The Zika virus (ZIKV) is a vector-borne flavivirus that has been detected in 87 countries worldwi... more The Zika virus (ZIKV) is a vector-borne flavivirus that has been detected in 87 countries worldwide. Outbreaks of ZIKV infection have been reported from various places around the world and the disease has been declared a public health emergency of international concern. ZIKV has two modes of transmission: vector and non-vector. The ability of ZIKV to vertically transmit in its competent vectors, such as Aedes aegypti and Aedes albopictus, helps it to cope with adverse conditions, and this could be the reason for the major outbreaks that occur from time to time. ZIKV outbreaks are a global threat and, therefore, there is a need for safe and effective drugs and vaccines to fight the virus. In more than 80% of cases, ZIKV infection is asymptomatic and leads to complications, such as microcephaly in newborns and Guillain–Barré syndrome (GBS) in adults. COVID-19 is a respiration-related disease caused by SARS-CoV-2 and was identified in China’s Wuha... more COVID-19 is a respiration-related disease caused by SARS-CoV-2 and was identified in China’s Wuhan city. More than 223 countries are affected by the disease worldwide. The new variants of the COVID-19 virus are causing problems, from average to life-threatening pneumonia and acute respiratory distress syndrome (ARDS). Presently, there are 170 vaccine candidates, out of which 10 have been approved by the WHO for vaccination, such as Ad26.COV2.S, Pfizer/BioNTech, COVISHIELD, Covovax, Moderna, KoviVac, and some other vaccines to combat the deadly SARS-CoV-2 infection. From all these vaccines, Pfizer/BioNTech and Moderna are showing the highest efficacy against COVID-19. These vaccines are highly efficient against COVID-19 disease, but their potentiality against new variants remains a question. Transmission blocking vaccines mainly focus on the target antigens which are immunogenic in natur... more Transmission blocking vaccines mainly focus on the target antigens which are immunogenic in nature and play an important role in parasite development. Anopheles SGU (Secretory Glycoconjugate of Unknown function) is the peritrophic matrix specific protein, involves in parasite infection and development in midgut. Here, SGU protein of Anopheles stephensi is targeted to develop a vector based Transmission Blocking vaccine (TBVs). Presently various immune-bioinformatics approaches are used to design the epitope based vaccine construct based on consensus antigenicity prediction and various physiochemical criteria like transmembrane domain, allergenicity, GRAVY index, toxicity and its stability index that can activate both humoral and acquired immunity. There are 4 B-cell, 5 cytotoxic T-cell and 8 T helper cell epitope were selected from IEDB online tool based on its immunogenicity and population coverage. The selected epitope are adjoin using linkers to formulate Transmission blocking vaccine construct. To enhance the immune response we use β-defensin as an adjuvant. The design vaccine construct is encoded for 392 amino acid and from molecular docking it confirmed that our vaccine construct bind efficiently with TLR8 & TLR9. MD simulation and immune simulation conformed that the designed vaccine construct is flexible and highly immunogenic in nature.
