Complementary DNA (cDNA)-oligodeoxythmidylate-celluloses were prepared from cDNA copies of polyso... more Complementary DNA (cDNA)-oligodeoxythmidylate-celluloses were prepared from cDNA copies of polysomal messenger RNA (mRNA) of Novikoff hepatoma, normal rat liver, and regenerating rat liver. cDNA synthesis with reverse transcriptase was approximately 46% with respect to input mRNA with oligodeoxythymidylate-cellulose primer. The cDNA's of normal liver, regenerating liver, and Novikoff hepatomas were used as affinity matrices for hybridization of different mRNA species. Under the conditions used, degradation of mRNA was not detected. After normalization for homologous hybridization efficiency, 53 and 65% of the Novikoff hepatoma mRNA bound to normal liver and regenerating liver cDNA's. Under these conditions an average of 82% of mRNA of normal liver bound to regenerating liver cDNA, and 92% of regenerating liver mRNA bound to normal liver cDNA. The bound and unbound mRNA's were analyzed by translation in the wheat germ system; 2-D gel analysis of the proteins synthesized in the wheat germ system indicated that the cDNA affinity columns selectively adsorbed some mRNA species.

CONSTITUTION: This molded article, having an improved thermal shock resistance to rupture of at least 600 cycles according to the more strict form of ANSI test Z 124,3, is prepd. by molding a composite compsn. comprising about 60-80 wt.% inorg. filler and about 20-40 wt.% unsatd. polyester. Such an improved thermal shock resistance is directly related to the use of an unsatd. polyester resin which in the unfilled state gives a cured item having a heat distortion temp. of about 68-78°C. The ability to induce new formation of capillaries in the cornea was tested for ceruloplasmin, th... more The ability to induce new formation of capillaries in the cornea was tested for ceruloplasmin, the copper carrier of serum, for fragments of the ceruloplasmin molecule with and without copper, for heparin, and for glycyl-L-histidyl-L-lysine, bound or not bound to copper ions. Male or female 2- to 3-kg New Zealand White rabbits were used. These experiments were prompted by the previous observation of copper accumulation in the cornea during angiogenesis and by the inability of copper-deficient rabbits to mount an angiogenic response. The results showed that the three different molecules were all able to induce angiogenesis provided that they were bound to copper. Fragments of the ceruloplasmin molecule also induced angiogenesis but only when copper was bound to the peptides. The data are interpreted to indicate that copper ions are involved in the sequence of events leading to angiogenesis and that the carrier molecules may be of quite a different nature.

Quantitatively, its concentration was highest in Novikoff hepatoma (150 microgram/g tissue) and Morris hepatoma 3924A (550 microgram/g tissue), which are rapid-growing tumors, less in Morris hepatoma 7794A (72 microgram/g tissue), which is of intermediate growth rate, and least in the slow-growing Morris hepatoma 9618A (25 microgram/g tissue). Utilization of the gelatin-agarose and Boyden chamber assays established that: (a) fragments or extracts of corneas treated with several effectors of angiogenesis in vivo acquired the capacity to mobilize the capillary endothelium in vitro; (b) this mobilization was selective for the capillary endothelium; endothelium from aorta and fibroblasts from human skin or rabbit cornea were unresponsive; and (c) among the effectors of angiogenesis utilized alone; i.e., without the intermediary action of the cornea, none were able to mobilize the capillary endothelium in vitro, except for the heparin-copper complex. The data are interpreted to indicate that new formation of capillaries in vivo is the end result of a cascade of events of which heparin and copper are important components.

The vascularization of the rabbit cornea was selected as an experimental approach that permits comparison of one cornea treated by the angiogenesis effector with the contralateral cornea of the same subject treated by the same molecule deprived of angiogenic capacity. Under these conditions, we observed that neovascularization was initiated by the appearance of a chemoattractant for the bovine capillary endothelium only in the cornea treated by the angiogenesis effector. The chemoattractant was purified about 150-fold by a single-step procedure, using gelatin:Sepharose affinity chromatography. Chemoattraction resulted from the combined effect of a chemotactic factor(s) and an activating factor(s). Protein 45/7.8 was isolated under nondenaturing conditions from Morris hepato mas 3924A and 9618A2 and Novikoff hepatoma in high purity by purification in three steps: ammonium sulfate fractionation; diethylaminoethyl cellulose chromatography; and hydroxylap- atite chromatography.
