Oxidative stress is universal in diabetes, being ultimately involved with the development complications. As a result of hyperglycemia, reactive oxygen/nitrogen species are produced in various tissues that leads to tissue damage with lipid peroxidation and protein oxidation, along with disruption in cellular homeostasis and accumulation of damaged molecules. Hence, supplementation with antioxidant compounds may offer some protection against diabetic complications. The pleiotropic effects of statins, including antioxidant and anti-inflammatory properties, represent an area of great interest in prevention and therapy of cardiovascular and neurological disorders. Using biomarkers of oxidative stress, in this study we examined the effect of non cholesterol lowering dose, long term fluvastatin treatment on oxidative stress in streptozotocin-diabetic rats. Experiments were conducted in 24 Wistar adult male rats. Diabetic and non-diabetic rats were treated orally for 6 months with fluvastatin (2 mg/kg/day, p.o) starting one week after streptozotocin injection (55 mg/kg, i.p.), (preventive study). In brain, heart, liver, pancreas and kidney homogenates malondialdehyde, lipid hydroperoxide, protein carbonyl content, advanced oxidation protein products, 3-nitrotyrosine levels and superoxide dismutase, catalase activities were measured. Hyperglycemia and dyslipidemia in diabetic groups remained unchanged after fluvastatin treatment. The drug act as antioxidant in the tissues. Hence, antioxidant property of fluvastatin, independent of cholesterol lowering effect, may play a role in prevention of diabetic complications. Clinical relevance of this effect of fluvastatin seems worthy of further studies.

Long term statin therapy can cause a modest increase in new-onset diabetes risk, and there is great interest in the mechanisms for this adverse effect. Main proposed mechanisms are increased insulin resistance and some defects in insulin secretion. Many factors can affect the risk including pre-existing diabetic risk, older age and potency of statin. But clearly, the benefits of these drugs in preventing cardiovascular disease outweigh the potential risk of diabetes.The aim of this review is to give underlying pathomechanisms and clinical relevance of diabetogenic effect of statins. Though flaxseed (FXS) is known for improving cardiovascular health, only limited studies are available on FXS concerning diabetic vascular reactivity. Hence, in this study, we studied vascular reactivity changes after FXS treatment on streptozotocin (STZ)-induced diabetic rat aortae. Female Wistar rats were divided into following four groups: control (C), FXS treated (CT), diabetic (D), and FXS treated diabetic (DT) groups. FXS (0,714 g/kg/day; orally) was started after one week of STZ injection and was given for 12 weeks, phenylephrine (Phe)-induced contractions were obtained on isolated aortic rings in the presence of indomethacin, L-NAME and superoxide dismutase (SOD), individually. Phe-responses were increased significantly in D group and completely improved after FXS intake, whereas FXS increased vascular reactivity to Phe in C group. Indomethacin incubation significantly attenuated Phe-induced contractions in all groups of aorta, particularly in D group. L-NAME incubation significantly increased Phe responses in all groups except D group. SOD incubation decreased the contractions efficiently in D group. The decreament was much lower in DT compared to D group, but reverse effects were observed in CT group. Our findings suggest FXS may provide beneficial effects on diabetes-induced vascular reactivity changes through NO and prostaglandin dependent pathways, but in healty condition FXS may have adverse effect probably via pro-oxidant activity.

It was also investigated whether insulin sensitizer thiazolidinedione drugs, pioglitazone and rosiglitazone, can directly affect the reactivity of IMA. Using organ bath techniques, cumulative concentration-response curves to phenylephrine (PE), KCl, cromakalim (CRO) and sodium nitroprusside (SNP) were constructed in diabetic and non-diabetic IMA rings. Means of maximal responses (% Emax) and pEC50 values (sensitivity) were compared. Emax values and the sensitivity to PE and KCl were increased while KATP-channel-mediated relaxations were reduced significantly in diabetic rings compared with non-diabetic rings (n = 5–12, P < 0.05). Diabetes mellitus, a metabolic disorder, is characterized by absolute or relative deficiencies in... more Diabetes mellitus, a metabolic disorder, is characterized by absolute or relative deficiencies in insulin secretion and/or insulin action associated with chronic hyperglycemia. The prevalence of diabetes is increasing worldwide, especially in developing countries. The diabetes treatment has higher costs, limited efficacy and side effects. As a result of these factors, patients often have used alternative forms of therapy such as herbal medicines. Plants often contain various amounts of phenolics, flavonoids and tannins and most of the studies are focused on the antidiabetic effects of these phytochemicals due to their antioxidant properties. In this review, the role of oxidative stress on diabetes and the effects of different phytochemicals (limonene, sinnamic acid and ursolic acid) to diabetes mellitus therapy will be discussed.

This study investigated the effects of the powerful antioxidant SMe1EC2 (2-ethoxycarbonyl-8-methoxy-2,3,4,4a,5,9b-hexahydro-1H-pyrido[4,3-b] indolinium dichloride) on pentose phosphate pathway (PPP) and glutathione-dependent enzyme activities in aged diabetic and aged matched control rats. Diabetes was induced by streptozotocin injection in rats aged 13-15 months. Diabetic and control rats were divided into two subgroups, one untreated and one treated with SMe1EC2 (10 mg/kg/day, orally) for 4 months. SMe1EC2 ameliorated body weight loss, but not hyperglycemia of aged diabetic rats. Amaç: Metabolik ve kronik bir hastalık olan diyabetin birçok ülkede önemli sağlık sorunu olma dur... more Amaç: Metabolik ve kronik bir hastalık olan diyabetin birçok ülkede önemli sağlık sorunu olma durumu artış göstermektedir. Diyabetin artmış serbest radikallerin üretimi ve azalmış antioksidan kapasiteyle ilişkili olduğu iddia edilmektedir. Diyabette oluşan oksiatif stres dokularda DNA hasarına neden olabilir. Antioksidan, antikanser, antiülser özellikler gibi sağlığa yararlı etkileri nedeniyle geleneksel Çin halk tıbbında yaygın olarak kullanılan ursolik asit çok iyi tanımlanmış pentasiklik triterpendir. Bu çalışmanın amacı, streptozotosin ile diyabet oluşturulmuş Wistar albino sıçanların böbreklerine ursolik asidin etkisini değerlendirmektir. Gereç ve Yöntemler: Bu amaçla, böbrek hücrelerindeki DNA hasarı alkali comet yöntemiyle araştırılmıştır. KAT, SOD, GR ve GSH-Px enzim aktiviteleri ve toplam GSH ve MDA düzeyleri gibi oksidatif stres parametreleri de ölçülmüştür. Bulgular: Ursolik asit tedavisinin, DNA hasarını, GR enzim aktivitelerini ve MDA seviyelerini önemli ölçüde azalttığı ve diyabetik sıçanlarda GSH düzeylerini ve CAT, SOD ve GSH-Px enzim aktivitelerini önemli ölçüde artırdığı bulundu. Sonuç: Sonuçlarımıza göre, ursolik asidin diyabetin neden olduğu böbrek hasarında yararlı olabileceği görülmüştür. It has been claimed that diabetes is associated with the increased formation of free radicals and decreased in antioxidant potential. Oxidative stress formed in diabetes may cause DNA damage in the tissues. Ursolic acid, a well-known pentacylic triterpene, is commonly used in traditional Chinese medicine due to its beneficial health effects such as antioxidant, anticancer, and antiulcer properties. The aim of this study was to investigate the effects of ursolic acid in the kidneys of Wistar albino rats with streptozotocin-induced diabetes. Materials and Methods: DNA damage was evaluated in the kidney cells of rats using alkaline comet assays. Oxidative stress parameters such as CAT, SOD, GR, and GSH-Px enzyme activities and total GSH and MDA levels were also evaluated. Results: Ursolic acid treatment was found to significantly decrease DNA damage, GR enzyme activities, and MDA levels, and significantly increase GSH levels and CAT, SOD and GSH-Px enzyme activities in diabetic rats. Conclusion: According to our results, it seems that ursolic acid may be beneficial against diabetes-induced renal damage.

Diyabetin artmış serbest radikallerin üretimi ve azalmış antioksidan kapasiteyle ilişkili olduğu iddia edilmektedir. Diyabette oluşan oksiatif stres dokularda DNA hasarına neden olabilir. Antioksidan, antikanser, antiülser özellikler gibi sağlığa yararlı etkileri nedeniyle geleneksel Çin halk tıbbında yaygın olarak kullanılan ursolik asit çok iyi tanımlanmış pentasiklik triterpendir. Bu çalışmanın amacı, streptozotosin ile diyabet oluşturulmuş Wistar albino sıçanların böbreklerine ursolik asidin etkisini değerlendirmektir. Gereç ve Yöntemler: Bu amaçla, böbrek hücrelerindeki DNA hasarı alkali comet yöntemiyle araştırılmıştır. KAT, SOD, GR ve GSH-Px enzim aktiviteleri ve toplam GSH ve MDA düzeyleri gibi oksidatif stres parametreleri de ölçülmüştür. Bulgular: Ursolik asit tedavisinin, DNA hasarını, GR enzim aktivitelerini ve MDA seviyelerini önemli ölçüde azalttığı ve diyabetik sıçanlarda GSH düzeylerini ve CAT, SOD ve GSH-Px enzim aktivitelerini önemli ölçüde artırdığı bulundu. Sonuç: Sonuçlarımıza göre, ursolik asidin diyabetin neden olduğu böbrek hasarında yararlı olabileceği görülmüştür. It has been claimed that diabetes is associated with the increased formation of free radicals and decreased in antioxidant potential. Oxidative stress formed in diabetes may cause DNA damage in the tissues. Ursolic acid, a well-known pentacylic triterpene, is commonly used in traditional Chinese medicine due to its beneficial health effects such as antioxidant, anticancer, and antiulcer properties. The aim of this study was to investigate the effects of ursolic acid in the kidneys of Wistar albino rats with streptozotocin-induced diabetes. Materials and Methods: DNA damage was evaluated in the kidney cells of rats using alkaline comet assays. Oxidative stress parameters such as CAT, SOD, GR, and GSH-Px enzyme activities and total GSH and MDA levels were also evaluated. Results: Ursolic acid treatment was found to significantly decrease DNA damage, GR enzyme activities, and MDA levels, and significantly increase GSH levels and CAT, SOD and GSH-Px enzyme activities in diabetic rats. Conclusion: According to our results, it seems that ursolic acid may be beneficial against diabetes-induced renal damage.
