Aims To investigate the expression of matrix metalloproteases (MMPs) and their inhibitors (TIMPs)... more Aims To investigate the expression of matrix metalloproteases (MMPs) and their inhibitors (TIMPs) in patients who develop local recurrence (LR) after mastectomy. Methods We analyzed the expressions of MMP-1,-2,-7,-9,-11,-13,-14, TIMP-1,-2, and-3, using immunohistochemical techniques, in primary tumors from patients without tumoral recurrence (n = 50), patients who developed distant metastasis (n = 50), and from patients who develop LRs (n = 25). LRs of the latter group were also analyzed for MMPs expression. All the patients underwent mastectomy. Results Score values for all MMPs and TIMPs were signiWcantly higher in primary tumors of patients with distant metastasis. Primary tumors from patients with LR have lower expressions of MMPs and TIMPs compared with those from patients who developed distant metastasis, and with patients without recurrence for some MMPs. Remarkably, however, primary tumors from patients with LR showed signiWcantly higher percentage of TIMP-1 and 2 expression in stromal cells compared to primary tumors from patients with distant metastasis or primary tumors from patients without tumoral progression. Furthermore, LRs had signiWcantly higher MMP-9 expression than their corresponding primary tumors. Conclusions Our data indicate diVerences in MMPs/ TIMPs expression between primary tumors of patients with LRs and of those with distant metastasis, both after mastectomy for breast cancer.

Th... more Some studies have investigated the association between motor development and cognitive skills. The present study aims to correlate motor development and academic performance in 79 students between 7 to 9 years old from a private school in São Paulo-Brazil. We used the Movement Assessment Battery for Children - Second Edition (MABC-2) to assess the motor development and the results of school report in Portuguese and Mathematics as a measure of academic performance. Pearson's correlation analysis showed a positive association between motor development and cognitive skills, evidencing that students who had better academic performance, also obtained better scores in motor performance. The association between MABC-2 total score and Portuguese grade, for example, obtained a significant correlation of p <0.014. Further studies should be carried out to understand if motor development improve can modulate cognitive skills.

We have measured the energy spread of the Booster beam at its injection energy of 400 MeV by thre... more We have measured the energy spread of the Booster beam at its injection energy of 400 MeV by three different methods: (1) creating a notch of about 40 nsec wide in the beam immediately after multiple turn injection and measuring the slippage time required for high and low momentum particles for a grazing touch in line-charge distribution, (2) injecting partial turn beam and letting it to debunch, and (3) comparing the beam profile monitor data with predictions from MAD simulations for the 400 MeV injection beam line. The measurements are repeated under varieties of conditions of rf systems in the ring and in the beam transfer line.

The Fermilab Proton Source has an upgrade plan called the Proton Improvement Plan (PIP) [1]. The ... more The Fermilab Proton Source has an upgrade plan called the Proton Improvement Plan (PIP) [1]. The PIP has several key objectives which were laid out by laboratory management in 2011. The plan should address the necessary hardware modifications for increased repetition rate to 15 Hz, capable of delivering 2.25E17 protons/hour while maintaining 2012 activation levels, ensuring viable operation of the proton source through 2025 with availability greater than 85%. Fermilab believes the PIP goals are necessary to ensure that the laboratory delivers on the commitment to the physics program. PIP is therefor considered an urgent and a high-priority activity. The PIP effort started in 2011 and official task management and funding schedule approval in 2012. Largely an accelerator effort, several other laboratory divisions are playing critical roles. The progress of PIP is going to be discussed in this paper.

Recently, three single nucleotide polymorphisms (SNPs) were identified at exons 2, 3, and 5 of the granzyme B gene, resulting in a haplotype in which three amino acids of mature protein Q48P88Y245 are changed to R48A88H245, which leads to loss of cytotoxic activity of the protein. In this study, we evaluated the frequency of these polymorphisms in Brazilian populations. METHODS: We evaluated the frequency of these polymorphisms in Brazilian ethnic groups (white, Afro-Brazilian, and Asian) by sequencing these regions. Cytotoxic T lymphocytes and natural killer cells play a crucial role in eliminating tumour and vi... more Cytotoxic T lymphocytes and natural killer cells play a crucial role in eliminating tumour and virus-infected cells. The perforin is a key part of the arsenal that these cells use to destroy their targets. In this study, we characterized single-nucleotide polymorphisms (SNPs) located in the promoter region of the perforin gene among distinct Brazilian ethnic groups. The study was carried out by sequencing this region in three groups: European, African and Asian descents. We demonstrated for the first time the occurrence of three new polymorphisms in the promoter region of gene PRF1: 494A/G (rs78058707), 720G/A (rs75925789) and 1176C/T (rs75183511). Three other SNPs already described in the literature 63A/G (rs35401316), 112A/ G (rs10999428) and 1012C/T (rs35069510) were also detected. The SNPs are distributed differently in the ethnic groups studied. The 112G allele was observed at high frequency, especially among Asian descents (48.1%). The 1012T allele was detected only among European descents, the 494G allele only among Asian descents and 1176T allele only in African descents. Based on the association between the polymorphisms described, ten new haplotypes were originated. In functional analysis, we noticed that SNPs present in most common haplotypes cannot induce significant differences in expression levels of perforin alone. In conclusion, this study demonstrates for the first time the existence of three new polymorphisms in perforin promoter and, contrary to what was stated, the presence of these SNPs does not alter the levels of protein expression.
