This study was conducted to determine the effects frying, boiling and freezing processes on the l... more This study was conducted to determine the effects frying, boiling and freezing processes on the levels of sulfachlorpyridazine (SCP) and trimethoprim (TMP) in broiler chest meat and liver. Male broiler chicks were assigned to two groups as control and treatment groups. Animals were fed on commercial diet not containing SCP and TMP for 35 days. At 30 th day, experimental group received suspensions of SCP-TMP mixture (30 mg/kg SCP and 6 mg/kg TMP) via craw by gavage once daily for 5 days. At the end of 35 th day, the chickens were sacrified and right chest tissue and the liver were taken out. A portion of the tissues were stored in-20°C freezer for 30 and 45 days. After sacrifiying, the raw tissues were exposed to frying and boiling processes. SCP and TMP analysis of tissues were performed by HPLC-DAD detector and reverse phase column. In conclusion, it has been determined that boiled and grilled processes caused a reduction of SCP and TMP residues at different rates in broiler tissues; while storing in the deep freezer did not cause a significant change on SCP and TMP residues.

A total of 66 male Wistar rats were used and six groups (control: 10 animals and experimental: 12... more A total of 66 male Wistar rats were used and six groups (control: 10 animals and experimental: 12 animals) were formed. While a separate control group was established for each study period, mad honey application to the animals in the experimental group was carried out with a single dose (12.5 g kg À1 body weight (b.w.); acute stage), at a dose of 7.5 g kg À1 b.w. for 21 days (subacute stage), and at a dose of 5 g kg À1 b.w. for 60 days (chronic stage). Tissue and blood oxidative stress markers (malondialdehyde (MDA), nitric oxide (NO), 4-hydroxynonenal (HNE), superoxide dismutase, catalase, glutathione (GSH) peroxidase, and glucose-6phosphate dehydrogenase), hepatic chemical metabolizing parameters in the liver (cytochrome P450 2E1, nicotinamide adenine dinucleotide (NADH)-cytochrome b5 reductase, nicotinamide adenine dinucleotide phosphate (NADPH)-cytochrome c reductase (CYTC), GSH S-transferase (GST), and GSH), and micronucleus and comet test in some samples were examined. Findings from the study showed that single and repeated doses given over the period increased MDA, NO, and HNE levels while decreasing/increasing tissue and blood antioxidant enzyme activities. From hepatic chemical metabolizing parameters, GST activity increased in the subacute and chronic stages and CYTC activity increased in the acute period, whereas GSH level decreased in the subacute stage. Changes in tail and head intensities were found in most of the comet results. Mad honey caused oxidative stresses for each exposure period and made some significant changes on the comet test in certain periods for some samples obtained. In other words, according to the available research results obtained, careless consumption of mad honey for different medical purposes is not appropriate.

This study was carried out to determine effect of purslane (Portulaca oleracea L.) addition to in... more This study was carried out to determine effect of purslane (Portulaca oleracea L.) addition to in quail diets on the carcass, serum lipid profile and antioxidant activity. A total of 140 one-day-old quail chicks allocated into 4 treatment group with 5 replicate. The treatment groups as follows: (1) Control (C, no purslane seed addition), (2) 2.5% purslane addition to C diet, (3) 5% purslane addition to C diet, (4) 10% purslane addition to C diet. The treatment groups did not affect the slaughter weight, carcass, and carcass part yields (p>0.05). However, in the 2.5% purslane supplemented group the total intestinal weight were lower than those of other groups (p 0.05). The purslane addition at 10% ratio in diet caused an increase to MDA level compared to C and 2.5% purslane supplemented groups (p<0.05). In the purslane addition at 10% ratio in diet increased the SOD, CAT, GSH-Px and NO concentrations that of C and 2.5 and 5% purslane supplemented groups (p<0.05). In conclusion, purslane addition to quail diets may affect antioxidant activity in the serum without significant change in carcass and blood lipid profile.

Bu çalışma, bıldırcın rasyonlarına semizotu (Portulaca oleracea L.) tohumu ilavesinin karkas, kan... more Bu çalışma, bıldırcın rasyonlarına semizotu (Portulaca oleracea L.) tohumu ilavesinin karkas, kan lipid profili ve antioksidan parametreleri üzerine etkisini belirlemek amacıyla yapılmıştır. Çalışmada, bir günlük yaşta toplam 140 adet bıldırcın kullanılmış ve bıldırcınlar 4 muamele grubuna 5 tekerrürlü olarak dağıtılmışlardır. Muamele grupları: (1) Kontrol (K, semizotu tohumu yok), (2) kontrol yemine % 2.5 semizotu tohumu ilavesi, (3) kontrol yemine % 5 semizotu tohumu ilavesi ve (4) kontrol yemine % 10 semizotu tohumu ilave edilen gruplardan oluşturulmuştur. Muameleler grupların kesim ağırlığı, karkas ve karkas parçaları oranı, kalp, karaciğer, taşlık ve bezel mide ağırlıklarını önemli olarak etkilememiştir (P>0.05). Ancak, rasyona %2.5 semiz otu tohumu ilavesi toplam barsak ağırlığını diğer gruplara göre azaltmıştır (P<0.05). Serum trigliserid, kolesterol, yüksek yoğunluklu lipoprotein (HDL) ve düşük yoğunluklu lipoprotein (LDL) konsantrasyonları muamelelerden etkilenmemiştir (P>0.05).Rasyona % 10 semizotu tohumu ilavesi kan malondialdehit (MDA) düzeyini K ve %2.5 semizotu tohumu katılan gruplara göre önemli düzeyde artırmıştır (P<0.05). Rasyona %10 düzeyinde semizotu tohumu ilavesi süperoksit distmutaz (SOD), katalaz (CAT), glutatyon peroksidaz (GSH-Px) ve nitrik oksit (NO) değerlerini K, %2.5 ve %5 semizotu tohumu içeren gruplara göre önemli derecede artırmıştır (P<0.05). Sonuç olarak, bıldırcın rasyonlarına semizotu tohumu ilavesi karkas özellikleri ve serum lipid profilini değiştirmeksizin serum antioksidan düzeylerini etkileyebileceğini göstermiştir.

The toxicokinetics of single-dose intravenous and intracrop administration of cypermethrin has be... more The toxicokinetics of single-dose intravenous and intracrop administration of cypermethrin has been described in broiler chickens. Twenty male broiler chickens were distributed in two groups of ten animals each. The animals in the groups mentioned were given cypermethrin in both routes at a single dose of 7.5 mg/kg b.w. Blood samples were collected by the wing vein of chickens at 0.083, 0.25, 0.50, 0.75, 1, 2, 4, 6, 8, 12, 18, 24 and 36 hours after administration of cypermethrin. Determination of serum cypermethrin levels was performed by the gas chromatography-electron capture detection. When the serum concentration-time graph of cypermethrin was evaluated, it was found that this pesticide showed a more convenient distribution tendency in the two-compartment open model. The values of t(1/2 beta), MRT and AUC(0 ->infinity)were 7.35 +/- 0.91 hours, 8.64 +/- 1.59 hours and 8741.98 +/- 1996.66 ng.h/ml, respectively, after intravenous administration of cypermethrin. On the other hand, C-max, t(max), t(1/2a),t(1/2 beta), MRT and AUC(0 ->infinity) values calculated for intracrop applied cypermethrin were 422.75 +/- 99.59 ng/ml, 0.80 +/- 0.15 hours, 0.24 +/- 0.05 hours, 10.97 +/- 2.11 hours, 15.39 +/- 2.56 hours and 4266.51 +/- 967.65 ng.h/mL, respectively. The systemic bioavailability of cypermethrin administered intracrop was 48.80%. From the results obtained, bioavailability of cypermethrin and the MRT at the same time in the body are moderate. These results were considered to be an important indicator in the evaluation and treatment of cypermethrin poisoning in broiler chickens

The purpose of this study is to investigate the effectiveness of the application of colostrum in ... more The purpose of this study is to investigate the effectiveness of the application of colostrum in aflatoxin toxication. For this aim, a total of 48, 3-4 month old Balb C female mice were used, and 4 groups were established. The animals of group 1 were the control group used. Groups 2, 3 and 4 were given 500 mg/kg bw/day bovine colostrum, 750 mu g/kg bw/day aflatoxin (approximately 77% AFB(1), 15% AFB(2), 5% AFG(1) and 3% AFG(2)) and 500 mg/kg bw/day bovine colostrum plus 750 mu g/kg bw/day aflatoxin, respectively, for 21 days by gavage. The MDA levels and the activities of CAT, SOD and GSH-Px were measured in the blood. Furthermore, levels/activities of the glucose, triglyceride, cholesterol, total protein, albumin, BUN, bilirubin, AST and ALP were performed in serum. These results showed that AF caused negative changes in all the oxidative stress parameters and in some biochemical parameters (triglyceride, total protein, AST and ALP), and applications of colostrum alleviated AF toxicity. It was concluded that colostrum is one of the compounds that can be used as a supportive agent in case of aflatoxin intoxication

A total of 140 one-day-old quail chicks allocated into 4 treatment groups with 5 replicates. The treatment groups as follows: (1) Control (C, without purslane seed addition), (2) 2.5% purslane seed addition to the control diet, (3) 5% purslane seed addition to the control diet, (4) 10% purslane seed addition to the control diet. The treatments did not affect the slaughter weight, carcass, and carcass part yields (p>0.05) of the birds in the treatment groups. However, the total intestinal weights of the birds in the 2.5% purslane supplemented group were lower than those of the other groups (p 0.05). The dietary inclusion of 10% purslane seed caused an increase to MDA level compared to C and 2.5% purslane seed supplemented groups (p<0.05). The toxicokinetics of single-dose intravenous and intracrop administration of cypermethrin has be... more The toxicokinetics of single-dose intravenous and intracrop administration of cypermethrin has been described in broiler chickens. Twenty male broiler chickens were distributed in two groups of ten animals each. The animals in the groups mentioned were given cypermethrin in both routes at a single dose of 7.5 mg/kg b.w. Blood samples were collected by the wing vein of chickens at 0.083, 0.25, 0.50, 0.75, 1, 2, 4, 6, 8, 12, 18, 24 and 36 hours after administration of cypermethrin. Determination of serum cypermethrin levels was performed by the gas chromatography-electron capture detection. When the serum concentration-time graph of cypermethrin was evaluated, it was found that this pesticide showed a more convenient distribution tendency in the two-compartment open model. The values of t(1/2 beta), MRT and AUC(0 ->infinity)were 7.35 +/- 0.91 hours, 8.64 +/- 1.59 hours and 8741.98 +/- 1996.66 ng.h/ml, respectively, after intravenous administration of cypermethrin. On the other hand, C-max, t(max), t(1/2a),t(1/2 beta), MRT and AUC(0 ->infinity) values calculated for intracrop applied cypermethrin were 422.75 +/- 99.59 ng/ml, 0.80 +/- 0.15 hours, 0.24 +/- 0.05 hours, 10.97 +/- 2.11 hours, 15.39 +/- 2.56 hours and 4266.51 +/- 967.65 ng.h/mL, respectively. The systemic bioavailability of cypermethrin administered intracrop was 48.80%. From the results obtained, bioavailability of cypermethrin and the MRT at the same time in the body are moderate. These results were considered to be an important indicator in the evaluation and treatment of cypermethrin poisoning in broiler chickens

The purpose of this study is to investigate the effectiveness of the application of colostrum in ... more The purpose of this study is to investigate the effectiveness of the application of colostrum in aflatoxin toxication. For this aim, a total of 48, 3-4 month old Balb C female mice were used, and 4 groups were established. The animals of group 1 were the control group used. Groups 2, 3 and 4 were given 500 mg/kg bw/day bovine colostrum, 750 mu g/kg bw/day aflatoxin (approximately 77% AFB(1), 15% AFB(2), 5% AFG(1) and 3% AFG(2)) and 500 mg/kg bw/day bovine colostrum plus 750 mu g/kg bw/day aflatoxin, respectively, for 21 days by gavage. The MDA levels and the activities of CAT, SOD and GSH-Px were measured in the blood. Furthermore, levels/activities of the glucose, triglyceride, cholesterol, total protein, albumin, BUN, bilirubin, AST and ALP were performed in serum. These results showed that AF caused negative changes in all the oxidative stress parameters and in some biochemical parameters (triglyceride, total protein, AST and ALP), and applications of colostrum alleviated AF toxicity. It was concluded that colostrum is one of the compounds that can be used as a supportive agent in case of aflatoxin intoxication
