Persistent and recurrent infection of hepatitis B virus (HBV) represents one of the most common a... more Persistent and recurrent infection of hepatitis B virus (HBV) represents one of the most common and severe viral infections of humans, and has caused a formidable health problem in the affected countries. Currently used antiviral drugs have a very limited success on controlling HBV replication and infection. RNA interference (RNAi), a process by which double-stranded RNA (dsRNA) directs sequence-specific degradation of target mRNA in mammalian and plant cells, has recently been used to knockdown gene expression in various species. In this study, we sought to determine whether RNAi-mediated silencing of HBV viral gene expression could lead to the effective inhibition of HBV replication. We first developed RNAi vectors that expressed small interfering RNA (siRNA) and targeted the HBV core or surface gene sequence. Our results demonstrated that these specific siRNAs efficiently reduced the levels of corresponding viral RNAs and proteins, and thus suppressed viral replication. Treatment with siRNA gave the greatest reduction in the levels of HBsAg (92%) and in HBeAg (85%) respectively in the cultured cell medium. Our findings further demonstrated that the RNAi-mediated antiviral effect was sequence-specific and dose-dependent. Therefore, our findings strongly suggest that RNAi-mediated silencing of HBV viral genes could effectively inhibit the replication of HBV, hence RNAi-based strategy should be further explored as a more efficacious antiviral therapy of HBV infection.

Abstract Numerical calculations were conducted to simulate the flow and mass transfer in narrow m... more Abstract Numerical calculations were conducted to simulate the flow and mass transfer in narrow membrane channels equipped with delta winglets, which are often used as longitudinal vortex generators to enhance heat transfer in heat exchanger applications. The channel consists of an impermeable solid wall and a membrane. The delta winglets are attached to the solid wall surface to enhance the mass transfer near the membrane surface and suppress the concentration polarization. The winglet performance was evaluated in terms of concentration polarization factor versus consumed pumping power. Calculations were implemented for NaCl solution flow in a membrane channel having a height of 2.0 mm for Reynolds numbers ranging from 400 to 1000. The delta winglets were optimized under equal pumping power condition, and the results of optimization suggest winglet height of 5/6 of the channel height, aspect ratio of 2.0, attack angle of 30°, and a winglet interval equal to the channel height. The optimal delta winglets were compared with the optimal rectangular winglets we found previously, and it is shown that the rectangular winglets yield a somewhat better performance than the delta winglets.

However, its biological function in hepatocellular carcinoma still remains unclear. Here, we investigated the effects of Rab23 on proliferation and migration in hepatocellular carcinoma cell and its potential mechanisms. We found over-expression of Rab23 promoted the Hep3B hepatocellular carcinoma cell migration, which could be reversed by Rab23 silencing. Rab23 induced Rac1 activation and followed progression of epithelial-mesenchymal transition (EMT) along with upregulation of N-cadherin, snail as well as vimentin and downregulation of E-cadherin via upregulating Transforming Growth Factor-β (TGF-β). Silencing Rac1 significantly attenuated Rab23-induced HepG2 migration and TGF-β. Moreover, knockdown of TGF-β effectively attenuated Rab23-induced EMT. Nowadays, the world demand for natural gas as a source of energy has been increased. Since gas pr... more Nowadays, the world demand for natural gas as a source of energy has been increased. Since gas production is constant during the year and its consumption is almost seasonal, underground gas storage (USG) is a common activity in countries with major transport and distribution gas pipeline infrastructures, which allows to efficiently resolve demand seasonality problem. In this paper, UGS was studied on a gas driving depleted reservoir through numerical simulation. A mass conservation equation based 3-D single-phase gas flow and a PVT equation were improved to construct the USG simulator equipped with the functions for calculating physical phenomena: 1) cyclic gas injection and continuous gas injection, 2) the average pressure change of the formation, 3) the gas storing capacity of the UGS, 4) the distribution of saturation after the gas injection. In addition, the Forchheimer equation was introduced to modify the Darcy equation, which included the influence of inertial force and made the simulation more accurate and available.
