Radnoj skupini za zakon o patentima detaljno je predstavljen je Nacrt Prijedloga Novog Zakona o p... more Radnoj skupini za zakon o patentima detaljno je predstavljen je Nacrt Prijedloga Novog Zakona o patentima, te je dana mogucnost eventualnih prijedloga i izmjena Nacrta Zakona o patentima. Novost u Zakonu je pojam konsenzualnog oatenta koji traje 10 godina, a isto tako postavljeno je pitanje tko je dovoljno strucan prevoditi inozemne patentne prijave koje stižu u Državni zavod za intelektualno vlasnistvo. Novim Zakonom o patentima želi se uskladiti visok stupanj usklađenosti s međunarodnim standardima kao sto su TRIPS-Agreement on trade related aspects of intellectual property rights, PCT- Patent Cooperation Treaty i Pariska konvencija za zastitu industrijskog vlasnistva.

The techniques of molecular biology and the application of biological markers have led to the introduction of cell-based assays and these have revolutionised the screening process. Stanicni modeli mogu biti od velike prednosti za ispitivanje mehanizama, metabolizma i potencijal... more Stanicni modeli mogu biti od velike prednosti za ispitivanje mehanizama, metabolizma i potencijalnih toksicnih ucinaka ksenobiotika. Potrebna je malena kolicina tvari koja se ispituje, metoda je relativno jednostavna i brza, a daje vrijedne znanstvene informacije bez suvisnih troskova. Primarna kultura stanica zadržava ekspresiju određenih stanicnih funkcija u usporedbi sa stanicnim linijama koje ekspresiraju samo djelomicno originalan fenotip. Nasi raniji rezultati su pokazali (1) da in vivo tretman stakora s atrazinom uzrokuje produženi ciklus karakteriziran produženim diestrusom. Cilj ove studije je istražiti metabolicke ucinke te citotoksicke, morfoloske i ultrastrukturalne promjene u uterusu stakora za vrijeme estrus, diestrus i proestrus faze ciklusa poslije tretmana s atrazinom. Aujeszky's disease (AD) is enzootic in swine in many parts of the world and one of the most i... more Aujeszky's disease (AD) is enzootic in swine in many parts of the world and one of the most important disease of the pig industry today manifested as respiratory disease, nervous signs and reproductive failure. Mortality and morbidity can be high in new born pigs. To reduce economic looses and to eradicate AD, pigs are now vaccinated with live attenuated vaccines lacking the gE protein. We developed a new subunit gE- vaccine from Tween 80-solubilised AD virus propagated by own procedure, in BHK-21 C13 cells growing in suspension. The vaccine was prepared in the form of water-in-oil-in-water emulsion. The efficacy of the vaccine was previously tested on pigs and in order to reduce cost we have tried to use rats. The pituitary gland as the main gland of the neuroendocrine system plays a key role in the mainte... more The pituitary gland as the main gland of the neuroendocrine system plays a key role in the maintenance of homeostasis. There are at least three levels of feedback control in the hypothalamus and the pituitary gland at the neuroendocrine level: long, short and ultrashort feedback systems. Releasing factors are released from the end of the axons of the neurosecretory cells into the blood supply to the AP to regulate hormone production. GH produced and stored in the AP stimulates protein synthesis and carbohydrate metabolism. GH controls the rate at which tissues can mobilise nutrients from the blood. GH directly inhibits the deposition and stimulate cell division, protein synthesis and bone growth. High density plating procedure was used to evaluate the effect of atrazine on anterior pituitary ... more High density plating procedure was used to evaluate the effect of atrazine on anterior pituitary cells of rats in monolayer culture. Collagenase-dispersed pituitary cells plated in suspension with medium-199 and 10% foetal calf serum attached quantitatively to plastic surfaces within 24 hours. Electron microscopy showed subpopulations of different cell types. After prolonged cultivation, most cells established small colonies with extensive contacts among them. Cell-to-cell formation of aggregates was significant and the colonies manifested morphological changes. The cells retained their enzymatic activity, converting testosterone into 5alfa-dihydrotestosterone by enzyme 5alfa-reductase. Immunohistochemical techniques facilitated differentiation of gonadotrophs producing follicle stimulating (FSH) and luteinising hormones (LH). Atrazine in concentrations of 5 to 50 µg/ml of medium was associated with a significant reduction in the number of viable cells within 72 hours. The results s...

Slices of rat pituitary gland, hypothalamus, amygdala, cerebral cortex and prostate have been inc... more Slices of rat pituitary gland, hypothalamus, amygdala, cerebral cortex and prostate have been incubated in vitro with labelled testosterone; the metabolites formed have been identified. Testosterone is converted into l7/3-hydroxy-So-androstan-3-one (androstanolone, dihydro testosterone, DHT) by all tissues examined. The prostate is the structure which effects such a conversion to the greatest extent; the pituitary gland and the hypothalamus are also very active; the cerebral cortex and the amygdala are also able to transform testosterone into DHT, but the rate of conversion is not as great as that found in the tissues previously mentioned. Androstenedione. 5a-androstan-3,17-dione and So-androstan-3a, 17/3-diol are also formed by some of the tissues. Castration increases and treatment with exogenous testosterone decreases the transformation of testosterone into DHT at the pituitary and the hypothalamic level. Both at the hypothalamic and the pituitary level, the addition in vitro of progesterone, 1 l-deoxycorticosterone. I I-deoxycortisol and corticosterone reduces the transformation of testosterone into DHT; on the contrary, the addition of pituitary FSH increases the conversion of testosterone into its 'active' metabolite. The ability to transform testosterone into DHT is much higher in all structures examined (with the exception of the prostate) in prepuberal than in adult rats.

OBJECTIVE: To give more insight in the progesterone metabolism in rat after the treatment with th... more OBJECTIVE: To give more insight in the progesterone metabolism in rat after the treatment with the progestin ethynodiol diacetate. METHODS: Urinary excretion of the metabolites of subcutaneously administred (4-14C)-progesterone was studied in female rats. After an acid hydrolysis and extraction of urine the metabolites were analysed by thin layer chromatography and by gas chromatography-mass spectrometry. RESULTS: The most of radioactivity was excreted during the first 24 h, and total of 8.36 % has been recovered within four days. The excreted metabolites in urine were found as glucuronides and free steroids (80.72 %), and 19.28 % were determined as sulphates. Among detected metabolites, 5alpha-pregnane-3,20-dione, 3alpha-hydroxy-5alpha-pregnan-20-one and A-homo-3-oxa-5alpha-pregnane-4,20-dione were determined in the urinary extracts. The metabolism of testosterone in the rat ventral prostate, anterior pituitary, basal hypothalamu... more The metabolism of testosterone in the rat ventral prostate, anterior pituitary, basal hypothalamus and amygdala was studied in vitro under the influence of vitamin B6 compounds. The influence of these compounds on the activity of 5 alpha-reductase (5 alpha-R), 3 alpha- and 17 beta-hydroxysteroid dehydrogenase (3 alpha-HSD, 17 beta-HSD) was determined for all the examined tissues. Pyridoxine hydrochloride significantly increased the activity of 5 alpha-R, 3 alpha- and 17 beta-HSD, but pyridoxal hydrochloride had an inhibitory influence on 5 alpha-R and showed no effect on 3 alpha-HSD activity at the prostate level. Male rat anterior pituitary, basal hypothalamus or amygdala incubated with pyridoxal phosphate and pyridoxal hydrochloride showed modified enzymatic activities. Pyridoxal hydrochloride showed an inhibitory effect on 5 alpha-R in the rat pituitary and basal hypothalamus as well as in the rat prostate.

After treatment with LH-RH one pregnancy occurred and in 3 more cases indirect evidence indicated that ovulation had occurred.

Herbicide atrazine (A) effect on reproductive processes in male rat has been examined at anteroir... more Herbicide atrazine (A) effect on reproductive processes in male rat has been examined at anteroir pituitary (AP), testis (T), prostate (P) and seminal vesicles (SV). In the present study A in corn oil was administred to 90 days old rats i.p. at 120 mg/kg b.w./each 72h/60 days. Exposure of rats had induced changes in LH and FSH synthesis, changes of spermatogenesis and structural changes at all selected tissues. LH and FSH were detected by immunohistochemical method, structural changes by light and electron microscopy, while spermatogenesis has been followed by measuring concentration, total number, motility and progressive motility of sperms. A-males in AP have decreased number of immunoreactive cells for LH 25%, while FSH increased for 70%. In T control FSH is dominantly present on the perifery and is concentrated with the spermatocytes and spermatogonia. In A-rats FSH presence is within the whole irregular organized seminiferous tubules. Histochemistry of P tissue shows the tubular alveolar gland with large irregular cavities in A-rats. Our previous research on DHT-receptors in prostate showed the reduction of DHT specific binding sites on receptor molecules. At SV evidently mucosal folds are more tighty packed in A-rats vs. control. The weight of SV in A-rats was much lower and a typical atrophy of the gland is present. Inhibition of spermatogenesis in A-treated rats was in sperm count from 14.3x106 per ml vs. 17.06106 in controls. At the same time the motile sperm count has been reduced from 43% on 35%, respectively.

METHODS: Urinary excretion of the metabolites of subcutaneously administred (4-14C)-progesterone was studied in female rats. After an acid hydrolysis and extraction of urine the metabolites were analysed by thin layer chromatography and by gas chromatography-mass spectrometry. RESULTS: The most of radioactivity was excreted during the first 24 h, and total of 8.36 % has been recovered within four days. The excreted metabolites in urine were found as glucuronides and free steroids (80.72 %), and 19.28 % were determined as sulphates. Among detected metabolites, 5alpha-pregnane-3,20-dione, 3alpha-hydroxy-5alpha-pregnan-20-one and A-homo-3-oxa-5alpha-pregnane-4,20-dione were determined in the urinary extracts. The metabolism of testosterone in the rat ventral prostate, anterior pituitary, basal hypothalamu... more The metabolism of testosterone in the rat ventral prostate, anterior pituitary, basal hypothalamus and amygdala was studied in vitro under the influence of vitamin B6 compounds. The influence of these compounds on the activity of 5 alpha-reductase (5 alpha-R), 3 alpha- and 17 beta-hydroxysteroid dehydrogenase (3 alpha-HSD, 17 beta-HSD) was determined for all the examined tissues. Pyridoxine hydrochloride significantly increased the activity of 5 alpha-R, 3 alpha- and 17 beta-HSD, but pyridoxal hydrochloride had an inhibitory influence on 5 alpha-R and showed no effect on 3 alpha-HSD activity at the prostate level. Male rat anterior pituitary, basal hypothalamus or amygdala incubated with pyridoxal phosphate and pyridoxal hydrochloride showed modified enzymatic activities. Pyridoxal hydrochloride showed an inhibitory effect on 5 alpha-R in the rat pituitary and basal hypothalamus as well as in the rat prostate.

The women had been previously treated with Clomid, but according to indirect evidence this drug either failed to induce ovulation or the women had insufficient luteal phase. After treatment with LH-RH one pregnancy occurred and in 3 more cases indirect evidence indicated that ovulation had occurred.

Herbicide atrazine (A) effect on reproductive processes in male rat has been examined at anteroir... more Herbicide atrazine (A) effect on reproductive processes in male rat has been examined at anteroir pituitary (AP), testis (T), prostate (P) and seminal vesicles (SV). In the present study A in corn oil was administred to 90 days old rats i.p. at 120 mg/kg b.w./each 72h/60 days. Exposure of rats had induced changes in LH and FSH synthesis, changes of spermatogenesis and structural changes at all selected tissues. LH and FSH were detected by immunohistochemical method, structural changes by light and electron microscopy, while spermatogenesis has been followed by measuring concentration, total number, motility and progressive motility of sperms. A-males in AP have decreased number of immunoreactive cells for LH 25%, while FSH increased for 70%. In T control FSH is dominantly present on the perifery and is concentrated with the spermatocytes and spermatogonia. In A-rats FSH presence is within the whole irregular organized seminiferous tubules. Histochemistry of P tissue shows the tubular alveolar gland with large irregular cavities in A-rats. Our previous research on DHT-receptors in prostate showed the reduction of DHT specific binding sites on receptor molecules. At SV evidently mucosal folds are more tighty packed in A-rats vs. control. The weight of SV in A-rats was much lower and a typical atrophy of the gland is present. Inhibition of spermatogenesis in A-treated rats was in sperm count from 14.3x106 per ml vs. 17.06106 in controls. At the same time the motile sperm count has been reduced from 43% on 35%, respectively.
