Background: The coagulation factor XIII is a pro-transglutaminase enzyme with tetrameric structur... more Background: The coagulation factor XIII is a pro-transglutaminase enzyme with tetrameric structure. An exchange of G for A in exon 2 of A subunit results in replacement of valine with leucine in amino acid 34. As a result of this substitution mutation, the clots produced are fragile and loose therefore it seems that FXIII Val34Leu polymorphism acts as a factor for individual protection against thrombosis Objectives: To determine the prevalence and role of FXIIIA Val34Leu polymorphism against deep vein thrombosis. Methods: This was a retrospective case-control study performed on 116 patients with DVT who were referred to Thrombosis and Homeostasis Laboratory affiliated to Iranian Blood Transfusion Organization. Also, 100 healthy individuals (blood donors) were recruited as control. Following DNA extraction and application of PCR and RFLP techniques in presence of restriction enzyme Cfo1, the genotypes of FXIII Val34Leu polymorphism were identified. The data were analyzed using chi square test as well as calculation of OD ratio and 95% confidence interval. Findings: The prevalence of FXIII Val34Leu polymorphism among the case and control groups was 22.4% and 37.4%, respectively. While the allele frequency of leucine in case group was 14.7% it was 20.2 % in control group. No significant correlation between polymorphism and sex was established. Conclusion: According to our data, no association between the FXIII Val34Leu polymorphism and protection against deep vein thrombosis was demonstrated. Therefore, it seems that this polymorphism occurs as a natural phenomenon and unaffected by gender.

Background: Hepatitis B virus (HBV) infection in patients who lack detectable hepatitis B surface... more Background: Hepatitis B virus (HBV) infection in patients who lack detectable hepatitis B surface antigen (HBsAg) is called occult hepatitis B infection. Such infec tions have been frequently identified in patients with chronic hepatitis C liver disease, but their prevalence is not known. Methods: 207 patients with chronic hepatitis C who were HCV-RNA and anti-HCV positive were studied for HBV-DNA by PCR, and for HBsAg and anti-HBc by ELISA. DNA was extracted by high pure nucleic acid kit (Roche-Germany). HBV DNA amplification was done with a set of primer directed to the pre-S region. HBsAg and anti-HBc were evaluated by a commercially available ELISA kit (Dade Behring). Results: 23 of 207 patients with chronic hepatitis C liver disease (11.1%) were positive for HBV-DNA (co-infection). Among this group 17 patients (8.2%) were HBsAg negative (occult infection). 8 of 17 patients with occult infection (47%) were anti-HBc positive and 9 were anti-HBc negative (53%). No significant difference was found in epidemiological and biochemical param eters in patients with HCV alone in comparison with HCV co-infected with occult hepatitis B (p= 0.453 for ALT and p= 0.498 for AST). Conclusion: Occult hepatitis B virus infections occur frequently in patients with chronic hepatitis C liver disease and may have clinical significance.

HPA genes frequency in Iran is not evaluated, so we invesgated the HPA-2, HPA-3, and HPA-5 frequency and their polymorphism. Materials and Methods: DNA from 120 Iranians (99 Iranian blood donors and 21 paents with platelet refractoriness) was isolated from peripheral blood mononuclear leucocytes and tested by restricted fragment length polymorphism analysis (RFLP). Results: The frequency of HPA phenotypes was determined as follows: (in donors) HPA-2a/2a:75.4%, HPA- 2a/2b:24.8%, HPA-3a/3a:8.1%, HPA-3a/3b:70.7%, HPA-3b/3b:21.2%, HPA-5a/5a:99 % and HPA-5a/5b:1%; (in paents) HPA-2a/2a:66.7%, HPA-2a/2b:33.3%, HPA-3a/3a:23.8%, HPA-3a/3b: 47.6%, HPA-3b/3b:28.6%, HPA-5a/5a:90.5%, and HPA-5a/5b: 9.5%. Background: Umbilical cord blood (UCB) is believed to be a highly valuable source of hematopoieti... more Background: Umbilical cord blood (UCB) is believed to be a highly valuable source of hematopoietic stem cells for transplantation. Objective: To investigate the prevalence of active and latent human cytomegalovirus (CMV) infection in UCB donors in Iranian population. Methods: A total of 825 UCB samples was collected under standard procedures and analyzed for the presence of CMV DNAs in buffy coat (latent infection) and plasma (active infection). DNA was extracted from buffy coat and plasma samples separately and tested with quantitative real-time PCR. All positive samples were checked by ELISA for IgG and IgM anti-CMV antibody. Results: Latent CMV infection was detected in 17 (2%) buffy coat samples with a low level of viral load, which indicated the presence of latent viral infection in donors.
