Class 3 semaphorins are involved in cancer progression. However, the roles of class 3 semaphorins in hepatocellular carcinoma (HCC) are unknown. We examined the expression levels of class 3 semaphorins in HCC cell lines with different metastatic potential and in carcinoma tissue samples. The results indicated that Semaphorin 3A expression was up-regulated in metastatic cell lines and in samples from patients with tumor recurrence. Cell functional studies revealed that Semaphorin 3A promoted HCC cell proliferation, migration, and invasion. Animal studies indicated that Semaphorin 3A overexpression enhanced tumor growth and lung metastasis. Semaphorin 3A also acted as a chemoattractant involved in direct recruitment of macrophages in vitro, and facilitated tumor-associated macrophage (TAM) infiltration in vivo. Neither the factors produced by tumor-associated neutrophils (TANs) nor their effects on tumor progression have been characterized. We investigated the roles of TANs in progression of hepatocellular carcinoma (HCC) using cell lines and immune cells isolated from patients. We performed studies with HepG2, PLC/PRF/5, MHCC97H and HCCLM3 human and Hepa1-6 and H22 mouse HCC cell lines; expression of chemokines and cytokines were knocked down with small hairpin RNAs. The role of these molecules in hepatocellular carcinoma (HCC), however, has not been fully elucidated. In this study, we employed miRNA-sequencing and identified 22 miRNAs involved in HCC metastasis. One of these, miR-28-5p, was downregulated in HCCs. This downregulation correlated with tumor metastasis, recurrence, and poor survival. Biofunctional investigations revealed that miR-28-5p deficiency promoted tumor growth and metastasis in nude mice without altering the in vitro biological characteristics of HCC cells. Through gene expression profiles and bioinformatics analysis, we identified interleukin-34 (IL-34) as a direct target of miR-28-5p, and the effects of miR-28-5p deficiency on HCC growth and metastasis was dependent on IL-34-mediated tumor-associated macrophage (TAMs) infiltration. ABSTRACT We report that a previously found fluorescent chemodosimeter for Hg(II) ions can also be... more ABSTRACT We report that a previously found fluorescent chemodosimeter for Hg(II) ions can also be used as a fluorescent probe for hypochlorous acid (HClO) in that its fluorescence intensity (excited at 520 nm and measured at 563 nm) is linearly proportional to the concentration of HClO in the 1-100 mu M range and with a 0.1 mu M detection limit. The probe can detect HClO with virtually no interference by Hg(II) and other metal ions. The mechanism for the high selectivity for HClO has been studied. The probe was applied to image hypochlorite in E. coli cells.

The development of novel selective probes with high sensitivity for the detection of Al(3+) is wi... more The development of novel selective probes with high sensitivity for the detection of Al(3+) is widely considered an important research goal due to the importance of such probes in medicine, living systems and the environment. Here, we describe a new fluorescent probe, N'-(4-diethylamino-2-hydroxybenzylidene)-2-hydroxybenzohydrazide (1), for Al(3+). Probe 1 was evaluated in a solution of acetonitrile/water (1:1 v/v). Compared with previously reported probes for Al(3+), probe 1 can be synthesized easily and in high yield. A Job plot confirmed that probe 1 is able to complex Al(3+) in a 1:1 ratio, and the binding constant was determined to be 4.25×10(8) m(-1). Moreover, the detection limit was as low as 6.7×10(-9) m, suggesting that probe 1 has a high sensitivity. Common coexistent metal ions, such as K(+), Co(2+), Ca(2+), Ba(2+), Ni(2+), Pb(2+), Hg(2+), Ce(2+), Zn(2+), Cd(2+), Fe(3+), showed little or no interference in the detection of Al(3+) in solution, demonstrating the high s...

As one of its ligands, CXCL5 is associated with neutrophil infiltration and poor prognosis in hepatocellular carcinoma (HCC). However, little is known about the role of the CXCR2/CXCL5 axis in the invasion and metastasis of HCC cells. In this study, we examined CXCR2 expression in human HCC cell lines and in three independent cohorts of HCC patients. The molecular effects of high expression levels of CXCR2 and CXCL5 in HCC cells were determined using qRT-PCR, western blot analysis, immunofluorescence, matrigel invasion assay, and xenograft mouse models. We found that high levels of CXCR2 correlated with progression and poor prognosis in human HCC. CXCR2/CXCL5 together promoted cell spreading by inducing the epithelial-mesenchymal transition (EMT) through activation of the PI3K/Akt/GSK-3β/Snail signaling pathway. Upon the addition of Fe(3+), the dramatic enhancement of both fluorescence and absorbance intensity, as well as the color change of the solution, could be observed. The detection limit of RAE for Fe(3+) was around 7.98 ppb. Common coexistent metal ions showed little or no interference in the detection of Fe(3+). Moreover, the addition of CN(-) could quench the fluorescence of the acetonitrile solution of RAE and Fe(3+), indicating the regeneration of the chemosensor RAE. The robust nature of the sensor was shown by the detection of Fe(3+) even after repeated rounds of quenching. As iron is a ubiquitous metal in cells and plays vital roles in many biological processes, this chemosensor could be developed to have applications in biological studies.

The RCM reaction modes depend on the length of polyether chain. Diethylene glycolic chain produced the dimer 7a and linear oligomer 7a′ with multi-cavities, whereas triethylene and tetraethylene glycolic chains allowed direct cyclization through intramolecular head-to-tail pattern to yield novel bridged biscalix[4]arenes 7b–c.

Three sulfonamide-amide hybridized molecules and one cyclic analogue were synthesized and their a... more Three sulfonamide-amide hybridized molecules and one cyclic analogue were synthesized and their assembling behaviors in the solid state were investigated by X-ray crystallography. The results showed that the hybridized molecules could be not only induced to take up helical secondary structures by a network of intramolecular hydrogen bonds, but also utilized as useful building blocks for assembling into 1D zigzag chains and superhelices, 2D layers and further 3D networks. Moreover, it was found that the multiple C-H/O]S hydrogen bonds played an important role in the assembling processes. q

Here, we describe a new fluorescent probe, N'-(4-diethylamino-2-hydroxybenzylidene)-2-hydroxybenzohydrazide (1), for Al(3+). Probe 1 was evaluated in a solution of acetonitrile/water (1:1 v/v). Compared with previously reported probes for Al(3+), probe 1 can be synthesized easily and in high yield. A Job plot confirmed that probe 1 is able to complex Al(3+) in a 1:1 ratio, and the binding constant was determined to be 4.25×10(8) m(-1). Moreover, the detection limit was as low as 6.7×10(-9) m, suggesting that probe 1 has a high sensitivity. As one of its ligands, CXCL5 is associated with neutrophil infiltration and poor prognosis in hepatocellular carcinoma (HCC). However, little is known about the role of the CXCR2/CXCL5 axis in the invasion and metastasis of HCC cells. In this study, we examined CXCR2 expression in human HCC cell lines and in three independent cohorts of HCC patients. The molecular effects of high expression levels of CXCR2 and CXCL5 in HCC cells were determined using qRT-PCR, western blot analysis, immunofluorescence, matrigel invasion assay, and xenograft mouse models. We found that high levels of CXCR2 correlated with progression and poor prognosis in human HCC. Upon the addition of Fe(3+), the dramatic enhancement of both fluorescence and absorbance intensity, as well as the color change of the solution, could be observed. The detection limit of RAE for Fe(3+) was around 7.98 ppb. Common coexistent metal ions showed little or no interference in the detection of Fe(3+). Moreover, the addition of CN(-) could quench the fluorescence of the acetonitrile solution of RAE and Fe(3+), indicating the regeneration of the chemosensor RAE. The robust nature of the sensor was shown by the detection of Fe(3+) even after repeated rounds of quenching. As iron is a ubiquitous metal in cells and plays vital roles in many biological processes, this chemosensor could be developed to have applications in biological studies.

The RCM reaction modes depend on the length of polyether chain. Diethylene glycolic chain produced the dimer 7a and linear oligomer 7a′ with multi-cavities, whereas triethylene and tetraethylene glycolic chains allowed direct cyclization through intramolecular head-to-tail pattern to yield novel bridged biscalix[4]arenes 7b–c.

The results showed that the hybridized molecules could be not only induced to take up helical secondary structures by a network of intramolecular hydrogen bonds, but also utilized as useful building blocks for assembling into 1D zigzag chains and superhelices, 2D layers and further 3D networks.
