By utilizing optimal conditions for maximizing siderophore production in shake flask cultures of P. fluorescens, we report successful characterization of the culture broth supernatant as a robust ferric ions biosensor. For characterizing the ferric ions biosensor, we tested the effects of pH, buffers, different ferric salts and possible interference by ferrous ions under different solution conditions. We find that the biosensor is very specific to ferric ions only with sensitivity to concentrations as low as 10 microM. Further, the response time of the biosensor is the shortest (approximately 5 min or smaller) for citrate as the accompanying anion with ferric ions. While the response time is longer than that expected of normal biosensors, it is well compensated by the simplicity and economics of the biosensor production. Extremely low standard deviations in several experimental repeats also highlight the robustness of the ferric ions biosensor. Most importantly, the biosensor is extremely easy to use due to its straightforward spectrophotometric applications. We also show the utility of the biosensor with the high resolution technique of fluorescence microscopy. Finally, we report a novel mechanistic finding that siderophores present in the culture broth supernatants have two distinct optically active sites on them, which can be monitored independently in presence or absence of ferric ions.

The electronic spectrum of a cold molecular beam of zirconium dioxide, ZrO(2), has been investiga... more The electronic spectrum of a cold molecular beam of zirconium dioxide, ZrO(2), has been investigated using laser induced fluorescence (LIF) in the region from 17,000 cm(-1) to 18,800 cm(-1) and by mass-resolved resonance enhanced multi-photon ionization (REMPI) spectroscopy from 17,000 cm(-1)-21,000 cm(-1). The LIF and REMPI spectra are assigned to progressions in the Ã(1)B(2)(ν(1), ν(2), ν(3)) ← X̃(1)A(1)(0, 0, 0) transitions. Dispersed fluorescence from 13 bands was recorded and analyzed to produce harmonic vibrational parameters for the X̃(1)A(1) state of ω(1) = 898(1) cm(-1), ω(2) = 287(2) cm(-1), and ω(3) = 808(3) cm(-1). The observed transition frequencies of 45 bands in the LIF and REMPI spectra produce origin and harmonic vibrational parameters for the Ã(1)B(2) state of T(e) = 16,307(8) cm(-1), ω(1) = 819(3) cm(-1), ω(2) = 149(3) cm(-1), and ω(3) = 518(4) cm(-1). The spectra were modeled using a normal coordinate analysis and Franck-Condon factor predictions. The structures, harmonic vibrational frequencies, and the potential energies as a function of bending angle for the Ã(1)B(2) and X̃(1)A(1) states are predicted using time-dependent density functional theory, complete active space self-consistent field, and related first-principle calculations. A comparison with isovalent TiO(2) is made.

The grouping of correlated classes into a package helps in better organization of modern object-o... more The grouping of correlated classes into a package helps in better organization of modern object-oriented software. The quality of such packages needs to be measured so as to estimate their utilization. In this paper, new package coupling metrics are proposed, which also take into consideration the hierarchical structure of packages and direction of connections among package elements. The proposed measures have been validated theoretically as well as empirically using 18 packages taken from two open source software systems. The results obtained from this study show strong correlation between package coupling and understandability of the package which suggests that proposed metrics could be further used to represent other external software quality factors.
