From 217 diabetic women over thirty-nine years of age attending a diabetes clinic, there were sel... more From 217 diabetic women over thirty-nine years of age attending a diabetes clinic, there were selected for study twenty-seven who had had an early (< 12 yrs.), and thirty-seven who had had a late (> 16 yrs.) menarche. Patients in both groups were obese, but those in the former were much more so than in the latter (85.4 kg. vs. 75.4 kg.; 156 cm. vs. 158 cm.). Hypertension and hypertensive vascular complications were more prevalent in the first group, while ketoacidosis and retinal microaneurysms were almost absent. The group with late menarche developed retinal microaneurysms much more rapidly than the other group and perhaps even than the general population of the clinic.

The method is compared with the indirect immunofluorescence technique and with an E-rosette test.

Platelet aggregation in response to 5-hydroxytryptamine was investigated in 40 normal subjects, i... more Platelet aggregation in response to 5-hydroxytryptamine was investigated in 40 normal subjects, in 45 patients with acute myocardial infarction, and in 65 patients with peripheral arterial obstructive disease. It was found that of the 110 patients with cardiovascular disease, 40% had a biphasic irreversible platelet aggregation, whereas this phenomenon occurred in only 7.5% of the normal population. A double-blind placebo-controlled study further showed that a subacute treatment with ketanserin, a selective 5-HT2-receptor antagonist both on platelets and on vascular tissue, efficiently abolished the irreversible platelet aggregation in patients hyperreactive to 5-hydroxytryptamine. In an additional open study, including 10 patients with peripheral arterial obstructive disease, a chronic treatment with ketanserin 40 mg t.i.d. for a period of 3 months significantly suppressed the primary platelet aggregation to 5-HT at 2 X 10(-5) M and at 2 X 10(-6) M and significantly lowered the plasma beta thromboglobulin levels. Since 5-HT is a potent mediator of vasospasm, treatment with ketanserin might be of therapeutic value in atherosclerotic diseases, where platelet activation is thought to be involved.

The use of oral oxatomide (30 to 360 mg per day) for the treatment of allergic conditions of the ... more The use of oral oxatomide (30 to 360 mg per day) for the treatment of allergic conditions of the bronchi, skin and gastro-intestinal tract was investigated in a total of 70 subjects. Therapeutic results were evaluated at intervals ranging between 1 and 16 months. Symptomatic improvements were scored globally, forced expiratory volume in 1 second was measured in asthmatic patients, and the lesions of atopic dermatitis were rated according to a modified Clendening scale. The results suggest that oxatomide was particularly effective in the treatment of allergic rhinitis and skin allergies. Allergic asthma responded fairly well in patients with a short history of disease and gastro-intestinal conditions reacted moderately, with haemorrhagic rectitis responding better than gastritis. These findings warrant further controlled trials of oxatomide in such conditions.

T-lymphocytes were counted by E-rosette tests and by the immunogold staining method with OKT3.PAN monoclonal antibody. Helper and suppressor T-lymphocytes were determined by the immunogold staining method with OKT4.IND and OKT8.SUP monoclonal antibody. The relative and absolute numbers of T-lymphocytes and helper T-lympohocytes in peripheral blood of patients with RA did not differ significantly from those in the blood of healthy subjects. However, the relative and absolute numbers of suppressor T-cells were significantly lower in patients with RA than in healthy subjects. The decrease of suppressor T-cells in the blood of patients with RA dit not correlate with the activity of the disease nor the presence of the rheumatoid factor.

In a first experiment, an acute intravenous administration of 10 mg R 41 468, a pure serotonin-re... more In a first experiment, an acute intravenous administration of 10 mg R 41 468, a pure serotonin-receptor blocking agent with high selectivity for blood vessels and thrombocytes and devoid of central effects, dramatically reduced systolic and diastolic blood pressure in 23 elderly hypertensive patients. Heart rate and cardiac output remained virtually unchanged. In a second double-blind placebo-controlled cross-over study a highly significant decrease of systolic and diastolic blood pressure was obtained in 14 elderly hypertensive patients during an 8-day oral treatment with 40 mg t.i.d. of R 41 468. No serious side-effects were observed. An oral maintenance therapy with R 41 468 for 3 weeks showed a further reduction of blood pressure, resulting in a normalization of blood pressure, taking into account the advanced age of the patients. However, patients with cardiovascular (CV) diseases have been reported to have enhanced platelet activity and to show hyperaggregability in response to common aggregators. Very little is known on 5-hydroxytryptamine (5-HT)-induced platelet aggregation in these patients. In a previous preliminary study we observed an increased sensitivity of platelets in response to 5-HT in patients with CV diseases. We further showed that ketanserin, a selective 5-HT receptor antagonist both on platelets and vascular tissue, abolished the 5-HT dependent hyperreactivity of platelets in patients with CV diseases. In a prospective study we investigated platelet aggregation in response to 5-HT at 2 x 10-5Mol in 405 patients with various CV diseases as compared with an age-matched control group of 110 apparently healthy donors. Red blood cell (RBC) filtration in platelet rich plasma (PRP) and platelet poor plasma (PPP) was ... more Red blood cell (RBC) filtration in platelet rich plasma (PRP) and platelet poor plasma (PPP) was equally decreased (p < 0.0001) in 120 patients with acute myocardial infarction (AMI) as compared to a control group. In a double-blind experiment 2 groups of 30 patients with AMI received an acute oral dose of 60 mg of ketanserin, a serotonin (5-HT) antagonist at 5-HT2-receptors, or placebo. Ketanserin treatment improved RBC filtration in PRP with an average increase of 30%. A similar experiment using PPP showed a significant increase of 10%. Filtration of plasma improved after ketanserin treatment in PRP, but not in PPP. Cross-exchange experiments showed the ketanserin-induced improvement of RBC filtration in PRP and PPP to be also plasmadependent. 5-HT in vitro at 10−9M deteriorated RBC filtration in PPP (p < 0.05), and ketanserin in vitro at 10−7M counteracted this phenomenon (p < 0.001). Serum hemolytic complement activity and its components C3, C4 and C1q, serum immunoglobulins A, M... more Serum hemolytic complement activity and its components C3, C4 and C1q, serum immunoglobulins A, M and G and peripheral E-rosette forming cells were determined in 11 patients with inflammatory acne. IgG levels were increased and the E-rosette formation was markedly impaired. Levamisole treatment restored the E-rosette formation and improved or cured the inflammatory lesions in 10 out of 11 patients.

The effects of thymopoietin pentapeptide (TP5) and levamisole on human peripheral T lymphocytes w... more The effects of thymopoietin pentapeptide (TP5) and levamisole on human peripheral T lymphocytes were investigated by means of the E-rosette formation. Different kinds of E-rosette forming cells (RFC) were measured, i.e. active RFC (RFCa), total RFC (RFCt) and azathioprine-inhibited RFCt. The effects of TP5 and levamisole on RFC were very similar. Azathioprine inhibition of RFCt was restored completely by levamisole and partially by TP5. Both substances enhanced RFCa of healthy subjects and RFCt of patients in which these were low. TP5 and levamisole may act via a common cyclic nucleotide second signal.
