So, it was intended to assess the antioxidant activity of natural extracts in order to attain their functional quality. It was observed that all the extracts (brown and green propolis, Ginkgo biloba and Isoflavin Beta®) and the standard used (quercetin) showed antioxidant activity in a dose-dependent manner with IC 50 values ranging from 0.21 to 155.28 μg mL-1 (inhibition of lipid peroxidation and scavenging of the DPPH • assays). We observed a high correlation (r 2 = 0.9913) among the antioxidant methods; on the other hand, the antioxidant activity was not related to the polyphenol and flavonoid content. As the DPPH • assay is a fast method, presents low costs and even has a high correlation with other antioxidant methods, it could be applied as an additional parameter in the quality control of natural extracts. RESUMEN. "Evaluación de la actividad antioxidante como parámetro adicional para lograr la calidad funcional de extractos naturales". Debido a las diferencias en la calidad funcional de extractos naturales, nosotros hemos advertido también diferencias en su efectividad, por lo que se pretende estimar la actividad antioxidante de extractos naturales para lograr su calidad funcional. Fue observado que todos los extractos (propolis marrón y verde, Ginkgo biloba e Isoflavin Beta®) y el patrón usado (quercetina) mostraron actividad antioxidante de una manera dosis-dependiente con valores de IC 50 que van de 0,21 a 155,28 al μg mL-1 (inhibición de la peroxidación lipídioca y captación de radicales libres del DPPH •). Se observó una alta correlación (r 2 = 0,9913) entre los métodos antioxidantes y por otro lado la actividad antioxidante no estuvo relacionada con el contenido del polifenoles ni de flavonoides. Como el análisis del DPPH • es un método rápido, presenta costos bajos e incluso tiene una correlación alta con otros métodos antioxidantes, este método podría utilizarse como un parámetro adicional en el control de calidad de extractos naturales.

Esquamosan inhibited the rat aortic ring contraction evoked by phenylephrine in a concentration-dependent manner and showed an inhibitory effect on vasocontraction of the depolarized aorta with high-concentration potassium. The vasorelaxant effect by esquamosan could be attributed mainly to the inhibition of calcium influx from extracellular space through voltage-dependent calcium channels or receptor-operated Ca2+ channels and also partly mediated through the increased release of NO from endothelial cells. This study investigated the potential use of topically and orally administered propolis extracts to prevent UV irradiation-induced oxidative stress in skin. The results illustrated that green propolis extract (GPE) contained greater amounts of polyphenols, coumaric acid, drupanin, baccharin and artepillin C than did brown propolis extract (BPE). GPE showed higher antioxidant activity than BPE when the IC50 (concentration that caused 50 % inhibition) values were compared. Interesting, the oral treatment of hairless mice demonstrated a recovery of 30.0 % for GPE and 22.8 % for BPE with respect to UV irradiation-induced GSH depletion. A literatura relata propriedades antioxidante, anti-inflamatória e antidiabética para esta planta. Sua utilização é por meio do chá, obtido da infusão de partes da planta, precisamente seu cálice seco. Devido a suas propriedades, o consumodeste chá no Brasil vem crescendo. The toxicity in cell culture and the potential skin irritation resulting from topical application of this formulation were also investigated. Methods: Mouse dorsal surfaces were treated topically with 300 mg of the unloaded and quercetin-loaded (0.3%, w/w) microemulsions before and after exposure to UVB (2.87 J/cm2) irradiation. The untreated control groups irradiated and non-irradiated were also evaluated. En este estudio, la tecnica de cromatografia liquida de alta resolucion en fase reversa para la d... more En este estudio, la tecnica de cromatografia liquida de alta resolucion en fase reversa para la determinacion de la rutina en el extracto Erythroxylum suberosum fue desarrollada y validada. Se realizo un analisis de regresion, con la observacion de una buena linealidad (r = 0,9997). Los valores obtenidos para la precision y la determinacion de la precision estan de acuerdo con las directrices ICH. La deteccion y cuantificacion de los limites de la rutina fueron 0,19 µg / mL y 0,60 µg / mL, respectivamente. Los resultados demostraron que el metodo desarrollado es una tecnica fiable de HPLC para la determinacion de la rutina en el extracto de E. suberosum. Ademas, se investigo la aplicabilidad de este metodo en los estudios de estabilidad y la estandarizacion de hierbas.

Therefore, this study aimed to investigate the use of a cream gel formulation containing the UV filters benzophenone-3, octyl methoxycinnamate, and octyl salicylate to prevent skin damage from a single dose of UVR (2.87 J/cm2). This protective effect was evaluated in vivo by measuring the following biochemical parameters: reduced glutathione levels, secretion of matrix metalloproteinases, and myeloperoxidase activity. Proteases hydrolyze the peptide bonds of proteins into peptides and amino acids, being found in a... more Proteases hydrolyze the peptide bonds of proteins into peptides and amino acids, being found in all living organisms, and are essential for cell growth and differentiation. Proteolytic enzymes have potential application in a wide number of industrial processes such as food, laundry detergent and pharmaceutical. Proteases from microbial sources have dominated applications in industrial sectors. Fungal proteases are used for hydrolyzing protein and other components of soy beans and wheat in soy sauce production. Proteases can be produced in large quantities in a short time by established methods of fermentation. The parameters such as variation in C/N ratio, presence of some sugars, besides several other physical factors are important in the development of fermentation process. Proteases of fungal origin can be produced cost effectively, have an advantage faster production, the ease with which the enzymes can be modified and mycelium can be easily removed by filtration. The production of proteases has been carried out using submerged fermentation, but conditions in solid state fermentation lead to several potential advantages for the production of fungal enzymes. This review focuses on the production of fungal proteases, their distribution, structural-functional aspects, physical and chemical parameters, and the use of these enzymes in industrial applications.

Propolis, which is a natural product widely consumed in the folk medicine, is a serious candidate... more Propolis, which is a natural product widely consumed in the folk medicine, is a serious candidate to be applied topically due to its outstanding antioxidant properties. So, the purpose of this study was to develop stable topical formulations added with propolis extract in an attempt to prevent and/or treat the diseases occurring in skin caused by UV radiation. The antioxidant activity using a chemiluminescent method was used to evaluate the functional stability and the permeation/retention in skin of these formulations. In the long-term stability study, the formulations were stored at 25+/-2 degrees C/AH and at 40+/-2 degrees C/70% RH for 360 days. It was found in this study, that the formulations prepared with Polawax showed functional and physical stability in the period of study. In addition, this formulation presented good results in the percutaneous study, allowing the antioxidant compounds present in the propolis extract to reach lower layers in pig ear skin and in the whole hairless mice skin (retention=0.12 and 0.13 microL of propolis/g of skin, respectively). In the in vivo study, it was also suggested that this formulation may be effective in protecting skin from UVB photodamage, nevertheless other assays need to be done in order to have a complete understanding of the protective effect of formulations added with propolis extract.
In this study, a reverse phase-high performance liquid chromatography (RP-HPLC) technique for det... more In this study, a reverse phase-high performance liquid chromatography (RP-HPLC) technique for determination of rutin in Erythroxylum suberosum extract was developed and validated. A regression analysis was performed, with the observation of good linearity (r = 0.9997). The values obtained for precision and accuracy determination are in agreement with ICH guidelines. The detection and the quantitation limits of rutin were 0.19 µg/mL and 0.60 µg/mL, respectively. The results demonstrated that the developed method is a reliable HPLC technique for determination of rutin in E. suberosum extract. In addition, the applicability of this method in stability studies and standardization of herbs was investigated.

An analytical procedure to quantify 3-benzophenone, octylmethoxycinnamate and octylsalicylate was... more An analytical procedure to quantify 3-benzophenone, octylmethoxycinnamate and octylsalicylate was validated and employed to assess these ultraviolet filters in sunscreen formulations and from skin penetration studies. The effect of the vehicle on the skin retention of these filters was investigated. HPLC and extraction procedure were found to be reliable when obtaining data for the sunscreen formulations and for evaluation skin penetration. The results demonstrated that a cream gel generated higher epidermal concentrations of these filters than a lotion or cream-based formulation. Additionally, when comparing the skin retentions of each filter using the same formulation, 3-benzophenone showed the highest skin retention.

(droga vegetal, tintura e xarope) e Cordia verbenacea (droga vegetal, tintura e pomada). Além disso, foi investigada a possível interferência ocasionada pelos metabólitos secundários presentes nos dois derivados vegetais. A contaminação microbiana foi determinada seguindo o método de semeadura em profundidade. O estudo da interferência no método de avaliação da qualidade microbiológica foi realizado por determinação da recuperação de microrganismos viáveis, utilizando as cepas Escherichia coli (ATCC 25922) e Candida albicans (ATCC 90028). Na avaliação da qualidade microbiológica, as folhas secas oriundas de C. verbenacea e Mikania sp. apresentaram 4,5 x 104 UFC/g e 2,08 x 104 UFC/g de bactérias, respectivamente. A chromatographic technique for determination of rutin and narcissin in marigold extract and topi... more A chromatographic technique for determination of rutin and narcissin in marigold extract and topical formulations was developed and validated. The method shows linearity over the concentration range of 0.2-6.0 mg/mL of rutin (r = 0.9986) and 0.8-12.0 µg/mL of narcissin (r = 0.9951). The values obtained for precision and accuracy are in agreement with ICH guidelines. Both the formulation excipients and the porcine ear skin samples did not interfere with the flavonoids determination. The recovery of rutin and narcissin in skin samples added with marigold extract was 81.41% and 83.35%, respectively, which demonstrate the applicability of this method to perform skin penetration studies.

The increased amount of melanin leads to skin disorders such as age spots, freckles, melasma and ... more The increased amount of melanin leads to skin disorders such as age spots, freckles, melasma and malignant melanoma. Tyrosinase is known to be the key enzyme in melanin production. Plants and their extracts are inexpensive and rich resources of active compounds that can be utilized to inhibit tyrosinase as well as can be used for the treatment of dermatological disorders associated with melanin hyperpigmentation. Using in vitro tyrosinase inhibitory activity assay, extracts from 13 plant species from Brazilian Cerrado were evaluated. The results showed that Pouteria torta and Eugenia dysenterica extracts presented potent in vitro tyrosinase inhibition compared to positive control kojic acid. Ethanol extract of Eugenia dysenterica leaves showed significant (p,0.05) tyrosinase inhibitory activity exhibiting the IC 50 value of 11.88 mg/ mL, compared to kojic acid (IC 50 value of 13.14 mg/mL). Pouteria torta aqueous extract leaves also showed significant inhibitory activity with IC 50 value of 30.01 mg/mL. These results indicate that Pouteria torta and Eugenia dysenterica extracts and their isolated constituents are promising agents for skin-whitening or antimelanogenesis formulations.

Proteolytic enzymes have potential application in a wide number of industrial processes such as food, laundry detergent and pharmaceutical. Proteases from microbial sources have dominated applications in industrial sectors. Fungal proteases are used for hydrolyzing protein and other components of soy beans and wheat in soy sauce production. Proteases can be produced in large quantities in a short time by established methods of fermentation. The parameters such as variation in C/N ratio, presence of some sugars, besides several other physical factors are important in the development of fermentation process. Proteases of fungal origin can be produced cost effectively, have an advantage faster production, the ease with which the enzymes can be modified and mycelium can be easily removed by filtration. The production of proteases has been carried out using submerged fermentation, but conditions in solid state fermentation lead to several potential advantages for the production of fungal enzymes. This review focuses on the production of fungal proteases, their distribution, structural-functional aspects, physical and chemical parameters, and the use of these enzymes in industrial applications.

Propolis, which is a natural product widely consumed in the folk medicine, is a serious candidate... more Propolis, which is a natural product widely consumed in the folk medicine, is a serious candidate to be applied topically due to its outstanding antioxidant properties. So, the purpose of this study was to develop stable topical formulations added with propolis extract in an attempt to prevent and/or treat the diseases occurring in skin caused by UV radiation. The antioxidant activity using a chemiluminescent method was used to evaluate the functional stability and the permeation/retention in skin of these formulations. In the long-term stability study, the formulations were stored at 25+/-2 degrees C/AH and at 40+/-2 degrees C/70% RH for 360 days. It was found in this study, that the formulations prepared with Polawax showed functional and physical stability in the period of study. In addition, this formulation presented good results in the percutaneous study, allowing the antioxidant compounds present in the propolis extract to reach lower layers in pig ear skin and in the whole hairless mice skin (retention=0.12 and 0.13 microL of propolis/g of skin, respectively). In the in vivo study, it was also suggested that this formulation may be effective in protecting skin from UVB photodamage, nevertheless other assays need to be done in order to have a complete understanding of the protective effect of formulations added with propolis extract.
In this study, a reverse phase-high performance liquid chromatography (RP-HPLC) technique for det... more In this study, a reverse phase-high performance liquid chromatography (RP-HPLC) technique for determination of rutin in Erythroxylum suberosum extract was developed and validated. A regression analysis was performed, with the observation of good linearity (r = 0.9997). The values obtained for precision and accuracy determination are in agreement with ICH guidelines. The detection and the quantitation limits of rutin were 0.19 µg/mL and 0.60 µg/mL, respectively. The results demonstrated that the developed method is a reliable HPLC technique for determination of rutin in E. suberosum extract. In addition, the applicability of this method in stability studies and standardization of herbs was investigated.

An analytical procedure to quantify 3-benzophenone, octylmethoxycinnamate and octylsalicylate was... more An analytical procedure to quantify 3-benzophenone, octylmethoxycinnamate and octylsalicylate was validated and employed to assess these ultraviolet filters in sunscreen formulations and from skin penetration studies. The effect of the vehicle on the skin retention of these filters was investigated. HPLC and extraction procedure were found to be reliable when obtaining data for the sunscreen formulations and for evaluation skin penetration. The results demonstrated that a cream gel generated higher epidermal concentrations of these filters than a lotion or cream-based formulation. Additionally, when comparing the skin retentions of each filter using the same formulation, 3-benzophenone showed the highest skin retention.

(droga vegetal, tintura e xarope) e Cordia verbenacea (droga vegetal, tintura e pomada). Além disso, foi investigada a possível interferência ocasionada pelos metabólitos secundários presentes nos dois derivados vegetais. A contaminação microbiana foi determinada seguindo o método de semeadura em profundidade. O estudo da interferência no método de avaliação da qualidade microbiológica foi realizado por determinação da recuperação de microrganismos viáveis, utilizando as cepas Escherichia coli (ATCC 25922) e Candida albicans (ATCC 90028). Na avaliação da qualidade microbiológica, as folhas secas oriundas de C. verbenacea e Mikania sp. apresentaram 4,5 x 104 UFC/g e 2,08 x 104 UFC/g de bactérias, respectivamente. A chromatographic technique for determination of rutin and narcissin in marigold extract and topi... more A chromatographic technique for determination of rutin and narcissin in marigold extract and topical formulations was developed and validated. The method shows linearity over the concentration range of 0.2-6.0 mg/mL of rutin (r = 0.9986) and 0.8-12.0 µg/mL of narcissin (r = 0.9951). The values obtained for precision and accuracy are in agreement with ICH guidelines. Both the formulation excipients and the porcine ear skin samples did not interfere with the flavonoids determination. The recovery of rutin and narcissin in skin samples added with marigold extract was 81.41% and 83.35%, respectively, which demonstrate the applicability of this method to perform skin penetration studies.

Tyrosinase is known to be the key enzyme in melanin production. Plants and their extracts are inexpensive and rich resources of active compounds that can be utilized to inhibit tyrosinase as well as can be used for the treatment of dermatological disorders associated with melanin hyperpigmentation. Using in vitro tyrosinase inhibitory activity assay, extracts from 13 plant species from Brazilian Cerrado were evaluated. The results showed that Pouteria torta and Eugenia dysenterica extracts presented potent in vitro tyrosinase inhibition compared to positive control kojic acid. Ethanol extract of Eugenia dysenterica leaves showed significant (p,0.05) tyrosinase inhibitory activity exhibiting the IC 50 value of 11.88 mg/ mL, compared to kojic acid (IC 50 value of 13.14 mg/mL). Pouteria torta aqueous extract leaves also showed significant inhibitory activity with IC 50 value of 30.01 mg/mL. These results indicate that Pouteria torta and Eugenia dysenterica extracts and their isolated constituents are promising agents for skin-whitening or antimelanogenesis formulations.
