The products were evaluated for physicochemical, microbiological, and sensory properties during cold storage (7 °C, 28 days). Probiotic products showed higher acidity (lower pH value and higher lactic acid content), better fatty acid profile (higher concentration of linoleic acid), and a higher concentration of bioactive compounds (⍺-tocopherol and phenolics) than the control product. Inulin-added products showed the best performance, with higher probiotic viability and consistency index. Furthermore, they presented a higher concentration of bioactive compounds (β-sitosterol, stigmasterol, and total phytochemicals) and improved anti-diabetic properties (45.4% of inhibition of ⍺-amylase). Fermented milk appearance, acid aroma, and typical flavor were the main sensory attributes of the inulin-added products. Probiotic counts were higher than 106 CFU/mL during shelf life and simulated gastrointestinal conditions in all products added with probiotics. Synbiotic FBs processed with WSEBA were obtained and characterized for the first time. In conclusion, L. casei and prebiotics improve the technological, functional, and sensory properties of FBs processed with WSEBA, being advisable to utilize L. casei and inulin.

Crystalline mesoporous molecular sieves ILM-Al have been prepared in strongly acidic ionic liquid (IL) media, and used as catalysts for the self-oxidation of alkali lignin to aromatic acids. In the Page 1 of 40 ACS Paragon Plus Environment ACS Sustainable Chemistry & Engineering preparation of the new type of mesoporous molecular sieves, the imidazolium-based acidic functional IL system served as both solvent and structure-directing agent. The ILM-Al exhibited higher catalytic performance and selectivity for aromatic acids in the self-oxidation of lignin than in that of fossil fuels, and the main products were homoveratric acid and homovanillic acid. The catalyst could be recycled and reused with negligible loss in activity over five cycles, and the IL could also be recycled for further use. Moreover, Zr and Sn were also successfully incorporated into the framework of ordered crystalline mesoporous materials by the same simple method. The obtained ILM-Zr and ILM-Sn also showed excellent catalytic activity for the self-oxidation of alkali lignin.

Functional analyses were complemented within silicoprediction of mutation pathogenicity based on the recently crystallized human CYP21A2 structure. Mutated proteins were transiently expressed in COS-1 cells and enzyme activities towards 17-hydroxyprogesterone and progesterone were determined. Residual enzyme activities between 43% and 97% were obtained for p.Arg16Cys, p.Ser101Asn, p.Ser202Gly, p.Pro267Leu, and p.Thr450Met, similar to the activities of the well-known nonclassic mutations p.Pro453Ser and p.Pro482Ser, whereas the p.Leu12Met variant showed an activity of 100%. Conversely, the novel p.Ser113Phe, p.Gln389_Ala391del, and p.Thr450Pro mutations drastically reduced the enzyme function below 4%. TheKmvalues for all novel vari...

Research paper thumbnail of Vertebral osteomyelitis associated with single and mixed bacterial infection in broilers

The objective of this study was to determine the frequency and aetiology of VO in broilers in a highly productive broiler region. For this, 608 broilers with locomotory problems were analysed from 18 farms. Clinical signs were recorded, necropsy was performed and samples were collected from vertebral bodies with gross changes for molecular and histopathological analysis and for bacterial isolation. From broilers with locomotory changes, 5.1% (31/608) had VO and, of these, 93.5% were 40 days old or older and 89.7% were males. The birds with VO presented varying degrees of limited mobility and this was related to the level of compression to the spinal cord. Bacterial species of the genus Enterococcus (DNA detected in 53.6%) were the aetiological agents involved in most VO cases. Enterococcus faecalis was detected most frequently (35.7%), but Enterococcus hirae was also present in some lesions (7.1%). Escherichia coli was detected in 35.7% of vertebral lesions and co-infection with E. faecalis was confirmed in 7.1% cases. Staphylococcus aureus was involved in 14.3% of the cases, being 7.1% in co-infection with Enterococcus spp. or E. hirae. Our study has indicated that, in Brazil, VO in broilers may not be caused by a single infectious agent and has a lower frequency than recently reported in other countries. This study suggests that there are geographical differences between Brazil and other countries concerning the frequency and aetiology of VO.

The water of the beaches 1, 2 and 3 was considered unsuitable for bathing (40%, 60% and 80%, respectively). Enterococcus faecalis was the predominant species (63.5%) in the water of the three beaches, followed by E. faecium (36.5%). Most lineages (36.5%) were isolated in beach 2; 34.6% were isolated in beach 1, and 28.8% in 3. High antimicrobial resistance to nalidixic acid (94.3%), tetracycline (26.4%) and amicacine (26.0%) was observed. All strains showed 100% susceptibility to ampicillin and imipenem. Enterococcus faecalis was also susceptible to vancomycin and E. faecium to penicillin. Multiple resistance profile was observed in 88.5% (MAR >0.18) of the strains, three of which were resistant to six antimicrobials. Considering the resistance increase of autochthonous bacteria observed in the marine environment, special attention should be paid to the multiple resistance profile of the isolates.

The potential impact of human respiratory coronavirus (CoV) infections was underestimated until the severe acute respiratory syndrome (SARS-CoV) outbreak in 2003, which showed that new, highly pathogenic coronaviruses could be introduced to humans, highlighting the importance of monitoring the circulating coronaviruses. The use of sensitive molecular methods has contributed to the differential diagnosis of viruses circulating in humans. Our study aim was to investigate the molecular epidemiology of human CoV strains circulating in Arkansas, their genetic variability and their association with reported influenza-like symptoms. We analyzed 200 nasal swab samples, collected by the Arkansas Department of Health in 2010, for influenza diagnosis. All samples were from patients showing acute respiratory symptoms while testing negative for influenza. Samples were pre-screened, using a quantitative reverse transcriptase polymerase chain reaction (qRT-PCR) multiprobe for coronavirus, and subjected to confirmatory pancoronavirus and/or strain-specific reverse transcriptase (RT)-PCR followed by sequence analysis. Unfortunately, most are inappropriately used leading to increased multi-resistant bacteria. It is well known that initial empirical therapy with broad-spectrum antibiotics reduce mortality rates. However the prolonged and irrational use of antimicrobials may also increase the risk of toxicity, drug interactions and diarrhea due to Clostridium difficile. Some strategies to rational use of antimicrobial agents include avoiding colonization treatment, de-escalation, monitoring serum levels of the agents, appropriate duration of therapy and use of biological markers. This review discusses the effectiveness of these strategies, the importance of microbiology knowledge, considering there are agents resistant to Staphylococcus aureus andKlebsiella pneumoniae, and reducing antibiotic use and bacterial resistance, with no impact on mortality.

Research paper thumbnail of Involvement of astroglial fibroblast growth factor-2 and microglia in the nigral 6-OHDA Parkinsonism and a possible role of glucocorticoid hormone on the glial mediated local trophism and wound repair

We have observed in previous studies that 6-hydroxydopamine (6-OHDA)-induced lesions in the nigro... more We have observed in previous studies that 6-hydroxydopamine (6-OHDA)-induced lesions in the nigrostriatal dopamine (DA) system promote increases of the astroglial basic fibroblast growth factor (FGF-2, bFGF) synthesis in the ascending DA pathways, event that could be modified by adrenosteroid hormones. Here, we first evaluated the changes of microglial reactivity in relation to the FGF-2-mediated trophic responses in the lesioned nigrostriatal DA system. 6-OHDA was injected into the left side of the rat substantia nigra. The OX42 immunohistochemistry combined with stereology showed the time course of the microglial activation. The OX42 immunoreactivity (IR) was already increased in the pars compacta of the substantia nigra (SNc) and ventral tegmental area (VTA) 2 h after the 6-OHDA injection, peaked on day 7, and remained increased on the 14th day time-interval. In the neostriatum, OX42 immunoreactive (ir) microglial profiles increased at 24 h, peaked at 72 h, was still increased at 7 days but not 14 days after the 6-OHDA injection. Two-colour immunofluorescence analysis of the tyrosine hydroxylase (TH) and OX42 IRs revealed the presence of small patches of TH IR within the activated microglia. A decreased FGF-2 IR was seen in the cytoplasm of DA neurons of the SNc and VTA as soon as 2 h after 6-OHDA injection. The majority of the DA FGF-2 ir cells of these regions had disappeared 72 h after neurotoxin. The astroglial FGF-2 IR increased in the SNc and VTA, which peaked on day 7. Two-colour immunofluorescence and immunoperoxidase analyses of the FGF-2 and OX42 IRs revealed no FGF-2 IR within the reactive or resting microglia. Second, we have evaluated in a series of biochemical experiments whether adrenocortical manipulation can interfere with the nigral lesion and the state of local astroglial reaction, looking at the TH and GFAP levels respectively. Rats were adrenalectomized (ADX) and received a nigral 6-OHDA stereotaxical injection 2 days later and sacrificed up to 3 weeks after the DA lesion. Western blot analysis showed time-dependent decrease and elevation of TH and GFAP levels, respectively, in the lesioned versus contralateral midbrain sides, events potentiated by ADX and worsened by corticosterone replacement. ADX decreased the levels of FGF-2 protein (23 kDa isoform) in the lesioned side of the ventral midbrain compared contralaterally. The results indicate that reactive astroglia, but not reactive microglia, showed an increased FGF-2 IR in the process of DA cell degeneration induced by 6-OHDA. However, interactions between these glial cells may be relevant to the mechanisms which trigger the increased astroglial FGF-2 synthesis and thus may be related to the trophic state of DA neurons and the repair processes following DA lesion. The findings also gave further evidence that adrenocortical hormones may regulate astroglial-mediated trophic mechanisms and wound repair events in the lesioned DA system that may be relevant to the progression of Parkinson&amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;#39;s disease.

Research paper thumbnail of Continuous production of fatty acid ethyl esters from sunflower oil using supercritical ethanol

Fuel, 2012

The production of first generation biofuels has increased in the last few years because of the ri... more The production of first generation biofuels has increased in the last few years because of the rising price of fossil fuels and environmental policies. Biodiesel process alternatives have been proposed in order to achieve higher yields, non-catalytic and more environmentally friendly processes, a greater profit from by-products and particularly the possibility of using low quality and cheaper feedstocks. In this regard, one of the most studied technologies has been the non-catalytic supercritical transesterification of fats and raw vegetable oils. This work reports results on the continuous production of fatty acid ethyl esters (FAEEs) from the noncatalytic supercritical ethanolysis of sunflower oil. The reaction was carried out in the following range of operating conditions: (i) ethanol-to-oil molar ratio of 40:1; (ii) temperature and pressure range, 573-618 K and 165-200 bar and (iii) mass flow rates varying from 3 to 16 g/min. Given the high sensitivity of the specific density in supercritical mixtures, the mixture residence time in the reactor was estimated based on experimental data of the reactive mixture density. Moreover, we show here that overlooking the use of this experimental data and assuming ideal solution behavior for density calculation, which is usually done, may lead to important deviations in the kinetic model parameters. The mixture densities were correlated with the Peng-Robinson equation of state (PR-EoS). In the range of operating conditions studied in this work, up to 90% by mass fraction of fatty ester was obtained.

Fuel, 2012
