To establish an index system for rapid assessment of environment with high transmission risk of s... more To establish an index system for rapid assessment of environment with high transmission risk of schistosomiasis in marshland and lake regions. The alternative indices were established preliminarily by the documentary method, and then the Delphi method was used two rounds to select the indices and establish the index system. The degree of familiar and authority of the experts as well as the weights of all indices were assessed. A total of 3 primary indices namely morbidity in domestic animals, morbidity in humans and Oncomelania snail status were established, among which the weight of morbidity in domestic animals (0.68) was the highest. Totally 16 secondary indices were established, among which the combinative weights of the positive rate of wild feces (0.09), the infection rate of livestock (0.09), the infection rate of humans (0.07), the number of cases with acute schistosomiasis (0.07), the area with infected snails (0.07), and the density of infected snails (0.07) were the highe...

To explore an effective therapy for pregnant Toxoplasma gondii infection by using acetyl spiramyc... more To explore an effective therapy for pregnant Toxoplasma gondii infection by using acetyl spiramycin combined with azithromycin. ELISA and PCR were used to diagnose and evaluate the therapy efficiency to toxoplasmosis in pregnant women. The serological test showed that the positive rates of specific antibodies IgM and IgG to Toxoplasma gondii in 285 pregnant women were 1.05% (3/285) and 5.97% (17/285), respectively. All the 3 cases of serum IgM positive pregnant women received the amniotic fluid PCR tests for Toxoplasma gondii DNA and 2 were positive, and they received spiramycin combined with azithromycin. After the therapy, their serum IgM antibody specific to Toxoplasma gondii and positive amniotic fluid PCR test for Toxoplasma gondii DNA turned to be negative. Acetyl spiramycin in combination with azithromycin is effective in the treatment of pregnant toxoplasmosis.

To explore the killing effect of different fractional proteins from Microtus fortis (Mf) serum to... more To explore the killing effect of different fractional proteins from Microtus fortis (Mf) serum to S. japonicum juveniles, and to find possible association of the proteins with the natural resistance to schistosome infection. The proteins in Mf serum were separated by means of ion-exchange column chromatography and molecular sieve column chromatography. After desalted by dialysis and lyophilized, the proteins were dissolved in DMEM medium which contained 300 U/ml penicillin and 300 microg/ml streptomycin, and the two-day old schistosomula were added in for in vitro cultivation (100 +/- 20/well). The killing activity of the fractional proteins to the juvenile worms was defined by mortality rate. 58 fractional proteins were separated from Mf serum, in which six proteins were confirmed to have a significant killing activity to schistosomula. DuIFN-gamma cDNA was cloned and expressed in COS-7 cells, and the antiviral activity of DuIFN-gamma was detected and neutralized by specific antibodies. Ducks were vaccinated with DHBpreS/S DNA alone or co-immunized with plasmid expressing DuIFN-gamma. DuIFN-gamma mRNA in peripheral blood mononuclear cells (PBMCs) from immunized ducks was detected by semi-quantitative competitive RT-PCR. Anti-DHBpreS was titrated by enzyme-linked immunosorbent assay (ELISA). DHBV DNA in sera and liver was detected by Southern blot hybridization, after ducks were challenged with high doses of DHBV. Staphylococcus epidermidis is one of the major causative agents for nosocomial infections. To rev... more Staphylococcus epidermidis is one of the major causative agents for nosocomial infections. To reveal the pathogenesis factors, we performed the comparative proteomic analysis of invasive ATCC35984 and commensal ATCC12228 strains by two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight-mass spectrometry. The differentially expressed proteins were involved in carbohydrate metabolism, sugar binding, lipid degradation and amino acid binding. In addition, we demonstrated that the trap gene was transcribed by 3.657 AE 0.156 (P o 0.01) -fold higher in ATCC35984 than in ATCC12228. Levels of accumulation-associated protein (AAP) were found to be low by the immunodot blotting assay in ATCC12228, which is unable to form biofilm. Our results suggest that the target of RNAIII activating protein and AAP may contribute to S. epidermidis virulence and biofilm formation.

The PCBS was gained through physical and chemical disposals to porcine cancellous bone and was implanted subcutaneously in rabbit. The histological investigation and the expression of Th1 (such as IL-2, IFN-gamma) and Th2 (such as IL-4, IL-10) mRNA were measured by HE, RT-PCR respectively in 1,2,4,8,12 week postoperation. HE revealed: Inflammation cells (lymphocytes and monocytes) were shown abundantly in the surrounding tissue in 1 week and were decreased gradually since 2 week. At 12 week, there were few inflammation cells in the surrounding tissue. The aim of present study is to investigate the molecular characteristic of Staphylococcus aureus isolates isolated from the pus samples from the patients with purulent skin and soft tissue infections in Wenzhou, China. Between December 2002 and June 2008, a total of 111 nonduplicate S. aureus isolates were collected from the pus samples of the patients with SSTIs in a teaching hospital in Wenzhou, China. All the tested isolates were confirmed as S. aureus using a Staph SPA agglutination kit, Gram's stain and a Vitek-60 microbiology analyzer. All the contestants were grouped by the operation standard, qualitative judgment, quantitative determination, and geographical location of Hunan and Hubei provinces, and their scores were statistically analyzed by SPSS 16.0 software. The total scores of the contestants of the two provinces were high and there was no significant difference between them. Among the professional persons, the scores of operation standard and qualitative judgment were significantly higher than those of quantitative determination. There were no significant differences among the scores grouped by the different genders, ages, professional titles and areas (all P > 0.05). To analyse the implications of different virulence factors in Staphylococcus epidermidis infections, the complete genome of Staphylococcus epidermidis strain ATCC 12228, a non-biofilm forming, non-infection associated strain used for detection of residual antibiotics in food products, was sequenced. This strain showed low virulence by mouse and rat experimental infections. The genome consists of a single 2499 279 bp chromosome and six plasmids. Therefore, the spread of high-level aminoglycoside resistance determinants has become a great concern. , 680 distinct Escherichia coli clinical isolates were collected from a teaching hospital in Wenzhou, China. PCR and DNA sequencing were used to identify 16S rRNA methylase and extended-spectrum β-lactamase (ESBL) genes, including armA and rmtB, and in situ hybridization was performed to determine the location of 16S rRNA methylase genes. Conjugation experiments were subsequently performed to determine whether aminoglycoside resistance was transferable from the E. coli isolates via 16S rRNA methylase-bearing plasmids. Homology of the isolates harboring 16S rRNA methylase genes was determined using pulse-field gel electrophoresis (PFGE).

Microtus fortis is a naturally resistant vertebrate host of Schistosoma japonicum by preventing c... more Microtus fortis is a naturally resistant vertebrate host of Schistosoma japonicum by preventing completion of parasite's life cycle. Sera of M. fortis were found to have anti-schistosome effect in vitro and in vivo. In order to identify genes associated with the anti-schistosome effect of M. fortis, we screened a M. fortis marrow cDNA expression library by expression cloning and identified a 331-bp clone gC14.75. It was the homologue of heat shock protein 90␣ (HSP90␣). Full-length of M. fortis HSP90␣ gene, Mf-HSP90␣, was amplified according to gC14.75 and Cricetulus griseus HSP90␣. To test the potential anti-schistosome function of Mf-HSP90␣, we prepared conditioned medium of Mf-HSP90␣ and added it to schistosomula cultured in vitro. It caused 27.0% schistosomula death rate in 96 h, which was considerably higher than that of negative control. We transferred Mf-HSP90␣ by retroviral expression vector pLXSN into mice to investigate its anti-schistosome effect in vivo. Compared with those of DMEM injection control, mice injected with Mf-HSP90␣ recombinant retrovirus had 40.8% worm burden reduction and 57.9% reduction in liver eggs per gram (LEPG) indicating its anti-schistosome effect in vivo. Taken together, our results suggested Mf-HSP90␣ as a novel anti-schistosome molecule in vitro and in vivo.

China) has reached a level where accurate methods are required for monitoring the national schistosomiasis control programme and to verify whether transmission has been interrupted. We have assessed the prevalence of active S. japonicum infection by use of an up-converting phosphor lateral-flow (UCP-LF) assay for determination of circulating anodic antigens (CAA) in urine and serum, and compared the findings with those of the Kato-Katz technique for egg detection in stool and an immunohaemagglutination assay (IHA) for specific antibodies in serum. The study was carried out in three villages located in a remaining S. japonicum-endemic area in P.R. China. Acetyl spiramycin in combination with azithromycin is effective in the treatment of pregnant toxoplasmosis.

The proteins in Mf serum were separated by means of ion-exchange column chromatography and molecular sieve column chromatography. After desalted by dialysis and lyophilized, the proteins were dissolved in DMEM medium which contained 300 U/ml penicillin and 300 microg/ml streptomycin, and the two-day old schistosomula were added in for in vitro cultivation (100 +/- 20/well). The killing activity of the fractional proteins to the juvenile worms was defined by mortality rate. 58 fractional proteins were separated from Mf serum, in which six proteins were confirmed to have a significant killing activity to schistosomula. Ducks were vaccinated with DHBpreS/S DNA alone or co-immunized with plasmid expressing DuIFN-gamma. DuIFN-gamma mRNA in peripheral blood mononuclear cells (PBMCs) from immunized ducks was detected by semi-quantitative competitive RT-PCR. To reveal the pathogenesis factors, we performed the comparative proteomic analysis of invasive ATCC35984 and commensal ATCC12228 strains by two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight-mass spectrometry. The differentially expressed proteins were involved in carbohydrate metabolism, sugar binding, lipid degradation and amino acid binding. In addition, we demonstrated that the trap gene was transcribed by 3.657 AE 0.156 (P o 0.01) -fold higher in ATCC35984 than in ATCC12228. Levels of accumulation-associated protein (AAP) were found to be low by the immunodot blotting assay in ATCC12228, which is unable to form biofilm. Our results suggest that the target of RNAIII activating protein and AAP may contribute to S. epidermidis virulence and biofilm formation.

All the contestants were grouped by the operation standard, qualitative judgment, quantitative determination, and geographical location of Hunan and Hubei provinces, and their scores were statistically analyzed by SPSS 16.0 software. The total scores of the contestants of the two provinces were high and there was no significant difference between them. Among the professional persons, the scores of operation standard and qualitative judgment were significantly higher than those of quantitative determination. Therefore, the spread of high-level aminoglycoside resistance determinants has become a great concern. , 680 distinct Escherichia coli clinical isolates were collected from a teaching hospital in Wenzhou, China. PCR and DNA sequencing were used to identify 16S rRNA methylase and extended-spectrum β-lactamase (ESBL) genes, including armA and rmtB, and in situ hybridization was performed to determine the location of 16S rRNA methylase genes. Conjugation experiments were subsequently performed to determine whether aminoglycoside resistance was transferable from the E. coli isolates via 16S rRNA methylase-bearing plasmids. Homology of the isolates harboring 16S rRNA methylase genes was determined using pulse-field gel electrophoresis (PFGE).

Microtus fortis is a naturally resistant vertebrate host of Schistosoma japonicum by preventing c... more Microtus fortis is a naturally resistant vertebrate host of Schistosoma japonicum by preventing completion of parasite's life cycle. Sera of M. fortis were found to have anti-schistosome effect in vitro and in vivo. In order to identify genes associated with the anti-schistosome effect of M. fortis, we screened a M. fortis marrow cDNA expression library by expression cloning and identified a 331-bp clone gC14.75. It was the homologue of heat shock protein 90␣ (HSP90␣). Full-length of M. fortis HSP90␣ gene, Mf-HSP90␣, was amplified according to gC14.75 and Cricetulus griseus HSP90␣. To test the potential anti-schistosome function of Mf-HSP90␣, we prepared conditioned medium of Mf-HSP90␣ and added it to schistosomula cultured in vitro. It caused 27.0% schistosomula death rate in 96 h, which was considerably higher than that of negative control. We transferred Mf-HSP90␣ by retroviral expression vector pLXSN into mice to investigate its anti-schistosome effect in vivo. Compared with those of DMEM injection control, mice injected with Mf-HSP90␣ recombinant retrovirus had 40.8% worm burden reduction and 57.9% reduction in liver eggs per gram (LEPG) indicating its anti-schistosome effect in vivo. Taken together, our results suggested Mf-HSP90␣ as a novel anti-schistosome molecule in vitro and in vivo.

China) has reached a level where accurate methods are required for monitoring the national schistosomiasis control programme and to verify whether transmission has been interrupted. We have assessed the prevalence of active S. japonicum infection by use of an up-converting phosphor lateral-flow (UCP-LF) assay for determination of circulating anodic antigens (CAA) in urine and serum, and compared the findings with those of the Kato-Katz technique for egg detection in stool and an immunohaemagglutination assay (IHA) for specific antibodies in serum. The study was carried out in three villages located in a remaining S. japonicum-endemic area in P.R. China.
