quite comparable.

Inequality of information among various participants is assumed as well as the existence of different incentives. The paper argues that regulation in general and a safety net in particular are necessary to make the financial service industry operate in a safe and appropriate way. From this vantage point, transition banking and, more closely, peculiarities of the Serbian system are analysed. Some guidelines for redesigning the existing safety net in Serbian banking are proposed.

A kinetic spectrophotometric method for measuring 2,4-dichlorphenoxyacetic acid (2,4-D) is descri... more A kinetic spectrophotometric method for measuring 2,4-dichlorphenoxyacetic acid (2,4-D) is described in this study. The proposed method is based on the inhibitory effect of 2,4-D on the oxidation of sulfanilic acid by hydrogen peroxide in the presence of Co(II) ion, which was monitored at 370 nm. The variables affecting the rate of the reaction were investigated and the optimum conditions were established. 2,4-D can be measured in the range of 0.07-2.35 μg mL-1 and from 2.35 to 23.50 μg mL-1 with the detection limit of 0.004 μg mL-1. This method was used to determined traces of 2,4-D in baby fruit juices. The HPLC method was used like a comparative method to verify the results. The results obtained by the proposed method were compared with those obtained by HPLC using UV-DAD detection and they showed a good agreement.

It is well documented that deregulation of MYC leads to tumor development, yet many aspects of th... more It is well documented that deregulation of MYC leads to tumor development, yet many aspects of this process are only partially understood. We have established a transgenic mouse model in which c-MYC is conditionally expressed in lymphoid cells using the tetracycline-regulated system of gene regulation. Mice with continuously expressed transgenic c-MYC died of invasive T- or B-cell lymphomas within 4 months. Lymphomas developing in transgenic mice were c-MYC dependent since doxycycline treatment led to tumor regression. Using transplantation of established tumor cell lines labeled with GFP, we followed the fate of neoplastic cells in recipients upon MYC inactivation. This approach allowed us to elucidate both apoptosis and differentiation as mechanisms of tumor elimination. Comparative genomic hybridization (CGH) and FISH analyses were performed in order to analyze possible chromosomal aberrations induced by c-MYC. We observed that overexpression of c-MYC is sufficient to induce recurrent patterns of genomic instability. The main observation was a gain of genomic material that corresponded to chromosome 15 in several T-cell tumors, which could be identified as trisomy.

Deletion of the inhibitor of differentiation 2 (Id2) gene, essential for generation of CD3-CD4 + lymphoid tissue-inducer (LTi) cells and development of secondary lymphoid organs, did not affect formation of tertiary lymphoid structures. Rather, mature CD3 + CD4 + T cells were critical for the development of tertiary lymphoid structures. The initial stages of this process involved interaction of CD3 + CD4 + T cells with DCs, the appearance of peripheral-node addressin-positive (PNAd +) vessels, and production of chemokines that recruit lymphocytes and DCs. These findings indicate that the formation of tertiary lymphoid structures does not require Id2-dependent conventional LTis but depends on a program initiated by mature CD3 + CD4 + T cells.
