The Aurora kinases represent a novel family of serine/threonine kinases critical for cell cycle c... more The Aurora kinases represent a novel family of serine/threonine kinases critical for cell cycle control. As knowledge of the roles that the Aurora kinases play in mitotic events continues to grow, so too does the interest in targeting these kinases therapeutically. In this report, we explore the use of irradiation in combination with an Aurora kinase inhibitor in prostate cancer cell lines. Materials/Methods: We use a commercially available Aurora kinase inhibitor (EMD Biosciences) to inhibit the activity of both Aurora A and Aurora B in human DU-145 and PC-3 prostate cancer models. We also use siRNA to knock down Aurora A and/or Aurora B in prostate cancer cells. Radiosensitivities are compared in Aurora kinase-inhibited cells and in control cells. Results: We demonstrate that use of an inhibitor that targets the activity of both Aurora A and Aurora B can sensitize the prostate cancer cell lines DU-145 and PC-3 to irradiation, as indicated by a decreased cell viability and a reduced clonogenic survival in Aurora kinase inhibitor treated cells compared with control cells. This inhibitor induces cell death through polyploidy resulting in mitotic catastrophe. Using siRNA we also show that the inhibitor phenotype is not duplicated with Aurora A knockdown, but is readily visible in Aurora B knockdown and double knockdown cells. Individual knockdown of Aurora A or Aurora B was found to increase radiation sensitivity in prostate cancer cells. Furthermore, it is by two different mechanisms that cell death occurs, with Aurora A knockdown triggering caspase-mediated apoptosis while Aurora B knockdown inducing caspase-independent mitotic catastrophe. We have also shown that use of an Aurora kinase inhibitor with irradiation can significantly reduce tumor formation in athymic nu/nu mice. Conclusions: Our data suggests that the Aurora kinases are potential therapeutic targets for radiation sensitization in prostate cancer.

We here-in assess the cosmetic and functional outcome with distal extension of the MIUP in grafted tubularised incised-plate urethroplasty (G-TIP) repair. This prospective study included the surgical experience of 263 cases of primary hypospadias operated upon between 2012 and 2015. The G-TIP technique included standard steps of Snodgrass urethroplasty, including degloving and harvesting of glans wings, followed by MIUP that was extended distally beyond the margins of the urethral plate (UP) into the glans. Background: One of the most frequent problems after thyroidectomies is an injury to the recurrent... more Background: One of the most frequent problems after thyroidectomies is an injury to the recurrent laryngeal nerve (RLN) that will lead to vocal cord palsy. This study aimed to assessed the outcomes of intraoperative repair of the recurrent laryngeal nerve. Patients and Methods: In the period of the study we retrived the data of patients who had been underwent thyroid surgery and they sufferd from RLN injery or scarification with immediate intraoperative nerve repair. This retrospective study has been conducted between 2012 and 2020, 813 patients underwent thyroid surgery, 27 (3.3%) of them diagnosed with RLN injury and divided into 2 groups: group A (n = 9), with intraoperative proof of iatrogenic injury of the RLN, and group B (n = 18), in which malignant invasion was diagnosed intraoperative or recognized during thyroidectomy with a therapeutic transection. Immediate microsurgical reconstruction of the RLN was performed. The evaluation was performed at 3, 6, and 9 months post-surgical repair utilizing aspiration and voice improvement as subjective tests, fiberoptic direct laryngoscopy as an objective test. Results: Aspiration significantly improved in both groups (p < 0.05). Voice quality improved in both groups but better in group A (p = 0.02). Conclusion: Immediate RLN reconstruction during thyroidectomies gave excellent postoperative voice quality especially after thyroidectomies in benign thyroid lesions.

As the tongue is a highly specialized tissue, restoring of the tongue is one of the major challenges facing oncological reconstructive surgeons. Patient and methods Since 2013, we have reconstructed 60 resections of the tongue using a free flap [radial forearm, anterolateral thigh (ALT)]. The complete mobile tongue resection (total and near-total tongue resection) represented a small group of 10 patients. We checked the records of the seven patients who had been reconstructed with an ALT flap with a cathedral design. The seven patients (five males and two females) underwent a complete mobile tongue reconstruction with an ALT free flap with a cathedral design. Background: Many non-surgical and surgical procedures have been used to treat patients with chron... more Background: Many non-surgical and surgical procedures have been used to treat patients with chronic recurrent temporomandibular joint dislocation (CRTMJD). Autologous blood injection (ABI) represents a promising approach. Aim of the work: To evaluate the effectiveness and safety of ABI in the treatment of patients with CRTMJD. Patients and Methods: This prospective comparative study involved patients with CRTMJD who were treated by arthrocentesis and ABI with those treated with arthrocentesis alone between January 2017 and January 2020. Analysis of clinical presentation, diagnosis, close observation of patients has been carried out. Results: 140 patients were included in this study, 87 were males and 53 were females. Their ages ranged between 16 to 82 years. They were randomly divided into two groups of 70 each. Group-1 received arthrocentesis followed by ABI and Group-2 received arthrocentesis alone. In group-1, only one ABI was required and was successful in 63 patients and reinjection was required in 7 patients. None of the patients need surgery due to re-injection failure. Conclusion: Positive results on this modality were observed, some questions are still noted about the effect of blood injection on the articular cartilage and on the development of fibrous or bony ankylosis.

Patients and Methods: This study included 126 patients (69 males and 57 females) their age ranged from 9 days to 16 years with cystic neck swellings and had been treated by Excision in surgical procedure in the Maxillofacial/Head and Neck Surgical Unit, Pediatric Surgery Unit at Sohag University Hospitals, in the period from March 2014 to March 2019. Analysis of the clinical presentation, the type of neck swelling was done. All patients were eligible for this study. Results: Total of 126 cases were included in this study, Majority of cases present as midline neck swelling 90 (71.4%) and 31 (24.6%) as lateral neck swelling and 5 (4%) as submandibular swelling, and as regard diagnosis thyroglossal cyst were 82 (65.1%) and 19 (15.1%) were cystic hygroma and 16 (12.7%) were branchial cyst (10 left and 5 right and 1 bilateral) and 8 (6.3%) were dermoid cyst, and one case (0.8%) were cystic teratoma Conclusion: Surgical resection is the best treatment option not only for esthetic purposes but also to get rid of the recurrent infection and risk of fistula formation and possible pressure on neck structures.

Methods: a prospective study of 80 patients suffering from unilateral primary inguinal hernia who underwent elective operation for hernia repair. As the tongue is a highly specialized tissue, restoring of the tongue is one of the major challenges facing oncological reconstructive surgeons. Patient and methods Since 2013, we have reconstructed 60 resections of the tongue using a free flap [radial forearm, anterolateral thigh (ALT)]. The complete mobile tongue resection (total and near-total tongue resection) represented a small group of 10 patients. We checked the records of the seven patients who had been reconstructed with an ALT flap with a cathedral design. The seven patients (five males and two females) underwent a complete mobile tongue reconstruction with an ALT free flap with a cathedral design. Background: Many non-surgical and surgical procedures have been used to treat patients with chron... more Background: Many non-surgical and surgical procedures have been used to treat patients with chronic recurrent temporomandibular joint dislocation (CRTMJD). Autologous blood injection (ABI) represents a promising approach. Aim of the work: To evaluate the effectiveness and safety of ABI in the treatment of patients with CRTMJD. Patients and Methods: This prospective comparative study involved patients with CRTMJD who were treated by arthrocentesis and ABI with those treated with arthrocentesis alone between January 2017 and January 2020. Analysis of clinical presentation, diagnosis, close observation of patients has been carried out. Results: 140 patients were included in this study, 87 were males and 53 were females. Their ages ranged between 16 to 82 years. They were randomly divided into two groups of 70 each. Group-1 received arthrocentesis followed by ABI and Group-2 received arthrocentesis alone. In group-1, only one ABI was required and was successful in 63 patients and reinjection was required in 7 patients. None of the patients need surgery due to re-injection failure. Conclusion: Positive results on this modality were observed, some questions are still noted about the effect of blood injection on the articular cartilage and on the development of fibrous or bony ankylosis.

In a prospective comparative analysis of open complete thyroidectomy, this work aimed to examine the effect of modern suture ligation procedures using the Harmonic ® process in comparison with conventional suture ligation.

Background: The aim of this research was to assess the pattern of presentation and outcome of Cys... more Background: The aim of this research was to assess the pattern of presentation and outcome of Cystic neck swelling in pediatric patients following surgery. Patients and Methods: This study included 126 patients (69 males and 57 females) their age ranged from 9 days to 16 years with cystic neck swellings and had been treated by Excision in surgical procedure in the Maxillofacial/Head and Neck Surgical Unit, Pediatric Surgery Unit at Sohag University Hospitals, in the period from March 2014 to March 2019. Analysis of the clinical presentation, the type of neck swelling was done. All patients were eligible for this study. Results: Total of 126 cases were included in this study, Majority of cases present as midline neck swelling 90 (71.4%) and 31 (24.6%) as lateral neck swelling and 5 (4%) as submandibular swelling, and as regard diagnosis thyroglossal cyst were 82 (65.1%) and 19 (15.1%) were cystic hygroma and 16 (12.7%) were branchial cyst (10 left and 5 right and 1 bilateral) and 8 (6.3%) were dermoid cyst, and one case (0.8%) were cystic teratoma Conclusion: Surgical resection is the best treatment option not only for esthetic purposes but also to get rid of the recurrent infection and risk of fistula formation and possible pressure on neck structures.
