We tested the germination responses of freshly harvested and afterripened (stored for 2 and 7 months; 25C at 50% relative humidity) seeds from three populations (P1, P2 and P3) in Iran at varying constant or alternating temperatures, with or without GA3 and in light and continuous darkness. No germination occurred in freshly harvested seeds incubated at any condition without GA3 application, indicating that all the seeds were dormant. Seeds from P1 and P2, which developed under relatively dry, warm conditions, germinated over a wider range of temperatures after 2 months of dry storage, indicating type 6 of non-deep physiological dormancy (PD). Seeds from P3, which developed under relatively wet, cool conditions, incubated at constant temperatures (especially on GA3), exhibited an increase in maximum temperature for germination, indicating type 1 of non-deep PD. Light improved germination of after-ripened seeds, and GA3 application substituted for the light requirement for germination. This is the first report that environmental conditions during seed development may be correlated with differences in the type of non-deep PD. We conclude that milk thistle seeds are positively photoblastic and photodormant and the germination responses of after-ripened seeds from different populations are different under darkness. Therefore, the impacts of genetic differences and maternal effects on the induction of dormancy during seed development should be considered in attempts to domesticate this medicinal plant.

Research paper thumbnail of Continuous Use of Tribenuron-Methyl Selected for Cross-Resistance to Acetolactate Synthase–inhibiting Herbicides in Wild Mustard (Sinapis arvensis)

Tribenuron-methyl (TM) has been used recurrently to control this species, thus selecting for resistant S. arvensis populations. The objectives were: (1) to determine the resistance level to TM of 14 putatively resistant (PR) S. arvensis populations, collected from winter wheat fields in Golestan province, Iran, in comparison to one susceptible (S) population; and (2) to characterize the resistance mechanisms and the potential evolution of cross-resistance to other classes of acetolactate synthase (ALS)-inhibiting herbicides in three populations (AL-3, G-5, and Ag-Sr) confirmed as being resistant (R) to TM. The TM doses required to reduce the dry weight of the PR populations by 50% were between 2.2 and 16.8 times higher than those needed for S plants. The ALS enzyme activity assays revealed that the AL-3, G-5, and Ag-Sr populations evolved cross-...

The present study has investigated the effect of silicon nutrition on alleviation of Zn toxicity in rice plants. Plants were cultured hydroponically with three different Zn concentrations 3 levels of Zn including 10, 100 and 150 gL Zn as zinc sulfate and two levels of Si including 0, 1.5 mM as sodium silicate. Excess Zn imposed a significant retardation in growth along with a reduction of chlorophyll, carotenoids, lignin and soluble proteins under 150 gL Zn. Also, Zn toxicity decreased the activity of catalase in shoot and soluble peroxidase in both root and shoot. Consequently, H2O2 content and lipid peroxidation level increased under Zn stress compared with the control plants. On the contrary, Si application reduced Zn accumulation, H2O2 content and lipid peroxidation level under 150 gL Zn. Therefore, the Si-nourished plants showed an increased amount of chlor...

Research paper thumbnail of Catalase and Metallothionein genes expression analysis in wheat cultivars under drought stress condition

Drought stress is one of the serious problems that restricted agronomic plant production worldwid... more Drought stress is one of the serious problems that restricted agronomic plant production worldwide. In molecular level, the harmful effect of drought stress is mostly caused by producing of large amount of reactive oxygen species (ROS). Catalase and Metallothionein genes have a crucial role to mope the hydrogen peroxide (H2O2) resulting reducing oxidative damage. In this research the gene expression pattern of Catalase and Metallothionein was studied in response to drought stress treatments. The treatments included - 0.3 bar, - 0.9 bar, - 8 bar and -12 bar and wheat varieties included Zagros (drought tolerant), Moghan (semi- tolerant) and Tajan (drought sensitive). The amount of cellular oxidative levels (TBARM) increased steady by intensify of drought stress levels. Real time PCR analysis showed different expression pattern for catalase and metallothionein encoded genes. Catalase gene expression was increased during drought stress up to -8 bar and reduced in -12 bar treatment, in a...

This herbicide plays a critical role in post-emergence weed management options for cereals especially in Golestan province in Iran. Recently, failures to control wild mustard (Sinapis arvensis L.) and turnipweed (Rapistrum rugosum L.) with recommended dose of tribenuron methyl applications have been reported for wheat fields (6 and 20). It motivated us to trace resistant populations and evaluate their degree of resistance and find a quick and cost-effective method to identify resistant populations in wild mustard and turnipweed . Materials and Methods: At first, seeds were collected from wheat fields in Golestan province. These fields had been treated annually with tribenuron-methyl for more than 5 years, so wild mustard and turnipweed control had become erratic with this herbicide in recent years. Seeds were also coll...
