The genetic improvement of any crop mainly depends on the amount of genetic variability present i... more The genetic improvement of any crop mainly depends on the amount of genetic variability present in the population. To explore this variability, an effort was made to classify, understand the nature and magnitude of genetic variability and diversity among 112 rice genotypes for 16 quantitative traits using Mahalanobis D 2 statistics. Analysis of variance revealed a wide and significant variation for all the 16 traits studied. A slight difference between PCV and GCV was found, suggesting that genetic control of traits is higher than environmental influence. Characters such as harvest index, days to first flowering, days to 50% flowering, and plant height showed high heritability coupled with high genetic advance as per cent of mean suggesting that selection for the improvement of these traits may be rewarding. Cluster analysis indicated that the 112 rice genotypes were grouped into 7 clusters, wherein cluster I had the highest number of genotypes (73) followed by cluster III with 22 genotypes. The maximum inter-cluster distance was recorded between clusters V and VII with a D 2 value of 58981.07, followed by clusters II and V (51875.13). The number of spikelets per panicle (52.17%) and the number of grains per panicle (27.26%) were found to be the most contributing traits towards total genetic diversity. Hence, information on the nature and degree of divergence would help the plant breeder in the selection and hybridization procedure for choosing the right type of parents to improve yield and other traits.

Assessment of genetic diversity is of utmost important in rice breeding from the perspective of selection, conservation and proper utilization. The present study was undertaken with an objective to assess the genetic diversity among 28 rice cultivars by use of 22 SSR markers. A total of 41 alleles were detected across 28 rice cultivars used. PIC values varied widely among SSR loci tested and it ranged from 0.753 to 0.067, with a mean value of 0.376. The 28 rice cultivars were grouped into two clusters i.e., cluster I and II with dissimilarity coefficient 0.42. Cluster I was sub divided into two minor sub-groups IA and IB having 6 and 11 genotypes respectively. IB was further subdivided into minor groups. Genetic Diversity in rice provides a great opportunity for the plant breeders to develop novel an... more Genetic Diversity in rice provides a great opportunity for the plant breeders to develop novel and improved cultivars with desirable characteristics. In the present study, a total of 30 SSR markers were used across twelve drought tolerant rice genotypes for their characterization and divergence analysis. Among the thirty SSR markers used, only twenty three SSR markers showed polymorphism and the PIC value ranged from 0.551 (RM 11) to 0.305 (RM 307) with a mean value of 0.38. Variation in the number of alleles ranged from 2 to 3 with an average of 2.217. A total of 51 microsatellite alleles were amplified from twelve genotypes and this demonstrates considerable variability among genotype. In the current study, a diverse set of 3 K sub-panel of rice consisting of 150 accessions was evaluated for resistant starch and predicted glycemic index, including nine other quality traits under transplanted situation. Significant variations were noticed among the accessions for the traits evaluated. Trait associations had shown that amylose content possess significant positive and negative association with resistant starch and predicted glycemic index. Genome-wide association studies with 500 K SNPs based on MLM model resulted in a total of 41 marker-trait associations (MTAs), which were further confirmed and validated with mrMLM multi-locus model. The experiment was conducted in alpha lattice design and observations were recorded in five randomly selected plants for sixteen traits. The data were analyzed in WINDOSTAT 9.3 ver. for correlation and path analysis. The correlation analysis showed very strong correlation of grain yield per plot with biomass yield per plot. The 22 rice genotypes were characterized based on 16 quantitative traits viz., days to first flowering, days to 50 per cent flowering, days to maturity, the total number of effective tillers per plant, plant height (cm), panicle length (cm), the number of spikelets per panicle, the number of grains per panicle, spikelet fertility percentage, grain weight per panicle (g), grain yield per plant (g) , 1000-grain weight (g), grain yield per plot (kg), grain yield per hectare (kg), grain L/B Ratio, grain zinc content (ppm or mg /kg) using Mahalanobis D 2 statistic. D 2 analysis distributed the 22 genotypes into six clusters, of which cluster I was the largest with 15 genotypes. Cluster II had maximum intra-cluster values of 5.16 and the maximum inter-cluster distance was observed between the clusters III and V (40.51) followed by cluster IV and V (28.39) indicating the importance of the genotypes present in these clusters for exploiting heterosis for the desirable traits of these clusters.

Fruit and shoot borer (FSB) Leucinode orbonalis is most serious and destructive pest causing heavy yield losses. Agrobacterium mediated genetic transformation was performed in two brinjal cultivars viz RHRB-35 and Manjarigota with Cry1Aabc gene. Several factors influencing transformation efficiency such as type of explants, Agrobacterium cell density, infection time, pre-culture duration, co-cultivation duration, overgrowth control antibiotics have been evaluated. Combination of cefotaxime and carbenicillin at 250 mg/l concentration effectively control Agrobacterium overgrowth. The present field experiment was conducted to study the diversity present in 29 local genotypes of rice using both morphological and molecular ways. Methodology: The experiment was conducted at Agricultural Research Farm, Banaras Hindu University, during Kharif-2017 in an augmented block design with 29 rice genotypes including 3 checks. Mahalanobis’ D2 analysis was carried out to assess the morphological diversity present among the genotypes and molecular analysis was done with 21 polymorphic SSR markers using the NTSYSpc software. The 22 rice genotypes were characterized based on 16 quantitative traits viz., days to first flowering, days to 50 per cent flowering, days to maturity, the total number of effective tillers per plant, plant height (cm), panicle length (cm), the number of spikelets per panicle, the number of grains per panicle, spikelet fertility percentage, grain weight per panicle (g), grain yield per plant (g) , 1000-grain weight (g), grain yield per plot (kg), grain yield per hectare (kg), grain L/B Ratio, grain zinc content (ppm or mg /kg) using Mahalanobis D 2 statistic. D 2 analysis distributed the 22 genotypes into six clusters, of which cluster I was the largest with 15 genotypes. Cluster II had maximum intra-cluster values of 5.16 and the maximum inter-cluster distance was observed between the clusters III and V (40.51) followed by cluster IV and V (28.39) indicating the importance of the genotypes present in these clusters for exploiting heterosis for the desirable traits of these clusters.

Fruit and shoot borer (FSB) Leucinode orbonalis is most serious and destructive pest causing heavy yield losses. Agrobacterium mediated genetic transformation was performed in two brinjal cultivars viz RHRB-35 and Manjarigota with Cry1Aabc gene. Several factors influencing transformation efficiency such as type of explants, Agrobacterium cell density, infection time, pre-culture duration, co-cultivation duration, overgrowth control antibiotics have been evaluated. Combination of cefotaxime and carbenicillin at 250 mg/l concentration effectively control Agrobacterium overgrowth. Two week old Cotyledon and three week old hypocotyls explants precultured for 2 days and then infected with Agrobacterium suspension (OD600= 0.2 to 0.4) for 1 min showed maximum transformation efficiency. Increasing trends in the occurrence of diabetes underline the need to develop low glycemic index ... more Increasing trends in the occurrence of diabetes underline the need to develop low glycemic index (GI) rice with preferred grain quality. In the current study, a diverse set of 3 K sub-panel of rice consisting of 150 accessions was evaluated for resistant starch and predicted glycemic index, including nine other quality traits under transplanted situation. Significant variations were noticed among the accessions for the traits evaluated. Trait associations had shown that amylose content possess significant positive and negative association with resistant starch and predicted glycemic index. Genome-wide association studies with 500 K SNPs based on MLM model resulted in a total of 41 marker-trait associations (MTAs), which were further confirmed and validated with mrMLM multi-locus model.
