In vivo 13C Nuclear Magnetic Resonance (NMR) spectroscopy was used to investigate the pathways of... more In vivo 13C Nuclear Magnetic Resonance (NMR) spectroscopy was used to investigate the pathways of glucose metabolism, non-invasively, in living cell suspensions of Propionibacterium freudenreichii subsp. shermanii. This species is the main ripening flora of the Swiss-type cheeses and is widely used as propionic acid and vitamin B12 industrial producer. The flow of labelled [1-13C]glucose was monitored in living cell suspensions and enrichment was detected in main products like [1-13C]glycogen, [6-13C]lycogen, [1-13C]trehalose, [6-13C]trehalose, [1-13C]propionate, [2-13C]propionate, [3-13C]propionate, [1-13C]acetate, [2-13C]acetate, [1-13C]succinate, [2-13C]succinate and [1-13C]CO2. alpha and beta glucose consumption could be examined separately and were catabolized at the same rate. Whole cells of Lactobacillus helveticus were immobilized in calcium-alginate beads to produce lac... more Whole cells of Lactobacillus helveticus were immobilized in calcium-alginate beads to produce lactic acid from cheese whey ultrafiltrate. Ca-alginate-entrapped cells were characterized by higher fermentation rates and optimum pH than free cells. No difference could be observed in the profile of cell activity against temperature for either type of cells. After a heat treatment, cell activity was higher for free cells than for immobilized cells. Continuous lactic acid fermentation using a packed bed reactor was investigated.

Dairy propionibacteria, and especially Propionibacterium freudenreichii subsp. shermanii, are imp... more Dairy propionibacteria, and especially Propionibacterium freudenreichii subsp. shermanii, are important in the food industry and biotechnology. Only a few investigations have focused on the complex physiology of this remarkable bacterium, while the physiology of dairy lactic acid bacteria has been extensively studied over the past decades. Here we report the progress of our whole genome sequencing project: 93% of the P. shermanii genome was assembled in 426 contigs with low overall redundancy. Our annotation strategy overlaps the sequence finishing step, thus improving it. Annotation of the incomplete genome is performed using ContigBrowser, a bioinformatic tool allowing data management, developed in our laboratory. This resulted in the detection of 2611 putative proteins (data May 2003). Our tool allows an expert annotation by manual verification and curation of functional protein categories after automatic assignment. Food processes use different microorganisms, from bacteria to fungi. Yeast strains have been exte... more Food processes use different microorganisms, from bacteria to fungi. Yeast strains have been extensively studied, especially Saccharomyces cerevisiae. However, to date, very little is known about the potential beneficial effects of molds on gut health as part of gut microbiota. We undertook a comprehensive characterization of five mold strains, Penicillium camemberti, P. nalgiovense, P. roqueforti, Fusarium domesticum, and Geotrichum candidum used in food processes, on their ability to trigger or protect intestinal inflammation using in vitro human cell models and in vivo susceptibility to sodium dextran sulfate-induced colitis. Comparison of spore adhesion to epithelial cells showed a very wide disparity in results, with F. domesticum and P. roqueforti being the two extremes, with almost no adhesion and 20% adhesion, respectively. Les fermentations tiennent une place privilégiée dès que l'on parle de bio-industries et sur ce , i;egistre, la Br, etagne était b'Îen peu pourvue. 11 fallait donc étudier, rassurer, poser les bonnes questions, se référer à l'existant, explorer le devenir. – The solubility and the selectivity of αS1-, β- and κ-casein isolation from an industri-ally pro... more – The solubility and the selectivity of αS1-, β- and κ-casein isolation from an industri-ally produced spray-dried sodium caseinate as a function of temperature (20–40 ◦C) and of ethanol volume fraction (0–0.6) at pH 5.0–7.5 were studied. Reverse-phase high-performance liquid chro-matography and nitrogen balance were used as analytical methods. At 20 ◦C, casein solubility as a function of pH (5–7) and of the ethanol volume fraction (0–0.6) is quite well represented by an equation describing the ionisation process. αS1-, β- and κ-casein solubility and β- and κ-casein ex-traction selectivity decrease both with a rise in the ethanol volume fraction to 0.45–0.50 and with temperature increase. For αS1-casein, the selectivity increases to 67 % for an ethanol volume fraction of 0.47 at 29 ◦C. This fraction could be used as an emulsifying agent or for cream liqueur production or further purification.

Le dispositif selon l'invention comprend avantageusement une cuve de fermentation (6) comportant deux boucles externes (9, 19) de circulation du substrat fermente: une boucle (9) de brassage du substrat fermente effectuant un prelevement et une reintroduction du substrat fermente dans la cuve (6) de fermentation, une boucle (19) d'extraction du melange (21) biologique a base d'acide propionique fabrique (21), incluant un module (22) de separation melange biologique fabrique (21)/bacteries (23). Le procede mis en ÷uvre est du type semi-continu, forme d'une succession de cycles de fermentation/extraction avec recyclage integral de la biomasse, puis recyclage partiel apres obtention d'une concentration en biomasse predeterminee.

The complex and diverse microbial ecosystem residing within humans contributes critically to these intergenerational impacts. Perinatal factors, including maternal nutrition, antibiotic use and maternal stress, alter the maternal gut microbiota during pregnancy, which can be transmitted to the offspring. In addition, gestational age at birth and mode of delivery are indicated frequently to modulate the acquisition and development of gut microbiota in early life. The early-life gut microbiota engages in a range of host biological processes, particularly immunity, cognitive neurodevelopment and metabolism. The perturbed early-life gut microbiota increases the risk for disease in early and later life, highlighting the importance of understanding relationships of perinatal factors with early-life microbial composition and functions.
