To facilitate automated validation of participant self-reports of meal timing, the present study focuses on the creation of a meal detection algorithm using continuous glucose monitoring (CGM), physiological monitors and machine learning. While most CGM-related studies focus on participants who are diabetic, this study is the first to apply machine learning to meal detection using CGM in metabolically healthy adults. Furthermore, the results demonstrate a high area under the receiver operating characteristic curve (AUC-ROC) and precision-recall curve (AUC-PR). A cold-start simulation using a random forest algorithm yields .891 and .803 for AUC-ROC and AUC-PR respectively on 110minutes data, and a non-cold start simulation using a gradient boosted tree model yields over .996 (AUC-ROC) and .964 (AUC-PR). Here it is demonstrated that CGM and physiological monitoring data is a viable tool for practitioners and scientists to objectively validate self-reports of meal consumption in healthy participants.

The mechanisms that modulate and limit the signaling output of adult stem cell niches remain poor... more The mechanisms that modulate and limit the signaling output of adult stem cell niches remain poorly understood. To gain further insights into how these microenvironments are regulated in vivo, we performed a candidate gene screen designed to identify factors that restrict BMP signal production to the cap cells that comprise the germline stem cell (GSC) niche of Drosophila ovaries. Through these efforts, we found that disruption of Wnt4 and components of the canonical Wnt pathway results in a complex germ cell phenotype marked by an expansion of GSC-like cells, pre-cystoblasts and cystoblasts in young females. This phenotype correlates with an increase of decapentaplegic (dpp) mRNA levels within escort cells and varying levels of BMP responsiveness in the germline. Further genetic experiments show that Wnt4, which exhibits graded expression in somatic cells of germaria, activates the Wnt pathway in posteriorly positioned escort cells. The activation of the Wnt pathway appears to be l...

However, more recently, Ding et al. (2012) have shown that specific types of lies (concealment vs. faking lies) have distinct neural correlates. Using Near-Infrared Spectroscopy, we examined whether action-vs. speech-based lies also have distinct neural correlates. Participants had three sessions: learning, recognition, and lying. In the learning session, participants produced a hand gesture or spoke aloud 20 sentences, respectively. In the recognition session, participants recognized whether they spoke or gestured a sentence via a key press. In the lying session, participants lied for 20 sentences and told the truth for another 20 sentences about whether they gestured or spoke via a key press. Our results revealed the right superior frontal gyrus was uniquely activated by action lies. This implies certain types of lies such as concealment, faking, and action lies activate unique regions of the brain.

We examined whether the higher-order cognitive functions are involved in the activations in the mirror neuron area when we perceive simplified pseudo-postures. We measured 14 participants' brain activation during the posture-recognition task using near-infrared spectroscopy. The participants' task was to observe five sequentially presented target pseudo-postures and judge whether a test pseudo-posture was identical to one of the preceding five target pseudo-postures. The results in the majority of participants (n = 10/14) revealed that the activity in the inferior frontal mirror neuron area is modulated by perception of human-likeness, but not in the remaining four participants (n = 4/14). These results suggest that the degree of the activation of higherorder cognitive functions, which may be engaged in the inhibitory and/or facilitative processing of human body or bodily movement, leads to the distinctive activities in the inferior frontal mirror neuron area.

However the mechanisms that govern the formation, size and signaling of in vivo niches remain poorly understood. Loss of the highly conserved histone demethylase Lsd1 in Drosophila escort cells results in increased BMP signaling outside the cap cell niche and an expanded germline stem cell (GSC) phenotype. Here we present evidence that loss of Lsd1 also results in gradual changes in escort cell morphology and their eventual death. To better characterize the function of Lsd1 in different cell populations within the ovary, we performed Chromatin immunoprecipitation coupled with massive parallel sequencing (ChIP-seq). This analysis shows that Lsd1 associates with a surprisingly limited number of sites in escort cells and fewer, and often, different sites in cap cells. These findings indicate that Lsd1 exhibits highly selective binding that depends greatly on specific cellular contexts. Lsd1 does not directly target the dpp locus in escort cells. Instead, Lsd1 regulates engrailed expression and disruption of engrailed and its putative downstream target hedgehog suppress the Lsd1 mutant phenotype. Interestingly, over-expression of engrailed, but not hedgehog, results in an expansion of GSC cells, marked by the expansion of BMP signaling. Knockdown of other potential direct Lsd1 target genes, not obviously linked to BMP signaling, also partially suppresses the Lsd1 mutant phenotype. These results suggest that Lsd1 restricts the number of GSC-like cells by regulating a diverse group of genes and provide further evidence that escort cell function must be carefully controlled during development and adulthood to ensure proper germline differentiation.

The main factors that contribute to the nature and special characteristics of sherry vinegar are the raw sherry wine, the traditional process of acetic acid fermentation in butts (the solera system), and the physicochemical activity during the aging process in the solera system. A number of chemical and biochemical changes that occur during sherry vinegar aging are similar to those that take place in sherry wine during its biological activity process (where the wine types obtained are fino and manzanilla) or physicochemical activity process (to give oloroso wines). Significant increase in acetic acid levels was observed during the biological activity phase. In addition, the concentrations of tartaric, gluconic, succinic, and citric acids increased during the aging, as did levels of amino acids and acetoin. A color change was also produced during this stage. Glycerol was not consumed by acetic acid bacteria, and levels of higher alcohols decreased because of the synthesis of acetates. On the other hand, in the physicochemical phase the microbiological activity was lower. Concentrations of some cations increased because of evaporation of water through the wood. A color change was also produced in this stage. Concentrations of different amino acids decreased because of reaction with carbonyl compounds. A precipitation of potassium with tartaric acid was also observed.

Research paper thumbnail of Resveratrol Content of Palomino fino Grapes:  Influence of Vintage and Fungal Infection

The objective of this study was to determine the influence of certain factors on the resveratrol ... more The objective of this study was to determine the influence of certain factors on the resveratrol content of Palomino fino grapes, cultivated in the Jerez-Xé rè z-Sherry area, at the moment of harvest. The results show that the resveratrol content is highly influenced by the climatic conditions prior to the period of maturation of the fruit. On the other hand, the gray mold pressure in the vineyard, a fungal infection caused by Botrytis cinerea, increased the resveratrol contents at the early stages of fungal development. When Botrytis development was extensive, the resveratrol content tended to decrease in the juice but tended to increase in the skin. Physiological stress of the plant leads to increases in the resveratrol content, caused as much by the climatic conditions of the vintage as by biotic factors. In this case resveratrol is present mainly in the glycosylated form.

These precursors, represented fundamentally by the glycosidically-bound aroma compounds, are present in the grape at higher concentrations than the free or volatile forms. Acid hydrolysis of these precursors is not a viable way to produce significant chemical modifications in terpenes, but enzymatic hydrolysis is the best technique to increase the concentration of the free aroma fraction in wines. As a result, current studies are focused on the use of β-glycosidase enzymes to hydrolyze and release these aroma precursors as a way to improve the characteristics and sensory quality of wines. This work describes the influence that the application of `Novarom G' β-glycosidase enzyme has on the aromatic composition of monovarietal wines from four white varieties: Palomino fino, Sauvignon blanc, Traminer, and Viura.

The grapes were cultivated in an experimental viney...
