Background: Maternal exposure to cigarette smoke during pregnancy and lactation might harmful for... more Background: Maternal exposure to cigarette smoke during pregnancy and lactation might harmful for the fetus. The smoke contains many free radicals that could be eliminated by antioxidant. This study aimed to investigate the effect of Spirulina platensis ethanol extract as antioxidant against cigarette smoke exposure during pregnancy until lactation by assessing oxidative stress markers in neonatal brain tissues. Methods: The experimental study used 26 offspring divided into four groups: (C) = offspring of maternal control group; (Cg) = offspring of maternal exposed to cigarette smoke; (CgSp) = offspring of maternal given spirulina and exposed to cigarette smoke; and (Sp) = offspring of maternal given spirulina only group, during gestation and 9 days lactation (30 days). Each group consisted of 6 offspring obtained from 2 adult females mated with male Sprague-Dawley rats. The exposure of cigarette smoke was 4 burn cigarettes/day for 30 days. Vitamin C (ascorbic acid) plays an exogenous antioxidant to protect biomolecules against cellular... more Vitamin C (ascorbic acid) plays an exogenous antioxidant to protect biomolecules against cellular redox-reactive species generation, including ROS, that is involved in the development of oxidative stress-related diseases. Therefore, some ascorbic acid studies have concentrated on its ROS scavenging activity. We designed a combination of vitamin C and octanoic acid as vitamin C derivatives to evaluate their antioxidant potential in silico study using molecular docking. Compound 1 demonstrated the receptor-ligand interaction in Lipoxygenase (LO) than Zileuton-ZIL with its lower binding free energy (ΔG) and inhibition constant (Ki) value. Meanwhile, 6 exhibited lower Gibs energy (ΔG) and inhibition constant (Ki) in NADPH oxidase (NO) receptor than Dextromethorphan-DEX. The results add to the evidence supporting antioxidant activities of 1 and 6 in two receptors (LO and NO, respectively) via ROS generation. Thus, further research is warranted into their efficiency in reducing oxidative stress for possible commercial antioxidants.

Th... more Cancer stem cells have been reported to maintain stemness under certain extracellular changes. This study aimed to analyze the effect of extracellular O2 level modulation on the glucose metabolism of human CD24-/CD44+ breast cancer stem cells (BCSCs). The primary BCSCs (CD24-/CD44+ cells) were cultured under hypoxia (1% O2) for 0.5, 4, 6, 24 and 48 hours. After each incubation period, HIF1α, GLUT1 and CA9 expressions, as well as glucose metabolism status, including glucose consumption, lactate production, O2 consumption and extracellular pH (pHe) were analyzed using qRT-PCR, colorimetry, fluorometry, and enzymatic reactions, respectively. Hypoxia caused an increase in HIF1α mRNA expressions and protein levels and shifted the metabolic states to anaerobic glycolysis, as demonstrated by increased glucose consumption and lactate production, as well as decreased O2 consumption and pHe. Furthermore, we demonstrated that GLUT1 and CA9 mRNA expressions simultaneously increased, in line with HIF1α expression. In conclusion, modulation of the extracellular environment of human BCSCs through hypoxia shifedt the metabolic state of BCSCs to anaerobic glycolysis, which might be associated with GLUT1 and CA9 expressions regulated by HIFlα transcription factor.

The clinical findings are; the duration of retinal detachment, retinal quadrants involvement, and the severity of proliferative vitreoretinopathy (PVR). A cross sectional study was conducted, with 34 consecutive vitreous sample were obtained from dr. Cipto Mangunkusumo National General Hospital (RSCM), from October 2015 to October 2016. The samples were taken from RRD patient who received vitrectomy in the hospital. There were no significant difference between the ICAM-1 and HIF-1a levels to the clinical findings that is measured; duration of retinal detachment (p>0.05), extent of retinal detachment area (p>0.05) and severity of PVR (p>0.05). Futsal sports have high explosive characteristics with moderate-heavy intensity. The increase of ... more Futsal sports have high explosive characteristics with moderate-heavy intensity. The increase of cortisol is a biomarker for high-stress situations desirable throughout futsal-that catabolic hormone secreted in a state of physical or psychological stress. Meanwhile, a person's emotions are harmful, such as stress, anxiety, fear, and frustration. The release of the cortisol hormone can activate the sympathetic nervous system, distinguished by an increase in heart rate, and reduce the ability to think and react. Additionally, it represents a role in decreasing mood and muscle fatigue. This study's persistence was to determine the differences in the frequency of futsal practice on physical stress in adolescents (27 boys). This investigation approach is an experimental study in adolescents (13,63±0,84 years old) who perform futsal sports with different futsal frequency groups for ten weeks, specifically 1-time weekly exercise frequency (F1), 3-times weekly exercise frequency (F3), and 5-times weekly exercise frequency (F5). This research had passed the research ethics review issued. The testing results on F1 found no significant improvement in cortisol (51,21±22,66 vs. 61,02±27,69). In F3, there was no significant increase at the cortisol level (57,71±15,45 vs. 61,82±13,57), and in F5 group, there was a significant increase at the cortisol level (51,02±9,38 vs. 70,33±16,36). Interestingly, only the F5 group showed a significant increase at the cortisol level in this study. Anticipated F5 physically attends to be extremely high in the intensity of futsal exercise and induces fatigue and muscle damage that can stimulate an improvement in the endocrine system, constant with enhancing cortisol levels in the blood. This cortisol release can activate the sympathetic nervous system, characterized by a high pulse rate, but it can reduce the ability to think.

The high activity of cells and tissues during regeneration requires high energy therefore oxygen demand is very high. Cytoglobin (Cygb) is a protein that plays a role to diffuse oxygen to mitochondria. Cygb is a molecule that has a high affinity to oxygen and expressed during hypoxic conditions, when the tissue lacks oxygen. The tissue in regeneration process experiences at hypoxia state, Cygb protein expression have an important role during the tissue regeneration process. The Spearman correlation test results the strong correlation between the expression of Cygb protein of immunoratio test in immunohistochemical staining with the growth of epidermal thickness of tissue (r = 0.67; p <0.05). The results of correlation test between Cygb protein expression with the growth of dermis thickness showed moderate correlation properties (r = 0.406; p <0.05).
