Interleukin-6 (IL-6) gene is a multifunctional cytokine which is expressed in lymphocytes, fibrob... more Interleukin-6 (IL-6) gene is a multifunctional cytokine which is expressed in lymphocytes, fibroblasts, macrophages, in response to different types of inflammatory stimuli. IL-6 also controls induction and expression of metallothioneins (MTs) which maintain homeostasis of zinc and copper. In human, IL-6 gene is located on chromosome 7p21 and −174 G/C polymorphism located in its promoter region. Recently, genetic studies showed that IL-6 −174 G/C promoter polymorphism influences IL-6 gene transcription and plasma cytokine levels. The aim of this study is to determine the IL-6 promoter polymorphism effect on trace element levels and toxic metal accumulation in the kidney and liver tissues. Kidney and liver tissues were collected from 122 autopsy cases in Ankara district. IL-6 promoter polymorphism was detected by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) analysis. The genotype frequencies were found as 54.9% homozygote typical (GG), 39.3% heterozygote (GC) and 5.7% homozygote atypical (CC). The allele frequencies in all subjects were consistent with Hardy-Weinberg equilibrium (2 = 0.179; p > 0.05). In order to assess the cadmium (Cd), lead (Pb), zinc (Zn) and copper (Cu) levels in the autopsy tissues, a dual atomic absorption spectrophotometer system was used. As a result, no statistical association was found between the IL-6 promoter polymorphism and Pb, Cd, and Cu (p > 0.05) levels in the kidney and liver tissues but statistically significant differences were detected with the Zn concentration (p < 0.05).

Glucose transporter 1 (Glut1) is located in basolateral side of cells and mediates the unidirecti... more Glucose transporter 1 (Glut1) is located in basolateral side of cells and mediates the unidirectional transport of glucose as non-insulin dependent glucose transporter protein. ıt is responsible for transportation of glucose especially to cancer cells and also transports arsenic compounds that is related with diabetes mellitus. The present study aimed to detect the allele and genotype frequencies of Glut1-2841a>t single nucleotide polymorphism (SNP) in turkish population. Blood samples were collected from unrelated 100 male and 110 female. Polymerase Chain reaction (PCr)-restriction Fragment length Polymorphism (rFlP) technique was used to analyze the blood samples. The distribution of-2841a>t SNP was 55.2%, 38.1% and 6.7% for aa, at and tt genotype, respectively. The frequency of the a allele was found as 74.3% and those of the t allele as 25.7%. The statistical evaluation represented that the allele frequencies was accordant with Hardy-Weinberg equilibrium (χ2 =0.002, p=0.97). This was the first study examining the frequencies of Glut1-2841a>t polymorphism in turkish population. ın our opinion, these results can be used in epidemiological studies both in turkey and other countries.

The purpose of this study is to investigate the association between DNA damage and blood arsenic ... more The purpose of this study is to investigate the association between DNA damage and blood arsenic levels in individuals occupationally exposed to arsenic (As). A hundred-twenty exposed individuals from silver mining plate workers were monitored in the way of DNA damage in blood lymphocytes using comet assay (single cell gel electrophoresis, SSGE). The DNA damage levels were measured by BAB Bs Comet Assay system and As levels were analyzed by the atomic absorption spectrometer (AAS) system. The mean blood arsenic level was 17.03±10.85 µg/L. A statistically significant positive correlations were found between the arsenic levels and comet parameters (tail intensity, comet length and tail length) (r=0.360, r=0.334, r=0.259, respectively, p<0.01). Our results showed that exposure to arsenic may cause DNA damage in peripheral lymphocytes of Turkish silver mining plate workers.

In order to investigate the role of lipid peroxidation in the paracetamol and aspirin hepatotoxic... more In order to investigate the role of lipid peroxidation in the paracetamol and aspirin hepatotoxicity, toxic doses of paracetamol (500 mg/kg) and aspirin (200 mg/kg) were given IP to rats. Malondialdehyde (MDA), a product of lipid peroxidation, was tested in both plasma and liver. Plasma MDA levels were found to be 16.33 0.92 in control group, 24.25. 0.86 in paracetamol group and 20.27 0.73 nmol MDA/ml in aspirin group. Hepatic MDA levels were found to be 348 15.8 in control group, 425 8.17 in paracetamol group and 406. 10.27 nmol MDA/g wet weight in aspirin group. Paracetamol and aspirin caused significantly increases in MDA levels in both plasma and liver (p<0.05). According to our results, lipid peroxidation may conclude an indicator of paracetamol and aspirin hepatotoxicity.

Toksik metal düzeylerinin taranması rutin bir prosedür değildir. Bu çalışmanın amacı, maruziyeti olmayan Türk bireylerde demir, çinko ve bakır gibi eser elementlerin ve kadmiyum, kurşun gibi toksik metallerin tam kan düzeylerini saptamak ve kronik metal toksisitesi riski bulunan bireyleri tanımlamaktır. Gereç ve Yöntem: Tam kan kurşun ve kadmiyum düzeyleri grafit fırınlı atomik absorpsiyon spectrometresi ile analiz edildi. In this study, 1-hydroxypyrene, 9-hydroxyphenantrene, and 3-hydroxybenzo[a]pyrene levels were determined in the urine of occupationally exposed coke-oven worker as markers of exposure to polycyclic aromatic hydrocarbons. The validated high-pressure liquid chromatography method with fluorescence detection was performed for metabolite analysis. Average values of 1-hydroxypyrene were 1.26 µmol/mol creatinine and of 9-hydroxyphenantrene 3.68 µmol/mol creatinine in the workers group, and 0.04 and 0.06 µmol/mol creatinine in the control group. While the average value of 3-hydroxybenzo[a]pyrene was 0.007 µmol/mol creatinine in the workers group, it was below the limit of detection in the control group.
