Low-frequency noise (LFN) leads to an abnormal proliferation of collagen and development of tissu... more Low-frequency noise (LFN) leads to an abnormal proliferation of collagen and development of tissue fibrosis. It has been shown that myocardial fibrosis in association with gap junction remodeling occurs in several cardiac diseases and can be implicated in the development of ventricular tachyarrhythmias. We previously reported a strong development of myocardial fibrosis induced by LFN in rats but it is not known whether LFN induces any modification on cardiac connexin43 (Cx43). The aim of this study was to evaluate modifications on cardiac Cx43 induced by LFN in Wistar rats. Two groups of rats were considered: A LFN-exposed group with 10 rats submitted continuously to LFN during 3 months and a control group with 8 rats. In recent years, type II NADH dehydrogenases (NDH-IIs) have emerged as potential drug targets for... more In recent years, type II NADH dehydrogenases (NDH-IIs) have emerged as potential drug targets for a wide range of human disease causative agents. In this work, the NDH-II enzyme from the Gram-positive human pathogen Staphylococcus aureus was recombinantly expressed in Escherichia coli, purified, crystallized and a crystallographic data set was collected at a wavelength of 0.873 Å. The crystals belonged to the orthorhombic space group P212121, with unit-cell parameters a = 81.8, b = 86.0, c = 269.9 Å, contained four monomers per asymmetric unit and diffracted to a resolution of 3.32 Å. A molecular-replacement solution was obtained and model building and refinement are currently under way.

RNA interference (RNAi), a valuable tool to specifically silence gene expression, is becoming an ... more RNA interference (RNAi), a valuable tool to specifically silence gene expression, is becoming an indispensable weapon in the arsenal of functional genomics, target validation and gene-specific therapeutic research. We have shown that Streptolysin-O (SLO) reversible permeabilization is an efficient method to deliver small interfering RNAs (siRNAs) to hard-to-transfect human myeloma cell lines. In addition, we have shown that transfection of siRNAs, using the SLO reversible permeabilization, specifically and efficiently induces gene silencing in myeloma cell lines.

Oxomolybdenum(VI) complexes containing diverse ligands from an electronic and topological point o... more Oxomolybdenum(VI) complexes containing diverse ligands from an electronic and topological point of view have been analysed by means of (95)Mo NMR in solution with the purpose of using this technique as a tool to study their coordination chemistry and reactivity. The relationship between the electronic density on the metal tuned by the electron-donor ability of the coordinated ligands and the (95)Mo chemical shift has been proved for mono- and bimetallic complexes showing a hexa- or hepta-coordination around the metal centre. The different origins of the signal broadening (associated either to the symmetry of the metallic polyhedron or to the presence of isomers or to the ligand de-coordination) have been also considered to rationalise the obtained data.

In the purple sulfur bacterium Allochromatium vinosum, the reaction is catalyzed by the periplasmic diheme c-type cytochrome thiosulfate dehydrogenase (TsdA). Here, we report the crystal structure of the 'as-isolated' form of A. vinosum TsdA to 1.98 Å resolution, and those of several redox states of the enzyme to different resolutions. The protein contains two typical class I c-type cytochrome domains wrapped around two hemes axially coordinated by His-53/Cys-96 and His-164/Lys-208. These domains are very similar suggesting a gene duplication event during evolution. A ligand switch from Lys-208 to Met-209 is observed upon reduction of the enzyme. NADH:quinone oxidoreductases (NDHs), constitute one of the electron entry points into membrane-bo... more NADH:quinone oxidoreductases (NDHs), constitute one of the electron entry points into membrane-bound respiratory chains, oxidising NADH and reducing quinones. Type-II NDHs (NDH-2) are functionally unable to translocate protons and are typically constituted by a single approximately 50 kDa subunit lacking iron-sulfur clusters and containing one flavin as the sole redox centre. No three dimensional crystal structure is yet available for NDHs. We describe the crystallisation and preliminary structure determination of a NDH-2 that contains a covalently bound FAD, isolated from the membrane fraction of Acidianus ambivalens, a hyperthermoacidophilic archaeon capable of growing at 80 degrees C and pH 2.0. NDH-2 was solubilised with the detergent n-dodecyl-beta-d-maltoside and crystallised using ammonium phosphate as precipitant. The structure was solved by MIRAS using Pt and I derivatives.

Despite the prevalent occurrence of tetrathionate formation and its well documented significance within the sulfur cycle, little is known about the enzymes that catalyze the oxidative condensation of two thiosulfate anions. To fill this gap, the thiosulfate dehydrogenase (TsdA) enzyme from the purple sulfur bacterium Allochromatium vinosum was recombinantly expressed in Escherichia coli, purified and crystallized, and a crystallographic data set was collected. The crystals belonged to the monoclinic space group C2, with unit-cell parameters a = 79.2, b = 69.9, c = 57.9 Å, β = 129.3°, contained one monomer per asymmetric unit and diffracted to a resolution of 1.98 Å.

Foi realizado um estudo seccional de base populacional, sobre a ocorrência da filariose linfática... more Foi realizado um estudo seccional de base populacional, sobre a ocorrência da filariose linfática bancroftiana no município do Cabo de Santo Agostinho, Pernambuco, nordeste do Brasil. Foram examinados 7.650 indivíduos de ambos os sexos, residentes em 1.416 domicílios, dos quais 7.644 indivíduos foram identificados como negativos para filariose e seis casos positivos. A idade dos indivíduos examinados variou de 0 a 98 anos (média de 26,6 anos), enquanto que a idade dos positivos variou de 11 a 29 anos, média de 22,5 anos. Cinco dos seis casos eram do sexo masculino. Os indivíduos microfilarêmicos eram residentes na localidade de Pista Preta (01 caso) e nos distritos de Ponte dos Carvalhos (04 casos) e de Pontezinha (01 caso), sendo esse ultimo autóctone. Entre as pessoas examinadas 109 (1,4%) apresentaram queixas clínicas relacionadas à filariose. Esses resultados apontam para a existêcia de transmissão da filariose no município do Cabo de Santo Agostinho, dado esse que determina a necessidade de se estabeceler atividades de controle da endemia, assim como vem colaborar no planejamento do programa nacional de eliminação, uma vez que consegue delimitar áreas contíguas na região metropolitana do Recife com e sem transmissão filarial.

NADH:quinone oxidoreductases (NDHs), constitute one of the electron entry points into membrane-bo... more NADH:quinone oxidoreductases (NDHs), constitute one of the electron entry points into membrane-bound respiratory chains, oxidising NADH and reducing quinones. Type-II NDHs (NDH-2) are functionally unable to translocate protons and are typically constituted by a single approximately 50 kDa subunit lacking iron-sulfur clusters and containing one flavin as the sole redox centre. No three dimensional crystal structure is yet available for NDHs. We describe the crystallisation and preliminary structure determination of a NDH-2 that contains a covalently bound FAD, isolated from the membrane fraction of Acidianus ambivalens, a hyperthermoacidophilic archaeon capable of growing at 80 degrees C and pH 2.0. NDH-2 was solubilised with the detergent n-dodecyl-beta-d-maltoside and crystallised using ammonium phosphate as precipitant. The structure was solved by MIRAS using Pt and I derivatives.

Despite the prevalent occurrence of tetrathionate formation and its well documented significance within the sulfur cycle, little is known about the enzymes that catalyze the oxidative condensation of two thiosulfate anions. To fill this gap, the thiosulfate dehydrogenase (TsdA) enzyme from the purple sulfur bacterium Allochromatium vinosum was recombinantly expressed in Escherichia coli, purified and crystallized, and a crystallographic data set was collected. The crystals belonged to the monoclinic space group C2, with unit-cell parameters a = 79.2, b = 69.9, c = 57.9 Å, β = 129.3°, contained one monomer per asymmetric unit and diffracted to a resolution of 1.98 Å.

Foi realizado um estudo seccional de base populacional, sobre a ocorrência da filariose linfática... more Foi realizado um estudo seccional de base populacional, sobre a ocorrência da filariose linfática bancroftiana no município do Cabo de Santo Agostinho, Pernambuco, nordeste do Brasil. Foram examinados 7.650 indivíduos de ambos os sexos, residentes em 1.416 domicílios, dos quais 7.644 indivíduos foram identificados como negativos para filariose e seis casos positivos. A idade dos indivíduos examinados variou de 0 a 98 anos (média de 26,6 anos), enquanto que a idade dos positivos variou de 11 a 29 anos, média de 22,5 anos. Cinco dos seis casos eram do sexo masculino. Os indivíduos microfilarêmicos eram residentes na localidade de Pista Preta (01 caso) e nos distritos de Ponte dos Carvalhos (04 casos) e de Pontezinha (01 caso), sendo esse ultimo autóctone. Entre as pessoas examinadas 109 (1,4%) apresentaram queixas clínicas relacionadas à filariose. Esses resultados apontam para a existêcia de transmissão da filariose no município do Cabo de Santo Agostinho, dado esse que determina a necessidade de se estabeceler atividades de controle da endemia, assim como vem colaborar no planejamento do programa nacional de eliminação, uma vez que consegue delimitar áreas contíguas na região metropolitana do Recife com e sem transmissão filarial.
